In order to study the interaction of the extracellular and intracellular osmoregulatory systems of the bivalve Rangia cuneata, we have measured blood osmotic and ionic concentrations together with intracellular free amino acid concentrations and total tissue water under identical salinity conditions. Like freshwater bivalves, the blood of R. cuneata is maintained hyperosmotic (50 mOsm) to the environment in salinities below 110 mosm by the regulation of Na+, Cl-, K+ and Ca2+ concentrations. On the other hand in company with marine bivalves, R. cuneata also regulates intracellular free amino acids (FAA) as a mechanism to control cellular volume during osmotic stress over the entire non-lethal salinity range (3 to 620 mOsm). Alanine is the predominant intracellular osmotic effector. Thus, by utilizing the osmoregulatory mechanisms of both marine and freshwater bivalves, R. cuneata is able to tolerate salinities ranging from freshwater to 25 ppt and to traverse the faunal salinity boundary, known as the horohalinicum (5 to 8 ppt), controlling cell volume throughout.Please address requests for reprints to Dr. S. Rangia cuneata were collected every six weeks for one year from February 1984 to January 1985 at the mouth of the Dog River, Alabama, USA and dosed with 200 ppb [9–14C] anthracene at 10, 20, and 30°C. Uptake rates of [9–14C] anthracene measured over a 15-h period were not significantly different (P>0.05), but depuration rates measured over a two-week period were significantly higher with increasing temperature. The seasonal changes in rates were related to the annual changes in biochemical composition of the clams associated with the reproductive cycle. Lipid levels were nearly constant over the year, averaging 88.0±2.0 mg/g dry wt. The level of carbohydrates peaked in July, a month before the maximal gonad index, and decreased steadily through September during spawning. The level of proteins increased from 76.6 mg/g dry wt prior to spawning to 138.3 mg/g dry wt after spawning. Uptake rates were highest in late summer during the peak of gametogenesis and decreased during spawning in the fall. Depuration rates remained low from March through August and rapidly increased to the highest levels during the fall spawning period. The results suggest that the stage of the reproductive cycle, with its associated biochemical changes, and temperature play an important role in the pattern of accumulation and release of anthracene in R. cuneata.  相似文献

The aim of this study is to elucidate the osmoregulatory capabilities of the intertidal pulmonate Onchidium tumidium. Our results indicate that O. tumidium could tolerate hyperosmotic stress more effectively than hypoosmotic stress. In 10% seawater (SW), it was capable of maintaining its plasma hyperosmotic and hyperionic to the external medium. However, it rapidly gained weight in 10% SW, and no restoration of body weight was observed. In contrast, in 90% SW it was able to recover its body weight partially. In a hyperosmotic medium, accumulation of intracellular free amino acids (FAA) was responsible for cell volume regulation. The accumulated FAA might originate from protein catabolism, which occurred in the internal organs (IO). In contrast, the relatively smaller amination:deamination ratio of glutamate dehydrogenase in the body wall (BW) and IO of specimens exposed to hypoosmotic stress might lead to a higher rate of amino acid catabolism. This was supported by the greater ammonia contents in the BW and IO of, and higher rate of ammonia excretion by, specimens exposed to 10% SW. Received: 11 March 1998 / Accepted: 2 May 1999  相似文献

The regulation of muscle fiber K+ and free amino acid (FAA) concentrations during hypoosmotic stress was investigated in the moderately euryhaline crab Cancer irroratus. After 6 h of exposure to 60% ASW, muscle fiber K+ concentration declined from 185 mM to 140 mM. Following this, the blood glycine levels began to increase, indicating an FAA efflux from the cells. These data indicate that both muscle fiber K+ and FAA contribute to cell volume regulation in C. irroratus. The early release of K+ limits the initial rate of cell hydration. The subsequent efflux of glycine accounts for the volume regulation response of the muscle fibers. The cell volume regulatory system of C. irroratus is a coordinated use of both inorganic ions and FAA.  相似文献

