For the first time, gender-related differences in some important functional parameters of haemocytes from the clam Ruditapes philippinarum have been studied. Clams were collected during the pre-spawning phase, when sex can be distinguished by microscopic observation of gonadal smears, in order to ascertain whether the two sexes reach the stressful spawning period with different degrees of immunosurveillance. Total haemocyte count (THC), haemocyte size frequency distribution, capability of haemocytes to assume the vital dye neutral red (NR, indicative of endocytotic activity), lysozyme-like and acid phosphatase activities in both haemocyte lysate (HL) and cell-free haemolymph (CFH), and superoxide dismutase (SOD) and catalase (CAT) activities in HL were all evaluated. No differences in THC values were found between the two sexes, but differing haemocyte size frequency distributions were observed: the fraction of larger haemocytes (7–9 μm diameter, 200 fl volume) was markedly increased in females and that of smaller haemocytes (<5 μm diameter, <200 fl volume) markedly increased in males. Interestingly, when haemocytes were observed under the light microscope, a significantly higher percentage of granulocytes was found in haemolymph from females, compared with that of males. Significantly increased NR uptake was recorded in female haemocytes. No significant variation in lysozyme-like activity was observed in HL, whereas in CFH, enzyme activity was significantly higher in females. HL acid phosphatase activity was significantly higher in males with respect to females; no significant variations in enzyme activity were observed in CFH. HL SOD and CAT activities were significantly higher in females than in males. Overall, on the basis of the cell parameters studied here, gender-related differences in immune parameters were found in R. philippinarum, indicating that females have more active haemocytes than males during the pre-spawning period.  相似文献

Fishes are sensitive indicators of pollutants present in water.These pollutants cause various physical and physiological alterations in fishes. In the present work alteration in the activity of acid and alkaline phosphatase was evaluated in testicular tissue of fresh water fish Heteropneustes fossilis exposed to LC50 value of linear alkyl benzene sulphonate (LAS) for different exposure periods [24 h, 48 h, 72 h and 96 h] With increase in the concentration of chemical LAS, the activity of acid phosphatase (ACP) was reported elevated while a significant fall in the activity of alkaline phosphatase (ACP) was recorded for same exposure period. Elevated activity of ACP, one of the important hydrolases of lysosomes, is quite suggestive of bringing about gross necrosis and dysarchitecture. ALP is involved in various metabolic activities including gonadal maturation and as such decreased activity of this enzyme is definitely one of the important causative factors for reproductive impairment of the fish.  相似文献

Alterations in the activity of enzymes Acid phosphatase (E.C.3.1.3.2) and Alkaline phosphatase (EC 3.1.3.1) in organs such as liver, gills and muscle of rohu following its exposure to surfactants viz. CTAB, SDS and Triton X-100 were analyzed. Different levels of exposure were given depending on the LC50 value of the surfactant used. Also, the influence of age and weight of the organisms was tested simultaneously. The activity of ACP in the tissues of fish exposed to all the three surfactants showed marked enhancement after exposure; the effect being highest in the liver followed by gill and muscle. Activity levels of ALP in different tissues of the fish exposed to the surfactants also showed an increase. Maximum increase was found in the liver followed by muscle, and gill. The increase in the levels of these enzymes indicates a stressful condition of the fish.  相似文献

The effect of linear alkylbenzene sulphonate (LAS) on copper and lead bioaccumulation in Ruditapes philippinarum was studied. The in vitro and in vivo effect of LAS on acid and alkaline phosphatase activities was also examined. For lead, significant differences (P<0.01) were observed between organisms exposed to lead and to the mixture of LAS and lead. In the case of copper, these differences were only significant in the digestive gland (P<0.05). In vitro acid phosphatase was inhibited by LAS and the mixture of LAS with copper showed the effect of decreasing the enzymatic activity. Nevertheless, in vivo no significant differences in the digestive gland and gills (P < 0.05) were observed.  相似文献

The extra-embryonic albuminous capsule fluid of the marine prosobranch Nucella lapillus L., collected from March to May 1988 in Brittany, France, has been investigated by sodium dodecylsulfate-polyacrylamide-gelelectrophoresis (SDS-PAGE) and biochemical assays. The fluid at the gastrula- and early veliger stage of the snail embryo was analysed for proteins on SDS-gels, i.e., for acid phosphatase, alkaline phosphatase, -glucuronidase and arylsulfatase activities. The protein and inorganic phosphorous contents were also determined. Electrophoresis revealed the presence of different proteins and a variation in the band pattern during early developmental stages. The total protein content of the capsule fluid decreases significantly from gastrula to early veliger stage. In the differentiated developmental stage the acid phosphatase activity was higher than the poorly differentiated gastrula stage. The detected activity of arylsulfatase indicates that acid mucopolysaccharides are present. In general, observed changes in the organic components of the intracapsular fluid of N. lapillus substantiate the nutritive value for developing embryos.  相似文献

