Specimens of farmed European sea bass (Dicentrarchus labrax L., 1758) were exposed to different cadmium (Cd) concentrations (4.47, 5.63, 7.08 and 8.91 mg l(-1)) for 24 and 48 h. The effects of Cd on numbers of some cell types and structures (i.e., chloride cells, CCs; macrophage aggregates, MAs; rodlet cells, RCs) and on structure and ultrastructure of the main organs (gill, liver, intestine, kidney) were studied with routine process for light and transmission electron microscopy. Following cadmium exposure, the numbers of branchial CCs as well as intestinal and renal RCs increased significantly within 24h. Increase in metal concentration did not affect the magnitude of the numerical increment of the aforementioned cells. Moreover, in treated fish (24 and 48 h) the numbers of MAs in both head kidney and spleen were significantly higher than in control conspecifics, whilst the global area of MAs was less influenced by the acute treatment. In exposed sea bass, all the examined organs exhibited cellular modifications which appeared time- and dose-dependent. The gills showed telangectasia, lamellar fusion, oedema, epithelial lifting and leukocyte infiltration. In the liver, kidney and intestine acute cell swelling and vacuolization were common. Ultrastructurally the alterations observed frequently in hepatocytes, tubular epithelial cells and enterocytes included presence of numerous myelinoid bodies, damaged mitochondria, dilatation of endoplasmic reticulum, high number of lysosomes and autophagolysosomes. In intestinal and kidney tubular epithelia of treated fish, rodlet cells displayed some anomalies like dilatation of nuclear envelope, cytoplasmic vacuolization, presence of myelinoid bodies, rodlets degeneration and extensive discharge activity. 相似文献

Polychlorinated biphenyls (PCBs) and dichlorodiphenyltrichloroethane (DDT) and metabolites were quantified in muscle and liver of sea bass (Dicentrarchus labrax) collected in Ria de Aveiro and in two fish farms. Sea bass from natural environment showed lower levels than fish from farming, which may be partly attributed to the higher lipid content of cultivated fish. PCB congener distribution in tissues of sea bass from the two farms resembled that of diet pellets suggesting that commercial diet is a major source of PCBs. However, fish in the two sites were fed with diet of similar PCB and DDT contamination but showed distinct levels in its tissues not explained by lipid content. 相似文献

The main goal of this study was to assess both mercury (Hg) accumulation and organs’ specific oxidative stress responses of gills, liver and kidney of Dicentrarchus labrax with emphasis on seasonality. Fish were collected in cold and warm periods in three stations: reference, moderated and highly contaminated sites. Our results showed that seasonal factors slightly influenced Hg accumulation between year periods (cold and warm) and strongly affected organs’ response basal levels. In contrast, seasonality seemed not to influence oxidative stress responses, since similar response patterns were obtained for both year periods, and moderate degree of antioxidant responses was obtained. Moreover, the oxidative stress profile may be attributed to Hg contamination degree, which showed organ-specific response and accumulation patterns. Hence, gills showed to be able to adapt to Hg contamination, and in opposition, kidney and liver demonstrated some vulnerability to Hg toxicity. The critical Hg concentrations indicated specific threshold limits for each organ. Overall, seasonality should be taken into account in monitoring programmes, helping to characterize the individuals’ reference values of response and thus to discriminate between the effects induced by natural causes or by contamination. 相似文献

The effect of chronic exposure to a low concentration (0.5 microg l(-1)) of cadmium ions was investigated on escape behaviour of sea bass, Dicentrarchus labrax, using video analysis. Observations were also performed on the microanatomy of lateral system neuromasts. When fish were exposed for 4h per day over 8 days to the cadmium ions, most of both types of neuromasts observed remained intact. However, some of them presented damaged sensory maculae. Whereas before cadmium exposure, fish responded positively to nearly all the lateral system stimulations, after exposure they decreased by about 10% their positive responses to stimulations. From the 15th day after the beginning of cadmium exposure, neuromasts presented progressively less damage, cadmium accumulation in gills and scales decreased significantly and fish escape behaviour had recovered. This study presents a new concept in ecotoxicology: using behavioural change to reveal the effects of pollution levels, scarcely detectable by currently used techniques (physiological responses). 相似文献

