Although a number of manufactured nanoparticles are applied for the medical and clinical purposes, the understanding of interaction between nanomaterials and biological systems are still insufficient. Using nematode Caenorhabditis elegans model organism, we here investigated the in vivo toxicity or safety of hydroxylated fullerene nanoparticles known to detoxify anti-cancer drug-induced oxidative damages in mammals. The survival ratio of C. elegans rapidly decreased by the uptake of nanoparticles from their L4 larval stage with resulting in shortened lifespan (20 d). Both reproduction rate and body size of C. elegans were also reduced after exposure to 100 μg mL−1 of fullerol. We found ectopic cell corpses caused by apoptotic cell death in the adult worms grown with fullerol nanoparticles. By the mutation of core pro-apoptotic regulator genes, ced-3 and ced-4, these nanoparticle-induced cell death were significantly suppressed, and the viability of animals consequently increased despite of nanoparticle uptake. The apoptosis-mediated toxicity of nanoparticles particularly led to the disorder of digestion system in the animals containing a large number of undigested foods in their intestine. These results demonstrated that the water-soluble fullerol nanoparticles widely used in medicinal applications have a potential for inducing apoptotic cell death in multicellular organisms despite of their antioxidative detoxifying property.  相似文献

Increasing use of nanoparticles in daily products is of great concern today, especially when their positive and negative impact on environment is not known. Hence, in current research, we have studied the impact of silver nanoparticle (AgNPs) and silver nitrate (AgNO3) application on seed germination, root, and shoot length of castor bean, Ricinus communis L. plant. Silver nanoparticles had no significant effects on seedling growth even at higher concentration of 4,000 mg L?1, while the silver in bulk form as AgNO3 applied on the castor bean seeds inhibited the seed germination. Silver uptake in seedlings of the castor seeds on treatment with both the forms of silver was confirmed through atomic absorption spectroscopy studies. The silver nanoparticle and silver nitrate application to castor seeds also caused an enhanced enzymatic activity of ROS enzymes and phenolic content in castor seedlings. High-performance liquid chromatography analysis of individual phenols indicated enhanced content of parahydroxy benzoic acid. These kinds of studies are of great interest in order to unveil the movement and accumulation of nanoparticles in plant tissues for assessing future applications in the field or laboratory.  相似文献

Two types of nano-scale zero-valent iron (nZVI-B prepared by borohydride reduction and nZVI-T produced by thermal reduction of iron oxide nanoparticles in H2) and a micro-scale ZVI (mZVI) were compared for PCB degradation efficiency in water and soil. In addition, the ecotoxicity of nZVI-B and nZVI-T particles in treated water and soil was evaluated on bacteria, plants, earthworms, and ostracods. All types of nZVI and mZVI were highly efficient in degradation of PCBs in water, but had little degradation effect on PCBs in soil. Although nZVI-B had a significant negative impact on the organisms tested, treatment with nZVI-T showed no negative effect, probably due to surface passivation through controlled oxidation of the nanoparticles.

As in vivo system, we propose Drosophila melanogaster as a useful model for study the genotoxic risks associated with nanoparticle exposure. In this study we have carried out a genotoxic evaluation of titanium dioxide (TiO2), zirconium oxide (ZrO2) and aluminium oxide (Al2O3) nanoparticles and their microparticulated forms in D. melanogaster by using the wing somatic mutation and recombination assay. This assay is based on the principle that loss of heterozygosis and the corresponding expression of the suitable recessive markers, multiple wing hairs and flare-3, can lead to the formation of mutant clones in treated larvae, which are expressed as mutant spots on the wings of adult flies. Third instar larvae were feed with TiO2, ZrO2 and Al2O3 nanoparticles, and their microparticulated forms, at concentrations ranging from 0.1 to 10 mM. Although a certain level of aggregation/agglomeration was observed in solution, it must be noted than the constant digging activity of larvae ensures that treated medium pass constantly through the digestive tract ensuring exposure. The results showed that no significant increases in the frequency of all spots (e.g. small single, large single, twin, total mwh and total spots) were observed, indicating that these nanoparticles were not able to induce genotoxic activity in the wing spot assay of D. melanogaster. Negative data were also obtained with the microparticulated forms. This indicates that the nanoparticulated form of the selected nanomaterials does not modify the potential genotoxicity of their microparticulated versions. These in vivo results contribute to increase the genotoxicity database on the TiO2, ZrO2 and Al2O3 nanoparticles.  相似文献