Water content in the tissues, tissue proportions, and content and concentrations of copper, zinc, iron, cadmium, manganese, and cobalt were determined in male shore crabs Carcinus maenas of different sizes. Average wet to dry weight ratios in midgut gland, gills, muscle, exoskeleton, stomach, heart, testes, hypodermis, and haemolymph were 5.3, 9.5, 4.1, 1.5, 4.8, 6.1, 3.8, 4.7, and 13, respectively. The water content in the soft tissues generally increased with increasing size of the crabs (except in testes). Dry midgut gland, gills, muscle, exoskeleton, stomach, heart, testes, hypodermis, and haemolymph constituted 4, 0.9, 20, 68, 0.5, 0.12, 0.5, 0.7 and 5.3%, respectively, of the body dry weight. Dry midgut gland, gills, stomach, hypodermis, and haemolymph constituted a larger proportion of the body weight in small than in large crabs, whereas the opposite was the case for exoskeleton. Protein concentrations in the haemolymph decreased with size whereas haemolymph volume increased. Seventy percent of the copper body burden was located in the haemolymph. The majority of zinc (61%) occurred in muscle, whereas the midgut gland was the major site of cadmium storage (89%). The exoskeleton contained the major part of the body burden of iron (71%) and manganese (95%). Most of the cobalt body burden was located in the exoskeleton (45%) and haemolymph (44%). As tissue hydration and tissue proportions changed with size, tissue metal concentrations did not necessarily vary identically with size when expressed on a dry weight compared to a wet weight basis. Tissue potassium concentrations were negatively correlated with tissue water contents, indicating that water in tissues with a high wet to dry weight ratio was of extracellular origin. However, comparison of tissue and haemolymph copper levels showed that this extracellular water could not be pure haemolymph as tissue copper concentrations would have been higher than the values actually found.  相似文献

以中华绒螯蟹(Eriocheir sinensis)作为暴露生物,检测了水体Hg2+对中华绒螯蟹肝胰腺、血淋巴谷丙转氨酶(GPT)、谷草转氨酶(GOT)、碱性磷酸酶(AKP)和酸性磷酸酶(ACP)活性的影响。结果表明,肝胰腺中GPT和GOT活性随水体中Hg2+浓度(0.01~0.30mg·L-1)升高而降低;中华绒螯蟹血淋巴中GPT和GOT活性变化趋于分化,0~0.05mg·L-1暴露组,随Hg2+浓度升高血淋巴GPT活性迅速上升,0.10~0.30mg·L-1暴露组,随Hg2+浓度升高血淋巴GPT活性迅速下降;GOT活性随着水体Hg2+暴露浓度的升高则持续上升,表现出"剂量-效应"正相关;两种组织中AKP和ACP活性随暴露浓度的升高都表现出"低-高-低"的变化趋势,并且最高值均出现在0.05mg·L-1的Hg2+处理组。4种酶活性对水环境中Hg2+反应灵敏,可作为中华绒螯蟹毒理学评价指标以及重金属监测指标。  相似文献

