Feeding, growth and bioluminescence of the thecate heterotrophic dinoflagellate Protoperidinium huberi were measured as a function of food concentration for laboratory cultures grown on the diatom Ditylum brightwellii. Ingestion of food increased with food concentration. Maximum ingestion rates were measured at food concentrations of 600 g C l-1 and were 0.7 g C individual-1 h-1 (1.8 D. brightwelli cells individual-1 h-1). Clearance rates decreased asymptotically with increasing food concentration. Maximum clearance rates at low food concentration were ca. 23 l ind-1 h-1, which corresponds to a volume-specific clearance rate of 5.9x105 h-1. Cell size of P huberi was highly variable, with a mean diameter of 42 m, but no clear relationship between cell size and food concentration was evident. Specific growth rates increased with food concentration until maximum growth rates of 0.7 d-1 were reached at a food concentration of 400 g C l-1 (1000 cells ml-1). Food concentrations as low as 10 g C l-1 of D. brightwellii (25 cells ml-1) were able to support growth of P. huberi. The bioluminescence of P. huberi varied with its nutritional condition and growth rate. Cells held without food lost their bioluminescence capacity in a matter of days. P. huberi raised at different food concentrations showed increased bioluminescence capacity, up to food concentration that supported maximum growth rates. The bioluminescence of P. huberi varied over a diel cycle, and these rhythmic changes persisted during 48 h of continuous darkness, indicating that the rhythm was under endogenous control.  相似文献

In order to assess the intake of lead and cadmium by consumers of home grown vegetables in urban areas, replicated experimental plots of uniform size, comprising summer and winter crops, were established in 94 gardens and allotments in nine towns and cities in England.The geometric mean lead and cadmium concentrations for the soils (n = 94) were 217 g g–1 (ranging from 27 to 1,676 g g–1) and 0.53 g g–1 (<0.2–5.9 g g–1), respectively. Compared with agricultural soils, the garden and allotment soils contained elevated levels of lead but not cadmium.Lead concentrations in the vegetables ranged from <0.25 g g–1 to 16.7 g g–1 dry weight and cadmium concentrations ranged from <0.025 g g–1 to 10.4 g g–1 dry weight. Lead concentrations were higher than reported background levels, although <1% exceeded the statutory limit for saleable food in the UK (1 g g–1 fresh weight). Cadmium concentrations were generally similar to background levels.  相似文献

The substrate analogue [14C]-methylammonium was used to study ammonium/methylammonium uptake by Symbiodinium microadriaticum (zooxanthellae). The value of the Michaelis constant (K m) for the uptake system was approximately 35 M with methylammonium as substrate; ammonium was a competitive inhibitor of methylammonium uptake, and the K m for ammonium uptake (determined as the inhibition constant, K i, for methylammonium) was 6.6 M. Methylammonium uptake by zooxanthellae was light-dependent. Methylammonium uptake rates of zooxanthellae which had been freshly isolated from the hermatypic coral Acropora formosa (0.85±0.05x10-10 mol min-1 cell-1) were lower than those of axenic cultures of the zooxanthellae from Montipora verrucosa (Acroporidae) grown under various nitrogen regimes (1.6 to 12x10-10 mol min-1 cell-1). Maximum uptake rates were found for ammonium-starved cultured M. verrucosa zooxanthellae (10.2 to 12x10-10 mol min-1 cell-1); M. verrucosa zooxanthellae growing with ammonium as nitrogen source and zooxanthellae which had been freshly isolated from A. formosa gave similar and considerably lower uptake rates (0.85 to 1.6x10-1 mol min-1 cell-1). These results suggest that either coral tissue contains sufficient ammonium to repress synthesis of the uptake system of the algal symbionts or, alternatively, there are additional barriers to ammonium transport for zooxanthellae in vivo.  相似文献

