The aim of this study was to determine the load of Escherichia coli transferred via drainage waters from drained and undrained pasture following a grazing period. Higher concentrations (ranging between 10(4) and 10(3) colony forming units [CFU] g(-1)) of E. coli persisted in soil for up to 60 d beyond the point where cattle were removed from the plots, but these eventually declined in the early months of spring to concentrations less than 10(2) CFU g(-1). The decline reflects the combined effect of cell depletion from the soil store through both wash-out and die-off of E. coli. No difference (P > 0.05) was observed in E. coli loads exported from drained and undrained plots. Similarly, no difference (P > 0.05) was observed in E. coli concentrations in drainage waters of mole drain flow and overland plus subsurface interflow. Intermittent periods of elevated discharge associated with storm events mobilized E. coli at higher concentrations (e.g., in excess of 400 CFU mL(-1)) than observed during low flow conditions (often <25 CFU mL(-1)). The combination of high discharge and cell concentrations resulted in the export of E. coli loads from drained and undrained plots exceeding 10(6) CFU L(-1) s(-1). The results highlight the potential for drained land to export E. coli loads comparable with those transferred from undrained pasture.  相似文献

Assessment methods for determining the presence and number of fecal bacteria and Escherichia coli (E. coli) in waters, foodstuffs, sewage effluent, and soils have evolved from multiple tube fermentations (MTF's) to membrane filtrations (MF's) to, most recently, defined substrate technologies (DST's). Mounting evidence indicates Colilert DST (IDEXX, Westbrook, ME) to be a versatile assessment technique for detecting and enumerating E. coli over a range of applications. This study compared the performance of Colilert DST with a confirmed standard MF technique using m-FC broth (Millipore, Bedford, MA) in assessing E. coli in ten different environmental water samples obtained monthly over a 3-year period from the upper Appomattox River, VA. For the duration of the study, E. coli counts measured by Colilert DST were positively correlated (Pearson's correlation coefficient=0.956; slope=0.979; p<0.0001) with E. coli counts measured by confirmed MF procedures. The results of a two-factor ANOVA revealed that Colilert DST counts compared equally to confirmed MF counts by year (p=0.974), by stream sampled (p=1.0), and by season (p=0.696). E. coli counts were significantly lower during cold season months (Dec/Jan/Feb) than during warm season months (Jun/Jul/Aug) for each year contributing to marked variation in sample quality. Counts obtained by Colilert DST compared equally to those obtained by MF across all samples and dates for the three years. Colilert DST presents a laboratory protocol that is simpler to manage, quicker to process, and easier to quantify results than MF. These factors, plus the enhanced precision and versatility of Colilert DST over the span of this three-year study attests to its suitability for testing ambient surface waters.  相似文献

Large and repeated manure applications can exceed the P sorption capacity of soil and increase P leaching and losses through subsurface drainage. The objective of this study was to evaluate the fate of P applied with increasing N rates in dairy wastewater or poultry litter on grassland during a 4-yr period. In addition to P recovery in forage, soil-test phosphorus (STP) was monitored at depths to 180 cm in a Darco loamy sand (loamy, siliceous, semiactive, thermic Grossarenic Paleudults) twice annually. A split-plot arrangement of a randomized complete block design comprised four annual N rates (0, 250, 500, and 1000 kg ha(-1)) for each nutrient source on coastal bermudagrass [Cynodon dactylon (L.) Pers.] over-seeded with ryegrass (Lolium multiflorum L. cv. TAM90). Increasing annual rates of N and P in wastewater and poultry litter increased P removal in forage (P = 0.001). At the highest N rate of each nutrient source, less than 13% of applied P was recovered in forage. The highest N rates delivered 8 times more P in wastewater or 15 times more P in poultry litter than was removed in forage harvests during an average year. Compared with controls, annual P rates up to 188 kg ha(-1) in dairy wastewater did not increase STP concentrations at depths below 30 cm. In contrast, the highest annual P rate (590 kg ha(-1)) in poultry litter increased STP above that of controls at depth intervals to 120 cm during the first year of sampling. Increases in STP at depths below 30 cm in the Darco soil were indicative of excessive P rates that could contribute to nonpoint-source pollution in outflows from subsoil through subsurface drainage.  相似文献

