Allozyme electrophoresis at five loci was used to clarify the species status of two morphs of the scleractinian coral Montipora digitata (Dana, 1846), using specimens collected from three locations off the Queensland coast between October 1991 and April 1993. The two morphs, occurring sympatrically, were distinguished by one fixed gene difference at Locus LT-2 and frequency differences at three other loci, indicating reproductive isolation between them. Populations of both morphs were generally in Hardy-Weinberg equilibrium, suggesting that sexual reproduction does occur. Despite sampling designed to reduce the chance of collecting clonemates, the genetic diversity ratio (G O:GE) indicated that asexual reproduction also occurs. This is in agreement with the known modes of reproduction for this species. There was significant genetic differentiation between populations of one of the morphs of M. digitata. This could have been influenced by greater degrees of asexual reproduction in this morph, selection within sites, or variation in larval survival and recruitment patterns. Relatively low F st (differentiation among populations) values found for a broadcast spawning species during this study are in accordance with the hypothesis that brooded planulae are adapted for rapid settlement, whereas planulae produced by broadcast spawners are adapted for widespread dispersal.  相似文献

Asexual propagation is an underappreciated way of increasing the sessile population of the metagenetic Scyphozoa. Our studies on Sanderia malayensis have shown a great variety of asexual reproduction modes. A new mode of polyp asexual propagation is described. By integrating earlier reported modes occurring in other species we established a classification which may serve as a standard tool to describe asexual propagation in Scyphozoa. The morphological characteristics and the population genetic structures of the fissiparous seastar Coscinasterias acutispina were investigated for eight sites in the Sea of Japan in order to clarify the presence of sexual and asexual reproduction. Morphological observation based on arm length showed that fission was common at all eight sites examined, indicating the likely production of clonal individuals. A random amplified polymorphic DNA (RAPD) marker was used to detect clones arising by fission and to assess gene flow among sites. A simulation approach using RAPD data revealed the presence of clonal individuals at almost all sites, suggesting the existence of asexual reproduction. The result of phylogenetic analysis according to RAPD genotype showed no relationship between genetic and geographic distances. Considering the limited movement ability of seastar species during the adult phase, these observations suggest the existence of marked gene flow among sites, due to dispersal of planktonic larvae produced by sexual reproduction. These observations suggest that multi-locus genotypic compositions depend on the relative amounts of recruitment from sexual and asexual reproduction in each population.  相似文献

Ophiomyxa brevirima is a direct-developing species of brittle-star found around the coast of New Zealand. The mode of reproduction of this ophiuroid was studied using isozyme electrophoresis of mother and brooded offspring. Multi-locus assessment of the genotypes of 35 adult females and their brooded offspring was undertaken. A minimum of 45% of broods assessed showed evidence of amictic reproduction, with all offspring in these broods possessing genotypes that were identical to the genotype of their heterozygous mother. In most other broods there was evidence of segregation. These observations are interpreted as evidence of both sexual and asexual reproduction in this ophiuroid species. Sponges display a variety of reproductive strategies that have the potential to influence population genetic structure. Histological examination of ten reproductive individuals of the Western Australian sponge Haliclona sp. showed that this species broods embryonic larvae that are potentially limited in dispersal capabilities. Because sponges have the potential to propagate in a number of modes, allozyme electrophoresis was used to assess the relative importance of asexual and sexual reproduction to recruitment, and to quantify genetic subdivision over different spatial scales. Tissue samples from 227 sponges were collected from reefs within two areas 400 km apart: Hamelin Bay and Rottnest Island. Contrary to expectations for highly clonal populations, genotypic diversity within sites was high, no linkage disequilibrium was found, and there was no evidence of genotypic clustering within reefs. There was no genetic evidence that asexual reproduction is important for the maintenance of populations. Genetic comparisons were consistent with mixing of sexually produced recruits within reefs, on a scale up to a few hundred metres, but significant genetic subdivision between reefs (FST=0.069 at Hamelin Bay, 0.130 at Rottnest Island) indicated that water gaps of several hundred metres are effective at preventing dispersal. Subdivision between the two areas, separated by 400 km, was moderately greater (FST=0.142) than within, but the same alleles were predominant in the two areas. The ahermatypic scleractinian Balanophyllia elegans has served as a model of limited larval dispersal in an aclonal species. However, other species from the same family (Dendrophylliidae) produce larvae asexually, and closely connected polyps of B. elegans, potentially the result of asexual reproduction, are commonly observed in the field. Here, we use a combination of laboratory experiments, controlled crosses, and genetic surveys of field-collected individuals to demonstrate (1) marker allozymes are inherited in accordance with Mendel's laws, (2) polyps that are connected in the field are not genetically identical, and (3) laboratory manipulation of a single polyp can produce a second polyp on the aboral side of the original, but not adjacent to the original. In combination, these results suggest that the larvae of B. elegans result from mictic sexual reproduction, and that connected polyps result not from asexual budding but rather from the fusion of genetically distinct individuals.  相似文献

