Azo dyes are recalcitrant and refractory pollutants that constitute a significant menace to the environment. The present study is focused on exploring the capability of Bacillus sp. strain UN2 for application in methyl red (MR) degradation. Effects of physicochemical parameters (pH of medium, temperature, initial concentration of dye, and composition of the medium) were studied in detail. The suitable pH and temperature range for MR degradation by strain UN2 were respectively 7.0–9.0 and 30–40 °C, and the optimal pH value and temperature were respectively 8.0 and 35 °C. Mg2+ and Mn2+ (1 mM) were found to significantly accelerate the MR removal rate, while the enhancement by either Fe3+ or Fe2+ was slight. Under the optimal degradation conditions, strain UN2 exhibited greater than 98 % degradation of the toxic azo dye MR (100 ppm) within 30 min. Analysis of samples from decolorized culture flasks confirmed biodegradation of MR into two prime metabolites: N,N′dimethyl-p-phenyle-nediamine and 2-aminobenzoic acid. A study of the enzymes responsible for the biodegradation of MR, in the control and cells obtained during (10 min) and after (30 min) degradation, showed a significant increase in the activities of azoreductase, laccase, and NADH-DCIP reductase. Furthermore, a phytotoxicity analysis demonstrated that the germination inhibition was almost eliminated for both the plants Triticum aestivum and Sorghum bicolor by MR metabolites at 100 mg/L concentration, yet the germination inhibition of parent dye was significant. Consequently, the high efficiency of MR degradation enables this strain to be a potential candidate for bioremediation of wastewater containing MR.  相似文献

It is the first report in which a novel psychrotrophic Pseudomonas putida SKG-1 strain was evaluated for simultaneous bioremediation of pentachlorophenol and Cr6+ under various cultural and nutritional conditions. Pentachlorophenol (PCP) dechlorination products, bacterial structure, and functional groups were characterized by gas chromatography and mass spectrometry (GC–MS), scanning electron microscope and energy dispersive X-ray spectroscopy (SEM–EDS), and Fourier-transform infrared (FTIR) techniques. The strain was extremely tolerant to excessively higher individual concentration of PCP (1,400 mg l?1) and Cr6+ (4,300 mg l?1). Increasing concentration of PCP and Cr6+ exerted inhibitory effect on bacterial growth and toxicants’ removal. The strain exhibited growth, and concomitantly remediated both the pollutants simultaneously over a broad pH (7.0–9.0) and temperature (28–32 °C) range; maximum growth, PCP dechlorination (87.5 %), and Cr6+ removal (80.0 %) occurred at optimum pH 8.0 and 30 °C (from initial PCP 100 mg l?1 and Cr6+ 500 mg l?1) under shaking (150 rpm) within 72 h incubation. Optimization of agitation (125 rpm) and aeration (0.4 vvm) in bioreactor further enhanced PCP dechlorination by ~10 % and Cr6+ removal by 2 %. A direct correlation existed between growth and bioremediation of both the toxicants. Among other heavy metals, mercury exerted maximum and cobalt minimum inhibitory effect on PCP dechlorination and Cr6+ removal. Chromate reductase activity was mainly associated with the supernatant and cytosolic fraction of bacterial cells. GC–MS analysis revealed the formation of tetrachloro-p-hydroquinone, 2,4,6-trichlorophenol, and 2,6-dichlorophenol as PCP dechlorination products. FTIR spectrometry indicated likely involvement of carbonyl and amide groups in Cr3+ adsorption, and SEM–EDS showed the presence of chromium on P. putida surface. Thus, our promising isolate can be ecofriendly employed for biotreatment of various industrial wastes contaminated with high PCP and Cr6+ concentrations.  相似文献