Sea anemones (Aiptasia pulchella) containing zooxanthellae (Symbiodinium microadriaticum) were maintained in a long-term laboratory culture on a 12 h light (100 E m-2 s-1):12 h dark cycle. Photosynthetic oxygen production was measured for the symbiotic association and for freshlyisolated zooxanthellae. Light utilization efficiencies () were similar for both sets of zooxanthellae, suggesting negligible shading of zooxanthellae by animal tissue in this association. Whereas freshly-isolated zooxanthellae were photoinhibited at high irradiances (800 to 1 800 E m-2 s-1), zooxanthellae in the host continued to function at photosynthetic capacity. Time of day may influence photosynthetic measurements in symbiotic organisms, as it was found that photosynthesis in A. pulchella followed a diel periodicity at both light-saturating (1 200 E m-2 s-1) and subsaturating (150 E m-2 s-1) irradiances. There was a peak period of photosynthesis between 12.00 and 14.00 hrs. Light stimulated dark respiration rates of A. pulchella. Dark respiration of sea anemones increased somewhat towards the end of the light cycle and was always greater after exposure to high irradiances.  相似文献

An investigation on the abundance and distribution of trace metals (Fe, Cu, Zn, Mn, Cr, Cd and Pb) in water, and nine species of fish samples from Calabar river was carried out in 1992. The concentrations of iron (6000–7240gl–1), zinc (4910–7230gl–1), and cadmium (3–7gl–1) showed moderate pollution while those of copper (420–630gl–1), manganese (23–48gl–1), chromium (<10–20gl–1) and lead (<1–10gl–1) in water were well below WHO permissible levels. Significant seasonal changes (0.001p0.25) were obtained for iron, copper, zinc, manganese and cadmium in water. Furthermore, iron, zinc and cadmium showed statistically significant spatial changes (0.005p0.10). Of the nine fish species studied, no statistically significant relationship between body weight and the concentrations of the metals was observed. The concentrations of the metals per mean total body weight apparently decreases in the order Fe>Zn>Cu>Mn>Pb>Cd=Cr and were within the limits that were safe for consumption.  相似文献

Aquaria containing clean sand received a continuous supply of seawater from Santa Rosa Sound, Florida, USA, mixed with known concentrations (7, 76 and 622 g l-1) of pentachlorophenol (PCP). After 9 weeks, nematodes accounted for 87% of the total meiofauna. Nematode biomass and densities were greatest in aquaria exposed to 76 g PCP l-1 and were least in aquaria exposed to 622 g PCP l-1. Epistrate feeders were abundant in control aquaria and aquaria exposed to 7 and 76 g PCP l-1, but not in aquaria exposed to 622 g PCP l-1. Selective deposit feeders were not abundant in the control aquaria and aquaria exposed to 7 g PCP l-1, but comprised 19% of the nematodes in aquaria exposed to 76 g PCP l-1 and 61% in aquaria exposed to 622 g PCP l-1.  相似文献

Mussels, Mytilus edulis planulatus, were collected from Port Phillip Bay, Victoria, Australia, in February 1984, to test the assumption that they are integrators of cadmium pollution. Groups of mussels were subjected to the same average dose of cadmium (21 g l-1), administered according to different dosing regimes over 4 wk (Regimes 1, 2, 3, 5); other groups received twice the average dose (42 g l-1) in half the time (Regime 4). During each regime, the mussels were exposed to the different cadmium concentrations for one week at each concentration. Nominally, the regimes were: (1) 36 g (Wk 1) to 26 g (Wk 2) to 16 g (Wk 3) to 6 g (Wk 4) Cd l-1; (2) 6 to 16 to 26 to 36 g Cd l-1; (3) 21 to 21 to 21 to 21 g Cd l-1; (4) 42 to 42 g Cd l-1; (5) 42 to 42 g Cd l-1 to background (<0.5 g) to background. Differences in cadmium accumulation by mussels from Regimes 1 and 4 were not statistically significant, nor were differences in accumulation between mussels from Regimes 2 and 3. However, mussels from Regimes 1 and 4 had accumulated significantly more cadmium than had mussels from Regimes 2 and 3. Accumulation by mussels from Regime 5 was not significantly different from that by mussels from any of the other regimes. These results suggest that, at least for cadmium, the assumption that mussels are integrators of pollution should be treated with caution. They also have implications with regard to the quantitative biological monitoring of pollution. For example, even in a carefully controlled monitoring program, using mussels of standard size and condition, significant differences in cadmium content between mussels need not indicate exposure to different levels of contamination. Rather, these differences could reflect differences in the regime by which the contamination was received.  相似文献

