When measurements of in-vivo fluorescence are used to estimate photosynthesis in the field, the marked temporal and spatial variations in phytoplankton populations, and their nutrient and light histories, have produced varied results. Natural phytoplankton populations in large, flow-through mesocosms with different controlled nutrient and sewage sludge additions were sampled weekly from June to September 1984. Good correlations were observed between the increase in fluorescence upon the addition of DCMU (F) and both in-situ production and the parameters (Pm and ) of the photosynthesis-irradiance curve for these phytoplankton populations. Good correlations were also obtained between DCMU-enhanced fluorescence (FDCMU) and chlorophyll a concentrations. The relationship between F and in-situ 14C production was consistent among mesocosms even in the face of major shifts from diatom-dominated to dinoflagellate-dominated populations. On the other hand, the FDCMU:Chl a relationship was significantly different between mesocosms and related to species composition. It was concluded that F offers the possibility of rapidly and accurately indexing both in-situ production and the photosynthetic capacity of mixed phytoplankton populations.This study was supported by EPA cooperative agreement 810265-03 and NOAA grant NA-83-ABD-00008  相似文献

Diel periodicity in parameters of photosynthesis-irradiance (P-I) curves was incorporated into calculations of integral daily phytoplankton production for the Santa Barbara Channel off southern California (USA). Model equations of the relationship between photosynthesis and light were used in combination with observed slope () and asymptote (P max) values presented in the preceding paper. Primary production was always 19 to 39% less than comparable estimates obtained with the assumption of constant maximum daily and P max values. Regardless of which P-I formulation was used of 6 tested, observed production (using a temporal series of simulated in situ incubations) ranged from 13% less to 25% more than estimates from constant midday and P max values. The amplitude and timing of diel oscillations differed somewhat among 3 field stations. Maximum to minimum ratios ranged from approximately 3 to 5 for , and 4 to 6 for P max. The differences in amplitude and timing of oscillations in P-I curves both contributed to errors in calculating phytoplankton production. Thus, photosynthetic periodicity in the upwelling area of the Santa Barbara Channel influences phytoplankton production. There were oscillations in both and P max, and the time-dependence of these parameters should be considered to improve the accuracy of predictive models of primary productivity.  相似文献

Diel oscillations in the photosynthesis-irradiance (P-I) relationship are described for marine phytoplankton assemblages at 6 stations in an upwelling area off the southern California coast (USA) between May and August 1980. The initial slope () and asymptote (P max) of P-I curves changed significantly over the day; both parameters were in phase and had similar changes in amplitude. The diel oscillations in photosynthesis appeared unrelated to changes in chlorophyll a concentrations. Amplitudes of daily variations in photosynthesis ranged from approximately 3 to 9, as measured by the maximum to minimum ratio for photosynthetic capacity (P max). Diatom-rich samples collected during an upwelling event and those dominated by dinoflagellates both had midday to early afternoon maxima in and P max. Samples from other locations had peak photosynthetic activity later in the afternoon. The relationship between and P max was consistent in all phytoplankton samples analyzed, with a surprisingly high correlation considering the spatial and temporal scales encompassed in this study. These results indicate that the photosynthesis-irradiance (P-I) relationship is time-dependent and, moreover, that changes in and P max are closely coupled for a variety of natural phytoplankton assemblages.  相似文献