A native Bacillus cereus isolate has been employed, for the first time, for simultaneous decolorization, dechlorination of chloroorganics, and Cr6+ remediation from the real tannery effluent. Most of the physicochemical variables in 3:1 diluted effluent were well above the standard prescribed limits, which decreased substantially upon microbial treatment. The extent of bioremediation was better in diluted (3:1) as compared to undiluted effluent supplemented with nutrients and augmented with B. cereus isolate. Maximum growth, effluent decolorization (42.5 %), dechlorination (74.1 %), and Cr6+ remediation (34.2 %) were attained with 4.0 % (v/v) inoculum, 0.8 % glucose, and 0.2 % ammonium chloride in 3:1 diluted effluent at natural pH (8.1) within 72 h of incubation. The efficiency of bioremediation in a bioreactor was higher as compared to a flask trial during 72 h of incubation: decolorization (47.9 %) was enhanced by 5.4 %, dechlorination (77.4 %) by 3.3 %, and Cr6+ removal (41.7 %) by 7.5 % at an initial color of 286 Pt-Co units and initial concentration of 62 mg chloride ions and 108 mg l?1 Cr6+. Immobilized biomass of Pseudomonas putida and B. cereus coculture enhanced the extent of Cr6+ remediation (51.9 %) by 10.2 % compared to the bioreactor trial. Chromate reductase activity and reduced Cr directly correlated and were mainly associated with soluble fraction of B. cereus plus effluent natural microflora. The GC-MS analyses revealed the formation of metabolites such as acetic acid and 2-butenoic acid in bacterially treated effluent. The supplementation of nutrients along with B. cereus augmentation is required for efficient effluent bioremediation.  相似文献

Four efficient Cr(VI)-reducing bacterial strains were isolated from rhizospheric soil of plants irrigated with tannery effluent and investigated for in vitro Cr(VI) reduction. Based on 16S rRNA gene sequencing, the isolated strains SUCR44, SUCR140, SUCR186, and SUCR188 were identified as Bacillus sp. (JN674188), Microbacterium sp. (JN674183), Bacillus thuringiensis (JN674184), and Bacillus subtilis (JN674195), respectively. All four isolates could completely reduce Cr(VI) in culture media at 0.2 mM concentration within a period of 24–120 h; SUCR140 completely reduced Cr(VI) within 24 h. Assay with the permeabilized cells (treated with Triton X-100 and Tween 80) and cell-free assay demonstrated that the Cr(VI) reduction activity was mainly associated with the soluble fraction of cells. Considering the major amount of chromium being reduced within 24–48 h, these fractions could have been released extracellularly also during their growth. At the temperature optima of 28 °C and pH?7.0, the specific activity of Cr(VI) reduction was determined to be 0.32, 0.42, 0.34, and 0.28 μmol Cr(VI)?min?1?mg?1 protein for isolates SUCR44, SUCR140, SUCR186, and SUCR188, respectively. Addition of 0.1 mM NADH enhanced the Cr(VI) reduction in the cell-free extracts of all four strains. The Cr(VI) reduction activity in cell-free extracts of all the isolates was stable in presence of different metal ions tested except Hg2+. Beside this, urea and thiourea also reduced the activity of chromate reduction to significant levels.  相似文献

The equilibrium of the structure and functioning of freshwater ecosystems is dependent of detritivores that link all the other functional groups. The preference for feeding leaves with different diameters (particle size) and leaves with metal contamination (several concentrations of the essential metals copper and zinc) were determined for two detritivores, the decapod Atyaephyra desmarestii and the amphipod Echinogammarus meridionalis. Several no-choice and multi-choice assays were done to determinate which leaf diameter the amphipod and the decapod species would eat when they had or not had alternatives available and include a set of dual-choice assays with contaminated and uncontaminated foods. No significant preference was shown by either species relative to the diameter of leaves, either on no-choice or multi-choice assays. The presence of essential metals on food did not had any influence on the feeding choice of these organisms over the concentration range studied. Both showed no preference on ingesting food spiked with these essential metals, except E. meridionalis which preferred ingesting leaves with 2.19 μg.l?1 of copper instead of uncontaminated leaves. For further works, despite no preference for leaves with a certain diameter, the leaves with 0.70 cm (0.385 cm2of area) and with 0.50 cm (1.767 cm2 of area) should be used for A. desmarestii and E. meridionalis, respectively. Furthermore, to maintain E. meridionalis, the diet should include some percentage of copper in order to accomplish metabolic needs.  相似文献

