Diel photosynthetic production of water-extractable and residual carbohydrates in natural phytoplankton populations was investigated by analyzing monosaccharide components with combined gas chromatography/mass spectrometry (GC/MS) and13C tracer methods. The applicability of this technique for measuring the photosynthetic production of natural phytoplankton populations was examined. Phytoplankton was collected in August 1984 from Kinu-ura Bay, Japan, and incubated under natural light and dark conditions for 24 h. Cell density and concentration of chlorophylla increased about four- and three-fold, respectively, during the experimental period. Production of glucose, galactose, mannose, rhamnose, ribose, fucose, xylose and arabinose was measured in the water-extractable and residual carbohydrate fractions of the phytoplankton. Most of the monosaccharides in both fractions were produced the monosaccharides in both fractions were produced mainly during the daytime. Glucose was a major component in the water-extractable carbohydrate fraction produced by the phytoplankton, and decreased markedly at night. Thus, most of the glucose in the water-extractable carbohydrates seemed to be a constituent of the storage glucan of algal cells. Most monosaccharide components of the water-extractable carbohydrate fraction except glucose did not decrease significantly at night. Ribose of the water-extractable carbohydrate fraction was synthesized at a maximum rate in the morning, prior to the other monosaccharide constituents of this fraction. The major monosaccharide constituent in the residual carbohydrate fraction produced by the phytoplankton was glucose, which decreased significantly at night. Diel change in the production of monosaccharide constituents in residual carbohydrates indicated wide variability in the monosaccharide composition of cell-wall material in algae. The ratios of production of water-extractable and residual carbohydrates to POC production ranged from 8.8 to 28% and from 3.7 to 5.9%, respectively, throughout the daytime and nighttime.  相似文献

The chemical composition of the mucus from three hard corals (Acropora formosa, Pachyseris speciosa and Fungia fungites) and three soft corals (Sarcophyton sp., Lemnalia sp., and Cespitularia sp.) collected on the Great Barrier Reef (1982–1985) was determined. Significant variation exists in the composition and structure of the six mucus samples, indicating the absence of a common structure for coral mucus. In all cases protein and/or carbohydrate polymers are the major components of the mucus, and lipids are present only in small amounts. The glycose composition varied between species, with fucose (F. fungites and Lemnalia sp.), arabinose (A. formosa), galactose (P. speciosa) and N-acetyl glucosamine (Sarcophyton sp.) being present in high concentrations. With the exception of Sarcophyton sp. and Lemnalia sp., all mucus samples were acidic and contained significant sulphate but no uronic or sialic acids. The amino acid composition of the mucus samples was not unusual, apart from A. formosa, which contained a high percentage of serine and threonine, and F. fungites, which had high levels of glutamic acid present.  相似文献

We have studied carbohydrate metabolism in the marine environment by means of the isolation and characterization of carbohydrates of particulate matter from various depths. The present work is especially concerned with water-extractable carbohydrates of the particulate samples from 11 stations of the northwest Pacific Ocean. Water-extractable carbohydrate content in the particulate matter of the surface waters was measured; it ranged from 19.2 to 36.0% of the total particulate carbohydrate. These values decreased, ranging from 6.5 to 15.5% at 50 m depth. Such rapid vertical change of the concentration of the carbohydrate indicated that it was lost from the particulate matter much faster than the other particulate organic constituents such as protein and water-insoluble carbohydrate. This process left water-insoluble carbohydrate in the deep-water particulate matter as previously observed in the Sagami Nada, off eastern Honshu, Japan. Upon acid hydrolysis, water-extractable carbohydrate revealed glucose ranging from 67.0 to 84.2%. Rhamnose, fucose, ribose, arabinose, xylose, mannose and galactose were also identified as components of the carbohydrate fraction. Carbohydrate studies on the laboratory cultured diatoms clearly indicated that the water-extractable carbohydrates were food reserve materials of the diatoms. In view of this fact, the metabolism of the water-extractable carbohydrate in the euphotic zone and its underlying layer is discussed.  相似文献