Despite some progress in reducing the average lead level in the USA, the streets of Cincinnati, Ohio, are still contaminated by heavy metals. High levels of heavy metals will have significant unequivocal ecological impacts and pose a potential health hazard. This study evaluates the level of heavy- metal contamination in household dust and examines its relationships with the external environment. Samples of outdoor and indoor dust were collected from middle-income residential homes in the Greater Cincinnati Metropolitan District and the metal content was analysed by inductively coupled plasma atomic emission spectrophotometry. Results showed that the mean levels of lead in outdoor and indoor dusts are 650 g g-1 and 377 g g-1 and the copper levels are 253 g g-1 and 510 g g-1, respectively. The median levels are 156 g g-1 and 139 g g-1 for outdoor and indoor lead dusts and 35 g g-1 and 124 g g-1 for outdoor and indoor copper dusts. The degree of contamination may be ascribable to the age of the dwelling unit and the neighbourhood, the time when the unit was last painted, the presence of pets and the type of space heating. In addition, car exhausts seems to be a possible source of contamination.  相似文献

Adult male Neanthes arenaceodentata were exposed to 0.10±0.015 mg l-1 copper in the seawater of a continuous-flow bioassay system in the presence of a sand, a mud, a mixture of the sand and mud, and no sediment, to assess the influence of sediment type on Cu-induced mortality. The sediment type did influence mortality. The time to 50% mortality was 7.8 days without sediment, 36.5 days with sand, 54.5 days with the mixture, and 50.0 days with mud. There was no threshold Cu body burden that caused death. The mean Cu concentration per gram of N. arenaceodentata (24 h after death) without sediment was 270 g, in sand 994 g, in the mixture 1047 g, and in mud 1464 g. The differences in the toxic responses are discussed.  相似文献

The distribution of cyanobacteria in the surface waters of the North Sea was measured during July 1987. Numbers of cyanobacteria ranged from 2.5x106 to 1.7x108 cells 1-1. In the majority of stations, cyanobacterial numbers were highest in the near-surface water and a subsurface maximum was found at only one station. The distribution of 14C among the end-products of photosynthesis was determined for picoplankton (<1 m) and other phytoplankton >1 m throughout the North Sea. The majority of label was found in the protein fraction of both picoplankton and >1 m phytoplankton; incorporation into lipids and polysaccharides plus nucleic acids was much lower. We interpret the large incorporation into protein to be a consequence of nutrient limitation of these natural assemblages. Photosynthetic parameters of the two size fractions were also determined. Assimilation number (P m B ) and initial slope were greater for the picoplankton fraction than for phytoplankton >1 m but there was no evidence of significant photoinhibition of either fraction at irradiances up to 1 000 E m-2 s-1.  相似文献

Growth characteristics and nutrient uptake kinetics were determined for zooxanthellae (Gymnodinium microadriaticum) in laboratory culture. The maximum specific growth rate (max) was 0.35 d-1 at 27 °C, 12 hL:12 hD cycle, 45 E m-2 s-1. Anmmonium and nitrate uptake by G. microadriaticum in distinct growth phases exhibited Michaelis-Menten kinetics. Ammonium half-saturation constants (Ks) ranged from 0.4 to 2.0 M; those for nitrate ranged from 0.5 to 0.8 M. Ammonium maximum specific uptake rates (Vmax) (0.75 to 1.74 d-1) exceeded those for nitrate (0.14 to 0.39 d-1) and were much greater than the maximum specific growth rate (0.35 d-1), suggesting that ammonium is the more significant N source for cultured zooxanthellae. Ammonium and nitrate Vmax values compare with those reported from freshly isolated zooxanthellae. Light enhanced ammonium and nitrate uptake; ammonium inhibited nitrate uptake which was not reported for freshly isolated zooxanthellae, suggesting that physiological differences exist between the two. The shortterm (10–22 d) effect of Zn, Hg, Cu, Cd, Pb, and Ni on the length growth of Mytilus edulis is studied. Significant reductions of growth rate was found at 0.3 g Hgl-1, 3 g Cul-1, 10 g Znl-1, and 10 g Cdl-1 added to the local sea water, while concentrations of up to 200 gl-1 of Pb and Ni had no effect on the growth. With exposure to Cu and Zn, there was a linear reduction in growth rate with increasing metal concentration up to about 6 g Cul-1 and 100 g Znl-1. Above these levels, growth stopped with Cu, while with Zn it was stabilized at about 20% of control growth. When Hg and Cd were added, a curvilinear relationship between growth and metal concentration is indicated. With Hg, growth rate is nearly zero above 3–4 g Hgl-1, while the growth rate was 50% of control after 10 d of exposure to 100 g Cdl-1. At 2 g Cdl-1 there was a significant stimulation of length increase. Observed EC50-values for growth were 0.3–0.4 g Hgl-1, 3–4 g Cul-1, 60 g Znl-1, and 100 g Cdl-1.  相似文献