Chemical treatment of animal manure with Al, Fe, and Ca salts appears capable of concentrating P in a smaller volume, thereby providing increased manure management options. However, little information is available on the fate of nutrients in soils receiving chemically treated manure. An incubation study (1 d to 2 yr) was conducted with three soils (Soils I, II, and III with 12, 66, and 94 mg kg(-1) Bray-1 P, respectively) and four manure treatments (one untreated and three chemical including Al-, Fe-, and Ca-treated) at two rates (12.5 and 25 mg P kg(-1)), and a control (no manure). Subsamples were analyzed for Bray-1 P and water-extractable phosphorus (WEP) after eight incubation time periods. Phosphorus distribution among different fractions (soluble and loosely bound; Al-, Fe-, and Ca-bound; organic P; and residual) was also determined after 1 d and 1 yr. Water-extractable P increased when soils received untreated or Ca-treated manure in proportion to P application rate. Water-extractable P, however, decreased (compared with control) for Soils II and III or slightly increased for Soil I with addition of Al- or Fe-treated manure. Water-extractable P decreased sharply between 1 d and 1 to 2 wk and then remained relatively constant or increased slightly up to 2 yr depending on treatment and soil type. Bray-1 P increased for all treatment types and soils in the following order: Ca-treated > Al-treated >/= untreated > Fe-treated > control. Within each treatment, Bray-1 P decreased between 1 d and 1 to 2 wk and then gradually increased for up to 3 mo (Soils II and III) or 6 mo (Soil I). Application of Al- or Fe-treated manure decreased P solubility with the effect being more pronounced in soils with high background P. Since the application of Ca-treated manure increased both WEP and Bray-1 P, it should be recommended for soils where the objective is to increase P availability. Several years of P input through fertilizer and manure contributed mainly to aluminum-bound phosphorus (Al-P) and to a lesser degree to other fractions. Only soluble and loosely bound P (all soils) and Al-P (Soil I) exhibited treatment-type effects after receiving chemically treated manure. The study results will help bridge the gap between our knowledge of chemical treatment systems for animal manure and the ultimate fate of P when the treated manure is land-applied.  相似文献

Poultry litter provides a rich source of nutrients for perennial forages, but the usual practice of surface-applying litter to pastures can degrade water quality by allowing nutrients to be transported from fields in surface runoff, while much of the NH4-N volatilizes. Incorporating litter into the soil can minimize such problems in tilled systems, but has not been used for perennial forage systems. In this study, we minimized disturbance of the crop, thatch, and soil structure by using a knifing technique to move litter into the root zone. Our objective was to determine effects of poultry litter incorporation on quantity and quality of runoff water. Field plots were constructed on a silt loam soil with well-established bermudagrass [Cynodon dactylon (L.) Pers.] and mixed grass forage. Each plot had 8 to 10% slopes, borders to isolate runoff, and a downslope trough with sampling pit. Poultry litter was applied (5.6 Mg ha(-1)) by one of three methods: surface-applied, incorporated, or surface-applied on soil-aeration cuts. There were six treatment replications and three controls (no litter). Nutrient concentrations and mass losses in runoff from incorporated litter were significantly lower (generally 80-95% less) than in runoff from surface-applied litter. By the second year of treatment, litter-incorporated soils had greater rain infiltration rates, water-holding capacities, and sediment retention than soils receiving surface-applied litter. Litter incorporation also showed a strong tendency to increase forage yield.  相似文献

The Missouri Department of Natural Resources (MDNR) has closed or posted advisories at public beaches at Lake of the Ozarks State Park in Missouri because of Escherichia coli (E. coli) concentration exceedances in recent years. Spatial and temporal patterns of E. coli concentrations, microbial source tracking, novel sampling techniques, and beach‐use patterns were studied during the 2012 recreational season to identify possible sources, origins, and occurrence of E. coli contamination at Grand Glaize Beach (GGB). Results indicate an important source of E. coli contamination at GGB was E. coli released into the water column by bathers resuspending avian‐contaminated sediments, especially during high‐use days early in the recreational season. Escherichia coli concentrations in water, sediment, and resuspended sediment samples all decreased throughout the recreational season likely because of decreasing lake levels resulting in sampling locations receding away from the initial spring shoreline as well as natural decay and physical transport out of the cove. Weekly MDNR beach monitoring, based solely on E. coli concentrations, at GGB during this study inaccurately predicted E. coli exceedances, especially on weekends and holidays. Interestingly, E. coli of human origin were measured at concentrations indicative of raw sewage in runoff from an excavation of a nearby abandoned septic tank that had not been used for nearly two years.  相似文献