We investigated the effect of different densities of the burrowing deposit-feeding amphipod Monoporeia affinis on the potential for recruitment of zooplankton from benthic resting eggs. Hatching of resting eggs was induced in the laboratory on sliced and resuspended 1-cm depth-sections of sediment cores, collected at six stations in an archipelago area of the Gulf of Bothnia, Baltic Sea. The uppermost 5 cm of the sediment was studied. The most common species that hatched was Eurytemora affinis (Copepoda). Individuals from another copepod genus, Acartia, hatched in significant numbers only in the cores from two stations with low amphipod abundance. Cores from stations with high amphipod densities showed a deeper distribution of emerging E. affinis nauplii compared with stations with few amphipods; the oxidised sediment layer was also deeper at high M. affinis densities than at low. Total (0 to 5 cm strata pooled) number of hatched E. affinis nauplii was independent of amphipod density. This indicates that the effect of M. affinis on E. affinis eggs involves deeper burial due to bioturbation, rather than predation. Decreased benthic recruitment of zooplankton at localities with high M. affinis density is suggested, since more deeply positioned eggs are less likely to hatch. When hatching was induced in intact, non-sliced cores from one station, the number of E. affinis nauplii that hatched was on average 43% of the number that hatched in the upper centimetre of the sliced cores from the same station. This fraction (43%), if applied to the other stations, implied a potential for benthic recruitment of up to 80 000 ind m−2 for E. affinis. We experimentally investigated the effect of different densities of the burrowing, deposit-feeding amphipod Monoporeia affinis on the recruitment of zooplankton from benthic resting eggs. Intact sediment cores with in situ density and species composition of zooplankton resting eggs and benthic fauna were collected in the northern Bothnian Sea, part of the Baltic Sea. We removed as many M. affinis as possible from the cores and then added different numbers of M. affinis to the cores to generate a range of densities. The cores were exposed to different densities of M. affinis for either 3 or 40 days, after which the hatched zooplankton was registered. One subset of the cores were initially incubated under low temperature (2–3 °C, to prevent hatching) for 37 days (the resting phase), to allow for effects of M. affinis on unhatched resting eggs. These cores were then incubated under higher temperature (13 °C) for 3 days (the hatching phase), to induce hatching and allow for effects on hatching or hatched specimens. In a second subset of cores with the same time and temperature schedule, the M. affinis density was experimentally reduced at the start of the hatching phase, to evaluate the effect of M. affinis during the hatching phase. To a third subset of cores, we immediately initiated the hatching phase, without an experimental resting phase, to evaluate the effects induced during the resting phase. The most common zooplankton species that hatched was Eurytemora affinis (Copepoda), followed by Bosmina longispina maritima (Cladocera). In all cores that were subjected to a resting phase, the numbers of hatched E. affinis were log-linearly negatively related to density of M. affinis. An increase of M. affinis density from 1,000 to 5,000 individuals m−2, normal field densities, reduced the hatching by 60–70%. The negative impact was mainly exerted during the hatching phase, suggesting predation on, burial of or physical injury of hatching nauplii or eggs in a late development stage as likely mechanisms. Also, the number of B. longispina maritima that hatched was reduced by M. affinis during the hatching phase, but no clear relation to density of M. affinis could be identified. The results show that M. affinis can reduce recruitment to zooplankton from benthic resting eggs. Such impact by the benthos on resting stages of zooplankton is therefore a potentially significant link between the benthic and pelagic systems. The cell walls of diatoms consist of a silica frustule encased in an organic coating. Biochemical characterization of this coating should allow insight into: (1) the mechanism of silicification; (2) taxonomy and evolution of diatoms; (3) preservation of fossil frustules. The amino acid and sugar composition of cell walls from 6 diatom species have been elucidated. When compared to cellular protein, cell-wall protein is enriched in serine plus threonine and glycine, and depleted in acidic, sulfur-containing and aromatic amino acids. The sugars of the cell-wall carbohydrates are quite variable and fucose tends to replace glucose in estuarine species. Condensation of silicic acid, in epitaxial order, on a protein template enriched in serine and threonine, is suggested as the Si-depositing mechanism in diatoms. The nature of this template and the polysaccharides in the cell wall may determine the solubility of diatom frustules in various environments. There is sufficient variability in cell-wall amino acids to warrant further investigation of their taxonomic utility. Aplanochytrium sp. 2400/57 was isolated from samples of sargasso weed collected in the Sargasso Sea on April 4, 1979. The monocentric eucarpic thallus of this organism produced aplanospores by successive bipartitioning. The cell wall contained only 20% polysaccharide, composed of fucose, galactose, xylose, glucose, and galacturonic acid. The guanine+cytosine content of the DNA (mol% G+C) was 37.6%. L-isoleucine, L-leucine, and L-proline were utilized as nitrogen sources in synthetic media of defined composition.  相似文献