The reef coral Pocillopora damicornis (Linnaeus) was grown for 8 wk in four nutrient treatments: control, consisting of ambient, unfiltered Kaneohe Bay seawater [dissolved inorganic nitrogen (DIN, 1.0 M) and dissolved inorganic phosphate (DIP, 0.3 M)]; nitrogen enrichment (15 M DIN as ammonium); phosphorus enrichment (1.2 M DIP as inorganic phosphate); and 15 M DIN+1.2 M DIP. Analyses of zooxanthellae for C, N, P and chlorophyll a after the 8 wk experiment indicated that DIN enrichment increased the cellular chlorophyll a and excess nitrogen fraction of the algae, but did not affect C cell-1. DIP enrichment decreased both C and P cell-1, but the decrease was proportionally less for C cell-1. the response of cellular P to both DIN and DIP enrichment appeared to be in the same direction and could not be explained as a primary effect of external nutrient enrichment. The observed response of cellular P might be a consequence of in situ CO2 limitation. DIN enrichment could increase the CO2 (aq) demand by increasing the net production per unit area. Particulate organic carbon (POC) and nitrogen in sea water were measured in samples collected in the adjacent seas of the Pacific Ocean during the cruises of T. S. Oshoro-Maru (1969, 1970) and the R. V. Hakuho-Maru (KH-70-4, KH-72-1). High values were obtained in the northern North Pacific and the Bering Sea, the concentration of particulate carbon in the upper 50-m layer ranged from 35 to 550 g Cl-1. In the deep waters of these area, values above 50 g Cl-1 were frequently observed. The lowest values in the surface layer and deeper layers were obtained in the Japan Sea (23 gCl-1) and in the South China Sea (7 g Cl-1) respectively. A consistent minimum was located in the intermediate waters (100–400 m) throughout the entire region studied. Variation with depth was generally irregular with marked peak values in different layers. The POC distribution consited of these peak values and a relatively uniform background concentration. These background values slightly decreased with increasing depth and were different locally. Correlation analysis between carbon concentration and apparent oxygen utilization (AOU) of ambient water for the samples in the Japan Sea and the Sulu Sea showed that there was no systematic decrease of particulate carbon with increasing AOU. In these areas, the carbon concentration scattered in the higher AOU domain ranged from 10–100 g Cl-1. These observations support the conception that downward transport of particulate matter from the overlying surface layer in the adjacent seas of the Pacific Ocean may be fairly rapid.  相似文献

Ultrastructural evidence is presented of a chromatophoresystem in the zooxanthellae containing hermatypic, deep-water coral Leptoseris fragilis (Milne Edwards and Haime). It consists of multilobed cells which mainly occupy the intercellular space of the oral gastrodermis. The cellular processes are filled with electron-dense granules up to 1-m-long and 0.5-m-wide. Within the cytoplasm an elaborate system of microtubules is established. The ramifications of the pigment cells, containing the pigment granules, form a dense and nearly continuous layer close to the overlying zooxanthellae. It is speculated that host pigments may transform the violet portion of the incident light into longer wavelengths, thus increasing the photosynthetic efficiency of the zooxanthellae.  相似文献