Concentrations of polychlorinated biphenyls (PCBs), and organochlorine pesticides (OCPs) were determined in two fish species, mullet (Mugil cephalus) and sea bass (Dicentrarchus labrax), collected from Bizerte Lagoon and the Mediterranean Sea. In all samples, PCBs were found in higher concentrations than OCPs. The highest concentrations of OCPs and PCBs were found in sea bass, and in Bizerte Lagoon. Concentrations of DDTs and PCBs detected in this study were generally comparable or slightly higher than those found in studies from other Mediterranean and non-Mediterranean regions subject to a high anthropogenic impact. ∑PCBs, ∑HCHs and HCB levels were negatively correlated with lipid content, while no such correlation was seen for ∑DDTs. A significant correlation between levels and length and between levels and weight existed only for ∑PCBs. The daily intake of PCBs and OCPs ingested by people living in Bizerte through the studied fish species was estimated and compared with those observed in other areas. 相似文献

Chronic concentrations of polycyclic aromatic hydrocarbons (PAHs) have been commonly detected in international estuaries ecosystems. Reliable indicators still need to be found in order to properly assess the impact of PAHs in fish. After an in vivo chronic exposure to hydrocarbons, the enzymatic activity of 7-ethoxyresorufin O-deethylase (EROD) and the antioxidant defense system were assessed in sea bass, Dicentrarchus labrax. A total of 45 fish were exposed to the water-soluble fraction of Arabian crude oil, similar to a complex pollution by hydrocarbons chronically observed in situ, while 45 other control fish sustained the same experimental conditions in clean seawater. Fish samples were made after a 21-day exposure period and after a 15-day recovery period in clean fresh water. Throughout the experiment, liver EROD activity was significantly higher in contaminated fish than in control fish. In addition, nonenzymatic (total glutathione) and enzymatic (GPx, SOD, and CAT) antioxidant defense parameters measured in liver were not significantly different in fish. Furthermore, in gills, glutathione content had significantly increased while SOD activity had significantly decreased in contaminated fish compared to controls. On the other hand, CAT and GPx activities were not affected. Chronic exposure to PAHs disturbing the first step (SOD) and inhibiting the second step (GPx and CAT) could induce oxidative stress in tissues by the formation of oxygen radicals. After the postexposure period, there was no significant difference between control and contaminated fish in any of the antioxidant defense parameters measured in gills, attesting to the reversibility of the effects. 相似文献

The aim of this research was to assess the degree of heavy metal (cadmium, lead and mercury) contamination of wild boars from central Croatia and thereby conduct further tests to connect the contamination to oxidative stress occurrence. The highest cadmium concentration was noticed in the kidneys of older boars, the lead concentration was approximately the same in the liver and kidneys, but it was much higher than the concentration in the muscles, while the highest mercury concentration was measured in the kidneys of all wild boars. The correlation between the malondialdehyde (MDA) and the heavy metal concentrations was higher in tissue samples taken from younger animals (1 to 3 years of age) than in the tissue samples taken from older animals (5 to 6 years of age). In the kidneys of all wild boars there was a statistically significant connection between the MDA and cadmium concentration; in the kidneys of younger wild boars there was a statistically significant connection between the MDA and lead concentration. Further research on this topic should focus on MDA potential to find its use, not only as a contamination biomarker in the area of ecotoxicology, but also in the evaluation of the hygienic acceptability of animal products. 相似文献