Summary Feeding by the homopteranPsylla pyricola on leaves of pear trees induces the production of volatile compounds, such as (E,E)--farnesene and methyl-salicylate, as well as the production of polyphenols. The inference on induction is based on GC-MS and HPLC chromatograms from the same samples ofPsylla infested leaves, leaves from the same pear tree beforePsylla infestation and uninfested leaves from other pear trees.Psylla infestation greatly enhanced the production of volatiles ((E,E)--farnesene, methyl-salicylate and others) and triggered the production of new polyphenols, characterized by much longer retention times.However, the responses to infestation depend critically on leaf age (defined by leaf distance to apex). With respect to the leaf volatiles it appears that infested, old leaves produce fewer compounds and lower amounts of the volatiles than infested, young leaves. Moreover, there seem to be differences in pattern. Relative to (E,E)--farnesene, methyl-salicylate was found in much lower amounts in heavily infested, old leaves. With respect to polyphenols it was found that infested old leaves collected in August have polyphenols with the same retention times, but more or less equal amounts as uninfested young leaves collected in May. This shows thatPsylla infestation causes the induced response mostly in young leaves.The induced leaf volatiles may act as synomones to heteropteran bugs. As shown elsewhere,Anthocoris nemoralis responds significantly to (E,E)--farnesene and methyl-salicylate when offered in pure form against clean air in a Y-tube olfactometer. The effect of polyphenols on the performance ofP. pyricola is not yet known. Hence, a role in direct defence is still to be investigated.  相似文献

Two suites of phytoplankton samples have been collected in consecutive years at various times over a day from selected depths within vertically mixed and stratified water columns in the western Irish Sea, in order to provide a range of possible light histories within the populations collected. Values for the maximum rate of 14C retention (P max) and the initial slope of the 14C retention: light intensity curve () were obtained. Supra-thermocline samples from the stratified water exhibited higher P max values than corresponding subthermocline samples. Higher values of were also generally associated with samples from the supra-thermocline zone of the stratified region. Differences in the depth distribution of P max and in the mixed water were small, except in the presence of a shallow thermocline. In one suite of samples from the stratified water, a diurnal increase in the P max values of the supra-thermocline samples was observed. P max values obtained from the samples from the mixed water were interpreted in relation to the distribution obtained from the samples from the stratified zone. Data from both the contrasting sites visited for one sample suite demonstrated a two-phase relationship between the chlorophyll a concentration and both P max and . The rates of 14C retention of the first suite of samples were estimated by two techniques. The average differences in the retention were greater in samples from the sub-as opposed to suprathermocline zone. No trends were apparent in the smaples from the mixed waters.  相似文献

The effects of light exposure on the photosynthetic activity of kleptoplasts were studied in the sacoglossan mollusc Elysia viridis. The photosynthetic activity of ingested chloroplasts was assessed in vivo by non-destructively measuring photophysiological parameters using pulse amplitude modulation (PAM) fluorometry. Animals kept under starvation were exposed to two contrasting light conditions, 30 μmol photons m−2 s−1 (low light, LL), and 140 μmol photons m−2 s−1 (high light, HL), and changes in photosynthetic activity were monitored by measuring the maximum quantum yield of photosystem II (PSII), F v/F m, the minimum fluorescence, F o, related to chlorophyll a content, and by measuring rapid light-response curves (RLC) of relative electron transport rate (rETR). RLCs were characterised by the initial slope of the curve, αRLC, related to efficiency of light capture, and the maximum rETR level, rETRm,RLC, determined by the carbon-fixation metabolism. Starvation induced the decrease of all photophysiological parameters. However, the retention of photosynthetic activity (number of days for F v/F m > 0), as well as the rate and the patterns of its decrease over time, varied markedly with light exposure. Under HL conditions, a rapid, exponential decrease was observed for F v/F m, αRLC and rETRm,RLC, F o not showing any consistent trend of variation, and retention times ranged between 6 and 15 days. These results suggested that the retention of chloroplast functionality is limited by photoinactivation of PSII reaction center protein D1. In contrast, under LL conditions, a slower decrease in all parameters was found, with retention times varying from 15 to 57 days. F v/F m, αRLC and rETRm,RLC exhibited a bi-phasic pattern composed by a long phase of slow decrease in values followed by a rapid decline, whilst F o decayed exponentially. These results were interpreted as resulting from lower rates of D1 photoinactivation under low light and from the gradual decrease in carbon provided by photosynthesis due to reduction of functional photosynthetic units.  相似文献