The effect of ionizing radiation on the iron-linked protein (haemoglobin) of the marine lamellibranchs Anadara granosa (Linn.) and Cardita antiquata (Lam.) from Bombay waters, India is discussed. Purified haemoglobin solutions were exposed to a 60Cobalt source delivering a dose of about 4,600 rad/min, at the sample irradiation point. Radiation damage or degradation was measured spectrophometrically by studying changes in the absorption spectra following irradiation in the presence and absence of oxygen. Exposure to ionizing radiation in general caused a decrease in absorption of both haemoglobins, irrespective of location, viz extra-versus intracellular, at Soret (412 m), (540 to 42 m) and (574 to 76 m) peaks, and an increase in absorption at 510 and 630 m. Upon exposure to a higher dose, O2Hb of C. antiquata showed an increase in absorption at the protein peak (280 m); exposure to a lower dose, however, resulted in decreased absorption. Furthermore, the changes following irradiation were dependent upon the initial state of the pigment. Oxyhaemoglobin, when exposed to radiation, oxidized to hemiglobin, and hemiglobin reduced to oxyhaemoglobin. The extracellular haemoglobin of high molecular weight (3x106) of the false cockle C. antiquata was found to be extremely radio-resistant, whereas intracellular haemoglobin of low molecular weight (74,000) of the arcid clam A. granosa was highly radiosensitive, since it could not be exposed to doses exceeding 18,000 r.  相似文献

The effect of phosphate on the giant clam Tridacna gigas and on its symbiotic dinoflagellate Symbiodinium sp. was compared with that on cultured Symbiodinium sp. originally isolated from the same clarn species. Incubation of whole clams in elevated phosphate (10 M) reduced their capacity for phosphate uptake, but the uptake capacity of the clam's zooxanthellae population was not influenced. In addition, there was no change in the zooxanthellae density and the N:P ratio, of these algae., On the other hand, cultured zooxanthellae were influenced by the phosphate regimen of their culture medium. Compared with controls (0 M P), addition of 10 M phosphate to the culture medium caused an increase of 100% in cell density and decreases of 50% in the N:P ratio, and 80% in the phosphate-uptake capacity of the zooxanthellae. Zooxanthellae freshly isolated from the clams exhibited properties similar to those of zooxanthellae cultured in the absence of phosphate. These results demonstrate that the zooxanthellae population of T. gigas have limited access to the inorganic phosphate in sea water and the phosphate reserves within the animal host.  相似文献

The photosynthetic characteristics of prokaryotic phycoerythrin-rich populations of cyanobacteriaSynechococcus spp. and larger eukaryotic algae were compared at a neritic frontal station (Pl), in a warm-core eddy (P2), and at Wilkinson's Basin (P3) during a cruise in the Northwest Atlantic Ocean in the summer of 1984.Synechococcus spp. numerically dominated the 0.6 to 1 m fraction, and to a lesser extent the 1 to 5 m size fractions, at most depths at all stations. At P2 and P3, all three size categories of phytoplankton (0.6 to 1 m, 1 to 5 m, and >5 m) exhibited similar depth-dependent chages in both the timing and amplitude of diurnal periodicities of chlorophyllbased and cell-based photosynthetic capacity. Midday maxima in photosynthesis were observed in the upper watercolumn which damped-out in all size fractions sampled just below the thermocline. For all size fractions sampled near the bottom of the euphotic zone, the highest photosynthetic capacity was observed at dawn. At all depths, theSynechococcus spp.-dominated size fractions had lower assimilation rates than larger phytoplankton size fractions. This observation takes exception with the view that there is an inverse size-dependency in algal photosynthesis. Results also indicated that the size-specific contribution to potential primary production in surface waters did not vary appreciably over the day. However, estimates of the percent contribution ofSynechococcus spp. to total primary productivity in surface waters at the neritic front were significantly higher when derived from short-term incubator measurements of photosynthetic capacity rather than from dawn-to-duskin situ measurements of carbon fixation. The discrepancy was not due to photoinhibitory effects on photosynthesis, but appeared to reflect increased selective grazing pressure onSynechococcus spp. in dawn-to-dusk samples. Low-light photoadaptation was evident in analyses of the depth-dependency ofP-I parameters (photosynthetic capacity,P max; light-limited slope, alpha;P max alpha,I k ; light-intensity beyond which photoinhibition occurs,I b ) of the > 0.6 m communities at all three stations and was attributable to stratification of the water column. There was a decrease in assimilation rates andI k with depth that was associated with increases in light-limited rates of photosynthesis. No midday photoinhibition ofP max orI b was observed in any surface station. Marked photoinhibition was detected only in the chlorophyll maximum at the neritic front and below the surface mixed-layer at Wilkinson's Basin, where susceptibility to photoinhibition increased with the depth of the collected sample. The 0.6 to 1 m fraction always had lower light requirements for light-saturated photosynthesis than the > 5 m size fraction within the same sample. Saturation intensities for the 1 to 5 m and 0.6 to 1 m size fractions were more similar whenSynechococcus spp. abundances were high in the 1 to 5 m fraction. The > 5 m fraction appeared to be the prime contributor to photoinhibitory features displayed in mixed samples (> 0.6 m) taken from the chlorophyll maxima. InSynechococcus spp.-dominated 0.6 to 1 and 1 to 5 m size fractions, cellular chlorophylla content increased 50- to 100-fold with depth and could be related to increases in maximum daytime rates of cellularP max at the base of the euphotic zone. Furthermore, the 0.6 to 1 m and > 5 m fractions sampled at the chlorophyll maximum in the warm-core eddy had lower light requirements for photosynthesis than comparable surface samples from the same station. Results suggest that photoadaptation in natural populations ofSynechococcus spp. is accomplished primarily by changing photosynthetic unit number, occuring in conjuction with other accommodations in the efficiency of photosynthetic light reactions.  相似文献