Aqueous phenol solutions containing TiO(2) nanoparticles were irradiated with ultraviolet (UV), gamma-ray and electron beams. Organic compounds were fully removed by each type of radiation in the presence of the particles. The absorbed energy of the ionizing radiation (gamma-ray and electron beams) needed for removal was much lower than that of UV photocatalysis. Phenol was decomposed by the ionizing radiation in the absence of the nanoparticles and the addition of TiO(2) had no significant effect on phenol decomposition rate. Instead, total organic carbon (TOC) removal using the ionizing radiation was accelerated drastically by TiO(2). It is suggested that TiO(2) particles affect the intermediate compounds produced through the decomposition of phenol. The amount of removed TOC per absorbed energy were compared in the absence and the presence of TiO(2) nanoparticles. Radiolysis with the nanoparticles showed consistently high rate and high efficiency of TOC removal.  相似文献

Zinc oxide nanoparticles (ZnO NPs) are used in an array of products and processes, ranging from personal care products to antifouling paints, textiles, food additives, antibacterial agents and environmental remediation processes. Soils are an environment likely to be exposed to manmade nanoparticles due to the practice of applying sewage sludge as a fertiliser or as an organic soil improver. However, understanding on the interactions between soil properties, nanoparticles and the organisms that live within soil is lacking, especially with regards to soil bacterial communities. We studied the effects of nanoparticulate, non-nanoparticulate and ionic zinc (in the form of zinc chloride) on the composition of bacterial communities in soil with a modified pH range (from pH 4.5 to pH 7.2). We observed strong pH-dependent effects on the interaction between bacterial communities and all forms of zinc, with the largest changes in bacterial community composition occurring in soils with low and medium pH levels (pH 4.8 and 5.9). The high pH soil (pH 7.2) was less susceptible to the effects of zinc exposure. At the highest doses of zinc (2500 mg/kg dw soil), both nano and non-nano particulate zinc applications elicited a similar response in the soil bacterial community, and this differed significantly to the ionic zinc salt treatment. The results highlight the importance of considering soil pH in nanotoxicology studies, although further work is needed to determine the exact mechanisms controlling the toxicity and fate and interactions of nanoparticles with soil microbial communities.  相似文献

Nanoparticles offer the potential to improve environmental treatment technologies due to their unique properties. Adsorption of metal ions (Pb(II), Cd(II), Cu(II), Zn(II)) to nanohematite was examined as a function of sorbent concentration, pH, temperature, and exhaustion. Adsorption experiments were conducted with 0.05, 0.1, and 0.5 g/L nanoparticles in a pH 8 solution and in spiked San Antonio tap water. The adsorption data showed the ability of nanohematite to remove Pb, Cd, Cu, and Zn species from solution with adsorption increasing as the nanoparticle concentration increased. At 0.5 g/L nanohematite, 100 % Pb species adsorbed, 94 % Cd species adsorbed, 89 % Cu species adsorbed and 100 % Zn species adsorbed. Adsorption kinetics for all metals tested was described by a pseudo second-order rate equation with lead having the fastest rate of adsorption. The effect of temperature on adsorption showed that Pb(II), Cu(II), and Cd(II) underwent an endothermic reaction, while Zn(II) underwent an exothermic reaction. The nanoparticles were able to simultaneously remove multiple metals species (Zn, Cd, Pb, and Cu) from both a pH 8 solution and spiked San Antonio tap water. Exhaustion experiments showed that at pH 8, exhaustion did not occur for the nanoparticles but adsorption does decrease for Cd, Cu, and Zn species but not Pb species. The strong adsorption coupled with the ability to simultaneously remove multiple metal ions offers a potential remediation method for the removal of metals from water.  相似文献

