Copper and zinc interaction on clearance from water and distribution in different tissues was investigated for the freshwater mussel, Cristaria plicata, under laboratory conditions. Clearance rate of Cu or Zn from water was highly dependent on exposure concentration. Interaction effect was most evident at 300 ??g??L?1 Cu exposure and depressed the Zn clearance rate significantly (p<0.05). However, the presence of 100 ??g??L?1 and 300 ??g??L?1 Zn hardly affected the Cu clearance rate. The 300 ??g??L?1 Cu presence enhanced Cu accumulation in each tissue most significantly (p<0.01), but caused Zn content to decrease in the gills by 62% (p<0.05), viscera by 49% (p<0.05) and foot by 31% (p<0.05), and increase in the mantle by 97% (p<0.05) and the muscles by 243% (p<0.05) for different Zn exposure treatments. The response of metal accumulation in various tissues of the test mussels indicated that Zn transferred from the gills, viscera and foot to the mantle and muscles might be one of the important characteristics of the Zn regulatory mechanism by leading to a narrow range of Zn concentration in the different tissues.  相似文献

ABSTRACT

Lead (Pb) is one of the most toxic heavy metals that affect the physiological status of aquatic organisms. The present investigation evaluated the possible toxic effect of lead chloride (PbCl2) on biomarkers responses, DNA damage and histological alterations in Venus verrucosa gills and digestive gland. Three concentrations of PbCl2 (D1:1µgL?1, D2: 10µgL?1 and D3: 100µgL?1) were chosen for V. verrucosa exposure during six days. At the end of the trial, it was found that Pb tended to accumulate in both gills and digestive gland in a dose-dependent manner. However, gill tissues exhibited the highest metal burden. Our results showed an increase of malondialdehyde, protein carbonyls and advanced oxidation protein product levels in both organs following PbCl2exposure. The induction of both non-enzymatic and enzymatic antioxidant systems; as well as the decrease of the acetylcholinesterase activity and degradation of DNA structure was recorded in the gills and digestive gland. The histopathological alterations observed in gills (disruption of lamellas and cilia filaments?…) and digestive gland (lumens occlusion, necrosis and fibrosis) confirmed the aforementioned results. Our data highlighted the short-term toxicity effects of PbCl2 on V. verrucosa and pointed out a high sensitivity of gills towards this metal.  相似文献

The accumulations of Al, Cd, Cr, Cu, Fe, Ni, Pb and Zn in the exoskeleton, gills, hepatopancreas and abdominal muscles of crayfish Astacus leptodactylus (Eschscholtz, 1823) were determined. The strongest correlation observed was between Cr and Ni in the gills (r?=?0.904); moderate to strong correlations between Al, Cr, Fe, Ni and Cu were also observed in gill tissue. Disregarding the gills, the strongest correlation was found between Cu and Zn in the hepatopancreas (r?=?0.808); the correlation between these two metals might have been a result of metallothionein activity. The accumulation of Pb was found to correlate with that of Cd in the exoskeleton, Cd and Zn in the gills, Zn and Cu in the hepatopancreas and Cu in the abdominal muscle. None of these correlations were present in lakewater and sediment samples, suggesting that the crayfish metabolism may be responsible for the co-accumulation of metal–metal pairs. As all correlations in non-gill tissues are observed between divalent metals, a shared transporter such as divalent metal transporter 1 might be involved in the accumulation of these metals.  相似文献