Carbonic anhydrase II (CA II) can catalyze the reversible hydration reaction of CO2 at a maximum of 1.4?×?106 molecules of CO2 per second. The crude intracellular enzyme extract containing CA II was derived from Chlorella vulgaris. A successful CO2 capture experiment with the presence of calcium had been conducted on the premise that the temperature was conditioned at a scope of 30?C40?°C, that the biocatalyst-nurtured algal growth period lasted 3?days, and that pH ranged from7.5 to 8.5. Ions of K+, Na+, Ca2+, Co2+, Cu2+, Fe3+, Mg2+, Mn2+, and Zn2+ at 0.01, 0.1, and 0.5?M were found to exhibit no more than 30?% inhibition on the residual activity of the biocatalyst. It is reasonable to expect that calcification catalyzed by microalgae presents an alternative to geological carbon capture and sequestration through a chain of fundamental researches carried on under the guidance of sequestration technology.  相似文献

In order to investigate the treatment performance and microorganism mechanism of IVCW for domestic wastewater in central of China, two parallel pilot-scale IVCW systems were built to evaluate purification efficiencies, microbial community structure and enzyme activities. The results showed that mean removal efficiencies were 81.03 % for COD, 51.66 % for total nitrogen (TN), 42.50 % for NH4 +-N, and 68.01 % for TP. Significant positive correlations between nitrate reductase activities and TN and NH4 +-N removal efficiencies, along with a significant correlation between substrate enzyme activity and operation time, were observed. Redundancy analysis demonstrated gram-negative bacteria were mainly responsible for urease and phosphatase activities, and also played a major role in dehydrogenase and nitrate reductase activities. Meanwhile, anaerobic bacteria, gram-negative bacteria, and saturated FA groups, gram-positive bacteria exhibited good correlations with the removal of COD (p?=?0.388), N (p?=?0.236), and TP (p?=?0.074), respectively. The IVCW system can be used to treat domestic wastewater effectively.  相似文献

Intensive remediation of abandoned former organochlorine pesticides (OCPs) manufacturing areas is necessary because the central and surrounding soils contaminated by OCPs are harmful to crop production and food safety. Organochlorine and its residues are persistent in environments and difficult to remove from contaminated soils due to their low solubility and higher sorption to the soils. We performed a comprehensive study on the remediation of OCPs-contaminated soils using thermal desorption technique and solvent washing approaches. The tested soil was thermally treated at 225, 325, 400, and 500 °C for 10, 20, 30, 45, 60, and 90 min, respectively. In addition, we tested soil washing with several organic solvents including n-alcohols and surfactants. The optimal ratio of soil/solvent was tested, and the recycling of used ethanol was investigated. Finally, activities of polyphenol oxidase (PPO), urease (URE), alkaline phosphatase, acid phosphatase (ACP), and invertase (INV) were assayed in the treated soils. The tested soil was thermally treated at 500 °C for 30 min, and the concentration of contaminants in soil was decreased from 3,115.77 to 0.33 mg kg?1. The thermal desorption in soil was governed by the first-order kinetics model. For the chemical washing experiment, ethanol showed a higher efficiency than any other solvent. Using a 1:20 ratio of soil/solvent, the maximum removal of OCPs was achieved within 15 min. Under this condition, approximately 87 % of OCPs was removed from the soils. More than 90 % of ethanol in the spent wash fluid could be recovered. Activities of some enzymes in soils were increased after ethanol treatment. But ALP, ACP, and INV activities were decreased and PPO and URE showed slightly higher activities following remediation by thermal treatment. Both heating temperature and time were the key factors for thermal desorption of OCPs. The n-alcohol solvent showed higher removal of OCPs from soils than surfactants. The highly efficient removal of OCPs from soil was achieved using ethanol. More than 90 % of ethanol could be recovered and be reused following distillation. This study provides a cost-effective and highly efficient way to remediate the OCPs-contaminated soils.  相似文献

This paper investigates the potential of using the silver antibacterial properties combined with the metal ion exchange characteristics of silver-modified clinoptilolite to produce a treatment system capable of removing both contaminants from aqueous streams. The results have shown that silver-modified clinoptilolite is capable of completely eliminating Escherichia coli after 30-min contact time demonstrating its effectiveness as a disinfectant. Systems containing both E. coli and metals exhibited 100 % E. coli reduction after 15-min contact time and maximum metal adsorption removal efficiencies of 97, 98, and 99 % for Pb2+, Cd2+, and Zn2+ respectively after 60 min; 0.182–0.266 mg/g of metal ions were adsorbed by the zeolites in the single- and mixed-metal-containing solutions. Nonmodified clinoptilolite showed no antibacterial properties. This study demonstrated that silver-modified clinoptilolite exhibited high disinfection and heavy metal removal efficiencies and consequently could provide an effective combined treatment system for the removal of E. coli and metals from contaminated water streams.  相似文献