Patterns of nine intracellular free amino acids (FAA), which are utilized as organic osmolytes for salinity-induced cell volume regulation in marine osmoconformers, were compared in five Macoma balthica populations and seven Mytilus spp. populations along their European distribution. Three types of FAA patterns were classified within both taxa: a northern Baltic type, a southern Baltic type and an Atlantic/Mediterranean type which mainly differ regarding the share of alanine and taurine. Differences are discussed in relation to habitat salinity and population genetics. Along a salinity gradient, the total size of the intracellular FAA pool did not differ between sympatric M. balthica and Mytilus spp., and was significantly correlated with habitat osmolality in a range from 70 to 600 mmol kg−1 H2O (oligohaline to mesohaline) in both bivalves. In M. balthica, this correlation was mainly based on significant correlations of alanine (15–100 mmol kg−1 DW), glycine (30–100 mmol kg−1 DW) and taurine (0–70 mmol kg−1 DW) with habitat osmolality. In Mytilus spp., only glycine (25–100 mmol kg−1 DW) and taurine (4–180 mmol kg−1 DW) were significantly correlated with habitat osmolality. The concentration of alanine was three times lower in Mytilus spp. than in M. balthica and did not correlate with habitat osmolality. Within a habitat osmolality range from 600 to 1,100 mmol kg−1 H2O (mesohaline to marine) the concentration of FAA remained constant in both taxa. It is suggested that under marine conditions additional organic osmolytes must become more important for cell volume regulation in Macoma and Mytilus.  相似文献

Although the taxonomy of deep-sea protobranch bivalves is becoming better known, relatively little information is available on their reproductive biology and whether or not populations show reproductive periodicities. We have examined the reproductive biology of three common sympatric species as part of a long-term time-series of samples taken from 2900 m in the Rockall Trough from 1973 to 1983. Malletia cuneata Jeffreys, 1876 produces a maximum of 30 oocytes at any one time and these grow to a maximum size of 240 m. Maximum fecundity of Ledella pustulosa (Jeffreys, 1876) and Yoldiella jeffreysi (Hidalgo, 1877) is 174 and 360, respectively, and both species produce an egg of 120 m in diameter. These data indicate lecithotrophic early development in L. pustulosa and Y. jeffreysi, but direct development in M. cuneata; however, evidence from the prodissoconch length of M. cuneata suggests lecithotrophic development. L. pustulosa and Y. jeffreysi also differ from M. cuneata in having a distinct reproductive cycle with spawnout in the winter months. Both the continuously breeding M. cuneata and the seasonally breeding L. pustulosa ingest diatoms, coccoliths and foraminiferans, but whereas the diet of M. cuneata appears to be constant throughout the year there is an apparent reduction in the feeding activity of L. pustulosa concomitant with the deposition of phytodetritus on the deep-sea bed.  相似文献

Glycine was the most abundant free amino acid (FAA) in abdominal muscle of grass shrimp Palaemonetes pugio, accounting for over 50% of the total pool. Arginine, alanine, proline, taurine and serine were also present at relatively high concentrations. Following transfer from 17%. to 2 and 32%. S, new steady-state levels of total FAA were observed at 72 h. Total FAA and the sum of glycine, alanine and proline exhibited a positive correlation with salinity. Exposure to Aroclor 1254 did not have appreciable effects on total FAA levels, indicating that disruption of intracellular osmoregulation was not a major consequence of PCB toxicity. However, changes in individual amino acid concentrations in exposed shrimp were reflective of an altered metabolic state. Glycine, which did not show changes immediately following exposure, underwent large decreases after transfer to PCB-free water and indicated a delayed effect of PCB exposure. A correlation between salinity and the sum of glycine, alanine and proline did not exist in exposed shrimp.  相似文献

 Free amino acid (FAA) and protein content were measured in various developmental stages of Artemia franciscana, from cysts to Instar III metanauplii. In addition, decapsulated cysts of 15 Artemia populations from different localities were compared with respect to their FAA and protein content. Furthermore, the content and composition of the FAA pool were modulated by hatching the cysts at various salinities, and by enriching the nauplii with algae or a lipid-enrichment emulsion. The FAA content increased threefold from cysts to nauplii, and Instar III metanauplii contained nearly 50% taurine of total FAA. Cysts of A. franciscana were found to contain one-third the amount of FAA compared to the other Artemia species investigated. The content and pool composition of FAA was successfully modulated in 11 of 13 populations, where by the content of FAA was significantly increased when hatched at high salinity. Finally, enrichment elevated the content of FAA and changed the pool composition, thereby showing a dietary effect. Algal enrichment also increased the protein content. Despite the wide range of morphological diversity among the Acropora (Isopora) colonies on Heron Island (Great Barrier Reef, Australia), only two reproductively isolated species were present from 1977 to 1982: A. cuneata (Dana, 1846) and A. palifera (Lamarck, 1816). Both species released planulae lacking zooxanthellae and were simultaneous hermaphrodites with testes and ovaries occurring on separate mesenteries within the same polyp. Oogenesis preceded spermatogenesis. Seasonal cycles of gametogenesis, embryogenesis and planulation occurred in the two species. Colonies of A. cuneata developed two cycles of gametes. One cycle matured near the first quarter moon in April and the other on the same lunar phase in June. Planulae release occurred from about September to December each year and was not correlated with lunar phase. Gametes of A. palifera ripened only once per annum, a few days after the last quarter moon in November, and planulation occurred from about January to March. Embryos were brooded in the coelenteron of the polyps in both species. Ova were fertilized in the mesenteries and embryos were retained within an envelope of mesoglea and gastrodermis, remaining attached to the mesentery by a stalk until the larvae matured and were released. A. palifera and A. cuneata were abundant in the unpredictable reef flat environment. However, their life-history traits, e.g. seasonal reproduction, delayed sexual maturity, large colony size and fairly long life span, were more specialised than had been predicted for this type of environment.  相似文献