Uptake of Pb, Cu and Zn by different epiphytic moss species Floribundaria floribunda (Doz. &; Molk.) Fleisch., Taxiphyllum giraldii (C. Muell.) Fleisch. and Thuidium sparsifolium (Mitt.) Jaeg., an epilithic moss Thuidium delicatulum (L.) Mitt. and a leafy liverwort Ptychanthus striatus (Lehm. &; Linderb.) Nees. was studied experimentally. The plant bodies were treated with single metal and mixed metal solutions of Pb(NO3)2, CuCl2 and ZnCl2 at different concentrations ranging from 10?10 to 10?2 mol L?1. Higher uptake of Pb, Cu and Zn by T. giraldii, T. sparsifolium and P. striatus than by F. floribunda and T. delicatulum was shown in both treatments. Compared with the single metal treatment, the uptake of Zn was very low at higher concentrations in the mixed metal treatment. A slight depletion of Zn was noticed in the long-term exposure. Leaching of naturally accumulated Zn from the mosses and leafy liverwort was also observed when treated with higher concentrated (10?4, 10?2 mol L?1) Pb(NO3)2 and CuCl2 solutions. Similarly, the leaching of naturally accumulated Cu was observed in leafy liverwort P. striatus with 10?2 mol L?1 Pb(NO3)2 and ZnCl2 treatments. However, insignificant changes were observed on naturally accumulated Pb and Cu in mosses. From these experiments, the epiphytic mosses T. giraldii and T. sparsifolium, which have high Cation exchange capacity and large leaf surface area, have been found to be suitable for assessing heavy metal concentration in a moderately or slightly contaminated environment.  相似文献

Individuals of Mytilus edulis (collected from Sequim Bay, Washington, in April and August, 1980) were exposed to 5 g l-1 mercury as HgCl2 for 28 d. Gill mercury accumulation, mercury incorporation into the soluble fraction and low molecular weight, mercury-binding proteins of gills, and induction of these mercury-binding proteins were determined as a function of time. Short-term mercury uptake rates of excised gills were also determined for mussels sampled at intervals during exposure. Gill mercury accumulation occurred in three phases, represented by net uptake phases initially (up to Day 4) and toward the end of the exposure (Days 15 to 28), and an intermediate stable phase (Days 4 to 15). The stable phase was associated with induction of the predominant mercury-binding proteins and mercury incorporation into the proteins. After Day 15, the mercury-binding proteins were saturated and spillover of mercury into high molecular weight proteins had occurred. This was associated with saturation of the soluble fraction, increases of mercury on particulate fractions, and a loss of the ability of gills to maintain stable mercury concentrations. Mercury uptake rates of gills were not affected by the 28 d exposure of the whole organism.  相似文献

Although aquaculture is considered the fastest growing food production industry, nevertheless there is little information regarding pollutant levels in cultured fish. Samples of cultured sea bass – Dicentrarchus labrax (Linnaeus, 1758), sea bream – Sparus aurata (Linnaeus, 1758), and sharpsnout sea bream – Diplodus puntazzo (Walbaum, 1792) from three fish farms located in the Eastern Mediterranean which utilized antifouling paints on the nets were analyzed for quantitative determination of zinc (Zn), chrome (Cr), and copper (Cu) in the gills, the liver, and the muscle separately for each tissue and fish. The results show that the highest levels of zinc (up to 29.6 mg kg?1 dry wt) were found in the gills of all samples of sea bream and sharpsnout sea bream while in the samples of sea bass, the highest levels of zinc and chrome were found in the liver (up to 39.3 and 13.8 mg kg?1 dry wt, respectively). Insofar, as copper is concerned the highest exposure was found in the liver of sharpsnout sea bream (up to 5.49 mg kg?1 dry wt). The results indicate that the use of antifouling paints in aquaculture practices are a significant source of chemical pollution in cultured fish and poses a risk to the exposed organisms.  相似文献

The acid-base status of two sea urchins, Psammechinus miliaris (Gmelin) and Echinus esculentus (L.) during experimental emersion has been investigated. Sea urchins were collected from the Firth of Clyde between August and September 1987. The carbon dioxide capacity of the coelomic fluid of P. miliaris was greater than that of E. esculentus, although both were low and only marginally greater than that of sea water. The pH of the coelonic fluid was also low (7.05 to 7.17) and was influenced mainly by the internal partial pressure of CO2 (PCO 2). Acid-base disturbance in the coelomic fluid of both species during emersion, although minimal, was more pronounced in E. esculentus than in P. miliaris and was due primarily to an increase in the internal PCO 2, although there was an increase in the concentration of L-lactate in the coelomic fluid of E. esculentus. The coelomic fluid of both species was in a state of perfectly compensated respiratory acidosis. An increase in the concentration of divalent ions (Ca2+ and Mg2+) may be related to the dissolution of the test as a source of carbonate buffer.  相似文献