During 24 and 48 hr of exposure, the digestive glands of Lymnaea treated with a lethal concentration of 0.038 mgl(-1) CuSO4 revealed intense activity of acid phosphatase in perilobular margin. On the other hand, same area of the gland showed moderate activity of ATPase during 24 and 48 hr of exposure. However, alkaline phosphatase showed average activity in perialveolar region and perilobular margin during 24 and 48, and 72 hr of exposure respectively The changes in the activity of these enzymes were nonsignificant in alveolar margin and perialveolar region of the gland. The concentration of glutamine in Tridacna gigas haemolymph increased >35-fold following exposure to sea water supplemented with ammonium (20 μM), but no increase was observed with nitrate (20 μM). Lack of a diel cycle, no decrease in haemolymph glucose levels, the expression patterns of glutamine synthetase in zooxanthellae and host, and the lack of glutamine release in response to nitrate supplementation all support the proposition that the increase in haemolymph glutamine is a product of the host and not the zooxanthellae. Unlike ammonium, nitrate accumulates rapidly in the haemolymph. It has no effect on the concentration of glutamine in the haemolymph, but there is an increase in arginine, histidine and lysine in the haemolymph, suggesting the release of these essential amino acids from zooxanthellae. Glutamine synthetase (GS) activity decreased markedly in the gill and less so in the mantle over a period of 6 d exposure to elevated ammonium (20 μM). In contrast, GS activity in zooxan- thellae doubled. The response of zooxanthellae in situ was confirmed by incubating freshly isolated zooxanthellae for 4 d in ammonium, which resulted in a ten-fold increase in GS activity. Comparison of the in situ response of zooxanthellae with that obtained in vitro indicates that the symbionts are likely to be exposed to ammonium concentrations lower than that found in the haemolymph. Tissue concentrations and contents of copper and zinc were determined at 11 different stages of the moult cycle in male and female shore crabs Carcinus maenas. Metal concentrations in haemolymph, gills, midgut gland, muscle, and exoskeleton of males and haemolymph, gills, and midgut gland of females were determined, as were haemocyanin concentrations and haemolymph volumes (using 14C-inulin) in males. The changes in tissue Cu and Zn concentrations and contents that occur throughout the moult cycle can be attributed to muscle breakdown in late premoult, the period of starvation in late premoult and early postmoult, the resorption from and shedding of the old exoskeleton, and the dilution of the haemolymph caused by water uptake around the time of ecdysis. The present study demonstrates that whole-body Cu and Zn contents remain constant during a large part of the moult cycle of male and female C. maenas. This state of whole-body trace metal homeostasis is maintained in spite of major changes in tissue proportions and tissue Cu and Zn concentrations and contents. Previous studies have not carried out the necessary analysis to move from theoretical estimates to quantitative determination of the changes in tissue metal distribution associated with moulting in crustaceans; the data presented illustrate the necessity of measuring both tissue concentrations and contents of metals to avoid misinterpretation of either.  相似文献

The influence of copper(II)chloride on selected key enzymes and metabolites in intermediary metabolism and the energy charge potential of the hyperregulating shore crab, Carcinus maenas (L.) were investigated. Crabs, collected in Odense Fjord, Denmark between September 1989 and May 1990, were exposed to 10 ppm copper(II)chloride, in 10 ppt salinity seawater at 15°C for up to 1 wk. Hexokinase activity was 77% lower in the posterior gills and 60% lower in the midgut gland than in control crabs. Phosphofructokinase activity was reduced in the midgut gland by 82% but was unaffected in the gills. Pyruvate kinase activity was undetectable in the gills of exposed crabs and reduced by 47% in the midgut gland. Citrate synthase activity was depressed by 30% in the posterior gills and unaffected in other tissues. Copper exposure had no apparent effect on tissue cytochrome c oxidase activity. Chela muscle was the only tissue in which copper exposure did not bring about reductions in the enzyme activities tested. As a result of 1 wk of copper exposure the average lactate level increased by 3.5-fold in haemolymph, 9.3-fold in posterior gills and 6.5-fold in midgut gland. The average glucose level was raised by a factor of 6 in the haemolymph, 3.4 in the posterior gills and 1.2 in the midgut gland. Energy charge potential was at a very high level (ca. 0.9) and was unaffected by copper exposure.  相似文献

The sublethal toxicity of sodium arsenite on protein metabolism was investigated in teleost fish, Tilalpia mossambica at the end of 24, 48, 72 and 96 h of exposure. Total protein content, free amino acid content and activities of the enzymes aspartate amino transferase (AAT) and alanine amino transferase (ALAT) in liver, gill, brain and muscle exhibited significant (P<0.05) alterations throughout the investigation in relation to that of control. It is suggested that the fish is able to respond to the stressful situations by gearing up the metabolic activity as revealed by the elevated protein, amino acid content and the activities of AAT and ALAT.  相似文献

Potassium tetranitrodiammine cobaltate(III) K[Co(NH3)2(NO2)4] is a coordination complex having Co(III) as the central atom. Cobalt(III) compounds are being advocated for their anticarcinogenic potential. In the present study, we have for the first time evaluated this compound for its toxicity to rat RBC. The parameters studied include the effects of potassium tetranitrodiammine cobaltate(III) on GSH, GST, catalase, G6PD, acid phosphatase, GOT, and GPT. Scanning electron microscopy of RBCs of potassium tetranitrodiammine cobaltate(III) treated rats has also been carried out for evaluating its influence on the red cell morphology. The data showed that potassium tetranitrodiammine cobaltate(III) treatment causes marked biochemical changes in the rat RBCs along with changes in their shapes characterized by the formation of acanthocytes. The results thus suggest that acute administration of potassium tetranitrodiammine cobaltate(III) is potentially toxic to rat RBCs.  相似文献