Mercury (Hg) concentrations were monitored in wild and cultured fish collected from fresh and coastal waters in the Korean peninsula from April 2006 to August 2008 nationwide. Total Hg concentrations were reported for 5043 fish samples, including 78 species from 133 locations. Significant interspecies variation was noted in the Hg levels. The average Hg concentration in each fish species ranged from 6.31 μg kg−1 for mullet (Mugil cephalus) to 200 μg kg−1 for mandarin fish (Siniperca scherzeri). Among the species collected, the maximum concentration of Hg, 1720 μg kg−1, was measured in an Amur catfish (Silurus asotus). Only wild freshwater fish exceeded the WHO ingestion standard. Wild freshwater piscivorous fish samples from a large artificial upstream lake contained the highest Hg levels. Hg concentrations were compared between fish groups categorized as wild and farmed fish from freshwater and coastal waters. Although the wild freshwater fish had similar size ranges, their Hg concentrations were higher than those of the other groups. Compared to the feed of farmed marine and freshwater fishes, the prey of wild freshwater fish had a higher Hg concentration, and the total Hg concentrations in freshwater and associated sediment samples were higher than those in coastal water and associated sediment samples. In the freshwater environment, piscivorous fish bioaccumulated two times more Hg than carnivorous and omnivorous fish and four times more than planktivorous fish. The difference in Hg concentrations among trophic groups might have been due to differences in the size of fish, in addition to the variations among different trophic groups. Stable isotopes of carbon and nitrogen (delta(13)C and delta(15)N) have been determined in wild and farmed gilthead sea bream (Sparus aurata) samples of white and red muscle, liver, gills and gonads. First, delta(13)C and delta(15)N values were determined in samples with and without lipid removal to check the possible effect of lipid content on the stable isotope values of the different tissues studied. Differences were found for delta(13)C in all tissues studied apart from white muscle of wild fish, the tissue with the lowest lipid content. For delta(15)N values no differences were found in wild fish tissues. Liver from farmed fish showed lower delta(15)N value after lipid removing. Further conclusions were based on results obtained from lipid-free samples. delta(13)C of cultured fish tissues showed a mean depletion of 2.9+/-0.4 per thousand compared to wild specimens, suggesting different sources of carbon in the diet, probably due to the feed used during sea-cage culture. Cultured gilthead sea bream tissues were significantly more enriched in nitrogen than wild specimens by an average of 1.5+/-0.2 per thousand in white muscle, indicating a slight increase in the trophic level. Determination of stable isotope signatures of gilthead sea bream tissues allows clear discrimination between wild and cultured sea bream, and characterisation of differences in diet and feeding conditions in any tissue studied. 相似文献

We analyzed polychlorobiphenyls (PCBs), perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA) in the edible part of farmed sea bass reared in two fish farms in Liguria (Northern Italy). The aim was to determine the contamination levels and human exposure to these chemicals through fish consumption. Concentrations of “dioxin-like” PCBs (DL-PCBs) ranged from 0.033 to 0.759 pg ΣTEQ-PCB g−1 whole weight (w.w.) in fish farm 1 and from 0.032 to 1.60 pg ΣTEQ-PCB g−1 w.w. in fish farm 2, and the six indicators of “non-dioxin-like” (NDL-PCBs) from 0.538 to 9.33 ng Σ6PCB g−1 w.w. and from 1.62 to 27.6 ng Σ6PCB g−1 w.w. Concentrations were generally lower in farm 1 than in farm 2. One reason for this difference might be the proximity of farm 2 to the seaport of La Spezia, which could be a punctual source of pollutants influencing the contamination of the water in the farm. Principal component analysis (PCA) showed differences also in the congeners profiles for the two sites, with higher-chlorinated PCBs more abundant in farm 1, and lower-chlorinated PCBs were more abundant in farm 2. Most of the concentrations of PFOS and PFOA were below the limit of detection (LOD 0.05 ng g−1 w.w.). Only about 10% of the samples analyzed had levels slightly higher than the LOD. Assessments of exposure using these data showed that consumption of farmed fish may contribute significantly to PCBs through the diet, whereas the contribution of PFOS and PFOA seems to be low. 相似文献

