Although polychlorinated biphenyls (PCBs) have been banned in many countries for more than three decades, exposures to PCBs continue to be of concern due to their long half-lives and carcinogenic effects. In National Institute for Occupational Safety and Health studies, we are using semiquantitative plant-specific job exposure matrices (JEMs) to estimate historical PCB exposures for workers (n?=?24,865) exposed to PCBs from 1938 to 1978 at three capacitor manufacturing plants. A subcohort of these workers (n?=?410) employed in two of these plants had serum PCB concentrations measured at up to four times between 1976 and 1989. Our objectives were to evaluate the strength of association between an individual worker’s measured serum PCB levels and the same worker’s cumulative exposure estimated through 1977 with the (1) JEM and (2) duration of employment, and to calculate the explained variance the JEM provides for serum PCB levels using (3) simple linear regression. Consistent strong and statistically significant associations were observed between the cumulative exposures estimated with the JEM and serum PCB concentrations for all years. The strength of association between duration of employment and serum PCBs was good for highly chlorinated (Aroclor 1254/HPCB) but not less chlorinated (Aroclor 1242/LPCB) PCBs. In the simple regression models, cumulative occupational exposure estimated using the JEMs explained 14–24 % of the variance of the Aroclor 1242/LPCB and 22–39 % for Aroclor 1254/HPCB serum concentrations. We regard the cumulative exposure estimated with the JEM as a better estimate of PCB body burdens than serum concentrations quantified as Aroclor 1242/LPCB and Aroclor 1254/HPCB.  相似文献

A histopathological analysis of the testis exposed to diazinon, an organophosphorus pesticide was conducted. The testis from the control fish exhibited a normal histological structure with a round, oval or somewhat rectangular shaped with closely associated seminiferous tubules. The seminiferous tubules were surrounded by a layer of connective tissue with myoid cells, Leydig cells, blood vessels, lymph vessels and nerves. Upon exposure for 24 h, the overall structure of the testis looked disrupted and more often the sperm cells appeared to be somewhat larger, but spermatid were smaller than that of the control group. The 48 h exposure caused more dense and closely associated seminiferous tubules. The 72 h exposure induced dilation of the seminiferous tubules and their lumen. Ninety-six hours exposure induced more loose and disrupted structures. In this group (96 h), there was a significant reduction in both lumen and seminiferous tubule size in comparison with the control, 24, 48, and 72 h groups. After 2 weeks of exposure hardly any lumen was seen, which imparted more dense and compacted structures. The biometric analysis gave a highly significant increase in the diameter of the seminiferous lumen. After 48 h of exposure a significant reduction was seen in diameter in comparison with that of the control group. Seventy-two hour exposure indicated an increase in diameter, whereas 96 h exhibited another decrease. One week caused the diameter to increase whereas 2 weeks of exposure caused them to decrease again. The increase in diameter of the seminiferous tabule may be caused by weakening of the surrounding connective tissue and myoid cells, due to exposure to diazinon. The change in the diameters of the seminiferous tabules was very irregular. No apparent co-relation was found between the size of the fish, body weight and weight of the testis after they have been subjected to different exposure periods. There was significant change in the germ cells' diameters. As the germ cells are the essential first step in the process of spermatogenesis, their size reduction may hinder the production of viable spermatozoa, endangering the population dynamics. In order to find out the pathways of this pesticide effects, we propose to study the histopathology of pituitary gland a vital organ of the reproductive axis.  相似文献

In this study, the green-lipped mussel, Perna viridis (L.), was exposed to two concentrations of benzo[a]pyrene (B[a]P) (0.3 microg l(-1); 3 microg l(-1)) and two concentrations of Aroclor 1254 (0.5 microg l(-1); 5 microg l(-1)). In addition, a mixture of the contaminants was used (0.3 microg l(-1) B[a]P+0.5 microg l(-1) Aroclor 1254; 3 microg l(-1) B[a]P+5 microg l(-1) Aroclor 1254). All concentrations were nominal. A suite of enzymes [glutathione S transferase (GST), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (GR)], glutathione (GSH) level and lipid peroxidation (LPO) in the mussel gill and hepatopancreas were monitored over 18 days. CAT and GSH in gill tissue were positively correlated with concentration of Aroclor 1254. Activity of hepatic GST and SOD was significantly related to body burden of Aroclor 1254. LPO, GR and GPx in gill and hepatopancreas and hepatic GST were positively correlated with B[a]P concentration. The results indicate the importance of using biomarkers specific to the type of contaminant(s) that are likely to be present. Controlled laboratory experiments, such as this study, are useful in ascertaining biomarkers suitable for use with complex contaminant mixtures in the marine environment.  相似文献