With global climate change, ocean warming and acidification occur concomitantly. In this study, we tested the hypothesis that increasing CO2 levels affect the acid–base balance and reduce the activity capacity of the Arctic spider crab Hyas araneus, especially at the limits of thermal tolerance. Crabs were acclimated to projected oceanic CO2 levels for 12 days (today: 380, towards the year 2100: 750 and 1,120 and beyond: 3,000 μatm) and at two temperatures (1 and 4 °C). Effects of these treatments on the righting response (RR) were determined (1) at acclimation temperatures followed by (2) righting when exposed to an additional acute (15 min) heat stress at 12 °C. Prior to (resting) and after the consecutive stresses of combined righting activity and heat exposure, acid–base status and lactate contents were measured in the haemolymph. Under resting conditions, CO2 caused a decrease in haemolymph pH and an increase in oxygen partial pressure. Despite some buffering via an accumulation of bicarbonate, the extracellular acidosis remained uncompensated at 1 °C, a trend exacerbated when animals were acclimated to 4 °C. The additional combined exposure to activity and heat had only a slight effect on blood gas and acid–base status. Righting activity in all crabs incubated at 1 and 4 °C was unaffected by elevated CO2 levels or acute heat stress but was significantly reduced when both stressors acted synergistically. This impact was much stronger in the group acclimated at 1 °C where some individuals acclimated to high CO2 levels stopped responding. Lactate only accumulated in the haemolymph after combined righting and heat stress. In the group acclimated to 1 °C, lactate content was highest under normocapnia and lowest at the highest CO2 level in line with the finding that RR was largely reduced. In crabs acclimated to 4 °C, the RR was less affected by CO2 such that activity caused lactate to increase with rising CO2 levels. In line with the concept of oxygen and capacity limited thermal tolerance, all animals exposed to temperature extremes displayed a reduction in scope for performance, a trend exacerbated by increasing CO2 levels. Additionally, the differences seen between cold- and warm-acclimated H. araneus after heat stress indicate that a small shift to higher acclimation temperatures also alleviates the response to temperature extremes, indicating a shift in the thermal tolerance window which reduces susceptibility to additional CO2 exposure.  相似文献

The functions of the various gut regions of Oikopleura dioica (oesophagus, left and right gastric lobes, vertical intestine, mid-intestine and rectum) were investigated by means of histochemical, histoenzymatic and immunohistochemical techniques at light and electron microscopes. Ciliary food progression is evidenced by the presence of ATPases on cilia, along the entire gut, with the exception of the cardiac valve, a passive device controlling food direction. Absorptive processes (alkaline phosphatase), active transport (ATPases) and nitrogen excretion (D-amino acid oxidase) occur along the entire gut, in both ciliated microvillar and globular cells. The latter, typical of the left gastric lobe and rectum, are also involved in endocytotic processes (exogenous peroxidase as tracer) and intracellular digestion (!-amylase, aminopeptidase M, acid phosphatase, 5'-nucleotidase, non-specific esterase). The giant cells of the gastric band participate in extracellular digestion; they contain secretory granules positive to various hydrolytic enzymes, the activity of which is also recognisable in faecal pellets inside the intestinal lumen. Lipid storage occurs mainly in the right gastric lobe and vertical intestine, whereas protein storage takes place in the rectal granular cells. Epithelial transport and possible osmoregulation occur along the entire gut, especially at the level of diffuse baso-lateral interdigitations, which increase the plasmalemma surface enormously, are often associated with mitochondria and possess numerous ATPase pumps. Data extend previous histological observations and hypotheses on the physiological role of the various gut regions. The remarkable and specific location of enzymatic activities and nutrient storage are in agreement with the high capacity of O. dioica to process a great quantity of food very rapidly and efficiently.  相似文献