The intertidal sea anemone Oulactis muscosa (Drayton) is dioecious and most individuals are sexually mature throughout the year. Biochemical genetic evidence was used to determine the genetic structure of populations and to infer the relative contributions of sexual and asexual reproduction to recruitment. Data were collected for six enzyme-encoding loci from local populations spread along 735 km of the south east coast of Australia. The genetic structure of each of the nine local populations studied was consistent with recruitment by sexually produced individuals. In almost all cases, the observed single-locus genotypic frequencies closely matched those expected for hardy-Weinberg equilibria, however, consistent deficits of heterozygotes were detected for all loci. No apparent subdivision of the population was detected within the sampling area. Collections of about 50 individuals from each of five populations of the fissiparous holothurian species Stichopus chloronotus and four populations of Holothuria atra were made in 1999. These populations were located in the Torres Strait (western Pacific) and La Réunion (western Indian Ocean). Allozyme electrophoretic surveys of five (S. chloronotus) and six (H. atra) loci were conducted to compare patterns of asexual reproduction and to investigate connectivity between regions separated by large geographic distances. Deviations from genotype frequencies expected under Hardy-Weinberg equilibrium, mostly heterozygote excesses, were observed in all populations of both species. The maximum contribution of sexual reproduction (calculated as the maximum number of sexually produced individuals: sample size=N*/Ni) was similar for all S. chloronotus (58-64%) and H. atra (76-92%) populations, and on the same level as previously reported for midshelf reefs of the Great Barrier Reef. The higher values in the latter species indicated greater contributions of asexual reproduction to S. chloronotus populations. Variability was strongly reduced in S. chloronotus populations at La Réunion, with only one locus being variable in that population. When the dataset was reduced to one representative per multi-locus genotype per population to reduce the effect of asexual reproduction on calculations on gene flow, FST values were not significantly different from zero, suggesting high gene flow between these regions. However UPGMA cluster analyses using Rogers' genetic distance, roughly clustered populations by region. In the case of H. atra, pooled populations within each region were significantly different from those of the other region. Thus, although some restrictions in gene flow and greater genetic distances between the regions may exist, those differences are distinctly less than those reported in previous studies on echinoderms over similar geographic scales. Despite the importance of asexual reproduction for the maintenance of local population size, this study also confirmed that the potential for widespread dispersal mediated by sexually produced larvae is large.  相似文献

The reproductive cycles of two closely related calcareous sponges, Clathrina coriacea (Montagu, 1818) and C. blanca (Miklucho-Maclay, 1868), were studied to determine whether they were reproductively isolated. Populations of both species were sampled at Santa Catalina Island, California, USA, for 2.5 years. The reproductive period is from July to October in C. coriacea, and from April to August in C. blanca. Seawater temperature and habitat influence reproduction in both species. Fragmentation occurs in C. coriacea in the summer, while asexual reproduction by means of budding is seen in C. blanca throughout the year. The results of the study confirm that the two sponges are separate species.  相似文献

