Emerging evidence showed that 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) could induce expression of certain reactivation-associated genes in astrocytes, however, the consequent cellular effects and molecular mechanisms are still unclear. During the process of astrocyte reactivation, migration is a critical cellular event. In the present study, we employed wound-healing assay and Transwell® motility assay to explore the effects of TCDD on cell migration in primary cultured rat cortical astrocytes. We found that upon TCDD treatments at relative low concentrations (10? 10 and/or 10? 9?mol/L), the ability of primary astrocytes to migrate horizontally and vertically was promoted. In line with this cellular effect, the mRNA expression of two pro-migratory genes, including cell division cycle 42 (CDC42) and matrix metalloproteinase 2 (MMP2) was induced by TCDD treatment. Dioxin exerts its toxic effects mainly through aryl hydrocarbon receptor (AhR) pathway. So the role of AhR pathway in the pro-migratory effects of TCDD was examined using an AhR antagonist, CH223191. We found that application of CH223191 significantly reversed the pro-migratory effects of TCDD. Interestingly, the basal ability of horizontal migration as well as basal levels of CDC42 and MMP2 expression were dramatically reduced suggesting a possible physiological role of AhR in maintaining the endogenous migration ability of the primary astrocytes. These findings support the notion that dioxin promotes astrocyte reactivation at molecular and cellular levels.  相似文献

枯草芽孢杆菌对土臭素和2-甲基异冰片的降解动力学特性   总被引:1,自引:1,他引:0  
土臭素(geosmin,GSM)和2-甲基异冰片(2-methylisoborneol,2-MIB)是2种普遍存在于养殖水体中的土腥异味化合物,微生物降解是去除这两种物质的有效途径.本文研究了枯草芽孢杆菌(Bacillus subtilis)降解GSM和2-MIB的动力学特性.结果表明,枯草芽孢杆菌对GSM(1 000~2 000 ng·L-1)和2-MIB(1 000~2 000 ng·L-1)具有较好的降解性能,降解率均达90%以上;枯草芽孢杆菌对这些异味化合物的降解符合伪一级反应动力学,其生物降解速率常数(K)范围为0.14~0.41,降解速率不随GSM和MIB初始浓度的变化而变化;枯草芽孢杆菌对GSM降解的最大比生长速率umax为0.311,Monod常数KS为1.73,而在降解MIB过程中枯草芽孢杆菌的生长不符合Monod方程(R2=0.781).  相似文献

在五氯苯酚(PCP)与邻氯苯酚(2-CP)和2,4-二氯苯酚(2,4-DCP)3种物质单一毒性试验结果的基础上,按照等毒性单位设计,采用相加指数法,对五氯苯酚(PCP)与邻氯苯酚(2-CP)或2,4-二氯苯酚(2,4-DCP)以及这3种物质共存对斑马鱼(Brachydanio rerio)的联合毒性进行了试验研究.结果表明,邻氯苯酚(2-CP)和2,4-二氯苯酚(2,4-DCP)与五氯苯酚的联合毒性效应均表现为协同作用,使五氯苯酚的毒性剧增.由于邻氯苯酚或2,4-二氯苯酚的存在以及3种物质共存,使五氯苯酚对斑马鱼的48 h LC50由原来的0.102 mg/L,分别下降到0.031,0.029及0.022 mg/L.因此可见,水体污染评价目前只考虑单一毒物的影响还远远不够,由此制定的控制标准对水体的保护不利,必须从联合毒性的角度对水质污染进行综合评价.   相似文献