To investigate the transfer of lipophilic organochlorine compounds to the gonads in gilthead sea bream females, liver and oocytes from wild and farmed females, just before spawning, were analysed by gas chromatography coupled to tandem mass spectrometry after clean-up of the fatty extracts by normal phase HPLC. This analytical methodology allowed us to achieve limits of detection of around 0.1 ng/g. Data obtained show that the chemicals studied are transferred from the liver to oocytes in ratios up to 0.51. In general this means that less than a half of pollutant load is transferred from liver to oocytes. A significant correlation (p<0.05) between log K(ow) and ratios (oocytes/liver) was found. Compounds with larger Log K(ow) present lower lipid-based concentration ratios (oocytes/liver). We have inferred that the reason the fugacity model is not accomplished is specifically dependent on the vitellogenesis process, which involves the transfer of lipids from the liver to oocytes in teleostei. The acute toxicity (96 h) of pyrene (PY) to European seabass (Dicentrachus labrax) juveniles assessed in a semi-static bioassay (SSB) with medium renewal at each 12 h, and in a static bioassay (SB) without medium renewal was compared in laboratorial conditions (water PY concentrations: 0.07-10 mg L−1). Main findings in the SSB that assessed mainly the toxicity of PY and its metabolites were: increased levels of bile PY metabolites in good agreement with the profile of lipid peroxidation levels (LPO) in exposed fish relating PY exposure and oxidative damage; increased levels of PY-type compounds in the brain indicating their ability to cross the blood-brain barrier; increased levels of these substances in liver and muscle which are edible tissues for humans thus raising concern on potential adverse effects on consumers of fish from PY contaminated areas; a significant inhibition of glutathione S-transferase activity suggesting its involvement in PY detoxication as toxicant scavenger; finally, an almost complete impairment of the swimming velocity at all the PY concentrations linking sub-individual to higher population level effects. In the SB, where the overall toxicity of PY, its metabolites and environmental degradation products was evaluated, 19% and 79% of PY decay in test media was found at 12 and 96 h, respectively. In general, the effects were similar to those of SSB but with significant effects being induced at higher PY concentrations indicating that the parental compound is more toxic than its environmental degradation products. The other main differences relatively to the SSB were: increased levels of PY-type substances in the liver suggesting more accumulation in this organ. Therefore, these findings highlight the need of carefully considering experimental design options when assessing the toxicity of readily degradable substances to marine fish, and stress the importance of taking into consideration the toxicity of environmental degradation products in addition to toxic effects of the parental substance and its metabolites for marine ecological risk assessment. 相似文献

This paper reports concentrations of mercury in muscle tissue of flounder (Platichthys flesus L.) from various estuaries and coastal waters of north Wales and north-west England that encompass the north-east Irish Sea. Mercury concentrations were highest within and offshore the Ribble Estuary (0.476 +/- 0.037 mg kg(-1)) but were also high within the Mersey (0.389+/-0.042 mg kg(-1)) and Dee Estuaries (0.295+/-0.065) and at coastal sites in the vicinity. Values for all three estuaries and their near coastal sites were significantly higher than for sites in the north and central sectors of the study area. Regressions of mercury concentration against fish length showed significant relationships and age accumulation for the majority of sites, particularly within or in close proximity to the Mersey, Dee and Ribble Estuaries. 相似文献

Data gathered in this study suggested the exposure of rats and Algerian mice, living in an abandoned mining area, to a mixture of heavy metals. Although similar histopathological features were recorded in the liver and spleen of both species, the Algerian mouse has proved to be the strongest bioaccumulator species. Hair was considered to be a good biological material to monitor environmental contamination of Cr in rats. Significant positive associations were found between the levels of this element in hair/kidney (r=0.826, n=9, p<0.01) and hair/liver (r=0.697, n=9, p=0.037). Although no association was found between the levels of As recorded in the hair and in the organs, the levels of this element recorded in the hair, of both species, were significantly higher in animals captured in the mining area, which met the data from the organs analysed. Nevertheless, more studies will be needed to reduce uncertainty about cause-effect relationships. 相似文献

In order to elucidate marine pollution by butyltins and organochlorine residues in coastal waters of Thailand, green mussels (Perna viridis) were collected in 1994 and 1995 and analyzed. Butyltin compounds, such as tributyltin (TBT), dibutyltin (DBT) and monobutyltin (MBT), were detected in most mussel samples, ranging from 4 to 800 ng g(-1) wet wt (as total). The composition of butyltin derivatives in green mussel was in the order of TBT > DBT > MBT. The results indicated that butyltin contamination was widespread, particularly in high boating areas and in coastal aquaculture facilities. Concentrations of organochlorine residues were much lower than of butyltin compounds. Among organochlorine residues examined, dichlorodiphenyltrichloroethane compounds (DDTs) were found to be the highest, followed by polychlorinated biphenyls (PCBs) > chlordane compounds (CHLs) > hexachlorocyclohexanes (HCHs) > hexachlorobenzene (HCB). To our knowledge, this is a first report on the detection of butyltin residues in Thailand coastal waters. 相似文献