Adaptive cluster sampling (ACS) is a targeting sampling method that provides unbiased abundance estimators for populations of rare species that may be inadequately sampled with simple random sampling (SRS). ACS has been used successfully to estimate abundances of rockfish and sardine larvae from shipboard surveys. In this study, we describe the application of ACS for subtidal macroalgae. Using SCUBA, we measured abundances of Codium mamillosum, C. pomoides, and Halimeda cuneata at three islands and two levels of wave exposure. The three species were relatively patchy and could be sampled with ACS at one site per dive. Their distributions differed among islands and with exposure to wave energy, with H. cuneata found at only one island. ACS is a useful tool for understanding the spatial distribution and abundance of populations of rare benthic species, but, as was the case in this study, may not be as efficient as sampling with SRS with comparable replication.  相似文献

Effects of food availability and season on the free amino acid (FAA) and total protein content of the copepod Calanus finmarchicus females were investigated in two mesocosm experiments on the Norwegian west coast in spring and autumn. Starved C. finmarchicus females showed no change in total FAA content, but the FAA pool composition changed drastically. During the first 10 days of starvation the protein content showed a moderate decline (<2 µg ind -1); however, during the following 21 days the total content was drastically reduced, from 63 to 9 µg ind -1. This supports the notion of a sequential catabolism of endogenous nutrients during starvation. In females at high food concentrations, the body protein content increased during spring, but decreased during autumn. The FAA pool composition of females differed between spring and autumn in 14 of the 18 FAA investigated. Reduced body protein content and increased proportion of essential free amino acid were observed during starvation. Similar changes were observed in females sampled at the end of the mesocosm experiments in the autumn. The results suggest that mature C. finmarchicus females are in a negative protein balance during autumn, despite high food concentrations, contributing to a lower fitness than in females maturing during the spring.  相似文献

Although the three examined species of Argyrotheca are quite common in the Mediterranean, little is known about their reproductive cycles. This study shows that Argyrotheca cordata and Argyrotheca cistellula have continuous breeding activity, indicated by the occurrence of ripe eggs and larvae in their brood pouches throughout the year. The absolute number of larvae found in the brood pouches of both species corresponds to the adult's size. The third species examined, Argyrotheca cuneata, tends to breed in autumn, with larvae only present in September and November, but the sample-size was small. Larval development is very similar in all three Argyrotheca species, each embryo going through a gastrula stage, a two-lobed and a three-lobed stage. Before leaving the brood pouch, the larvae have an apical lobe with a girdle of elongated cilia, a mantle lobe with a midventral band of cilia and a pedicle lobe without ciliation. The larvae of A. cordata and A. cuneata possess four bundles of larval setae, whereas A. cistellula shows no setae at all. Received: 6 November 1998 / Accepted: 15 March 1999  相似文献

Naturally spawned and fertilized eggs of Atlantic halibut,Hippoglossus hippoglossus L., were analysed for protein, free amino acids (FAA), ammonium ions and energy content. The chemical composition was found to be size-dependent but varied little during egg development. Ammonium ions did, however, accumulate during the late embryonic stage, and the trend in FAA content was downward during the same period. Rates of O2 uptake and NH3 excretion followed exponential patterns. A total of 1µmol O2 was consumed and 120 nmol NH3 excreted between the time intervals of fertilization and 1 d post hatch. We assessed the ontogenetic changes in protein content and free amino acids (FAA) in eggs and early larvae of Engraulis ringens (anchoveta) off central Chile on different dates during the spawning season. On all sampling dates, a reduction in embryonic yolk-sac volume, proteins and FAA concentrations occurred during development. Protein electrophoresis (SDS–PAGE) of eggs and larvae showed at least 22 protein bands: 11 were consumed early and not detected after hatching. The proportion of essential FAA (EFAA) was higher than the proportion of non-essential FAA (NEFAA) in early eggs and in 7 day-old larvae (82.5-73% EFAA respectively). During egg development, the FAA pool was dominated by leucine, alanine and lysine, three amino acids contributing 35–44% of the total FAA in eggs. During larval development, histidine was the most abundant FAA. In July, total FAA constituted 13–18% of the egg dry weight. A similar proportion (45–51%) occurred in July between protein plus FAA and total lipids. The differences in egg size during the spawning season along with variability in batch composition suggests that the female spawning condition is a major factor determining egg quality and early offspring success.  相似文献