The influence of two neonicotinoids, i.e., imidacloprid (IMI) and acetamiprid (ACE), on soil microbial activities was investigated in a short period of time using a combination of the microcalorimetric approach and enzyme tests. Thermodynamic parameters such as Q T (J g?1 soil), ?H met (kJ mol?1), J Q/S (J g?1 h?1), k (h?1), and soil enzymatic activities, dehydrogenase, phosphomonoesterase, arginine deaminase, and urease, were used to evaluate whole metabolic activity changes and acute toxicity following IMI and ACE treatment. Various profiles of thermogenic curves reflect different soil microbial activities. The microbial growth rate constant k, total heat evolution Q T (expect for IMI), and inhibitory ratio I show linear relationship with the doses of IMI and ACE. Q T for IMI increases at 0.0–20 μg g?1 and then decreases at 20–80 μg g?1, possibly attributing to the presence of tolerant microorganisms. The 50 % inhibitory ratios (IC50) of IMI and ACE are 95.7 and 77.2 μg g?1, respectively. ACE displays slightly higher toxicity than IMI. Plots of k and Q T against microbial biomass-C indicate that the k and Q T are growth yield-dependent. IMI and ACE show 29.6; 40.4 and 23.0; and 23.3, 21.7, and 30.5 % inhibition of dehydrogenase, phosphomonoesterase, and urease activity, respectively. By contrast, the arginine deaminase activity is enhanced by 15.2 and 13.2 % with IMI and ACE, respectively. The parametric indices selected give a quantitative dose-response relationship of both insecticides and indicate that ACE is more toxic than IMI due to their difference in molecular structures.  相似文献

The extracellular polymeric substance (EPS) extracted from waste activated sludge (WAS) after short-time aerobic digestion was investigated to be used as a novel biosorbent for Cu2+ removal from water. The EPS consisted of protein (52.6 %, w/w), polysaccharide (30.7 %, w/w), and nucleic acid (16.7 %, w/w). Short-time aerobic digestion process of WAS for about 4 h promoted the productivity growth of the EPS for about 10 %. With a molecular weight of about 1.9?×?106 Da, the EPS showed a linear structure with long chains, and contained carboxyl, hydroxyl, and amino groups. The sorption kinetics was well fit for the pseudo-second-order model, and the maximum sorption capacity of the EPS (700.3 mg Cu2+/g EPS) was markedly greater than those of the reported biosorbents. Both Langmuir model and Freundlich model commendably described the sorption isotherm. The Gibbs free energy analysis of the adsorption showed that the sorption process was feasible and spontaneous. According to the complex results of multiple analytical techniques, including scanning electron microscopy, Fourier transform infrared spectroscopy, atomic force microscopy, etc., the adsorption process took place via both physical and chemical sorption, but the electrostatic interaction between sorption sites with the functional groups and Cu2+ is the major mechanism.  相似文献

The effects of the organic loading rate (OLR) on the performance and the granular sludge characteristics of an expanded granular sludge bed (EGSB) reactor used for treating real traditional Chinese medicine (TCM) wastewater were investigated. Over 90 % of the COD removal by the EGSB reactor was observed at the OLRs of 4 to 13 kg COD/(m3 day). However, increasing the OLR to 20 kg COD/(m3 day) by reducing the hydraulic retention time (HRT 6 h) reduced the COD removal efficiency to 78 %. The volatile fatty acid (VFA) concentration was 512.22 mg/L, resulting in an accumulation of VFAs, and propionic acid was the main acidification product, accounting for 66.51 % of the total VFAs. When the OLR increased from 10 to 20 kg COD/(m3 day), the average size of the granule sludge decreased from 469 to 258 μm. There was an obvious reduction in the concentration of Ca2+ and Mg2+ in the granular sludge. The visible humic acid-like peak was identified in the three-dimensional excitation-emission matrix (EEM) fluorescence spectra of the soluble microbial products (SMPs). The fatty acid bond, amide II bond, amide III bond, and C–H bond bending were also observed in the Fourier transform infrared (FTIR) spectra of the SMPs. Methanobacterium formicicum, Methanococcus, and Bacteria populations exhibited significant shifts, and these changes were accompanied by an increase in VFA production. The results indicated that a short HRT and high OLR in the EGSB reactor caused the accumulation of polysaccharides, protein, and VFAs, thereby inhibiting the activity of methanogenic bacteria and causing granular sludge corruption.  相似文献