Treatment with B-naphthoflavone (B-NF) (80mg/Kg/day x2), Pyrazole (200mg/Kg/day x1) or indole (400mg/kg/day x1) produce no change in the activity of either enzyme. It, seems therefore, that Aroclor (1254) and chlorpromazine may cause disordered kynurenine metabolism through alterations in the activities of its metabolizing enzymes. This, in turn, might affect nicotinamide adenine dinucleotide biosynthesis and/or the accumulation of some tryptophan metabolites suspected of being carcinogenic or co-carcinogenic.  相似文献

The toxicity of the PCB substitute Ugilec 141, a mixture of tetrachlorobenzyltoluenes (TCBTs), is compared with the toxicity of a commercial mixture of polychlorobiphenyls (Aroclor 1254) and with the model toxic PCB-congener 3,3',4,4',-tetrachlorobiphenyl (PCB-77) as a positive control. Alterations in liver weight, hepatic cytochrome P450 content and EROD and PROD activity, plasma thyroxin and retinol level, hepatic retinoid level and liver and thyroid pathology, have been studied in Ah-responsive and Ah-nonresponsive mice. Ugilec 141 proved to induce similar toxicological changes, qualitatively and quantitatively, to Aroclor 1254. Therefore Ugilec may pose a similar environmental and health risk as PCBs. The criteria for acceptance of new substances, like Ugilec 141, on the European market are discussed.  相似文献

The possibility of exposure of large segments of the population to complex, multifrequency microwave radiation in the environment is now a reality. It is necessary, therefore, to determine the safe level of exposure for the general population so as to prevent any occurrence of harmful effects without unduly restricting the beneficial uses of microwaves.

The biological effects generated by exposure to microwave radiation are usually designated as thermal or non-thermal (specific) in nature. Thermal effects are those interactions which are caused by the heating of the biological specimen and can be duplicated using conventional heating techniques. Nonthermal or specific effects are due to the direct interaction of the electromagnetic field of the incoming microwave radiation and the biological specimen.

The two organs of the body which are particularly sensitive to elevation in temperature are the testicles and the eyes. These organs, therefore, are the most sensitive to exposure to microwave radiation. Research on dogs, rabbits, and rats has shown that at 10 mW/cm, (power density in milliwatts per square centimeter) pathological damage to the testes include a degeneration of the epithelium lining of the seminiferous tubules, and a sharp reduction in the number of maturing spermatocytes. The reduction in testicular function due to the heating effect at 10 mW/cm2 appears to be temporary and reversible.

Cataracts have been produced in the eyes of experimental animals. Several investigators have used the eyes of rabbits to establish threshold for cataract formation. For CW radiation, the threshold in rabbits for long-term ex-posure, was measured to be approximately 100 mW/cm2. Several cases of eye cataracts in man due to microwave exposure have been reported at power densities of the same order of magnitude. More research is needed before threshold values for cataract formation in humans can be specified with certainty.

Nonthermal or specific effects are more difficult to detect than the thermal effects. This difficulty is due to the nature of the response of the biological specimen and the lack of explanation of the mechanism causing the effect. The most often reported effects of low level microwave radiation are neurological in nature. Effects on animals include changes in EEG patterns, changes in the conditioned reflexes, alterations in sensitivity to light, sound, and olfactory stimuli, alterations in the biocurrents of the cerebral cortex, and changes in behavior. Many subjective symptoms in humans working around microwave equipment have been reported by investigators from the U.S.S.R. and Eastern European researchers.