Weathering of bedrock and pedogenic processes can result in elevated heavy metal concentrations in the soil. Small-scale variations in bedrock composition can therefore cause local variations in the metal content of the soil. Such a case was found in the remote alpine area of the Swiss National Park. Soil profiles were sampled at an altitude of about 2,400 m, representing soils developed above different bedrocks. The concentration of lead in the profiles was found to be strongly dependent on the metal content in the bedrock underlying the soil and was strongly enriched in the top 10 cm. The dolomitic bedrock in the study area contains elevated lead concentrations compared with other dolomites. Dissolution of dolomite and accumulation of weathering residues during soil formation resulted in high lead concentrations throughout the soil profile. The enrichment of lead in the topsoil, however, is largely attributed to atmospheric input. The isotopic signature of the lead clearly indicates that it is mainly of natural origin and that atmospheric deposition of anthropogenic lead contributed to about 20 to 40% to the lead concentration in the topsoil on the bedrock with elevated lead concentrations. In the soils on bedrock with normal lead concentrations, the anthropogenic contribution is estimated to be about 75%. Also, zinc was very strongly enriched in the topsoil. This enrichment was closely correlated with the organic matter distribution in the profiles, suggesting that recycling through plant uptake and litter deposition was a dominant process in the long-term retention of this metal in the soil.  相似文献

Natural and synthetic steroidal hormones can be carried to agricultural soil through fertilization with municipal biosolids, livestock manure, or poultry manure. The persistence and pathways of dissipation of [4-(14)C]-testosterone and of [4-(14)C]-17beta-estradiol in organic-amended soils were investigated using laboratory microcosms. Testosterone dissipation was investigated over a range of amendment concentrations, temperatures, and soil types. Under all conditions the parent compound and transformation products were dissipated within a few days. Addition of swine manure slurry to soil hastened the transformation of testosterone and 17beta-estradiol to the corresponding less hormonally active ketones, 4-androstene-3,17-dione and estrone. Two other testosterone transformation products, 5alpha-androstan-3,17-dione and 1,4-androstadiene-3,17-dione, were also detected. Experiments with sterilized soil and sterilized swine manure slurry suggested that the transformation of (14)C-labeled hormonal parent compounds was mainly caused by microorganisms in manure slurry, while mineralization of the hormones to (14)CO(2) required viable soil microorganisms. Organic amendments transiently inhibited the mineralization of [4-(14)C]-testosterone, perhaps by inhibiting soil microorganisms, or by enhancing sorption and reducing the bioavailability of testosterone or transformation products. Overall, organic amendments influenced the pathways and kinetics of testosterone and estradiol dissipation, but did not increase their persistence.  相似文献

Trace element mobility in soils depends on contaminant concentration, chemical speciation, water movement, and soil matrix properties such as mineralogy, pH, and redox potential. Our objective was to characterize trace element dissolution in response to acidification of soil samples from two abandoned incinerators in the North Carolina Coastal Plain. Trace element concentrations in 11 soil samples from both sites ranged from 2 to 46 mg Cu kg(-1), 3 to 105 mg Pb kg(-1), 1 to 102 mg Zn kg(-1), 3 to 11 mg Cr kg(-1), < 0.1 to 10 mg As kg(-1), and < 0.01 to 0.9 mg Cd kg(-1). Acidified CaCl2 solutions were passed through soil columns to bring the effluent solution to approximately pH 4 during a 280-h flow period. Maximum concentrations of dissolved Cu, Pb, and Zn at the lowest pH of an experiment (pH 3.8-4.1) were 0.32 mg Cu L(-1), 0.11 mg Pb L(-1), and 1.3 mg Zn L(-1) for samples from the site with well-drained soils, and 0.25 mg Cu L(-1), 1.2 mg Pb L(-1), and 1.4 mg Zn L(-1) for samples from the site with more poorly drained soils. Dissolved Cu concentration at pH 4 increased linearly with increasing soil Cu concentration, but no such relationship was found for Zn. Dissolved concentrations of other trace elements were below our analytical detection limits. Synchrotron X-ray absorption near edge structure (XANES) spectroscopy showed that Cr and As were in their less mobile Cr(III) and As(V) oxidation states. XANES analysis of Cu and Zn on selected samples indicated an association of Cu(II) with soil organic matter and Zn(II) with Al- and Fe-oxides or franklinite.  相似文献