Female Penaeus esculentus Haswell were collected by 15 to 20 min duration trawls during 1990. Carotenoids were analysed in the digestive gland, abdominal muscle, the remainder of the body (hereafter called integument) and ovary of prawns in Stage 2 through Stage 4 (fully mature) of maturation. The only oxycarotenoids (xanthophylls) identified were astaxanthins or astaxanthin esters; occasionally low levels of -carotene were detected in the digestive gland. The concentrations of astaxanthin monoesters (AM) and diesters (AD) were highest, with only minor amounts of free astaxanthins (Ast), except in the maturing ovaries, where free astaxanthins predominated (up to 80% of the total carotenoid). Of the total carotenoid, 82 to 94% was in the integument, but at maturity the digestive gland contained 10.7±3.4% and the ovary 5.6±0.9% of the total carotenoid. Only the ovary increased in mass during maturation, reaching up to 5.2% of total prawn mass. During this period, digestive gland concentrations of AM, AD and Ast all increased (tota 20 to 120 g g-1); levels in the muscle and integument varied little throughout maturation (total 0.4 and 100 g g-1, respectively); ovary AM levels remained low throughout (1.5 to 1.2 g g-1), AD increased from only 2 to 5 g g-1, but Ast increased from 2 to 34 g g-1. Apart from the ovary, AM concentrations were the most variable. In common with other decapod Crustacea, the maturing ovary of P. esculentus contained high levels of carotenoids, indicating that these may have an important role in early development. The natural diet of P. esculentus includes a variety of carotenoids, but except for a little -carotene, the digestive gland, where absorption occurs, contained astaxanthins, with only an occasional trace of -carotene. This suggests that the conversion of dietary carotenoids to astaxanthin occurs soon after ingestion.  相似文献

Past mining activities in the county of Cornwall in the United Kingdom, has exposed the local environment to raised levels of As. Previous research has shown an increased concentration of As in crops grown in such areas and the subsequent take-up of As through the food chain may lead to an increased body burden in man. In an attempt to evaluate this, a pilot study was carried out using Particle Induced X-ray Emission to compare the hair As concentration of a group of Cornish residents to that of a control group consisting of residents in the counties of Oxfordshire and Wiltshire. The null hypothesis being that there is no significant difference between the hair As concentrations of the two groups.Each group comprised thirty six age and sex matched subjects. The mean hair As concentration for the Cornish group was 2.51 g g-1, which is significantly greater than that of 0.7 g g-1 found in the control group (p = 0.004). The World Health Organisation suggests a hair As value greater than 1 g g-1 as evidence of increased exposure to As and 5 g g-1 is quoted by the Canadian Government as evidence of significant ingestion of As. Twenty one Cornish subjects registered a value at or above 1 g As g-1 compared to nine in the control group (p = 0.0003). Five subjects in the Cornish group had values that exceed 5 g As g-1, compared to only two in the control group (p = 0.23). We use the results of this study as evidence to support our continued work in this area in order to evaluate possible health implications.  相似文献