The present paper studied the influence of different food regimes on the free amino acid (FAA) pool, the rate of protein turnover, the flux of amino acids, and their relation to growth of larval turbot (Scophthalmus maximus L.) from first feeding until metamorphosis. The amino acid profile of protein was stable during the larval period although some small, but significant, differences were found. Turbot larvae had proteins which were rich in leucine and aspartate, and poor in glutamate, suggesting a high leucine requirement. The profile of the FAA pool was highly variable and quite different from the amino acid profile in protein. The proportion of essential FAA decreased with development. High contents of free tyrosine and phenylalanine were found on Day 3, while free taurine was present at high levels throughout the experimental period. Larval growth rates were positively correlated with taurine levels, suggesting a dietary dependency for taurine and/or sulphur amino acids. Reduced growth rates in Artemia-fed larvae were associated with lower levels of free methionine, indicating that this diet is deficient in methionine for turbot larvae. Leucine might also be limiting turbot growth as the different diet organisms had lower levels of this amino acid in the free pool than was found in the larval protein. A previously presented model was used to describe the flux of amino acids in growing turbot larvae. The FAA pool was found to be small and variable. It was estimated that the daily dietary amino acid intake might be up to ten times the larval FAA pool. In addition, protein synthesis and protein degradation might daily remove and return, respectively, the equivalent of up to 20 and 10 times the size of the FAA pool. In an early phase (Day 11) high growth rates were associated with a relatively low protein turnover, while at a later stage (Day 17), a much higher turnover was observed. A study was undertaken to establish the role of free amino acids (FAA) in aerobic energy dissipation in embryos of turbot (Scophthalmus maximus) which contain an oil globule in the egg. Laboratory-reared developing eggs and larvae (15°C, 34 salinity) were measured for oxygen uptake, ammonia excretion, contents of FAA, protein, and ammonium, and volumes of yolksac and oil globule. Newly spawned eggs from different batches contained 55 to 90 nmol egg–1 of FAA. Resorption of FAA occurred in parallel with the consumption of yolk. Resorption of the oil globule, however, occurred predominantly after hatching and mainly after yolk resorption. The combined data suggest that approximately 70% of the FAA are utilized as an energy substrate, while the rest are polymerized into body proteins. FAA become a significant energy substrate in the early egg stage and account for 100% of the aerobic energy dissipation 2 d after Fertilization then decrease to ca. 60% at the time of hatching. Lipids derived from the oil globule seem to be the main fuel after hatching and account for ca. 90% of the energy dissipation at the onset of first-feeding. Thus, the energetics of fish embryos which contain an oil globule seems to be different from those that depend exclusively on the nutritional reserves of the yolk.  相似文献

In the larvae of many marine teleosts, the stomach is absent until they approach or attain metamorphosis. Consequently, the formation of chyme containing specific free amino acids from the gastric digestion of protein, which are believed to be signals initiating the release of the digestive hormone cholecystokinin (CCK), is lacking. CCK, when secreted into the blood circulation from specialized intestinal cells, stimulates gallbladder motility and is a key factor causing the release of pancreatic digestive enzymes into the gut lumen. Using first-feeding Atlantic herring larvae (Clupea harengus) as a model, the aim of the present study was to determine if a CCK response together with tryptic activity could be elicited in larvae ingesting dietary protein and/or FAA. Larvae were tube fed single lamellar liposome vesicles (SLV) containing: (1) physiological saline (PS), (2) bovine serum albumin (BSA), (3) specific free amino acids (FAA), or (4) a ratio (1:1) of BSA and FAA. The CCK and trypsin levels were then assayed (radio-immunoassay) at 0, 15, 60 and 120 min after tube feeding. A marked CCK response was elicited in all treatments compared to the PS control at 15 and 30 min and was significant (p<0.05) at 120 min after tube feeding. Larvae tube fed the FAA treatment exhibited CCK levels that increased linearly from 1.6 to 5.6 fmol mg-1 dry weight (DW) after 2 h of digestion, although this response was below the BSA and BSA:FAA treatments. The BSA and BSA:FAA treatments, after 15 min of digestion, showed a rapid CCK increase, over the PS and the FAA liposome treatments, to 8.1 and 5.4 fmol mg-1 DW, respectively. At the end of the assay, BSA and BSA:FAA demonstrated similar levels (10.2 and 9.2 fmol mg-1 DW, respectively). Larvae tube fed the PS control or the FAA liposome treatment did not demonstrate any appreciable increase in tryptic activity during the 2 h digestion period (0.03-0.071 and 0.03-0.048 mU mg-1 DW, respectively). In contrast, the BSA:FAA treatment increased from 0.03-0.148 mU mg-1 DW 1 h after feeding, which was significantly (p<0.05) higher than the PS and FAA liposomes, and then decreased markedly (0.085 mU mg-1 DW) after 2 h of digestion. The larvae tube fed BSA liposomes, however, demonstrated steadily increasing tryptic activity throughout the sampling period, attaining 0.255 mU mg-1 DW after 2 h, which was significantly (p<0.05) more than all the other treatments. The results showed that ingested liposomes containing FAA or the protein BSA or a combination of these two nutrients effectively stimulated CCK production in first-feeding herring larvae. In contrast, liposomes containing only physiological saline did not elicit a CCK response. In addition, liposomes containing BSA stimulated tryptic activity in herring larvae, which was not observed in fish fed liposomes that included only FAA or PS. This suggests that a suitable protein substrate is required to regulate protein digestion.  相似文献