The aim of this work was to determine the optimal conditions for the decolorization and the detoxification of two effluents from a textile industry—effluent A (the reactive dye bath Bezactive) and effluent B (the direct dye bath Tubantin)—using a laccase mediator system. Response surface methodology (RSM) was applied to optimize textile effluents decolorization. A Box–Behnken design using RSM with the four variables pH, effluent concentration, 1-hydroxybenzotriazole (HBT) concentration, and enzyme (laccase) concentration was used to determine correlations between the effects of these variables on the decolorization of the two effluents. The optimum conditions for pH and concentrations of HBT, effluent and laccase were 5, 1 mM, 50 % and 0.6 U/ml, respectively, for maximum decolorization of effluent A (68 %). For effluent B, optima were 4, 1 mM, 75 %, and 0.6 U/ml, respectively, for maximum decolorization of approximately 88 %. Both effluents were treated at 30 °C for 20 h. A quadratic model was obtained for each decolorization through this design. The experimental and predicted values were in good agreement and both models were highly significant. In addition, the toxicity of the two effluents was determined before and after laccase treatment using Saccharomyces cerevisiae, Bacillus cereus, and germination of tomato seeds.  相似文献

This study aims to synthesize 2-hydroxyethyl acrylate (HEA) and 2-acrylamido-2-methylpropane sulfonic (AMPS) acid-based hydrogels by gamma radiation and to investigate their swelling behavior and heavy metal ion adsorption capabilities. The copolymer hydrogels prepared were characterized via scanning electron microscopy, Fourier transformed infrared spectra, thermal gravimetric analysis, and X-ray photoelectron spectroscopy. The research showed that the copolymer hydrogel was beneficial for permeation due to its porous structure. In addition, the experimental group A-2-d [70 % water volume ratio and (n (AMPS)/n (HEA))?=?1:1] was an optimal adsorbent. The optimal pH was 6.0 and the optimal temperature was 15 °C. Pb2+, Cd2+, Cu2+, and Fe3+ achieved adsorption equilibriums within 24 h, whereas Cr3+ reached equilibrium in 5 h. Pb2+, Cd2+, Cr3+, and Fe3+ maximum load capacity was 1,000 mg L?1, whereas the Cu2+ maximum capacity was 500 mg L?1. The priority order in the multicomponent adsorption was Cr3+>Fe3+>Cu2+>Cd2+>Pb2+. The adsorption process of the HEA/AMPS copolymer hydrogel for the heavy metal ions was mainly due to chemisorption, and was only partly due to physisorption, according to the pseudo-second-order equation and Langmuir adsorption isotherm analyses. The HEA/AMPS copolymer hydrogel was confirmed to be an effective adsorbent for heavy metal ion adsorption.  相似文献