A sink for atmospheric methane (CH4) is microbial oxidation in soils. We report CH4 oxidation rates in freely and poorly drained soils on an intensively managed dairy farm. Following cattle urine application to half the plots (650 kg of nitrogen [N] ha(-1)) 31 chamber measurements were made over 100 d during autumn and winter. In the control plots, the freely and poorly drained soils' integrated CH4 oxidation rates averaged 1.8+/-0.2 and 0.6+/-0.1 kg CH4 ha(-1) yr(-1), respectively. In the poorly drained soil, the highest CH4 oxidation rates occurred when water-filled pore space (WFPS)<56% and CH4 oxidation rate declined by ninefold to near zero as WFPS increased from 56 to 68%. Urine application induced the freely and poorly drained soils' CH4 oxidation rates to decline for up to 2 mo by 0.7+/-0.2 and 0.4+/-0.1 kg CH4 ha(-1) yr(-1), respectively. The two soils' responses were thus not significantly different. After urine application, soil pore space CH4 concentration profiles suggested a simultaneous inhibition of bacteria that were CH4 oxidizers and stimulation of CH4 producers.  相似文献

In this study, a tracer bacteria was used to investigate the resuspension and persistence of sediment-associated bacteria in a small alluvial stream. The study was conducted in Swan Creek, located within the Grand River watershed of Ontario, Canada. A 1.1-m2 section of the bed was seeded with a strain of Escherichia coli resistant to nalidixic acid (E. coli NAR). The survival, transport, and redistribution of the tracer bacteria within a 1.7-km river section downstream of the source cell was assessed for a 2-mo period following the introduction of the tracer bacteria. This study has illustrated that enteric bacteria can survive in bed sediments for up 6 wk and that inactivation of the tracer bacteria resembled typical first-order decay. Critical conditions for resuspension, as well as resuspension rates, of sediment-associated bacteria were determined for several storm events. The critical shear stress for E. coli NAR resuspension in Swan Creek ranged from 1.5 to 1.7 N m(-2), which is comparable with literature values for critical shear stresses for erosion of cohesive sediments. Bacteria resuspension was primarily limited to the rising limb of storm hydrographs implying that a finite supply of sediment-associated bacteria are available for resuspension during individual storm events. The information presented in this paper will further the development of representative microbial water quality models.  相似文献

DOI: 10.1111/j.1752‐1688.2011.00528.x Abstract: Present models of the fate of enteric bacteria in surface water usually assume simple first‐order decay. However, observed decay kinetics are typically biphasic and there is increasing evidence for growth in the ambient environment. These features are similar to the stationary phase of long‐term batch cultures, where the dynamics are the result of repeated cycles of population takeovers by Growth Advantage in Stationary Phase (GASP) mutants able to scavenge nutrients released from dead cells. We investigated the possible role of this mechanism (i.e., growth of an adapted subpopulation) in surface water using laboratory experiments with various strains of Escherichia coli in different types of water. Addition of ampicillin (which only kills dividing cells) to these cultures caused a decrease in the total cell density, demonstrating the presence of a growing subpopulation. Furthermore, long‐term survivors from surface water cultures outcompete naïve cells and take over the population in surface water. This growth advantage is heritable (i.e., aged strains were isolated from a single cell) and consistent with the GASP mechanism, although the responsible loci were not identified. Our results show that E. coli populations in surface water are dynamic and consist of dying naïve cells and growing surface water adapted cells. These results can explain the apparent biphasic decay pattern observed in laboratory experiments. This mechanism may also be important in the ambient environment.  相似文献

Dairy manure application to soils can result in phosphorus (P)-related degradation of water quality. The P in these manure-impacted soils can be labile even years after abandonment and under conditions normally associated with high P stability. Failure of P to stabilize with time compounds the environmental consequences of dairy manure disposal, especially on sandy soils. The objectives of this study were to compare chemical characteristics of active and abandoned dairy manure-impacted soils and minimally impacted soils and to assess the continuous release of P in relation to sparingly soluble salts using repeated water extractions, X-ray diffraction, and speciation modeling of column leachates. Soil samples from Ap horizons were collected from nine highly manure-impacted (total P > 1000 mg P kg(-1) soil) areas on four active and five abandoned dairies and four minimally impacted soils (total P < 200 mg P kg(-1) soil). Soil extracts were analyzed for electrical conductivity (EC), soluble reactive phosphorus (SRP), Ca, Mg, Na, and K. The EC of the soil solutions decreased as active dairy > abandoned dairy > minimally impacted soils. Release of Mg and SRP were significantly correlated (r2 = 0.68) and did not decline after abandonment; Ca release was not correlated with SRP (r2 = 0.01), and declined significantly (p < 0.05) after abandonment. Speciation data from column leachates suggested that Mg-P phases and/or the most soluble Ca-P phases could control P solution activities. An implication of this study is that P stabilization via crystallization of calcium phosphates (even at near-neutral pH) may be preempted by Mg-P association. Thus, mechanisms to minimize P release may require P-retaining soil amendments or management of animal rations to eliminate Mg-P formation.  相似文献