The contents of free amino acids (FAA) and total protein, together with growth and gut-content, of turbot (Scophthalmus maximus L.) larvae reared at 14, 18 and 22 °C were studied from first-feeding to approximately 140 effective day-degrees post hatch (Deff ). Artemia franciscana nauplii and two species of rotifers were used as prey. Protein content accounted for about 42 and 26% of dry body mass in the A. franciscana nauplii and the rotifers, respectively. The FAA pool constituted 5.6 and 4.8% of the total amino acids in the same animals. The dry body mass of turbot larvae was exponentially related to Deff . Protein and FAA contents were linearly related to dry body mass, and were independent of rearing temperature between 14 and 18 °C. At the end of the experiment, however, turbot larvae at 22 °C had lower gut content values, retarded growth rates, and decreased FAA contents and concentrations. Thus, at this high temperature, turbot larvae seem unable to catch and ingest sufficient prey, or to sustain an amino acid assimilation rate from the intestine sufficient to meet metabolic demands. The objective of this study was to describe the seasonal variations in nucleic acid contents and amino acid profiles in the muscle of juvenile Nephrops norvegicus. RNA and protein contents, and RNA:protein and RNA:DNA ratios varied significantly between seasons, being highest in spring and lowest in autumn/winter ( P<0.05). Though DNA content increased significantly from autumn to summer ( P<0.05), protein:DNA ratio did not show significant seasonal variations ( P=0.05). In respect to protein-bound amino acid content (BAA), a significant increase was observed from winter to summer ( P<0.05). Both essential (EAA) and non-essential amino acids (NEAA) increased significantly (27.6% and 27.8%, respectively; P<0.05), mainly due to the considerable increase in arginine and proline (59.1% and 225.2%, respectively; P<0.05). A significant decrease was observed in the free amino acid content (FAA) from winter to summer ( P<0.05); and a higher percentage decrease occurred in free non-essential (FNEAA; 27.9%) in comparison to free essential amino acids (FEAA; 21.8%). The significant increase in RNA and BAA contents from winter to spring may be related to protein synthesis. On the other hand, the lowest values obtained in winter may be due to a reduction in feeding activity; in this period the muscle protein must be progressively hydrolysed, which is evident with the higher FAA content. The liberated amino acids enter the FAA pool and become available for energy production.  相似文献

Previous studies have demonstrated that cadmium can induce biochemical and physiological changes in yeast Saccharomyces cerevisiae. However, studies on the influence of cadmium on the ion balance in the cell and the interaction between cadmium and other ions are still relatively few in number. By using inductively coupled plasma-atomic emission spectrometry, the contents of some cations, including Zn2+, Ca2+, Fe3+, Cu2+, Mg2+, K+, and Na+ were measured. The data showed that the levels of Zn2+ and Fe3+ were increased, while those of Cu2+, K+, and Na+ were decreased after cadmium treatment. Afterwards, using the drop test assay, the interactions between cadmium and the selected ions were investigated. The results suggested that the cytotoxicity of cadmium could be attributable to the interference of cadmium with the intracellular cation homoeostasis. Calcium channel transporter Cch1 participates in the intracellular uptake of cadmium. Additionally, Zn2+, Ca2+, Fe3+, Mg2+, and K+ can rescue the toxic effect of cadmium in yeast.  相似文献