Blooms of the marine prymnesiophyte genus Phaeocystis link the oceanic and atmospheric compartments of the carbon and sulfur cycles. Modeling the fluxes of dimethylsulfide from the ocean to the atmosphere has been limited due to a lack of information on functional responses to environmental variables. In this study, the light-dependence of extracellular carbon production and dimethyl sulfide (DMS) production by non-axenic polar clones of Phaeocystis spp. was examined at different growth stages. Comparative experiments were run with non-axenic arctic clones of the diatoms Thalassiossira nordenskioeldii and Skeletonema costatum. A large portion of carbon incorporated by the colonial stage of Phaeocystis spp. is released extracellularly, in particular in stationary colonies. This extracellular production can be modeled as a function of irradiance, as for carbon incorporation. In Phaeocystis spp., cellular and extracellular carbon incorporation represent different uptake rates, indicating the formation of two distinct carbon pools. The release of extracellular carbon by polar Phaeocystis spp. was not a constant fraction of total production over the irradiance range used. We observed little extracellular carbon production by cells at high irradiance, and maximal rates were observed at intermediate irradiance. Newly incorporated carbon that accumulates in the mucilage of the colonial stage of antarctic Phaeocystis sp. during photosynthesis was not reutilized for cellular growth during the dark period, as observed for temperate clones. In contrast, only a minor fraction of the radiocarbon incorporated by the diatoms was released extracellularly for all growth stages. The production of DMS was an order of magnitude higher for Phaeocystis spp. than for diatoms. The chlorophyll-specific production of DMS and DMSP (dimethylsulphoniopropionate, the precursor to DMS) by Phaeocystis spp. showed a hyperbolic response to irradiance, while arctic diatoms (weak or non-producers of DMS), on the other hand, did not show any light-dependency of DMS production. An inverse relationship between DMS and DMSP production in stationary clones of arctic P. pouchetii was observed, but not for the exponentially growing antarctic clone. Stationary colonies also had higher DMS and dissolved DMSP production rates than exponentially growing ones. These relationships can be extrapolated to the field in areas where Phaeocystis spp. dominates.  相似文献

We measured development, growth and juvenile mortality of the common copepod Temora longicornis on 11 different monospecific diatom diets in order to estimate (1) how common the negative effects of diatoms are on the development of this copepod and (2) whether the arrested development is connected to deleterious polyunsaturated aldehydes (PUA) or food nutritional quality. Four diatom species (Thalassiosira weissflogii, Thalassiosira rotula CCMP1647, Leptocylindricus danicus CCPM469 and Skeletonema costatum CCMP1281) supported complete development, whereas development failed in or before metamorphosis on seven diatom species/strains (Chaetoceros affinis CCMP158, C. decipiens CCMP173, C. socialis, T. rotula CCMP1018, Thalassiosira pseudonana CCMP1010 and CCMP1335). However, four out of these seven species were not ingested by nauplii, either due to morphology (Chaetoceros spp.) or large size (T. pseudonana CCMP1010). The growth rate did not correlate with the ingestion rate of PUA, neither with ingestion of food mineral (nitrogen) nor with biochemical (polyunsaturated fatty acids, sterols) components. We show that, although some diatoms are of inferior food quality, this is unlikely to be connected to toxicity or due to a direct limitation by a single food nutritional compound.  相似文献