纳米TiO2对短裸甲藻的毒性效应   总被引:3,自引:2,他引:1  
为了揭示纳米TiO2对藻类的毒性作用和机制,研究了纳米TiO2对短裸甲藻的抑制特性及对酶活性、氧自由基等生理指标的影响.结果表明,纳米TiO2对短裸甲藻的生长有抑制作用,72 h半数致死浓度(EC50)为9.7 mg.L-1.在受试范围内随着纳米TiO2浓度的升高,超氧化物歧化酶(SOD)的活性显著性下降(P<0.05),随TiO2浓度增加,羟自由基的含量和过氧化氢酶(CAT)的活性显著性增加(P<0.05),超氧阴离子自由基含量也呈现升高的趋势.对照组羟自由基的含量是0.083U.mL-1,而在30 mg.L-1的纳米TiO2作用下,羟自由基的含量上升到1.1 U.mL-1.在20 mg.L-1纳米TiO2作用下,随着时间的延长,SOD活性、CAT活性和MDA(丙二醛)在暴露时间为12 h时达到最大,而48 h时降低到最小.12 h SOD的活性是0.14U.(107cell.min)-1,而48 h SOD的活性为0.01 U.(107cell.min)-1,而羟自由基只在48 h处呈显著性上升(P<0.05).纳米TiO2对抗氧化体系和自由基的影响可能与其抑藻机制有关.本研究为揭示纳米材料的环境生态效应提供了基础.  相似文献

阿散酸(p-arsanilic acid,ASA)是一种重要的有机砷化合物,较无机砷更加难以去除,目前关于去除水中ASA研究少有报道.本研究主要评价了TiO_2光催化剂(P25)对ASA的去除效果,考察了P25投量、pH和光照强度对其光催化氧化ASA的影响,探究了ASA光降解产物和主要机制.结果表明,在P25催化作用下,模拟自然光可氧化降解ASA为As(V),最终通过P25对As(V)的吸附作用将水中砷有效去除.当ASA初始浓度为2 mg·L~(-1),P25投量为1 g·L~(-1)时,光催化氧化-吸附0.5 h后,水中剩余砷的浓度约为0.34 mg·L~(-1).酸性条件下ASA的去除率远远高于碱性条件,最佳光照强度为68.5 mW·cm~(-2).P25光催化氧化ASA过程中羟基自由基起到了主要作用.  相似文献

应用阳离子交换树脂从活性污泥中提取了胞外聚合物(EPS),考察了EPS、Ca2 及pH值对生物絮凝作用的影响.结果表明,当pH≥7.5, EPS≥70mg/L,[Ca2 ]≥300mg/L时,有利于生物絮体的形成.通过正交试验,得出pH值、EPS和[Ca2 ]对生物絮体形成综合影响的回归方程: h浊度= -901.70 97.35pH 1.96[Ca2 ] 9.08EPS-0.11pH[Ca2 ]-0.91pHEPS-0.02[Ca2 ]EPS 0.0021pH[Ca2 ]EPS.回归分析表明,pH值对生物絮体形成影响最显著,其次是EPS和Ca2 .  相似文献

双酚A(BPA)普遍应用在工业制成品中,因需求量递增而产生的环境安全问题引发各方关注.与BPA对植物生长影响的研究工作相比,BPA影响植物生长的机理的研究甚少.光合作用是植物生产的重要生理过程,可通过叶绿素荧光测定技术探测和分析.基于此,本文利用叶绿素荧光测定技术,研究了BPA对大豆、玉米和水稻幼苗叶绿素荧光反应的影响.结果表明,3.0 mg·L-1BPA可降低大豆和玉米初始荧光F0,增加原初光能转化效率F v/F m、实际光能转化效率ΦPSⅡ、电子传递速率ETR、光化学猝灭系数qP和非光化学猝灭系数qN,对水稻各荧光参数无影响,即3.0 mg·L-1BPA可改善大豆和玉米光合PSⅡ系统,增强光能吸收,提高光合电子传递和光能转化效率;6.0 mg·L-1BPA可增加大豆和水稻的ΦPSⅡ、ETR和qP,降低F0,除增加qN外不影响玉米的其它荧光参数,表明6.0 mg·L-1BPA能改善大豆和水稻光合PSⅡ系统,提高光能转化和电子传递效率;除10 mg·L-1BPA对水稻各荧光参数无显著影响及17.2 mg·L-1BPA增加玉米和水稻的qN外,10 mg·L-1和17.2mg·L-1BPA可增加3种作物F0,抑制其它各荧光参数,即高剂量BPA引起作物光抑制,PSⅡ中心受损,光能转化和电子传递效率降低.此外,对比3种作物荧光参数变幅可知,BPA对大豆各荧光参数的影响玉米水稻.总之,BPA对3种作物叶绿素荧光反应的影响在方式和效果上存在差异.  相似文献