The mullet fish, Liza klunzingeri, commercially important and widely relished by Kuwaiti residents, and the stressed ecosystem in Kuwait Bay instigated us to conduct toxicity and bioaccumulation tests on heavy metals (Pb, Ni, V, Cu and Fe). Among five metals, Pb had the lowest observed effect concentration (LOEC) at 1 μg?l ?1. Using multi-factor Probit analysis, toxicity tests (72 h) on L. klunzingeri reared in filtered sea water in the laboratory showed Pb with maximum effect at median lethal concentration (LC50) followed by V, Ni, Cu and Fe. Their bioaccumulation factor (BAF) was in the sequence Pb>V>Fe>Cu and Ni. For fish exposed for 30 d, bioaccumulation exhibited increasing metal levels in liver followed by gills and muscles. These results suggest the potential use of L. klunzingeri as a bioindicator of metal pollution in the future.  相似文献

The uptake and effect of dissolved copper on regulation of hemolymph osmolality and Na+, K+, Cl-, Ca++, and Mg++ concentrations in the shore crab Carcinus maenas (L.) were determined at 400 mOsm (=14 S) ambient salinity. One mg Cu l-1 resulted in 50% mortality in 11 to 22 d; the highest sensitivity was observed around the moulting period. 0.25 and 0.5 mg Cu l-1 were not lethal after a onemonth exposure. Ten, 1, and 0.5 mg Cu l-1 altered regulation of osmolality, Na+, K+, and Cl- concentrations, while 0.25 mg Cu l-1 did not. Exposure to 1 mg Cu l-1 reduced hemolymph osmolality and Na+, K+, and Cl- concentrations to 80 to 90% of controls within 4 to 6 d and no further reduction was observed during a 21-d exposure. The effect of various copper concentrations on these four parameters were almost identical and the highest sensitivity was observed around the moulting period. Hemolymph calcium levels increased 20 to 80% in crabs exposed to 1 mg Cu l-1, while they decreased 20 to 30% in crabs exposed to 0.5 mg Cu l-1. Prolonged exposure to copper caused 20 to 70% reductions in hemolymph magnesium levels. Crabs exposed to 0.5 mg Cu l-1 for 29 d accumulated copper in hepatopancreas, gills, carapace, heart, testes, and hypodermis, but not in muscles and hemolymph. Increased copper levels in crabs exposed to 0.25 mg Cu l-1 were observed in hepatopancreas, gills, and carapace only. The present results suggest that effects of copper on ion regulatory processes in part explain why the toxicity of copper towards euryhaline invertebrates increases at low salinities.  相似文献

Studies concerning bioaccumulation kinetics and bioconcentration factor (BCF) of heavy metals like zinc (Zn), lead (Pb), chromium (Cr), cadmium (Cd), and copper (Cu) in earthworm Eudrilus eugeniae tissues including integument, gizzard, clitellum, and head region were undertaken. Calculated BCF, predicted K ow, and predicted K oc showed a significant correlation between heavy metals in different earthworm tissues, in substrate spiked with heavy metals. The regression coefficient (r 2) between heavy metal uptake concentration and exposure time varied between 0.73 and 0.99, indicating significant correlation. The K oc was a maximum of 13.9016 in case of Cu and integument at an exposure time of 100 days and a minimum of 0.1114 in case of Cr with respect to head at the same exposure time. Earthworms accumulated heavy metals following chronic exposure to municipal solid waste containing heavy metals. BCF and uptake rate kinetics of heavy metals were calculated and showed increased values in head tissue followed by integument.  相似文献