The pathways of cadmium (Cd) uptake and transfer within an estuarine planktonic community from the Patuxent River, Maryland, USA, were investigated using an assemblage of natural phytoplankton and the copepod Eurytemora affinis Poppe. The experiment was carried out in October 1992 in replicated 500-liter, flow-through, fiberglass tanks. Growth rate, species composition, and Cd loading affected the accumulation of Cd by the phytoplankton. Uptake of Cd by phytoplankton was proportional to the amount of Cd available in the water column. Partition coefficients (K d) for phytoplankton uptake averaged 4.4 × 104. As metal loading rates and phytoplankton species composition changed during the 12-d experiment, Cd partitioning declined. Transfer of Cd to E. affinis occurred from Cd-laden phytoplankton, with levels in the copepods being approximately the same as, or somewhat less, than in the phytoplankton. Some Cd uptake occurred in copepods exposed to dissolved Cd only; however, the uptake was considerably less than that seen from food. Thus, Cd content of higher trophic levels, such as copepods, can be affected by the degree of Cd incorporation in their food source, and by ecological factors regulating phytoplankton ingestion. Grazing impact of microzooplankton on phytoplankton was investigated on the Grand Bank, Newfoundland, Canada, in April, July and October 1984, using a seawater dilution method. In April a large proportion of chlorophylla was in the microplankton size fraction (> 20µm) while in mid-summer and fall most was in the nanoplankton size fraction (< 20µm). Diatoms were the dominant phytoplankters in April, while undetermined flagellates and coccolithophores were abundant in other seasons. Major grazers were oligotrichous ciliates in all seasons. Instantaneous grazing rates on nanophytoplankton, as measured by changes in chlorophylla, varied from 0.12 to 0.43 d–1 and those on microphytoplankton from 0.19 to 0.68 d–1. Grazing rates did not change over 24 and 48 h intervals. This level of grazing corresponded to a daily loss of about 20 and 30% of standing stock of chlorophylla and about 50 and 70% loss of potential production in the two size fractions respectively. Taxon-specific grazing rates, calculated from microscopic enumeration, showed that small diatoms were grazed heavily, and their growth was controlled by grazing in late spring. In late summer and fall, undetermined flagellates and coccolithophores were also grazed at high rates but their growth rates were higher than the grazing rates, and therefore, were not controlled by microzooplankton. In general, microzooplankton grazed on whatever appropriate sized food was dominant in the experimental water. Their potential ability to control the growth of certain food species may be one of the causes determining the species composition of phytoplankton communities.  相似文献

An Antarctic psychrophilic marine Vibrio species was isolated having the inducible ability to accumulate nonmetabolizable thiomethyl-14C--galactopyranoside (14C-TMG) through a galactose permease system. Induction of 14C-TMG uptake was found to have a salinity requirement which was higher than that required for uptake. At the optimum salinity, galactose and fucose were the primary inducers. Lactose produced a comparable induction but only at higher concentrations, whereas glucose did not cause induction. The initial rate of 14C-TMG uptake exhibited saturation kinetics with an apparent Km value of 4.8 x 10–6M. An amino acid, in addition to the inducer, was required for induction which could not be replaced by glycerol or galactose. Evidence is presented which indicates that the uptake of 14C-TMG is energy-dependent and that nutrient availability is more important than salinity for induction and uptake under conditions which would normally be found in the oceanic environment.Technical Paper No. 4911, Oregon Agricultural Experiment Station.  相似文献