以石油污染盐碱土壤为研究对象,利用磷脂脂肪酸(PLFAs)活性微生物标记法,分析紫茉莉(Mirabilis jalapa Linn.)根际土壤微生物群落结构的动态变化,探讨紫茉莉生长对根际土壤微生物与石油烃(TPH)降解的影响.结果表明,供试土壤中先后出现了22种微生物PLFAs,包括标识细菌的饱和脂肪酸(SAT)、标识革兰氏阳性菌(G+)的末端支链型饱和脂肪酸(TBSAT)、标识革兰氏阴性菌(G-)的单不饱和脂肪酸(MONO)和环丙脂肪酸(CYCLO)、标识真菌的多不饱和脂肪酸(PUFA)和标识放线菌的中间型支链型饱和脂肪酸(MBSAT)等六大类型.与未种紫茉莉土壤(CK)相比,根际土壤微生物PLFAs种类变异率在春、夏、秋季分别为71.4%、69.2%和33.3%;TPH降解率在春、夏、秋季分别提高了47.6%、28.3%、18.9%.相关性分析表明,石油烃的降解在CK土壤中与77.8%的PLFAs具有正相关关系(r0),55.6%的种类具有高度正相关关系(r≥0.8),其中,与SAT和MONO类群的相对含量正相关,相关系数分别为0.92、0.60;根际土壤中仅与42.1%的PLFAs正相关,21.1%的种类高度正相关,与TBSAT、MONO和CYCLO类群的相对含量正相关,相关系数分别为0.56、0.50、0.07.说明紫茉莉生长对根际土壤微生物群落结构及TPH降解速率均具有较大影响,且随生长季节的不同而有很大差异.该结果将为开展石油污染盐碱土壤的植物修复技术研究提供理论依据.  相似文献

利用传统细菌计数方法及分子生物学方法对新近发展起来的生物造粒流化床反应器与A2/O工艺的微生物特性进行比较研究.反应器中好氧菌、反硝化菌及反硫化菌的计数结果显示,单位质量的活性污泥中,生物造粒流化床中好氧菌的分布数是A2/O工艺好氧区的1~2倍,反硝化菌总数介于A2/O工艺的好氧区和缺氧区之间,而反硫化菌的总数介于A2/O工艺的缺氧区和厌氧区之间.用PCR-DGGE技术对生物造粒流化床反应器及A2/O工艺中的微生物进行多样性分析,结果显示,生物造粒流化床反应器的微生物群落比A2/O工艺丰富;DGGE指纹图谱聚类分析表明,生物造粒流化床反应器与A2/O工艺的微生物群落相似性为57.6%.  相似文献

Tri(2-chloroethyl) phosphate (TCEP) with the initial concentration of 5 mg/L was degraded by UV/H2O2 oxidation process. The removal rate of TCEP in the UV/H2O2 system was 89.1% with the production of Cl? and PO43? of 0.23 and 0.64 mg/L. The removal rate of total organic carbon of the reaction was 48.8% and the pH reached 3.3 after the reaction. The oxidative degradation process of TCEP in the UV/H2O2 system obeyed the first order kinetic reaction with the apparent rate constant of 0.0025 min?1 (R2=0.9788). The intermediate products were isolated and identified by gas chromatography-mass spectrometer. The addition reaction of HO? and H2O and the oxidation reaction with H2O2 were found during the degradation pathway of 5 mg/L TCEP in the UV/H2O2 system. For the first time, environment risk was estimated via the “ecological structure activity relationships” program and acute and chronic toxicity changes of intermediate products were pointed out. The luminescence inhibition rate of photobacterium was used to evaluate the acute toxicity of intermediate products. The results showed that the toxicity of the intermediate products increased with the increase of reaction time, which may be due to the production of chlorine compounds. Some measures should be introduced to the UV/H2O2 system to remove the highly toxic Cl-containing compounds, such as a nanofiltration or reverse osmosis unit.  相似文献