One of the problems associated with the use of ambient water quality standards in surface water regulation is the difficulty of identifying and regulating nonpoint source pollution, making such standards unenforceable, especially at the local level. We used the Escherichia coli indicator to locate the most contaminated reaches in rural South Carolina's Bush River watershed (297 km(2), 186 stream-km). We divided the watershed into 20 smaller reaches and sampled each reach multiple times, but restricted each sampling round to one day. We located four low order creek reaches, representing just nine stream-km, where we observed geometric mean E. coli densities of over 1250 E. coli/100 mL; in each case, the source of the contamination (riparian grazing of cattle) was easily identifiable. On the Bush River itself, we observed a step change in one reach where geometric means increased from 106 E. coli/100 mL to 565 E. coli/100 mL over the reach's 10 km length. In this case, the sources of contamination were not as obvious as in the lower order streams; in this case, more advanced Microbial Source Tracking techniques will be required to identify the sources. Nevertheless, this sampling protocol helped locate polluted reaches and provided decision-makers with reasonable justifications for concrete action in deciding where (or where not) to install conservation practices and where more sophisticated (and expensive) MST techniques were warranted.  相似文献

Use of the zebra mussel (Dreissena polymorpha) as an indicator of previously elevated bacteria concentrations in a watershed was examined. The ability of the zebra mussel to accumulate and purge Escherichia coli over several days was investigated in both laboratory and field experiments. In laboratory experiments, periodic enumeration of E. coli in mussels that had been exposed to a dilute solution of raw sewage demonstrated that (i) maximum concentrations of E. coli are reached within a few hours of exposure to sewage, (ii) the tissue concentration attained is higher than the concentration in the ambient water, and (iii) the E. coli concentrations take several days to return to preexposure concentrations when mussels are subsequently placed in sterile water. In field experiments conducted in southeast Michigan in the Clinton River watershed, brief increases in E. coli concentrations in the water were accompanied by increases in mussel concentrations of E. coli that lasted 2 or 3 d. The ability of mussels to retain and to concentrate E. coli made it possible to detect E. coli in the environment under conditions that conventional monitoring may often miss. Sampling caged mussels in a river and its tributaries may enable watershed managers to reduce the sampling frequency normally required to identify critical E. coli sources, thereby providing a more cost-effective river monitoring strategy for bacterial contamination.  相似文献

Attachment of bacteria to soil is an important component of bacterial fate and transport. Escherichia coli are commonly used as indicators of fecal contamination in the environment. Despite the fact that E. coli are derived exclusively from feces or manure, effect of the presence of manure colloids on bacteria attachment to agricultural soils was never directly studied. The objective of this work was to evaluate the magnitude of the effect of manure on E. coli attachment to soil. Escherichia coli attachment to soil was studied in batch experiments with samples of loam and sandy clay loam topsoil that were taken in Pennsylvania and Maryland. Escherichia coli cells were added to the water-manure suspensions containing 0, 20, and 40 g L(-1) of filtered liquid bovine manure, which subsequently were equilibrated with air-dry sieved soil in different soil to suspension ratios. The Langmuir isotherm equation was fitted to data. Manure dramatically affected E. coli attachment to soil. Attachment isotherms were closer to linear without manure and were strongly nonlinear in the presence of manure. The maximum E. coli attachment occurred in the absence of manure. Increasing manure content generally resulted in decreased attachment.  相似文献

Managing manure in no-till systems is a water quality concern because surface application of manure can enrich runoff with dissolved phosphorus (P), and incorporation by tillage increases particulate P loss. This study compared runoff from well-drained and somewhat poorly drained soils under corn (Zea mays, L.) production that had been in no-till for more than 10 yr. Dairy cattle (Bos taurus L.) manure was broadcast into a fall planted cover crop before no-till corn planting or incorporated by chisel/disk tillage in the absence of a cover crop. Rainfall simulations (60 mm h(-1)) were performed after planting, mid-season, and post-harvest in 2007 and 2008. In both years and on both soils, no-till yielded significantly less sediment than did chisel/disking. Relative effects of tillage on runoff and P loss differed with soil. On the well-drained soil, runoff depths from no-till were much lower than with chisel/disking, producing significantly lower total P loads (22-50% less). On the somewhat poorly drained soil, there was little to no reduction in runoff depth with no-till, and total P loads were significantly greater than with chisel/disking (40-47% greater). Particulate P losses outweighed dissolved P losses as the major concern on the well-drained soil, whereas dissolved P from surface applied manure was more important on the somewhat poorly drained soil. This study confirms the benefit of no-till to erosion and total P runoff control on well-drained soils but highlights trade-offs in no-till management on somewhat poorly drained soils where the absence of manure incorporation can exacerbate total P losses.  相似文献