Four species of estuarine benthic diatoms: Amphiprora c. f. paludosa W. Smith, Nitzschia c. f. dissipata (Kützing) Grunow, Navicula arenaria Donkin, and Nitzschia sigma (Kützing) W. Smith were grown in unialgal cultures. The growth rates of the diatoms were determined as the rate of increase of the chlorophyll a content of the cultures. The diatoms were cultured at different combinations of temperture, daylength, and quantum irradiance. The highest growth rates of Navicula arenaria occurred at 16° to 20°C; the other 3 species had their optimum at 25°C or higher. The small-celled species had higher growth rates at their optimum temperature, but at lower temperatures the growth rates of all 4 species became very similar. The minimum daily quantum irradiance that could effect light-saturated growth at 12° and 20°C ranged from 2.5 to 5.0 E.m-2.day-1. At 12°C, two species had their highest growth rates under an 8 h daily photoperiod. At 20°C, the three species tested all had highest growth rates under 16 h daily photoperiod. The growth response of the benthic diatoms is comparable to that of several cultures of planktonic diatoms, as described in the literature. The influence of temperature and quantum irradiance on the diatoms in the present investigation was comparable to the influence of temperature and light intensity on the 14C-fixation of marine benthic diatoms (Colijn and van Buurt, 1975).  相似文献

The response of phytoplankton to the presence of two mesozooplanktonic grazers was studied in a 44-h laboratory experiment from the phytoplankton perspective. The <45-μm-filtered plankton community, as well as females of the copepod Eurytemora affinis and nauplii of the barnacle Balanus improvisus originated from the northern Baltic Sea. The phytoplankton community was dominated by the dinoflagellate Heterocapsa triquetra, which was preferred as food by the mesozooplanktonic grazers, especially E. affinis. Cryptophytes were eaten by the B. improvisus nauplii, while heterotrophic nanoflagellates increased in the presence of B. improvisus nauplii, and colonial cyanobacteria increased in the presence of E. affinis. Because of the strong selective feeding on the dominant, large-sized phytoplankton species, the negative effect of the mesozooplanktonic grazers on total chlorophyll a was stronger than any cascade effect releasing phytoplankton from protozoan grazing.  相似文献

Chemical interactions among marine microalgae were studied in cultures of pennate diatoms on agar plates. Nine marine and freshwater pennate diatoms were surveyed as potential bioassay organisms; Cylindrotheca fusiformis was most favourable for assays because of its rapid and even growth on agar. Diatom and bacterial bioassays were used to screen cell and filtrate extracts of 14 microalgal cultures. A number of these algal species, which were grown axenically, produced extractable, intracellular and/or extracellular substances that inhibited the growth of C. fusiformis. Our results suggest that the culturing of pennate diatoms on a solid medium can provide a simple bioassay for screening algal extracts which potentially contain growth inhibitors involved in microalgal allelopathy.  相似文献

The relationship between the distribution of benthic diatoms and sediment characteristics of the Mangyung-Dongjin tidal flat on the west coast of Korea was investigated during June and July 1988. Diatoms were collected from the upper 5 mm of sediments at 60 sites along eight transect lines running perpendicular to the shore line. Of the 371 taxa encountered in the study area, 88% were pennate diatoms. Genera represented by the greatest number of species were Navicula, Nitzschia, Amphora, Cocconeis, Fragilaria and Achnanthes. The most abundant species were Paralia sulcata, Navicula sp. #1, N. arenaria and Cymatosira belgica; all were broadly distributed across the tidal flats. The 60 sites could be assigned to eight clusters with respect to similarity in species composition. Discriminant analysis showed that separation cluster was primarily related to the mean grain size of the sediment. The species could not be separated into groups based on similarities in occurrence; a high degree of spatial overlap was observed. The preferences of the more abundant species for grain size were, therefore, analysed by plotting numerical abundance against mean grain size. There were at least four patterns: species groups could be associated with finegrained sediments, those of intermediate size and coarser sediments and the last group showed no discernible pattern.  相似文献