Octopine dehydrogenase from the nemertean Cerebratulus lacteus was purified over 1000-fold to almost homogeneity. The enzyme does not bind to arginine Sepharose 4B. It has a monomeric structure with a relative molecular mass of 40000. Two isoenzymes were identified with isoelectric points of 5.6 and 5.4, whereas the purified isoenzymes of Pecten jacobaeus adductor mucles (which bind to arginine Sepharose 4B) had lower IEP's of 4.9 and 4.7. Apparent Km's of the nemertean ODH for arginine and pyruvate are dependent on the respective co-substrate concentration. This phenomenon may result in activation of ODH and, thus, production of octopine in locomotory highly active individuals while attacking food, especially when this takes place in a hypoxic habitat, such as decaying mud near the high-water mark. The apparent Km's for octopine (0.22 mM) and NAD+ (14 M) are low. Octopine is a substrate inhibitor for the reverse reaction above 2 mM, and a product inhibitor of the forward reaction by 50% at 1.2 mM. Therefore, only small amounts of octopine are likely to accumulate in vivo. Amino acid substrate specificity is limited to guanidino amino acids. We believe that the amino acid substrate specificity is not an evolutionary modification, but rather that it is narrowed to guanidino amino acids (or even specificity to arginine) in those species where ODH has a physiological function in maintaining redox balance during exercise. The specificity for keto acids is dependent on chain length, (-ketobutyrate>-ketocapronate); a second carboxyl group inactivates the enzyme.  相似文献

Quantitative data on digestive enzymes of bivalves are very limited and so this study was performed to obtain such information, and to find out whether some of these enzymes are well adapted to conditions in the digestive gland. The green musselPerna viridis was obtained from Clementi West Market in 1985. The results show that various carbohydrases are present in the digestive gland. The activities of-amylase, cellulase and-glucosidase are higher than that of other carbohydrases, whilst the activities of both lipase and protease, especially the former, are low. The pH optima of the following enzymes are also given:-amylase = 5.8; laminarinase = 5.5;-glucosidase = 5.8; trehalase = 4.8;-glucosidase = 5.5. The only enzyme which was not well adapted to conditions in the digestive gland (pH 6.2) was trehalase. The wide spectrum of enzymes present in this bivalve indicates that it has the ability to utilize a wide range of nutrients efficiently.  相似文献

The dynamics of 65Zn specific activity and total zinc in benthic fishes on the outer continental shelf off central Oregon (USA) were examined. A differential equation that relates specific activity of 65Zn in fish to that in fish food was used to estimate 's (zinc uptake-rate coefficients) for 3 different size classes of the flounder Lyopsetta exillis, a small predator of pelagic Crustacea, and for 1 size class of the flounder Microstromus pacificus, a large predator of infauna. The 's obtained for L. exilis were very close to the obtained in the laboratory for the flounder Pleuronectes platessa. The estimated for M. pacificus was very much smaller than the 's estimated for the other two species. A model that related to predicted weight-specific feeding rates suggested that the smaller of M. pacificus was caused by a low absorption efficiency of zinc from its prey. Sensitivity studies indicated that time histories of specific activity in the fishes are not sensitive to moderate changes in . The negative correlation between specific activity in the diet and in the weight of L. exilis was the major cause of the negative correlative between specific activity and weight in this species. In M. pacificus, where composition of diet does not vary with size, specific activity was independent of weight. The time history of specific activity in M. pacificus was very much lower than those in the different size classes of L. exilis, a result caused mainly by the much lower specific activity of the prey of M. pacificus. Differences in specific activity among other benthic fishes were also correlated with differences in specific activity of their prey. The food-web dynamics responsible for these patterns are discussed. Variation in total zinc concentrations among species was small. Within species of flounder, zinc concentration varied only slightly or not at all with weight.  相似文献

