Biomonitoring studies have increased as a consequence of risks and effects to human health on exposure to environmental contaminants, mainly air pollutants. Genetic biomarkers are useful tools for the early assessment of exposure to occupational and environmental pollution. The objective of the present study was to investigate genotoxic effects on people residing and/or working downwind from an oil refinery in southern Brazil and the mutagenic activity of airborne particulate matter (PM10). Samples of peripheral blood and buccal mucosa cells were evaluated using the single-cell gel electrophoresis assay (comet assay) and the micronucleus (MN) assay, respectively. PM10 samples were collected in the target site and the organic matter extraced with dichloromethane was assessed for mutagenic activity in the Salmonella/microsome assay. The exposed group (n = 37) was compared to a reference group (n = 37) of subjects living in an urban area with limited traffic and industrial influence, located far from the main industrial areas. All PM10 organic extracts showed mutagenic positive responses and the effect decreased in the presence of S9 mix indicating that the predominant compounds present were direct-acting mutagens. The responses of YGs strains are consistent with aromatic amines and nitroarenes being present in the PM10 extracts. The group in the area under the influence of the oil refinery (exposed group) showed significantly higher DNA damage in lymphocytes than the reference group. The MN frequencies in buccal mucosa were very low for both groups and no difference between groups was observed. No association was found between age and tobacco smoking habit and level of DNA damages measured by the comet assay. The results indicate that the comet assay was a sensitive tool to detect DNA damage in subjects under the influence of an oil refinery, with marked genotoxic activity in the atmospheric environment. 相似文献

To estimate the human health risk of inhaled diesel particles, it is necessary to know their deposition and retention in the respiratory tract and the rate of dissociation of mutagenic compounds associated with the particles. The deposition of a chain aggregate aerosol of 67Ga2O3 with size and shape characteristics similar to diesel exhaust particles has been evaluated using Beagle dogs. Approximately one-third of the inhaled activity is deposited in the respiratory tract with most of the particles deposited in the lung. The mutagenic activity present in dichloromethane, dog serum, dog lung lavage fluid, saline, dipalmitoyl lecithin (DPL) and albumin following incubation of these fluids with diesel exhaust particles was determined in the Ames Salmonella system. As observed by other investigators, large quantities of mutagenic activity were removed by dichloromethane. A very small amount of mutagenic activity was removed by the serum and lavage fluid over a 3-day incubation period. No activity was detected following elution with the other solvents. The finding that minimal mutagenic activity could be demonstrated in the biological media following incubation with diesel exhaust particles may be due to a lack of removal of mutagens from the particles or an inactivation of removed mutagens by protein binding or other processes. 相似文献

Mutagenic activity in diesel emission particulate extracts was detected by the Salmonella typhimurium/microsome assay. Direct-acting mutagens as well as promutagens requiring metabolic activation were detected. The extracts were fractionated into acidic, basic, and neutral fractions, and the neutral fraction was chromatographed into seven subfractions. Differences in the mutagenic potency of these fractions and subfractions were determined by the Salmonella assay. Fractions containing as yet unidentified compounds, but not polynuclear aromatic hydrocarbons, were found to make a major contribution to mutagenic activity of the extracts. 相似文献

Nitrobacter toxicity experiments were conducted by using a mixed culture obtained from biomass on rotary biological contactors. The culture was treated with inorganic and organic toxicants, and the toxic effect was expressed in nitrite consumption. Contact time was found to be an important factor in toxic effect. At low metal concentrations, a short exposure time resulted in stimulating effect to Nitrobacter, while prolonged exposure reversed the effect to an inhibiting one. For organic toxicants, the toxicity appeared to decrease as time increased. In general, the time effect can be delineated into three types, accumulative, acclimative, and stimulatory effects. Literature results and the results of this study are used to describe the time effect. 相似文献

One of the essential parameters influencing of the dose conversion factor is the ratio of unattached short-lived radon progeny. This may differ from the value identified for indoor conditions when considering special workplaces such as mines. Inevitably, application of the dose conversion factors used in surface workplaces considerably reduces the reliability of dose estimation in the case of mines.This paper surveyed the concentration of radon and its short-lived radon progeny and identified the unattached fraction of short-lived radon progeny. As well equilibrium factor during the month of August was calculated simultaneously at two extraction faces in a manganese ore mine.During working hours the average radon concentrations were 220 Bq m−3 and 530 Bq m−3 at Faces 1 and 2; the average short-lived progeny concentration was 90 Bq m−3 and 190 Bq m−3, the average equilibrium factors were 0.46 and 0.36, and the average unattached fractions were 0.21 and 0.17, respectively. The calculated dose conversion factor was between 9 and 27 mSv WLM−1, but higher values could also be possible. 相似文献