Food, especially animal origin food is the main source of polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs), and dioxin-like polychlorinated biphenyls (dl-PCBs) for human exposure. So, a simple, rapid and cheap bioassay method is needed for determination of dioxins in food samples. In this study, we used a new highly sensitive reporter cell line to determine the concentration of dioxins in 33 fish and seafood samples. The samples were extracted by shaking with water/isopropanol (1:1 v/v) and hexane and cleaned-up by a multi layered silica gel column and an alumina column, then analyzed using CBG 2.8D cell line. We compared the results obtained from the CBG 2.8D cell assay to those obtained from conventional High-Resolution Gas Chromatography–High Resolution Mass Spectrometry (HRGC–HRMS) analysis. Good correlations were observed between these two methods (r2=0.93). While the slope of regression line was 1.76, the bioanalytical equivalent (BEQ) values were 1.76 folds higher than WHO-TEQ values and the conversion coefficient was 0.568 (the reciprocal of 1.76). In conclusion, CBG 2.8D cell assay was an applicable method to determine dioxins levels in fish and sea food samples.  相似文献

3-羟基-3-甲基戊二酰辅酶A还原酶(3-hydroxy-3-methylglutaryl coenzyme A reductase,HMGR)是甲羟戊酸(mevalonic acid,MVA)途径中的第一个关键限速酶.克隆稀有鮈鲫HMGR基因全长,分析该基因在不同组织及雄鱼经不同浓度五氯酚(pentachlorophenol,PCP)暴露的表达差异,探索PCP对类固醇激素前体(胆固醇)合成水平基因转录调控的内分泌干扰机制.采用同源克隆策略及cDNA末端快速扩增技术(rapid-amplification of cDNA ends,RACE),从稀有鮈鲫肝脏中首次克隆得到3 101碱基(bp)的HMGR基因cDNA全长,基因命名为GrHMGR(GenBank accession No:KF885724).GrHMGR编码884个氨基酸,系统发育树分析表明,GrHMGR编码的蛋白与其他鱼类来源的HMGR氨基酸序列同源性较高.Real-time PCR分析表明GrHMGR的表达有组织特异性,在大脑,性腺和肝脏中均有表达,性腺中表达量最高.稀有鮈鲫雄鱼经不同浓度PCP暴露后,GrHMGR在大脑和性腺的表达随PCP浓度的增加显著下降,然而在肝脏组织中表达趋势有所不同.HMGR的表达下降会减少胆固醇的合成.说明PCP暴露会通过干扰稀有鮈鲫的胆固醇合成途径,进而对内分泌系统产生影响.  相似文献

四溴双酚A(TBBPA)是世界上产量和用量最大的溴代阻燃剂之一,在河流、底泥及污水中持久存在,严重威胁环境和人体健康.探讨了阳极为Ti/RuO_2-IrO_2电极的电化学法对溶液中TBBPA的降解效率,并考察了电流密度(5~20 m A·cm~(-2))、初始pH值(3~11)、极板间距(1~3 cm)及电解质浓度(1~40 mmol·L~(-1))对降解速率的影响.研究结果表明,TBBPA的降解遵循一级动力学方程,在溶液初始浓度为5 mg·L~(-1)、电流密度为10 mA·cm~(-2)、电解质浓度为5 mmol·L~(-1)及极板间距为1 cm的条件下,60 min后TBBPA的降解率可达97.2%,且降解速率常数和半衰期分别为0.060 min~(-1)和11.6 min,能量消耗为105.3 k Wh·m~(-3).同时运用HPLC-MS/MS检测出中间产物双酚A(BPA)和2-溴苯酚,并推断其降解途径主要为在羟基自由基(·OH)作用下连续的脱溴过程中,苯环与异丙基之间的C—C键断裂生成BPA和2-溴苯酚.电化学法是一种高效的、有潜力的降解难降解有机污染物的方法,本文将为今后实际废污水中TBBPA的去除提供基础数据和技术参数.  相似文献

多溴联苯醚(PBDEs)是广泛存在于环境中的一种新型持久性有机污染物.四溴联苯醚同分异构体中的BDE-47是多溴联苯醚中最重要的单体之一.试验采用人工土壤培养法,通过亚急性试验,研究了在不同暴露时间阶段下,不同BDE-47剂量对赤子爱胜蚓(Eisenia fetida)抗氧化酶(过氧化氢酶CAT)、代谢酶(谷胱甘肽转移酶GST)以及二者基因表达的影响.结果表明,在暴露14 d和28 d时,CAT活性被诱导上升并且差异显著;GST活性变化差异不显著;CAT基因表达水平在第14 d时呈现抑制效应,在后续的第28 d和第42 d基因表达水平上调;GST基因表达水平整体呈现诱导效应,并且差异显著,随着暴露时间的增加,低毒处理组(10、50 mg·kg-1)的基因表达水平逐渐下调至低于对照组的水平,高毒处理组(100、200 mg·kg-1)的基因表达量仍高于对照组水平;在BDE-47的暴露试验中,CAT与GST活性及其基因表达水平两项指标对低毒处理组较高毒处理组更为敏感.  相似文献