Due to the difficulties in testing for specific pathogens, water samples are tested for the presence of nonpathogenic indicator organisms to determine whether a water supply has been contaminated by fecal material. An implicit assumption in this approach is that where pathogenic microorganisms are present fecal indicator organisms are present as well; yet surprisingly few studies have been conducted that directly compare the transport of indicator organisms with pathogenic organisms in ground water environments. In this study we compared the cell properties and transport of Escherichia coli, a commonly used indicator organism, and Campylobacter jejuni, an important enteropathogen commonly found in agricultural wastes, through saturated porous media. Differences in cell properties were determined by measuring cell geometry, hydrophobicity, and electrophoretic mobility. Transport differences were determined by conducting miscible displacement experiments in laboratory columns. Under the experimental conditions tested, C. jejuni was much more negatively charged and more hydrophobic than E. coli. In addition, C. jejuni cells were slightly longer, narrower, and less spherical than E. coli. The variations in cell properties, primarily surface charge, resulted in significant differences in transport between these two microorganisms, with the transport of C. jejuni exceeding that of E. coli when conditions favored low attachment rates, thus calling into question the usefulness of using E. coli as an indicator organism for this important pathogen.  相似文献

Several genotypic methods have been developed for determining the host origin of fecal bacteria in contaminated waters. Some of these methods rely on a host origin database to identify environmental isolates. It is not well understood to what degree these host origin isolates are geographically variable (i.e., cosmopolitan or endemic). This is important because a geographically limited host origin database may or may not be universally applicable. The objective of our study was to use one genotypic method, ribotyping, to determine the geographic variability of the fecal bacterium, Escherichia coli, from one location in Idaho and three locations in Georgia for cattle (Bos taurus), horse (Equus caballus), swine (Sus scrofa), and chicken (Gallus gallus domesticus). A total of 568 fecal E. coli isolates from Kimberly, ID (125 isolates), Athens, GA (210 isolates), Brunswick, GA (102 isolates), and Tifton, GA (131 isolates), yielded 213 ribotypes. The percentage of ribotype sharing within an animal species increased with decreased distance between geographic locations for cattle and horses, but not for swine and chicken. When the E. coli ribotypes among the four host species were compared at one location, the percent of unshared ribotypes was 86, 89, 81, and 79% for Kimberly, Athens, Brunswick, and Tifton, respectively. These data suggest that there is good ribotype separation among host animal species at each location. The ability to match environmental isolates to a host origin database may depend on a large number of environmental and host origin isolates that ideally are not geographically separated.  相似文献

Soil biotic and abiotic factors strongly influence nitrogen (N) availability and increases in nitrification rates associated with the application of manure. In this study, we examine the effects of edaphic properties and a dairy (Bos taurus) slurry amendment on N availability, nitrification rates and nitrifier communities. Soils of variable texture and clay mineralogy were collected from six USDA-ARS research sites and incubated for 28 d with and without dairy slurry applied at a rate of ~300 kg N ha(-1). Periodically, subsamples were removed for analyses of 2 M KCl extractable N and nitrification potential, as well as gene copy numbers of ammonia-oxidizing bacteria (AOB) and archaea (AOA). Spearman coefficients for nitrification potentials and AOB copy number were positively correlated with total soil C, total soil N, cation exchange capacity, and clay mineralogy in treatments with and without slurry application. Our data show that the quantity and type of clay minerals present in a soil affect nitrifier populations, nitrification rates, and the release of inorganic N. Nitrogen mineralization, nitrification potentials, and edaphic properties were positively correlated with AOB gene copy numbers. On average, AOA gene copy numbers were an order of magnitude lower than those of AOB across the six soils and did not increase with slurry application. Our research suggests that the two nitrifier communities overlap but have different optimum environmental conditions for growth and activity that are partly determined by the interaction of manure-derived ammonium with soil properties.  相似文献