Evolutionary diversification of the broadly distributed copepod sibling species complex Eurytemora affinis has been documented in the northern hemisphere. However, the fine scale geographic distribution, levels of genetic subdivision, evolutionary, and demographic histories of European populations have been less explored. To gain information on genetic subdivision and to evaluate heterogeneity among European populations, we analyzed samples from 8 locations from 58° to 45°N and 0° to 23°E, using 549 base pairs of the mitochondrial cytochrome oxidase subunit I (COI) gene. We discovered three distinct lineages of E. affinis in Western Europe, namely the East Atlantic lineage, the North Sea/English Channel (NSEC) lineage, and the Baltic lineage. These geographically separated lineages showed sequences divergence of 1.7–2.1%, dating back 1.9 million years (CI: 0.9–3.0 My) with no indication of isolation by distance. Genetic divergence in Europe was much lower than among North American lineages. Interestingly, genetic structure varied distinctively among the three lineages: the East Atlantic lineage was divided between the Gironde and the Loire populations, the NSEC lineage comprised one single population unit spanning the Seine, Scheldt and Elbe rivers and the third lineage was restricted to the Baltic Proper (Sweden). We revealed high haplotype diversity in the East Atlantic and the Baltic lineages, whereas in the NSEC lineage haplotype diversity was comparatively low. All three lineages showed signs of at least one demographic expansion event during Pleistocene glaciations that marked their genetic structure. These results provide a preliminary overview of the genetic structure of E. affinis in Europe.  相似文献

Cyanobacterial blooms are a common phenomenon in the Baltic Sea, and the hepatotoxin nodularin has been frequently detected in certain Baltic Sea fishes and mussels. However, there is no knowledge about the naturally occurring concentrations of nodularin in Baltic Sea zooplankton. The aim of this study was to survey the concentrations of nodularin in natural zooplankton assemblages, and to study the depuration of nodularin in one common copepod species, Eurytemora affinis, experimentally. The nodularin concentrations in common zooplankton species were determined from field-collected samples from the northern Baltic Proper in 2001 and 2002, during cyanobacterial blooms, and the samples were analysed by ELISA immunoassay. Nodularin could be detected from the field-collected zooplankton, suggesting that during a natural bloom event toxins accumulate in their tissues. The concentrations were relatively low (0.07±0.01 μg g−1 ww), ranging from below detection limit to 0.62 μg g−1 ww. Some variation occurred in the concentrations between species and years; generally concentrations were higher in 2001 than in 2002. In the depuration experiment E. affinis copepods were fed with toxic Nodularia spumigena for 24 h, and their toxin contents were monitored for 24 h after transferring them to filtered seawater. A rapid decrease in nodularin concentrations occurred during the first 0.5–3 h after the exposure. However, after a 24-h depuration period in filtered seawater, nodularin could be still detected in E. affinis tissues, indicating that part of the accumulated nodularin, or its derivatives, could be transferred to planktivores.  相似文献

The effects of the water-soluble fraction of dead leaves of the eelgrass Zostera marina L. on the growth of 8 species of micro-algae (pennate and centric diatoms, dinoflagellates, and a green flagellate) and a bacterium were studied on agar plates and in liquid culture. The extracts of leaves which had been dead from a few days to 2 wk inhibited growth and often killed cells in all test organisms. Extracts were lethal even at concentrations equivalent to as little as 0.25 mg dry leaf ml-1, but inhibition decreased when extracts were prepared from leaves aged in the laboratory for 35 d (loss of anti-bacterial activity) or 90 d (loss of anti-algal activity). Extracts of leaves which had aged and dried several months in the field had no effect, except at very high concentrations (13 mg dry leaf ml-1) when the lag phase of growth was prolonged several days in a culture of the chlorophyte Platymonas sp. The active fraction in eelgrass leaves may be important in controlling initial growth of micro-organisms on eelgrass detritus, and it could determine the composition and activity of the epiphytic community on living leaves.  相似文献

In June and July 1984 phytoplankton distribution was investigated in the Fram Strait between Greenland and Svalbard. Chlorophylla, particulate organic carbon, nitrate and phytoplankton species composition were determined from six different depths in the upper 200 m of the water column. Multivariate analysis methods were applied to identify phytoplankton communities in relation to different hydrographic regimes. Three main domains could be distinguished in terms of both hydrography and biology: (1) the East Greenland shelf polynya with a high biomass mainly produced by chain-forming diatoms, (2) the ice-covered East Greenland Current with an extremely low standing stock dominated by flagellates and (3) the marginal ice zone with a biomass maximum in 20 to 40 m depth formed by diatoms, dinoflagellates andPhaeocystis pouchetii.  相似文献