目的提高La_2Ce_2O_7(LC)陶瓷材料的断裂韧性。方法采用固相反应方法合成Y_2O_3稳定ZrO_2(YSZ)掺杂的La_2Ce_2O_7(LC)陶瓷材料(LCZ),研究LCZ块材的弹性模量、断裂韧性等力学性能,以及热膨胀系数、热传导率等热物理性能。结果 10%YSZ摩尔分数掺杂的LCZ块材的断裂韧性约为1.4 MPa·m1/2,比LC提高了近10%。YSZ掺杂有效地抑制了LC在200~400℃温度区间热膨胀系数下降的现象。在200~1200℃温度范围内,LCZ块材的热膨胀系数为10×10-6~12×10-6K-1,在1200℃的热导率约为0.75W/(m·K),比YSZ降低了50%以上,显示了优异的隔热性能。结论 YSZ的掺杂使LC中部分Zr4+取代了La3+,降低了晶格中的O空位浓度,原子的横向剪切运动被削弱,有效地抑制了LC在200~400℃温度区间热膨胀系数下降的现象。由于t-ZrO_2→m-ZrO_2的相变对微裂纹的愈合作用,10%YSZ掺杂使LC块材的断裂韧性得到有效提高。  相似文献

根据2005年9月黄河口淡-咸水混合过程中表层水体CO2分压(pCO2)和盐度的同步观测资料,对黄河口淡-咸水混合过程中pCO2的变化规律进行了初步探讨.研究结果表明:黄河口pCO2与盐度总体上保持着良好的负相关关系,但在盐度5左右水体pCO2存在亏损现象,这种现象可能是由于河口水体受海水影响,其离子强度突然增加而导致黄河水体中的过饱和碳酸盐发生沉降作用引起的.  相似文献

Atmospheric CO2 concentration (Ca) is rising, predicted to cause global warming, and alter precipitation patterns. During 1994, spring barley (Hordeum vulgare L. cv. Alexis) was grown in a strip-split-plot experimental design to determine the effects that the main plot Ca treatments [A: Ambient at 370 μmol (CO2) mol−1; E: Enriched with free-air CO2 enrichment (FACE) at ∼550 μmol (CO2) mol−1] had on several gas exchange properties of fully expanded sunlit primary leaves. The interacting strip-split-plot irrigation treatments were Dry or Wet [50% (D) or 100% (W) replacement of potential evapotranspiration] at ample nitrogen (261 kg N ha−1) and phosphorous (29 kg P ha−1) fertility. Elevated Ca facilitated drought avoidance by reducing stomatal conductance (gs) by 34% that conserved water and enabled stomata to remain open for a longer period into a drought. This resulted in a 28% reduction in drought-induced midafternoon depression in net assimilation rate (A). Elevated Ca increased A by 37% under Dry and 23% under Wet. Any reduction in A under Wet conditions occurred because of nonstomatal limitations, whereas under Dry it occurred because of stomatal limitations. Elevated Ca increased the diurnal integral of A (A′) that resulted in an increase in the seasonal-long integral of A′ (A″) for barley leaves by 12% (P = 0.14) under both Dry and Wet - 650, 730, 905 and 1020 ± 65 g (C) m−2 y−1 for AD, ED, AW and EW treatments, respectively. Elevated Ca increased season-long average dry weight (DWS; crown, shoots) by 14% (P = 0.02), whereas deficit irrigation reduced DWS by 7% (P = 0.06), although these values may have been affected by a short but severe pea aphid [Acyrthosiphon pisum (Harris)] infestation. Hence, an elevated-Ca-based improvement in gas exchange properties enhanced growth of a barley crop.  相似文献