Along with the increasing utilization of engineered nanoparticles, there is a growing concern for the potential environmental and health effects of exposure to these newly designed materials. Understanding the behavior of nanoparticles in the environment is a basic need. The present study aims to investigate the distribution and fate of ceria nanoparticles in an aquatic system model which consists of sediments, water, hornworts, fish and snails, using a radiotracer technique. Concentrations of ceria in the samples at regular time intervals were measured. Ceria nanoparticles were readily removed from the water column and partitioned between different organisms. Both snail and fish have fast absorption and clearance abilities. Hornwort has the highest bioaccumulation factors. At the end of the experiment, sediments accumulated most of the nanoparticles with a recovery of 75.7 ± 27.3% of total ceria nanoparticles, suggesting that sediments are major sinks of ceria nanoparticles.  相似文献

Soil pollution by polychlorinated biphenyls (PCBs) arising from the crude disposal and recycling of electronic and electrical waste (e-waste) is a serious issue, and effective remediation technologies are urgently needed. Nanoscale zerovalent iron (nZVI) and bimetallic systems have been shown to promote successfully the destruction of halogenated organic compounds. In the present study, nZVI and Pd/Fe bimetallic nanoparticles synthesized by chemical deposition were used to remove 2,2′,4,4′,5,5′-hexachlorobiphenyl from deionized water, and then applied to PCBs contaminated soil collected from an e-waste recycling area. The results indicated that the hydrodechlorination of 2,2′,4,4′,5,5′-hexachlorobiphenyl by nZVI and Pd/Fe bimetallic nanoparticles followed pseudo-first-order kinetics and Pd loading was beneficial to the hydrodechlorination process. It was also found that the removal efficiencies of PCBs from soil achieved using Pd/Fe bimetallic nanoparticles were higher than that achieved using nZVI and that PCBs degradation might be affected by the soil properties. Finally, the potential challenges of nZVI application to in situ remediation were explored.  相似文献

In this study, the influence of the co-existence of TiO2 nanoparticles on the speciation of arsenite [As(III)] was studied by observing its adsorption and valence changing. Moreover, the influence of TiO2 nanoparticles on the bioavailability of As(III) was examined by bioaccumulation test using carp (Cyprinus carpio). The results showed that TiO2 nanoparticles have a significant adsorption capacity for As (III). Equilibrium was established within 30 min, with about 30% of the initial As (III) being adsorbed onto TiO2 nanoparticles. Most of aqueous As (III) was oxidized to As(V) in the presence of TiO2 nanoparticles under sunlight. The carp accumulated considerably more As in the presence of TiO2 nanoparticles than in the absence of TiO2 nanoparticles, and after 25-day exposure, As concentration in carp increased by 44%. Accumulation of As in viscera, gills and muscle of the carp was significantly enhanced by the presence of TiO2 nanoparticles.  相似文献

Several studies show that ultrafine particles have a larger surface area than coarse particles, thus causing a greater inflammatory response. In this study, we investigated chemical and biological oxidative effects of nanoparticles in vitro. Carbon black (CB) nanoparticles with mean aerodynamic diameters of 14, 56, and 95nm were examined. The innate oxidative capacity of the CB nanoparticles was measured by consumption of dithiothreitol (DTT) in cell-free system. The expression of heme oxygenase-1 (HO-1) in rat alveolar type II epithelial cell line (SV40T2) and alveolar macrophages (AM) exposed to CB nanoparticles was measured by ELISA. DTT consumption of 14nm CB was higher than that of other CB nanoparticles having the same particle weight. However, DTT consumption was directly proportional to the particle surface area. HO-1 protein in SV40T2 cells was significantly increased by the 14nm and 56nm CB, however, 95nm CB did not affect. HO-1 protein in AM was significantly increased by the 14, 56, and 95nm CB. The increase in HO-1 expression was diminished by N-acetyl-l-cysteine (NAC) treatment of each CB nanoparticles before exposure although the difference between the effects of NAC-treated and untreated 14nm CB did not achieve significant. In conclusion, CB nanoparticles have innate oxidative capacity that may be dependent on the surface area. CB nanoparticles can induce oxidative stress in alveolar epithelial cells and AM that is more prominent with smaller particles. The oxidative stress may, at least partially, be mediated by surface function of particles.  相似文献