It is known that asexual reproduction by fission is the dominant mode of propagation in the small (disc diameter <5.0 mm) ophiocomid brittle star Ophiocomella ophiactoides (H. L. Clark). This species is, however, able to reproduce sexually as well. Sexual reproduction occurs throughout much or all of the year in a population of O. ophiactoides at Jamaica. Gonads are often present in recently split individuals, indicating that sexual and asexual reproduction could occur simultaneously. Regeneration of new gonads in the new disc half is underway by the time an individual is competent to split again. Unsexable individuals are significantly smaller than males, which in turn are significantly smaller than females. The unsexable individuals appear to be below a threshold size for gonad production. Fecundity is low, with the largest females producing a maximum of about 7 400 eggs of small (mean diameter 80 m) size. Fertilized eggs develop into planktotrophic ophioplutei which are typical of the larvae of other species of ophiocomid brittle stars in morphology, rate of development and duration of planktonic period. Egg size and larval type for O. ophiactoides are characteristic of brittle stars with the planktotrophic mode of development, but egg number is uncharacteristically low. Fissiparity in O. ophiactoides may have evolved in association with small body size, concomitant low fecundity and a high-risk planktotrophic larva as a means of ensuring reliable recruitment. Available data, although sparse, suggest that this hypothesis may be applicable to fissiparous brittle stars in general. Fission in brittle stars has a broad systematic incidence at the familial level which suggests that it has evolved recurrently. The ability both to divide and broadcast appears to be a useful combination for O. ophiactoides: it can recruit reliably at large size to occupy favourable habitats quickly, but, it can still maintain low level dispersal of larvae which could provide escape from deteriorating habitats and the potential to colonize new habitats.Contribution No. 328 of the Discovery Bay Marine Laboratory, University of the West Indies  相似文献

Francis (1979) proposed that clonal and solitary forms of the anemone Anthopleura elegantissima are actually two species. In 1984 and 1985, samples from two to six California populations of all known forms and species of California Anthopleura were analyzed electrophoretically to determine their taxonomic relationships. Data from 14 enzymes and 18 loci, 17 of them polymorphic, show that the two forms of A. elegantissima are virtually identical electrophoretically, and there is no evidence of reduced gene flow between them. We conclude there are three species of Anthopleura in California: A. elegantissima (Brandt, 1835), A. xanthogrammica (Brandt, 1835) and A. artemisia (Dana, 1848). Genetic variation in the two species capable of asexual reproduction, A. elegantissima and A. artemisia, is extremely high, approximately 2.5 times that of the strictly sexual A. xanthogrammica.  相似文献

Asexual reproduction often predominates in populations of species experiencing range expansion. Locally selected genotypes can lead to the establishment of clonal populations that accumulate genetic diversity through time. Sexual reproduction has never been observed in extensive field and culture studies of the red alga Porphyra umbilicalis from Maine, USA, even though sexual reproduction predominates in this species in the eastern Atlantic (Europe). This suggests that Maine populations are indeed asexual and might consist of one or only a few genetic clones; we have tested this using AFLPs. Individuals were sampled at two sites in Maine (Cobscook Bay [n?=?25], Schoodic Point [n?=?26]) and compared to sexual individuals from England (Sidmouth [n?=?17]). The AFLP analysis determined that individuals at two sites in Maine containing putative asexuals were not strictly clonal; however, two multilocus lineages were sampled more than once. Two genetic clones, one at each Maine site, were comprised of 6 individuals each; the 39 additional Maine individuals had distinctive AFLP genotypes. However, when the individuals from Maine were compared with a known sexual population from Sidmouth, England, much greater genetic diversity was found within the sexual population in England. Finally, we examine how preparation of field-collected material for AFLP investigations can affect the inclusion of non-target DNA and demonstrate an in silico approach for removing some cryptic contaminants from analysis.  相似文献

The sexual and asexual phases of the reproductive cycle of the sponge Mycalecontarenii (Martens, 1824) were studied from samples collected over a period of 2 years (June 1994 to May 1996) in a Mediterranean coastal basin (Porto Cesareo, southwestern Apulia). The species is viviparous; year round it shows a discontinuous oocyte production. No males were found, possibly owing to the very short period of spermatogenesis. M.contarenii produces asexual buds during autumn and winter, and this process seems to be related to changes in water temperature. Even though, at present, we cannot evaluate the incidence of larval recruitment, we postulate that asexual reproduction in M.contarenii plays a crucial role in maintaining the sponge population. Reproductive cycle, asexual reproduction, and population dynamics of the fissiparous brittle star, Ophiactis savignyi, which inhabits the exhalant passages of the sponge Haliclona sp. were examined monthly from February 1991 to January 1992 at Wanlitung, southern Taiwan (22°N; 120°E). Mature gametes were found from March to December, but release was mainly in May and June. Sexual recruits were found from May to December, with the highest frequency (14.1% of population) in June. Sexual recruits composed 2.4% of the 1-yr sample. Sex ratio of male to female was 24:1. Fission occurred throughout the year, although the frequency of recently split individuals was lower from January to June (6 to 31%), and higher from July to December (42 to 52%). The occurrence of fission was highest after spawning. Regenerating individuals composed 48.2% of the 1-yr sample. Population density fluctuated greatly during summer due to recruitment by fission and mortality or dispersal due to the stressful environmental conditions. Both sexual and asexual reproduction of O. savignyi were successful at this site.  相似文献