A study was conducted to evaluate the effects of elevated concentrations of copper (Cu) and zinc (Zn) in a soil treated with biosolids previously spiked with these metals on Pinus radiata during a 312-day glasshouse pot trial. The total soil metal concentrations in the treatments were 16, 48, 146 and 232 mg Cu/kg or 36, 141, 430 and 668 mg Zn/kg. Increased total soil Cu concentration increased the soil solution Cu concentration (0.03–0.54 mg/L) but had no effect on leaf and root dry matter production. Increased total soil Zn concentration also increased the soil solution Zn concentration (0.9–362 mg/L). Decreased leaf and root dry matter were recorded above the total soil Zn concentration of 141 mg/kg (soil solution Zn concentration, >4.4 mg/L). A lower percentage of Cu in the soil soluble?+?exchangeable fraction (5–12 %) and lower Cu2+ concentration in soil solution (0.001–0.06 μM) relative to Zn (soil soluble?+?exchangeable fraction, 12–66 %; soil solution Zn2+ concentration, 4.5–4,419 μM) indicated lower bioavailability of Cu. Soil dehydrogenase activity decreased with every successive level of Cu and Zn applied, but the reduction was higher for Zn than for Cu addition. Dehydrogenase activity was reduced by 40 % (EC40) at the total solution-phase and solid-phase soluble?+?exchangeable Cu concentrations of 0.5 mg/L and 14.5 mg/kg, respectively. For Zn the corresponding EC50 were 9 mg/L and 55 mg/kg, respectively. Based on our findings, we propose that current New Zealand soil guidelines values for Cu and Zn (100 mg/kg for Cu; 300 mg/kg for Zn) should be revised downwards based on apparent toxicity to soil biological activity (Cu and Zn) and radiata pine (Zn only) at the threshold concentration.  相似文献

The low bioavailability of Pb and low number of Pb-tolerant plant species represent an important limitation for Pb phytoextraction. It was recently suggested that halophyte plant species may be a promising material for this purpose, especially in polluted salt areas while Pb mobility may be improved by synthetic chelating agents. This study aims to evaluate Pb extraction by the halophyte Sesuvium portulacastrum in relation to the impact of EDTA application. Seedling were cultivated during 60 days on Pb artificially contaminated soil (200, 400, and 800 ppm Pb) in the presence or in the absence of EDTA (3 g kg?1 soil). Results showed that upon to 400 ppm, Pb had no impact on plant growth. However, exogenous Pb induce a decrease in shoot K+ while it increased shoot Mg2+ and had no impact on shoot Ca2+ concentrations. Lead concentration in the shoots increased with increasing external Pb doses reaching 1,390 ppm in the presence of 800 ppm lead in soil. EDTA addition had no effect on plant growth but strongly increased Pb accumulation in the shoot which increased from 1,390 ppm in the absence of EDTA to 3,772 ppm in EDTA-amended plants exposed to 800 ppm exogenous Pb. Both Pb absorption and translocation from roots to shoots were significantly enhanced by EDTA application, leading to an increase in the total amounts of extracted Pb per plant. These data suggest that S. portulacastrum is very promising species for decontamination of Pb2+-contaminated soil and that its phytoextraction potential was significantly enhanced by addition of EDTA to the polluted soil.  相似文献

The utilization of sustainable and biodegradable lignocellulosic fiber to detoxify the noxious Cr(VI) from wastewater is considered a versatile approach to clean up a contaminated aquatic environment. The aim of the present research is to assess the proficiency and mechanism of biosorption on Ficus carica bast fiber via isotherm models (Langmuir, Freundlich, Temkin, Harkin’s–Jura, and Dubinin–Radushkevich), kinetic models, and thermodynamic parameters. The biomass extracted from fig plant was characterized by scanning electron microscopy and Fourier-transform infrared spectroscopy. To optimize the maximum removal efficiency, different parameters like effect of initial concentration, effect of temperature, pH, and contact time were studied by batch method. The equilibrium data were best represented by the Langmuir isotherm model, and the maximum adsorption capacity of Cr(VI) onto biosorbent was found to be 19.68 mg/g. The pseudo-second-order kinetic model adequately described the kinetic data. The calculated values of thermodynamic parameters such as enthalpy change (?H 0), entropy change (?S 0), and free energy change (?G 0) were 21.55 kJ/mol, 76.24 J/mol?K, and ?1.55 kJ/mol, respectively, at 30 °C which accounted for spontaneous and endothermic processes. The study of adsorbent capacity for Cr(VI) removal in the presence of Na+, Mg2+, Ca2+, SO 4 2? , HCO 3 ? and Cl? illustrated that the removal of Cr(VI) increased in the presence of HCO3? ions; the presence of Na+, SO 4 2? or Cl? showed no significant influence on Cr(VI) adsorption, while Ca2+ and Mg2+ ions led to an insignificant decrease in Cr(VI) adsorption. Further, the desorption studies illustrated that 31.10 % of metal ions can be removed from an aqueous system, out of which 26.63 % of metal ions can be recovered by desorption in first cycle and the adsorbent can be reused. The results of the scale-up study show that the ecofriendly detoxification of Cr(VI) from aqueous systems was technologically feasible.  相似文献