Tadpoles of the common freshwater Sunda toad, Duttaphrynus melanostictus (Amphibia, Bufonidae), were exposed for a 4-day period under laboratory conditions to copper (Cu), cadmium (Cd), zinc (Zn), lead (Pb), nickel (Ni), iron (Fe), aluminum (Al), and manganese (Mn) at various concentrations. Mortality was assessed and median times of death (LT50) and lethal concentrations (LC50) were calculated. LT50 and LC50 increased with the decrease in mean exposure times and concentrations for all metals. LC50 (96?h) for Cu, Cd, Zn, Pb, Ni, Fe, Al, and Mn were 0.03, 0.3, 4.2, 1.5, 8.8, 0.4, 1.9, and 39?mg?L?1, respectively. Cu was the most toxic to D. melanostictus, followed by Cd, Fe, Al, Pb, Zn, Ni, and Mn (Cu?>?Cd?>?Fe?>?Al?>?Pb?>?Zn?>?Ni?>?Mn). Duttaphrynus melanostictus is similarly sensitive to these metals as other amphibian tadpoles.

Hg、Cu、Cd、As、Zn对日本对虾胚胎LC50值分别为:0.0177 mg·L-1、0.1070 mg·L-1、1.6057 mg·L-1、3.3682 mg·L-1、8.2644 mg·L-1,致死效应最低可观测效用浓度(LOEC)分别为0.0037 mg·L-1、0.0166 mg·L-1、0.1302 mg·L-1、1.1572 mg·L-1、2.1522 mg·L-1;抑制发育LOEC分别为0.0062 mg·L-1、0.0176 mg·L-1、0.4002 mg·L-1、1.9497 mg·L-1、0.0617 mg·L-1。

Metabolic changes and toxic effects in Crangon crangon (collected during summer 1989 near Helsingør, Denmark and at Kulhuse in the Isefjord) were observed during exposure to different arsenate concentrations. Survival, respiration, and concentrations of hemolymph glucose, muscle glycogen andadenine nucleotides were measured. C. crangon was very tolerant of arsenate; survival was only affected at arsenate concentrations >25 ppm. Small individuals (0.2 to 0.29 g wet weight) were less tolerant than medium-sized and large individuals (0.5 to 0.59 g and 0.8 to 0.89 g wet weight), e.g. at 50 ppm the LT50 for small individuals was 180 h compared to 343 h and 360 h in middle-sized and large individuals. The relation between size and toxicity is probably due to the change in surface/volume ratio. Under clean conditions the respiration of C. crangon showed a clear relation between routine MO2 (O2 in mlg-1 h-1) and wet weight (r2=0.89; P<0.001). During shortterm exposure to arsenate (10 and 50 ppm), the respiratory rates (MO2) seemed to decrease in a size-dependent manner. The concentrations of blood glucose and muscle glycogen did not change in response to arsenate exposure. The concentrations of adenylate phosphates were high and constant throughout the experiments. C. crangon had maximal adenylate energy charge (AEC) values of ca. 0.8. No decrease in AEC due to the exposure to arsenate was observed.  相似文献

Metal pollution produces damage to marine organisms at the cellular level possibly leading to ecological imbalance. The present investigation focused on the acute and chronic toxicity of lead (Pb) and zinc (Zn) by examining the effects of biomarker enzymes in post-larvae of Penaeus monodon (Tiger prawn). Antioxidant biomarker responses such as lipid peroxidation (LPO) and catalase (CAT) activity for Pb and Zn were determined following chronic exposure. Acute Lethal Concentration 50 (LC50) values observed in the study at 96?h for Pb and Zn at 5.77?±?0.32?mg?L?1 and 3.02?±?0.82?mg?L?1, respectively. The estimated No Observed Effect Concentration and Lowest Observed Effect Concentration values for Pb were 0.014 and 0.029?mg?L?1 and that recorded for Zn was 0.011 and 0.022?mg?L?1, respectively. Among the two metals studied, toxicity of Zn was found to be greater to P. monodon than Pb. The activities of antioxidant defense enzymes and total protein content differed significantly from control following exposure to both metals. Overall, the biomarker studies demonstrated that alterations in antioxidant enzymes and induction of LPO reflect the consequences of heavy metal exposure in P. monodon.  相似文献