The reproductive response of sexual and asexual female Brachionus plicatilis (Muller) was examined over temperatures ranging from 20° to 40°C, salinities from 5 to 40 S, and food levels from 0.25 to 20 g Chlorella vulgaris dry-weight per ml. Reduced food levels, as well as temperature and salinity extremes, reduced reproduction of both sexual and asexual females, but did so differentially. Reproduction by sexual females was reduced to a greater extent at environmental extremes than asexual females. The broad, flat reproductive response curve of asexual females extended beyond the limits of the narrower, more sharply peaked curve of sexual females. Thus zones of exclusively asexual reproduction exist at environmental extremes where sexual reproduction is physiologically restricted. These results are corroborated by a comparison of the lifetime fecundity of individual sexual and asexual females over a 20°C temperature range. No differences in lifetime fecundity occurred between sexual and asexual females at 18° and 28°C. At 38°C, however, asexual female fecundity reached its highest level, while sexual female fecundity declined 15%. The appearance of sexual females in rotifer populations in the result of both inducible and repressible factors.  相似文献

Electrophoresis was used to provide genetic evidence of the mode of production of brooded planulae in each of four species of scleractinian coral collected from the central region of the Great Barrier Reef during September, October and November 1984. Comparisons were made of the multi-locus genotypes of planulae and their broodparents for two ahermatypic (non zooxanthellate) species,Tubastraea diaphana andT. coccinea and two hermatypic (zooxanthellate) species,Acropora palifera andSeriatopora hystrix. For both ahermatypic species, all planulae were found to be genetically identical to their broodparents, including 91 planulae which were heterozygous for at least one locus. These results are consistent with asexual (ameiotic) reproduction. In contrast, non-parental genotypes were detected in the majority of hermatypic broods, which is consistent with expectations for sexual reproduction with at least some outcrossing. These data confirmed that brooded planulae may be produced both sexually and asexually and countered the suggestion that electrophoretic studies of hermatypic corals may be weakened by the contaminating effect of the enzymes of their symbiotic xooxanthellae.Contribution No. 306 of the Australian Institute of Marine Science  相似文献

Nine species of ophiuroids — Amphipholis januarii, Axiognathus squamatus, Hemipholis clongata, Ophiocnida scabriuscula, Ophiactis lymani, Ophiactis savignyi, Ophionereis reticulata, Ophioderma appressum and Ophiothrix angulata —were collected in various habitats from the mid-littoral and upper infralittoral on the northern coast of the State of São Paulo, Brazil. The abundance of 6 species occurring in 28 species of algae was investigated. A total of 220 samples was collected at monthly intervals from April, 1969 to May, 1970. The distributional pattern of the populations, as determined by Mobisita's index, shows aggregation of the ophiuroids. No ophiuroid species was limited to one algal species. Each species prefers certain algal species and/or a different level on the shore. The most abundant species on algae was Ophiactis lymani, with a maximum density of 1,510 individuals/100 ml of algae. Four species of ophiuroids occurred inside sponges. The most abundant species in sponges was O. savignyi, with a maximum density of 1,892 individuals/100 g of dry sponge. Two species of ophiuroids occurred inside the bryozoan Schizoporella unicornis. Feeding observations have shown Axiognathus squamatus to capture its food by mucous nets. Decapod crustaceans fed on ophiuroids in aquaria. The copepod Thespesiopsyllus sp. was found in the body cavity of the 2 Ophiactis species. Simultaneous sexual and asexual reproduction were abserved in O. savignyi.  相似文献