Although amaranth is consumed in large quantities as a food dye, its mutagenicity and/or carcinogenicity is still controversial. Therefore, the investigation of the genetic activity induced by this dye might assist in the evaluation of its possible dangers to human health. Both bacteria and yeast strains were used for testing the mutagenic activity of amaranth. When the fluctuation test was used, amaranth was found to enhance both forward and reverse mutations in bacterial strains that contain R-plasmids. Amaranth was not found to induce any genetic activity in yeast cells at 28 °C but a significant increase in mitotic gene conversions was recorded at 37 °C in both acidic and neutral media. It has been postulated that amaranth has optimal action on microbial cells at 37 °C and should be considered a weak mutagen. 相似文献

Carcinogenic and mutagenic compounds, which were extracted from the particulates that adhered to inner surfaces of diesel and gasoline engine mufflers, were quantified by the series method of Soxhlet extraction, liquid-liquid partition, thin-layer chromatography, and spectrofluorometry. Mutagenic activity of their neutral and acidic fractions was tested in the improved Ames assay by the preincubation method with Salmonella typhimurium TA98 in the presence and absence of metabolic activation system (S-9 mix). The average content levels (μg/g tar) of polycyclic aromatic hydrocarbons from gasoline engine cars were greater than those from diesel engine vehicles. However, the levels of nitro derivatives of PAHs and polycyclic quinones from the diesel engines were greater than from the gasoline engines. Mutagenic activity of the diesel acidic fraction was the highest among the diesel and gasoline fractions, and was significantly higher in the absence of the S-9 mix. Furthermore, the relative value (Rc = 0) of infrared absorption of carbonyl stretching vibration to that of methylene asymmetric stretching vibration of the diesel acidic fraction was the highest among the diesel and gasoline fractions. These results strongly suggest that highly direct-acting mutagens in the acidic fraction are at higher levels in diesel emission particulates than those from gasoline, and that these mutagens are carboxylic acid, aldehyde, and alcohol derivatives of PAHs and NPAHs. 相似文献

Macro- and microeconomic evidence suggests a positive role of remittances in preparing households against natural disasters
and in coping with the loss afterward. Analysis of cross-country macroeconomic data shows that remittances increase in the
aftermath of natural disasters in countries that have a larger number of migrants abroad. Analysis of household survey data
in Bangladesh shows that per capita consumption was higher in remittance-receiving households than in others after the 1998
flood. Ethiopian households that receive international remittances seem to rely more on cash reserves and less on selling
household assets or livestock to cope with drought. In Burkina Faso and Ghana, international remittance-receiving households,
especially those receiving remittances from high-income developed countries, tend to have housing built of concrete rather
than mud and greater access to communication equipment, suggesting that they are better prepared against natural disasters. 相似文献

Data are presented on the mutagenicity of an organic extract of a composite sample of urban air particulates and of thirty PAH compounds in such samples, including four quinone derivatives, isolated quantitatively by thin-layer chromatography and identified by fluorescence or other spectral techniques. Mutagenic activity was determined by the Ames assay, using histidine auxotrophs of Salmonella typhimurium strains TA98, TA100, TA1537 and TA1538. All compounds were dissolved in dimethyl sulfoxide (DMSO) which was the least toxic of eight organic solvents tested.The mutagenic activity of a benzene extract of suspended particulates from the air of Hamilton, Ontario was significantly greater with strain TA98 than with strain TA100, suggesting the presence of more frame-shift acting mutagens. The mutagenic response of this extract was similar with and without S-9 activation.Mutagenic tests on the 30 PAH compounds indicated that only the benzo(a)pyrene quinones were direct acting mutagens. All of the chemical compounds were tested with and without S-9 activation. The following PAH showed unequivocal mutagenic activity, with S-9 activation: benz(a)anthracene, benzo-(a)pyrene, benzo(ghi)perylene, benzo(rst)pentaphene, benzophenanthrene, chrysene, 1, 2, 3, 4-dibenzanthracene, 2, 3, 6, 7-dibenzanthracene, and 3-methyl cholanthrene. The quinones of 1, 6-; 3, 6-; and 6, 12-benzo(a)pyrenes showed weak mutagenic activity but 3, 6-benzo(a)pyrene elicited also a photodynamic response. 相似文献

Global mitigative and adaptive efforts have not been able to effectively address the adverse impacts caused by climate change. Therefore, a direct solution is needed to address the significant resulting loss and damage (L&D). During the United Nations Framework Convention on Climate Change held in Doha in 2012, the issue of responding to L&D arising from climate change gained sudden traction and became one of the key issues that affected the outcome of the convention. In this paper, a study on the definition and connotations of L&D arising from climate change was conducted, together with an analysis of its relationship with related concepts, namely impacts, vulnerability, and risks. This led to the proposal of an L&D conceptual model that is more comprehensive, with the recognition of the need to address the issue through effective supplementation of existing mitigative and adaptive efforts. A systematic elaboration of an L&D response mechanism was made based on politics, law, and the market, leading to a preliminary presentation of a possible format for an L&D mechanism. Potential academic research directions for L&D were also proposed that could serve as references for the establishment of international and national L&D response mechanisms and related research. 相似文献