We monitored the photosynthetic response of an estuarine epilithic microflora exposed to natural variations in water level and light intensity. The experimental community developed on an artificial substrate in the intertidal zone of the St. Lawrence estuary. Fragilaria striatula dominated the assemblage. Samples for the determination of the Photosynthesis-Irradiance curve were collected at intervals of 2 h over a period of 11 d. The initial slope of the curve (B) and the maximum photosynthetic rate (P m B ) per unit Chl a were estimated. During spring tide, wave-induced turbulence reaching the experimental substrate at low tide eroded the arborescent stratum of the cell mat. The physiological condition of the remaining prostrate stratum was poor (low Chl a/ phaeopigment ratio). The photosynthetic response of the community was weak and showed little variability. During neap tide, the arborescent stratum of the permanently inundated community persisted. The community showed a stronger and more variable photosythetic response. During this period fluctuations in the magnitude of B and P m B were dominated by a 24-h periodicity, but also presented a secondary semidiurnal rhythm. The ciradian periodicity in the photosynthetic response was best explained by postulating an endogenous control. Circatidal variations in P m B were perhaps related to tidal fluctuations in nutrient availability. Photosynthetic performance in the kelp Laminaria solidungula J. Agardh was examined from photosynthesis irradiance (P-I) parameters calculated from in situ 14C uptake experiments, using whole plants in the Stefansson Sound Boulder Patch, Alaskan Beaufort Sea, in August 1986. Rates of carbon fixation were determined from meristematic, basal blade, and second blade tissue in young and adult sporophytes. Differences in saturating irradiance (I k, measured as photosynthetically active radiation, PAR), photosynthetic capacity (P max), and relative quantum efficiency () were observed both between young and adult plants and between different tissue types. I k was lowest in meristematic tissue (20 to 30 E m–2 s–1) for both young and adult plants, but consistently 8 to 10 E m–2 s–1 higher in young plants compared to adults in all three tissues. Average I k for non-meristematic tissue in adult plants was 38 E m–2 s–1. Under saturating irradiances, young and adult plants exhibited similar rates of carbon fixation on an area basis, but under light limitation, fixation rates were highest in adult plants for all tissues. P max was generally highest in the basal blade and lowest in meristematic tissue. Photosynthetic efficiency () ranged between 0.016 and 0.027 mol C cm–2 h–1/E m–2 s–1, and was highest in meristematic tissue. The relatively lower I k and higher exhibited by L. solidungula in comparison to other kelp species are distinct adaptations to the near absence of light during the eight-month ice-covered period and in summer when water turbidity is high. Continuous measurement of in situ quantum irradiance made in summer showed that maximum PAR can be less than 12 E m–2 s–1 for several days when high wind velocities increase water turbulence and decrease water transparency.The Univeristy of Texas Marine Science Institute Contribution No. 695  相似文献

Short-term changes in phytoplankton photosynthetic activity were studied during different periods of the years 2009 and 2010 in the coastal waters of a macrotidal ecosystem (the Strait of Dover, eastern English Channel). During each sampling period, samples were taken every 1.45 h., from sunrise to sunset, during at least 5 days distributed along a complete spring–neap tide cycle. The photosynthetic parameters were obtained by measuring rapid light curves using pulse amplitude modulated fluorometry and were related to environmental conditions and phytoplankton taxonomic composition. The maximum quantum yield (F v/F m) showed clear light-dependent changes and could vary from physiological maxima (0.68–0.60) to values close to 0.30 during the course of 1 day, suggesting the operation of photoprotective mechanisms. The maximum electron transport rate (ETRm) and maximal light utilization efficiency (α) were generally positively correlated and showed large diel variability. These parameters fluctuated significantly from hour to hour within each day and the intraday pattern of variation changed significantly among days of each sampling period. Stepwise multiple linear regressions analyses indicated that light fluctuations explained a part of this variability but a great part of variability stayed unexplained. F v/F m, ETRm and α were not only dependent on the light conditions of the sampling day but also on those of the previous days. A time lag of 3 days in the effect of light on ETRm and α variation was highlighted. At these time scales, changes in phytoplankton community structure seemed to have a low importance in the variability in photosynthetic parameters. The photoacclimation index E k showed a lower variability and was generally different from the incident irradiance, indicating a limited acclimation capacity with a poor optimization of light harvesting during the day. However, in well-mixed systems such as the Strait of Dover, the short-term photoacclimation is disrupted by the high level of variability in environmental conditions. Also, the variability observed in the present study can be associated with a particular kind of photosynthetic response: the “E k-independent” variability. The physiological basis of this photosynthetic response is largely unresolved and further researches on this subject are still required to better explain the dynamics of phytoplankton activity in the Strait of Dover.  相似文献