Samples of the scleractinian coral Pocillopora damicornis were collected from six sites located around four islands in the Ryukyu Archipelago, southern Japan, and subjected to allozyme electrophoresis. Seven polymorphic loci were examined for their allelic patterns. The ratio of observed to expected genotypic diversity (0.30 < G o :G e  < 0.64), the ratio of the observed number of genotypes to the number of individuals (0.47 < N g :N i  < 0.75), and deviations from Hardy–Weinberg equilibrium indicated that asexual reproduction plays a major role in the maintenance of established populations. However, populations were not completely dominated by a single or a few clones, and most clones were represented by only a few individual samples. The high frequency of typhoons in the region suggests that, in P. damicornis, fragmentation caused through occasional exposure to powerful waves is a major mode of asexual reproduction, but asexual production of planulae may also be contributing to the maintenance of populations. A significant genetic differentiation (F ST) was found between the six populations examined (0.027 < F ST < 0.092, average F ST = 0.056). The moderate gene flow is discussed according to characteristics of the larval stage of the species, and to circulation patterns in the region. The anemone Amphianthus inornata is found at bathyal depths living on colonies of the gorgonian Acanella arbuscula. Previous studies of the morphology and reproductive stage of this anemone, during different times of the year, have indicated that it reproduces sexually on a seasonal basis. A small proportion of the study population were also reported to be undergoing asexual reproduction by fission. The anemone Kadosactis commensalis is also bathyal, but lives mainly on the holothurian Paroriza prouhoi. Previous morphological studies have indicated that K. commensalis is a protandrous hermaphrodite that exhibits non-seasonal sexual reproduction only. In the present study, allozyme electrophoresis was used to examine the prevalence and genetic consequences of asexual reproduction in a population of Amphianthus inornata from 2 200 m in the Rockall Trough, North Atlantic Ocean. Genetic evidence, from five randomly selected polymorphic enzyme loci, for asexual reproduction in this species was weak. Exact tests indicated that genotype frequencies did not differ significantly from those expected under Hardy–Weinberg equilibrium. F IS (correlation of homologous alleles with reference to local population, assuming random mating) values also did not differ significantly from zero, and observed heterozygosity (H o =0.446) and genotypic diversity (G o =17.0387) were very similar to Hardy–Weinberg expected frequencies (H e =0.446; G * e =17.0010). Evidence suggests that the contribution of asexual reproduction to recruitment in the study population of A. inornata is low. For a single population of K. commensalis from 4 850 m on the Porcupine Abyssal Plain, North Atlantic Ocean, the hypothesis that inbreeding due to reduced occurrence of outcrossing between anemones on a single holothurian host was examined by electrophoresis of ten randomly selected enzyme loci. Single-locus genotypic frequencies were significantly different from expected frequencies for one locus P≤ 0.05, hexokinase-1 (Hex-1)]. F IS values were significantly different from zero for two enzyme loci (Hex-1 and Hex-2, P≤ 0.01 and P≤ 0.05, respectively), and the overall observed heterozygosity was lower than the expected heterozygosity (H o =0.125, H e =0.140). The hypothesis of inbreeding could not be rejected by the present study, although sample size was small (N=55), leading to possible bias in tests of significance. Genetic variation in A. inornata was higher than that recorded for most eukaryotes, although interlocus sampling error for only five loci is high. High genetic variability has been found in other sea anemones, and has been related to high longevity and mixed reproductive modes. Hydrozoan species are renowned for flexible asexual reproduction, which may predispose them to be successful invaders. Polyps of the invasive hydrozoan Moerisia lyonsi (Boulenger, 1908) have very high rates of asexual production of both polyp and medusa buds. In order to determine how environmental factors affect asexual reproduction in M. lyonsi, the quantitative relationships between polyp bud and medusa bud production were studied in a 31-day laboratory experiment during August 2001. The combined effects of prey (4, 8, 12, 16 Acartia tonsa copepods polyp–1 day–1), temperature (20°C, 29°C), and salinity (5, 15, 25) were tested on the development times for polyp buds (DTp) and medusa buds (DTm), the total asexual reproduction rate (ARR, no. buds polyp–1 day–1), and the ratio of medusa bud to total bud production (Rm). Greater food consumption significantly and directly enhanced ARR and Rm and shortened DTp and DTm. A lower temperature (20°C) and higher salinity (25) reduced food consumption, lengthened development times, and decreased ARR and Rm, with opposite effects for the higher temperature (29°C) and lowest salinity (5). The patterns of variation of these reproductive parameters are more complex. DTm was most sensitive and was significantly and directly affected by all three measured factors. In addition to food consumption, direct effects were seen by temperature on DTp and by salinity on Rm. ARR was directly affected only by food consumption. Overall, DTp, DTm, and Rm were more sensitive to environmental differences than was ARR. More favorable conditions enhanced medusa bud production.