The effects of cadmium exposure and dietary status on cadmium accumulation, fatty acid (FA) content and profiles were investigated in two colour forms of the shore crab Carcinus maenas. Groups of shore crabs were either starved or fed with blue mussels, Mytilus edulis, during a 40 d exposure period to 2 or 6 μM Cd2+ (as CdCl2). Starved green individuals accumulated more cadmium in haemolymph and hepatopancreas than did red crabs and green crabs fed during the experiments. In the red colour form, no difference in cadmium accumulation was observed between starved and fed individuals. In both colour forms, hepatopancreas contained more FA than gills and muscle. The FAs often present in the largest amounts in the tissues were 16:0, 16:1ω7, 18:1ω7, 18:1ω9, 20:4ω6, 20:5ω3 and 22:6ω3. However, saturated (SAFA) and mono-unsaturated fatty acids (MUFAs) were dominant in hepatopancreas, whereas poly-unsaturated fatty acids (PUFAs) were dominant in gills and muscles. At the beginning of the experiment, the total FA content in the hepatopancreas was 111.6 mg g−1 (dry weight) for red crabs and 78.4 mg g−1 for green shore crabs. During the experiment, however, the FA content decreased in red crabs. This decrease was more pronounced for starved individuals than for fed individuals. Also, the decrease in FA content was more pronounced in crabs exposed to 6 μM cadmium compared to crabs exposed to 2 μM or crabs not exposed to cadmium. No change in FA content was observed in green shore crabs, irrespective of diet and cadmium exposure. For both colour forms, no change in FA content was observed for gills and muscle. In red crabs, a decrease was observed for all FAs in the hepatopancreas. This decrease, however, was more pronounced for SAFAs and MUFAs than for PUFAs, indicating that the metabolism of FAs during starvation and cadmium exposure is selective. The experiments indicate that green colour forms of shore crabs are more tolerant of natural stress such as starvation and anthropogenic stress, e.g. cadmium exposure, than are red colour forms of shore crabs. Effects of Cadmium on Nutrient Uptake and Translocation by Indian Mustard   总被引:1,自引:0,他引:1
Plants that hyperaccumulate metals are ideal subjects for studying the mechanisms of metal and mineral nutrient uptake in the plant kingdom. Indian Mustard (Brassica juncea) has been shown to accumulate moderate levels of Cd, Pb, Cr, Ni, Zn, and Cu. In this experiment, 10 levels of Cd concentration treatments were imposed by adding 10–190 mg Cd kg–1 to the soils as cadmium nitrate [Cd(NO3)2]. The effect of Cd on phosphorus (P), potassium (K), calcium (Ca), magnesium (Mg), and the micronutrients iron (Fe), manganese (Mn), copper (Cu), and zinc (Zn) in B. juncea was studied. Plant growth was affected negatively by Cd, root biomass decreased significantly at 170 mg Cd kg–1 dry weight soils treatment. Cadmium accumulation both in shoots and roots increased with increasing soil Cd treatments. The highest concentration of Cd was up to 300 mg kg–1 d.w. in the roots and 160 mg kg–1 d.w. in the shoots. The nutrients mainly affected by Cd were P, K, Ca, Fe, and Zn in the roots, and P, K, Ca, and Cu in the shoots. K and P concentrations in roots increased significantly when Cd was added at 170 mg kg–1, and this was almost the same level at which root growth was inhibited. Zn concentrations in roots decreased significantly when added Cd concentration was increased from 50 to 110 mg kg–1, then remained constant with Cd treatments from 110 to 190 mg kg–1. However, Zn concentrations in the shoots seemed less affected by Cd. It is possible that Zn uptake was affected by the Cd but not the translocation of Zn within the plant. Ca and Mg accumulation in roots and shoots showed similar trends. This result indicates that Ca and Mg uptake is a non-specific process.  相似文献