Acid and alkaline phosphatase activities have been partially characterized in Ruditapes philippinarum (Adams and Reeve, 1850). Two activity peaks at pH=4.5 and pH 10.5 were detected in the gill, digestive gland, mantle, siphon and foot. Acid phosphatase activity was higher than that of alkaline phosphatase. The highest activity for both enzymes was observed in the digestive gland and, in decreasing order, the gill, foot, siphon and mantle. Alkaline phosphatase activity was similar in the mantle, siphon and foot. K m values were determined for both enzymes in the gill and digestive gland. Hill coefficients were near 1, indicating no allosteric behaviour for either enzyme in the two organs. The optimum temperature was the same for acid phosphatase in both gill and digestive gland (50 °C), while for alkaline phosphatase it differed for these two organs (gill, 40 °C; digestive gland, 35 °C). The apparent activation energy was obtained from Arrhenius plots, and ranged from 8.61 kcal/mol for alkaline phosphatase in the gill, to 10.84 kcal/mol for acid phosphatase in the digestive gland. The effects of metals (1 mM conc) on both enzyme activities were assayed in vitro. Hg strongly inhibited the enzyme activities in the gill and digestive gland, probably because of its affinity for the sulphydryl group. Histochemically, acid phosphatase in the gill was located in a granular form throughout the gill cells, but was undetectable in the ciliate epithelium of the gill filaments. Alkaline phosphatase was located in the gill skeleton. Clam size and phosphatase activities were inversely related, probably reflecting a decrease in shell deposition with inereasing size. As a function of season, both enzymes were present in lowest amounts in winter, when undifferentiated sex cells were predominant in the germinative epithelium, and highest in summer, when ripe individuals of both sexes were more frequent.  相似文献

采用半静态毒性实验方法,将菲律宾蛤仔(Ruditapes philippinarum)分别暴露于0.2、2、20μg·L-1的全氟辛酸(perfluorooctanoic acid,PFOA)中,在处理后第1、3、6、10、15、21天分别取样,测定整体组织中超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)、谷胱甘肽-S-转移酶(GST)、7-乙氧基异吩噁唑酮脱乙基酶(EROD)活性和过氧化脂质(LPO)含量。酶活性分析结果显示:PFOA对菲律宾蛤仔组织SOD、CAT和POD活性均呈现先促进后抑制的作用;低浓度组SOD活性在暴露第1天达到最高,显著高于对照组(P<0.01);中高浓度组SOD活性在暴露第6天达到最低;暴露1~15 d,低浓度组CAT活性均显著高于对照组(P<0.05);高浓度组CAT活性在暴露第6天得到显著诱导,其余时间基本处于抑制状态;中浓度组POD活性在暴露第3天即达到最高,高浓度组POD活性基本一直处于抑制状态;随着PFOA暴露时间的延长,菲律宾蛤仔组织LPO含量呈现了先降低后升高的趋势;各浓度组中EROD的活力都显著被诱导(P<0.01),与处理浓度呈正相关;中高浓度组的GST活性在胁迫期间变化比较显著,呈现诱导-抑制的变化规律。研究表明,PFOA暴露能够引起菲律宾蛤仔组织抗氧化酶和生物转化酶的变化,可以与其他敏感性指标一起作为指示早期海洋PFOA污染的生物标志物。  相似文献

研究了阿维菌素长期暴露下鲤鱼肝脏和肌肉超氧化物歧化酶(SOD)活性的动态变化.结果表明:阿维菌素对SOD活性具有较大影响.低浓度组(3.2μg·L-1)SOD活性随暴露时间无显著变化(p>0.05);中浓度组(5.6μg·L-1和7.5μg·L-1)SOD活性先显著上升(p<0.05),随后又显著下降(p<0.05);高浓度组(10μg·L-1和18μg·L-1)SOD活性呈逐渐下降趋势,48h后极显著低于对照(p<0.01).解除污染胁迫10d,低浓度和中浓度组SOD活性能恢复到正常水平,但高浓度组SOD活性不能恢复到正常水平,说明低浓度阿维菌素对鲤鱼机体产生的损伤是可逆性的,而高浓度阿维菌素会对鲤鱼机体产生不可逆损伤.阿维菌素暴露浓度与其对鲤鱼肝脏和肌肉SOD活性抑制率之间具有显著剂量-效应关系,可以考虑将其作为水体中阿维菌素类药物污染的生物标志物;同时,由于正常鲤鱼(对照组)肌肉中SOD活性和受污染胁迫时SOD活性变化的显著性远低于肝脏,因此在考虑用SOD作为生物标志物对水体中阿维菌素污染进行监测时,肝脏是比较理想的取样器官.  相似文献