The full-length cDNA of catalase (EfCAT) from Eisenia fetida was cloned (GenBank accession no. JN617999). Sequence characterization revealed that EfCAT protein sequence contained proximal heme-ligand signature sequence (351RLFSYSDTH359), two glycosylation sites (N145 and N436), the proximal active site signature (61FDRERIPERVVHAKGAGA78), and 12 amino acids (N145, H191, F195, S198, R200, N210, Y212, K234, I299, W300, Q302, and Y355), which were identified as putative residues involved in NADPH binding. These conserved motifs and catalase signature sequences were essential for the structure and function of EfCAT. The present study also investigated the effect of the veterinary food additive zinc oxide on antioxidant processes in E. fetida, at different concentrations and exposure durations. A significant increase (by 106.0 % compared to controls) in CAT activity at 500 mg/kg was registered at day 15. The superoxide dismutase (SOD) activity at 500 mg/kg increased to the maximum value (by 44.0 %) measured at day 15. There was a significant increase in glutathione peroxidase (GPx) activity for all concentrations after 5 days. The results showed that dietary Zn (500 mg/kg) causes oxidative damage to earthworms. At early stages of earthworms exposed to ZnO, GPx is the main enzyme to impair the oxidative status; while at later stages the enzymes CAT and SOD were the main indicators of oxidative stress. The antioxidant enzymatic variations may be an adaptive response of earthworms to survive in contaminated soils.  相似文献

Cyanobacterial biofertilizers are affected by paddy field pesticides as nontarget organism. Carbaryl is a carbamate pesticide and is commonly used against rice thrip pest in paddy fields. In the present work, cellular changes caused by exposure of the cyanobacterial biofertilizer namely Calothrix brevissima to carbaryl were studied with special reference to fatty acids, electrolyte leakage, sulfur metabolism, and osmolytes. To study the toxic effect of carbaryl, the test cyanobacterium was exposed to varying concentrations of pesticide (0, 10, 20, 30, and 40 mg L?1) for biochemical analyses. At 40 mg L?1 carbaryl, polyunsaturated fatty acids were reduced by 32 % and membrane leakage was increased by 27 % suggesting that free radical-mediated lipid peroxidation took place. The sulfur-containing metabolites namely cysteine, cystine, and methionine were increased by 79, 64, and 52 %, respectively. The enzymatic and nonenzymatic antioxidants namely glutathione S-transferase, glutathione reductase, reduced glutathione, and oxidized glutathione were increased to 56, 71, 72, and 60 %, respectively. Osmolytes that serve as stress enzyme protectors as well as nonenzymatic free radical scavenger were also increased, indicating their protective nature in context with carbaryl-induced stress. The respective increase in mannitol, trehalose, and glycogen were 158, 98, and 159 %. In C. brevissima, carbaryl-induced membrane leakage was counteracted by increasing enzymatic and nonenzymatic parameters that helped in scavenging free radicals.  相似文献

This study evaluated the toxicity of herbicide atrazine, along with its bioaccumulation and biodegradation in the green microalga Chlamydomonas mexicana. At low concentration (10 μg L?1), atrazine had no profound effect on the microalga, while higher concentrations (25, 50, and 100 μg L?1) imposed toxicity, leading to inhibition of cell growth and chlorophyll a accumulation by 22 %, 33 %, and 36 %, and 13 %, 24 %, and 27 %, respectively. Atrazine 96-h EC50 for C. mexicana was estimated to be 33 μg L?1. Microalga showed a capability to accumulate atrazine in the cell and to biodegrade the cell-accumulated atrazine resulting in 14–36 % atrazine degradation at 10–100 μg L?1. Increasing atrazine concentration decreased the total fatty acids (from 102 to 75 mg g?1) and increased the unsaturated fatty acid content in the microalga. Carbohydrate content increased gradually with the increase in atrazine concentration up to 15 %. This study shows that C. mexicana has the capability to degrade atrazine and can be employed for the remediation of atrazine-contaminated streams.  相似文献