The temperate diatom Skeletonema costatum (Grev.) Cleve was grown in low temperature and/or low light conditions. The cultures were acclimatized for at least three months before experiments were begun. Our data indicate that the initial slope of the photosynthesis vs irradiance curve () is controlled predominantly by light history and the light-saturated photosynthesis (P max) by temperature. The number of divisions per day decreased with decreasing light intensity, but was identical for cultures grown at 3° or 18°C. The metabolic pathways of inorganic carbon fixation were not fundamentally affected by low temperature or low light intensity, but both these factors increased labelling of C3 compounds, synthesized by the Calvin-Benson cycle, and decreased that of phosphoenolpyruvate (PEP) and other metabolites. This indicates an enhancement of ribulose-1,5-bisphosphate (RuBP) carboxylase activity, which is the first step in the C3 pathway (3-phosphoglycerate and sugar phosphate synthesis); this may optimize cell functions. At low temperatures, a seven-fold increase in RuBP carboxylase activity per cell was observed. S. costatum is able to adapt to low irradiance by increasing and decreasing I k (the ratio of P max:, light intensity at onset of light saturation), and to low temperature by increasing its cellular chlorophyll a and RuBP carboxylase content. However, in the latter case, adaptation is not optimal. This study revealed two main features: (1) there is evidence that RuBP carboxylase has a key function in adjustment to high rates of photosynthesis at suboptimal temperatures or irradiances; (2) adaptive mechanisms are dynamic processes and the role of the time scale in physiological adaptation should be considered.  相似文献

Diel periodicity of photosynthesis in marine phytoplankton   总被引:3,自引:0,他引:3  
Short-term changes in photosynthesis were documented for 17 of 24 marine phytoplankton species, representing a range of taxonomic groups. Periodicity in phytoplankton photosynthesis on light-dark cycles (diel periodicity) was widespread but not universal for the species studied. The centric diatoms Lauderia borealis, Ditylum brightwellii, Stephanopyxis turris, Coscinodiscus rex, Chaetoceros gracile, and Biddulphia mobiliensis had strong diel periodicity in photosynthetic capacity (P max). Amplitudes of the daily variations ranged from 2.9 to >50, with maxima in the morning or near midday, and with minima during the dark period, and these variations were not dependent on changes in cell pigmentation. There was some evidence for sustained photosynthetic periodicity in constant conditions in several diatoms, and an endogenous rhythm may have been present. The photosynthesis-irradiance (P-I) relationship was time-dependent for representative marine diatoms, with both the initial slope () and the asymptote (P max) of P-I curves exhibiting significant synchronous diel oscillations. Moreover, detailed studies of the amplitude and timing of photosynthetic periodicity for the diatoms L. borealis and D. brightwellii demonstrated large temporal variations in photosynthesis with morning maxima. These P-I oscillations are discussed with reference to models of primary production which use the relationship between photosynthesis and light as a component of predictive equations for phytoplankton growth in the sea.  相似文献

Photosynthetic parameters for netplankton (>22 m) and nanoplankton (<22 m) varied over similar ranges but exhibited different seasonal and geographic patterns of variation. Nanoplankton a was relatively constant (0.06 mg C [mg Chl · h]-1 [E m-2 s-1]-1), but P m B (mg C [mg Chl · d]-1) was an exponential function of temperature independent of nutrient concentration and vertical stability in the euphotic zone. The temperature function gives a P m B of 24 at 25°C for nanoplankton growing in an estuarine environment characterized by high nutrient concentrations and a shallow, stratified euphotic zone. Variations in netplankton a and P m B were less predictable and were not correlated with temperature, nutrients or vertical stability. Chain forming diatoms with small cells were able to achieve high (0.10 to 0.15) and P m B (20 to 24) that were 3 to 5 times higher than large-celled diatoms and dinoflagellates were able to achieve.  相似文献

