The aim of this study was to follow contamination of ready-to-eat food with Listeria monocytogenes by using the Step One real time polymerase chain reaction (PCR). We used the PrepSEQ Rapid Spin Sample Preparation Kit for isolation of DNA and MicroSEQ? Listeria monocytogenes Detection Kit for the real-time PCR performance. In 30 samples of ready-to-eat milk and meat products without incubation we detected strains of Listeria monocytogenes in five samples (swabs). Internal positive control (IPC) was positive in all samples. Our results indicated that the real-time PCR assay developed in this study could sensitively detect Listeria monocytogenes in ready-to-eat food without incubation.  相似文献

A repository of 39 and 34 gaseous organic compounds in compressed gas cylinders at PPM (5 to 700) and PPB levels (1 to 10,000) respectively has been established by EPA for use in performance audits. In this program, to ensure that the concentration of each gas standard has not changed, each standard is periodically analyzed for stability. The gas mixtures were initially analyzed once or twice by the manufacturer before shipment to Research Triangle Institute (RTI). The gas mixtures were then analyzed at RTI upon receipt from the gas manufacturer and then reanalyzed at 2, 6 and 12 months, and annually thereafter, to determine any change in concentration.

An examination of the stability data shows that concentrations for all the organics tested usually varied by less than 10 percent over a period of several years. Ethylene oxide and propylene oxide and trans 1, 4-dichloro-2-butene below the 10 ppb level have been found to be unstable. Six other compounds at the PPM level (aniline, cyclohexanone, p-dichlorobenzene, ethylamine 1, 2-dibromoethylene, and formaldehyde) have been found to be unstable in compressed gas cylinders.  相似文献

Abstract

Loss of filtration efficiency in a fabric filter baghouse is typically caused by bag failure, in one form or another. The degree of such failure can be as minor as a pinhole leak or as major as a fully involved baghouse fire. In some cases, local air pollution regulations or federal hazardous waste laws may require estimation of the total quantity of particulate matter released to the environment as a result of such failures. In this paper, a technique is presented for computing the dust loading in the baghouse exhaust when one or more bags have failed. The algorithm developed is shown to be an improvement over a previously published result, which requires empirical knowledge of the variation in baghouse pressure differential with bag failures. An example calculation is presented for a bag-house equipped with 200 bags. The prediction shows that a small percentage of failed bags can cause a relatively large proportion of the gas flow to bypass the active bags, which, in turn, leads to high outlet dust loading and low overall collection efficiency from the baghouse.  相似文献

Solanum tuberosum L. cv Norchip plants were grown in open-top chambers in the summer of 1986. Plants were treated with charcoal-filtered air, nonfiltered air, or nonfiltered air supplemented with 33, 66, or 99% of the ambient ozone (O3) concentrations from 1000 to 2000 h eastern daylight time daily. In addition, plants received charcoal-filtered air plus 0, 0.15 (393 microg m(-3)), 0.34 (891 microg m(-3)), or 0.61 (1598 microg m(-3)) ppm sulfur dioxide (SO2) from 0900 to 1200 h once every 14 d for a total of four treatments. Ozone induced a linear reduction in number and weight of Grade One (> 6.35-cm diameter) potato tubers and in total weight of tubers. Ozone also induced linear reductions in the percentage of dry matter of tubers and linear decreases in glucose and fructose content of Grade One tubers. Sulfur dioxide induced a stimulation and then decline of the number, percentage of dry matter, and sucrose content of Grade One tubers. The SO2 response best fit a quadratic curve. No O3 x SO2 interactions were detected for any of the yield or quality functions measured.  相似文献