Genetic effects have been reported by some investigators. Exposure of chick embryos to microwave radiation induced abnormal development while conventional heating to the same temperature did not cause abnormalities. In general, the abnormalities appeared to be caused by the inhibition of growth and cell differentiation. Abnormal development of the “mealworm” beetle pupae has also been detected when exposed to microwave radiation. This study also indicates that the abnormalities were not due to thermal effects but due to the inhibition of cell differentiation caused by a direct interaction with the electromagnetic field.  相似文献

The measurement and induction of mixed function oxyigenases (MFOs) of limnetic gastropods were investigated to estimate their suitability as biomarkers. A determination of MFO activities was performed through the measurement of ethoxyresorufin-O-dealkylase (EROD), pentoxy-(PROD) and benzoxyresorufin-O-dealkylase-activity (BROD). Optimal measuring conditions of these activities were investigated in preparations of the digestive gland of 4 species of limnetic gastropods. Results indicate that MFO-activity inhibiting substances are accumulating in the microsomal pellet, the fraction commonly used for the measurement of MFO-activities. Therefore, the fraction used for induction studies was the postmitochondrial supernatant (PMS). EROD, PROD, and BROD activity ofPlanorbis planorbis andPlanorbis carinatus were measured after a treatment with Aroclor 1254 for 1–17 days. Maximal induction of EROD and PROD were 6 and 10 times the value in the control group ofP. carinatus, respectively induction inP. planorbis was lower. BROD-activity could not been measured inP. planorbis. InP. carinatus, BROD-activity increased to a maximum of 2 times after traetment with Aroclor 1254. The level of induction of EROD-activity is comparable to results described in analog studies with fish.  相似文献

We examined the accumulation of PCBs in ospreys (Pandion haleaetus) that were exposed to local sediment sources. Eggs, chick plasma, and sediment samples were collected over a range of 14 km (0.2-14.2 km) from a PCB source in Sturgeon Lake, ON. Sum PCB concentrations declined in chick plasma (range 422.5-58.3 ng/g) as distance from the PCB source increased, but there was a poor relationship with sum PCBs in eggs. Both tissues indicated an Aroclor 1248/1254 source. Aroclor 1254 comprised an average of 66.9% of sum PCBs in chick plasma from Sturgeon Lake, but comprised only from 27.0 to 44.4% in plasma from other Great Lake colonies. Dietary differences among osprey colonies were not sufficient to explain the PCB patterns observed. There was weak evidence that the ability to metabolize PCBs may differ between juveniles and adults, based upon the PCB profile in eggs and chick plasma.  相似文献

Nuclear magnetic resonance (NMR) - based metabolomics has the potential to identify toxic responses of contaminants within a mixture in contaminated soil. This study evaluated the metabolic response of Eisenia fetida after exposure to an array of organic compounds to determine whether contaminant-specific responses could be identified. The compounds investigated in contact tests included: two pesticides (carbaryl and chlorpyrifos), three pharmaceuticals (carbamazephine, estrone and caffeine), two persistent organohalogens (Aroclor 1254 and PBDE 209) and two industrial compounds (nonylphenol and dimethyl phthalate). Control and contaminant-exposed metabolic profiles were distinguished using principal component analysis and potential contaminant-specific biomarkers of exposure were found for several contaminants. These results suggest that NMR-based metabolomics offers considerable promise for differentiating between the different toxic modes of action (MOA) associated with sub-lethal toxicity to earthworms.  相似文献

The aromatic amine 3,4-dichloroaniline (DCA) is a model environmental contaminant, precursor for synthesis and degradation product of several herbicides, which is commonly found in European estuarine ecosystems. In this work, the possibility of using biochemical and histological markers to assess sub-lethal effects of DCA in natural populations of Pomatoschistus microps juveniles was investigated. Alterations on the activities of the enzymes acetylcholinesterase (AChE), lactate dehydrogenase (LDH) and glutathione S-transferase (GST) and histological alterations on spleen were investigated after 96 h of exposure to sublethal concentrations of DCA (0.50-1.49 mg/l). At the concentrations tested, DCA had no effect on AChE activity. LDH and GST activities were significant altered in treated animals when compared to control groups. As already described for mammals, DCA induced splenic histological alterations in P. microps, including expansion of red pulp and deposition of hemosiderin granules in a concentration-dependent manner. This suggests that DCA is a xenobiotic of concern in estuaries receiving agricultural effluents.  相似文献