The marine amphipodAllorchestes compressa Dana, fed on the seagrassHeterozostera tasmanica, was exposed to sublethal concentrations of Cd, Cr, Cu and Zn for 4 wk in flowing sea water, and the concentrations producing the minimum detectable decreases (the minimum effect concentrations, MECs) in average weight, survival and biomass (average weight × survival proportion) were estimated by interpolation from regression models. Survival and biomass were more sensitive than average weight as indicators of sublethal effects. The lowest values of MEC for Cd, Cr, Cu and Zn were 11, >250, 3.7 and 99µg 1–1, respectively. For Cu, this value fell below the minimum risk concentration (MRC) calculated from acute toxicity tests (LC50) and application factors (AF); for Cd, the MEC was similar to the MRC; for Cr and Zn, the MECs were well above the MRCs. The metal concentrations in the amphipods at the MECs were 46, >46, 364 and 139µg g–1 dry wt for Cd, Cr, Cu and Zn, respectively. Accumulation of the nutrient metals (Cr, Cu and Zn) showed some evidence of metabolic regulation, but the non-nutrient Cd was accumulated without regulation until the amphipods died. In general, those metals that were more highly accumulated by the amphipods were the more toxic.  相似文献

Heavy metal concentrations (Cd, Co, Cr, Cu, Fe, Mn, Ni, Pb and Zn) were determined in various tissues (hepatopancreas, branchial hearts, salivary gland, gills, genital tract, mantle, arms and skin) of Octopus vulgaris collected from three different contaminated sites in front of Alexandria (Egypt) during 2000. All collected tissues displayed high enrichment factors when compared to ambient levels. Heavy metal concentrations in most tissues displayed significant differences among sites, sizes and sex. This study suggests that hepatopancreas, and to a lesser extent branchial hearts, are better indicators of chronic Cu, Fe, Zn and Cd contamination than edible tissues. The enrichment factor (EF) for heavy metals in the hepatopancreas and in edible tissues allowed discriminating our samples into three main groups; (1) EF?>?55 (Cu), (2) EF ranging from 15 to 7.5 (Fe, Cd and Zn) and (3) EF?相似文献

The effect of cadmium introduced as inorganic salts (chloride, nitrate, sulfate, carbonate, acetate and iodide) or organic chelates (with EDTA, NTA and humic acid) on the physiological behaviour of six species of tropical lamellibranchs — arcid blood clamsAnadara granosa andA. rhombea, venerid clamsMeretrix casta andKatelysia opima, and mytilid musselsPerna viridis andP. indica, collected from various intertidal areas in southern India from 1986 to 1989 — was evaluated. Inorganic salts of Cd are reported to completely ionize upon introduction to full-strength sea water and form predominantly (97%) chloride complexes. Some inorganic Cd salts, therefore, would hardly affect physiological parameters. However, bioaccumulation of Cd was greatest upon exposure to Cd introduced as sulfate, followed by iodide, acetate, chloride, nitrate and carbonate; 96-h LC50's also varied, but in a different order, being highest for chloride (3.5µg ml–1), followed by nitrate, acetate, iodide and sulfate (1.8µg ml–1). Furthermore, Cd levels increased linearly with time of exposure and levels in medium, and were species-specific, being highest in the arcid clams, followed by the venerid clams and mytilid mussels. The presence of other metals such as Zn reduced bioaccumulation of Cd significantly (P < 0.001), but the reverse was not true. Upon exposure to Cd in the presence of Cu, on the other hand, accumulation of both metals was reduced by 15 to 20%. In the presence of organic chelators — EDTA, NTA and reduced glutathione (GSH) — Cd accumulation was significantly inhibited, by 1/2 to 1/3. Humic acid, on the other hand, had no effect. Cd levels in soft tissues were inversely related to ambient salinity in the range of 12 to 32. However, both Zn and Cu significantly inhibited Cd accumulation irrespective of salinity. Cd increased the rate of filtration inAnadara granosa over the first 24 h by 28%, but on prolonged exposure (96 h) that rate decreased significantly, by 50%. Zn alone and in the presence of Cd, on the contrary, initially reduced filtration activity, which subsequently increased to control levels upon continued exposure. Cu alone and in combination with Cd and Zn also significantly (P < 0.001) inhibited filtration relative to controls. This combination also progressively reduced respiratory activity with time, the maximum decrease being 80% after 96 h of exposure. Furthermore, impact of all three metals on both filtration rate and oxygen consumption was initially synergistic, but changed to antagonistic upon prolonged exposure up to 96 h.Deceased.