In August 1984, hourly measurements of photosynthetic characteristics were carried out during 96 h, at 5 and 10 m, on a natural population of phytoplankton in the St Lawrence Estuary. Synchronous circadian variations of similar amplitude (max./min.: 2 to 3) were observed at the two depths in both the photosynthetic capacity (P m B ) and the photosynthetic efficiency (B). Maximum values occurred at around noontime and minima during the night. Estimates of daily specific productivity were computed with and without the observed circadian variability. Seasonal differences in the locomotor activity pattern of Podarcis sicula held in constant conditions in the laboratory were found to be associated with systematic differences in both the freerunning period of locomotor rhythms () and the circadian activity time (). In order to establish whether the pineal played a role in the control of seasonal changes in circadian parameters, we examined the effects of pinealectomy in constant conditions on the locomotor behavior of lizards displaying the bimodal activity pattern typical of summer. In most lizards pinealectomy lengthened , shortened and abolished the bimodal activity pattern. Pinealectomy induced a sudden transition from the typical locomotor behavior of summer, characterized by a marked bimodal pattern, short and long , to the typical locomotor behavior of autumn, characterized by a unimodal pattern, a long and short . These results demonstrate that the pineal plays a central role in the seasonal reorganization of the circadian system that occurs in P. sicula.  相似文献

Availability of colloidal ferric oxides to coastal marine phytoplankton   总被引:5,自引:0,他引:5  
Cell growth of a coastal marine diatom, Phaeodactylum tricornutum (stock cultures), and two red tide marine flagellates, Heterosigma akashiwo and Gymnodinium mikimotoi (stock cultures), in the presence of soluble chelated Fe(III)-EDTA (1:2) and of four different phases of ferric oxide colloids were experimentally measured in culture experiments at 20°C under 3000 lux fluorescent light. Soluble Fe(III)-EDTA induced the maximal growth rates and cell yields. The short-term uptake rate of iron by H. akashiwo in Fe(III)-EDTA medium was about eight times faster than that in solid amorphous hydrous ferric oxide (Fe2O3·xH2O) medium. In culture experiments supplied with four different ferric oxide forms, the orders of cell yields are amorphous hydrous ferric oxide>-FeOOH (lepidocrocite)>Fe5O7(OH)·4H2O (hydrated ferric oxyhydroxide polymer >-FeOOH (goethite). The specific growth rates () at logarithmic growth phase in Fe(III)-EDTA, amorphous hydrous ferric oxide and -FeOOH media were significantly greater than those in Fe5O7 (OH)·4H2O and -FeOOH media. The thermodynamically stable forms such as Fe5O7(OH)·4H2O and -FeOOH supported a little or no phytoplankton growth. The iron solublities and/or proton-promoted iron dissolution rates of these colloidal ferric oxides in seawater at 20°C were determined by simple filtration techniques involving -activity measurements of 59Fe. The orders of solubilities and estimated dissolution rate constants of these ferric oxides in seawater were consistent with that of cell yields in the culture experiments. These results suggest that the availability of colloidal iron to provide a source of iron for phytoplankton is related to the thermodynamic stability and kinetic lability of the colloidal ferric oxide phases, which probably control the uptake rate of iron by phytoplankton.  相似文献

Species diversity and trophic structure of plankton communities have been considered as most important biological features. The Fisher () and Simpson () indices and the information index H=- pi log2 pi (Margalef) were used for evaluation of species diversity. The indices have been calculated within the group of filter feeders by numbers ( and H) and biomass ( and H) of the species; the best results were obtained in the latter case. The index H=- pi log2 pi is proposed for the quantitative evaluation of trophic structure, where a portion of an individual trophic group is taken as pi. The plankton material was collected by the R.V. Vityaz in the northern Indian Ocean in January through February, 1960. Analysis of characteristics of zooplankton distribution, species and trophic diversity in connection with characteristics of the water regime resulted in the definition of 3 types of the plankton communities: (a) those in intensive divergence zones; (b) those in poor divergence zones; (c) those in regions with stable water stratification or with feebly marked convergence, distiguished by different values of biomass, species diversity, and trophic group ratios.  相似文献