Lake Ontario coho salmon were known to contain a mixture of chemical contaminants. A previous study demonstrated that rats fed the Lake Ontario fish-supplemented diet for 28 days exhibited mild biochemical and histological changes. The purpose of the present study was to determine the effects due to a longer term of exposure and the reversibility of these effects. Growth rate and food consumption were not affected by feeding the animals with Lake Ontario or Pacific fish-supplemented diets for 13 weeks. No deaths were observed. Decreased spleen weights were observed in groups of males fed 1.45%, 5.8% Lake Ontario and 2.9% Pacific diet. After a 13 week recovery the spleen weights returned to normal. Decreased serum potassium was observed in male rats fed 2.9% Lake Ontario diet, and all levels of Pacific diet for 13 weeks, and was not evident following maintenance on normal diet. Serum glucose was not affected by the 13-week period of treatment, however; a reduction in this parameter occurred in male rats fed the two highest doses of Lake Ontario diet and all doses of Pacific diet following the 13-week recovery period. Minor hematological changes occurred only in the male rats fed either Lake Ontario or Pacific diet following a 13 week recovery period and included reduced marrow myeloid cells and myeloid/erythroid ratio. Hepatic microsomal ethoxyresorufin deethylase activity was significantly increased in rats ingesting Lake Ontario diet. Mild histological changes occurred in the liver and thyroid of the treated males, and in the liver and kidney of the treated females. These changes were attributed to the chemical residues and/or the fish diet. Data presented here indicated that the Lake Ontario fish-supplemented diet can cause mild biochemical, hematological and histological changes but most of these were reversible when exposure was terminated.  相似文献

Hepatic S9 preparations from Aroclor 1254 induced rats and 3-methylcholanthrene induced woodchucks were used to investigate, in vitro, the mutagenic potential of five amino acid conjugates of 2,4-Dichlorophenoxyacetic acid (alanine, aspartic acid, leucine, methionine and tryptophan). Five strains of Salmonella typhimurium (TA97, TA98, TA100, TA1535, TA1538) were utilized for this purpose. Dose-response effects producing a two-fold increase of revertants over spontaneous levels were not observed with either S9 preparation indicating that the amino acid conjugates are not promutagens in these assays.  相似文献

On exposure to sunlight or photolysis at λ >300nm, the dechlorination of a polychlorinated biphenyl mixture (Aroclor 1254) in the H donor alcohol 2-propanol under neutral conditions is enhanced by the presence of the photosensitizer hydroquinone. Dechlorination is strongly promoted by an increasingly aqueous solvent (1:1 water:alcohol) and by maintaining neutral conditions (pH 7.0 buffer) both in the presence and absence of sensitizer. Atmospheric oxygen (continuous aeration) retards the hydroquinone induced dechlorination less than the direct photolysis process.  相似文献

Treatment with Aldrin, an organochlorine insecticide, for 13 and 26 days caused suppression of 3beta-hydroxysteroid dehydrogenase (3beta-HSD) and 17beta-hydroxysteroid dehydrogenase (17beta-HSD) activities, along with accumulation of cholesterol in the testicular tissues of adult rats. The same treatment also resulted in a reduction in the nuclear diameter of Leydig cells (LCND) and diameter of seminiferous tubules. A decrease in the weight of testes, seminal vesicles and ventral prostate was also noted. HCG administration in the long-term (26 days) treatment restored the steroidogenic enzymes activity and the nuclear diameter of the Leydig cells. It also reduced the accumulation of tissue cholesterol towards the vehicle-injected controls. The inhibition of steroidogenesis in the testes possibly reflects a decrease in pituitary gonadotrophin release after the treatment with Aldrin.  相似文献

Aroclor 1254 (1860 mg/l) in alkaline 2-propanol and in the presence of acetone (4% v/v) was photodechlorinated to biphenyl at λ >300 nm with exceptionally high quantum yield (Φ= 18). Under similar conditions photodechlorination of extracts of Aroclor 1254 contaminated soil (730 mg/kg) proceeded with lower quantum yield Φ= 0.4. While oxygen severely quenched photolysis, dechlorination was accomplished under thermal conditions using di-t-butyl peroxide, t-BuOOBu-t, as free radical initiator. A free radical chain reaction is suggested in which acetone triplet, T1(n,π*), or t-BuO· radical abstracts H-atom from 2-propanol to give the ketyl radical, (CH3)2 OH, which after losing a proton to the alkaline medium gives the ketyl radical anion, (CH3)2CHO·−. The Aroclor in turn reacts with the latter species through an electron-transfer process giving unstable aryl radical anion, , which cleaves releasing the chloride anion, Cl and the aryl radical, A·  相似文献

Light microscopic study of different body organs of cat-fish following exposure to HgCl2 0.2 mg/L in water for 30 days revealed that focal degeneration of liver cells and disorganization of hepatic cords occurred at places. Furthermore, centrilobular atrophy and compensatory hypertrophy of some hepatic cells were also observed. In the kidneys disintegration of renal epithelium along with displacement of nuclei, shrinkage of glomeruli, breakdown of Bowman's capsule and heavy infiltration by inflammatory cells were observed. The histopathological changes noted in the intestine included degeneration of lining epithelium, and diminution of goblet cells. Microscopic section of ovaries exhibited reduction of ooplasm leading to formation of atypical oocytes. An increase in the occurrence of atretic oocytes and interfolicular spaces was also discernible. No histopathological lesions could be detected in testes of male fish probably because of the difference in the maturity of the control and experimental groups. The pathomorphological alterations in relation to mercury toxicity in spleen were associated with the disorganization of the splenic cords resulting in the displacement of lymphatic tissue cells within the substance of splenic pulp. Marked depletion of the red pulp was noticeable.  相似文献

The spatial distribution, degree of pollution and major sources of PCBs were evaluated in surficial sediments within the heavily urbanized and industrialized ?zmit Bay and its main freshwater inputs. ΣPCB concentrations range from 2.90 to 85.4ngg(-1) in marine sediments and from ND to 47.7ngg(-1) in freshwater sediments. Results suggest that high concentrations of ΣPCBs were localized around a chlor-alkali plant and an industry that handles bulk liquid, dry and drummed chemicals, and petroleum products in the Bay. Using a chemical mass balance receptor model (CMB), major sources of PCBs in the region were investigated. The CMB model identified Aroclor 1254 and 1260 to be the major PCB sources in marine sediments and the less chlorinated Aroclor 1248 and 1242 as the major PCB sources in freshwater sediments. The potential sources for the PCBs were briefly discussed in terms of their use in various industrial applications.  相似文献

The analysis of PCBs in fish tissues by immunoassay methods was evaluated using fish collected from a US monitoring program, the National Contaminant Biomonitoring Program of the US Department of Interior, Fish and Wildlife Service. Selected composite whole fish samples, which represented widely varying concentrations and sources of PCBs, were extracted and subjected to congener PCB analysis by gas chromatography (GC) and total PCB analysis using an ELISA (ePCBs) calibrated against technical Aroclor 1248. PCB congener patterns in these fishes were different from the patterns found in commercial Aroclors or their combinations as demonstrated by principal component analysis of normalized GC congener data. The sum of the PCB congeners measured by GC (total-PCBs) ranged from 37 to 4600 ng/g (wet weight). Concentrations of PCBs as determined by the ELISA method were positively correlated with total-PCBs and the ePCBs/total-PCBs ratios for individual samples ranged from 1 to 6. Ratios of ePCBs/total-PCBs for dilutions of Aroclors 1242, 1254, and 1260 and for matrix spikes range from 0.6 for 1242 to 2.5 for 1254 and 1260. These results suggest that higher chlorinated PCB congeners have higher affinity for the anti-PCB antibodies. Partial least squares with latent variable analysis of GC and ELISA data of selected Aroclors and fish samples also support the conclusion that ELISA derived PCB concentrations are dependent on the degree of chlorination.  相似文献