Iodine-131 reaches the marine environment through its excretion to the sewer by nuclear medicine patients followed by discharge through coastal and deepwater outfalls. 131I has been detected in macroalgae, which bio-accumulate iodine, growing near the coastal outfall of Cronulla sewage treatment plant (STP) since 1995. During this study, 131I levels in liquid effluent and sludge from three Sydney STPs as well as in macroalgae (Ulva sp. and Ecklonia radiata) growing near their shoreline outfalls were measured. Concentration factors of 176 for Ulva sp. and 526 for E. radiata were derived. Radiation dose rates to marine biota from 131I discharged to coastal waters calculated using the ERICA dose assessment tool were below the ERICA screening level of 10 μGy/hr. Radiation dose rates to humans from immersion in seawater or consumption of Ulva sp. containing 131I were three and two orders of magnitude below the IAEA screening level of 10 μSv/year, respectively. 相似文献

In the studies reported here, the micronucleus assay, a common cytogenetic technique, was used to examine the dose-responses in fibroblasts from three ungulate species (white-tailed deer, woodland caribou, and Indian muntjac) exposed to high doses of ionizing radiation (1-4 Gy of (60)Co gamma radiation). This assay was also used to examine the effects of exposure to low doses (1-100 mGy) typical of what these species experience in a year from natural and anthropogenic environmental sources. An adaptive response, defined as the induction of resistance to a stressor by a prior exposure to a small "adapting" stress, was observed after exposure to low doses. This work indicates that very small doses are protective for the endpoint examined. The same level of protection was seen at all adapting doses, including 1 radiation track per cell, the lowest possible cellular dose. These results are consistent with other studies in a wide variety of organisms that demonstrate a protective effect of low doses at both cellular and whole-organism levels. This implies that environmental regulations predicated on the idea that even the smallest dose of radiation carries a quantifiable risk of direct adverse consequences to the exposed organism require further examination. Cytogenetic assays provide affordable and feasible biological effects-based alternatives that are more biologically relevant than traditional contaminant concentration-based radioecological risk assessment. 相似文献

We analyze longtime series of annual snout positions of several valley glaciers in the northwestern Italian Alps, together with a high-resolution gridded dataset of temperature and precipitation available for the last 50 years. Glacier snout fluctuations are on average negative during this time span, albeit with a period of glacier advance between about 1970 and 1990. To determine which climatic variables best correlate with glacier snout fluctuations, we consider a large set of seasonal predictors, based on our climatic dataset, and determine the most significant drivers by a stepwise regression technique. This in-depth screening indicates that the average glacier snout fluctuations strongly respond to summer temperature and winter precipitation variations, with a delay of 5 and 10 year, respectively. Snout fluctuations display also a significant (albeit weak) response to concurrent (same year) spring temperature and precipitation conditions. A linear regressive model based on these four climatic variables explains up to 93 % of the variance, which becomes 89 % when only the two delayed variables are taken into account. When employed for out-of-sample projections, the empirical model displays high prediction skill, and it is thus used to estimate the average glacier response to different climate change scenarios (RCP4.5, RCP8.5, A1B), using both global and regional climate models. In all cases, glacier snout fluctuations display a negative trend, and the glaciers of this region display an accelerated retreat, leading to a further regression of the snout position. By 2050, the retreat is estimated to be between about 300 and 400 m with respect to the current position. Glacier regression is more intense for the RCP8.5 and A1B scenarios, as it could be expected from the higher severity of these emission pathways. 相似文献

BackgroundPhysical activity (PA) has beneficial, whereas exposure to traffic related air pollution (TRAP) has adverse, respiratory effects. Few studies, however, have examined if the acute effects of TRAP upon respiratory outcomes are modified depending on the level of PA.ObjectivesThe aim of our study was to disentangle acute effects of TRAP and PA upon respiratory outcomes and assess the impact of participants TRAP pre-exposure.MethodsWe conducted a real-world crossover study with repeated measures of 30 healthy adults. Participants completed four 2-h exposure scenarios that included either rest or intermittent exercise in high- and low-traffic environments. Measures of respiratory function were collected at three time points. Pre-exposure to TRAP was ascertained from land-use-modeled address-attributed values. Mixed-effects models were used to estimate the impact of TRAP and PA on respiratory measures as well as potential effect modifications.ResultsWe found that PA was associated with a statistically significant increases of FEV1 (48.5 mL, p = 0.02), FEV1/FVC (0.64%, p = 0.005) and FEF25–75% (97.8 mL, p = 0.02). An increase in exposure to one unit (1 μg/m3) of PMcoarse was associated with a decrease in FEV1 (− 1.31 mL, p = 0.02) and FVC (− 1.71 mL, p = 0.01), respectively. On the other hand, for an otherwise equivalent exposure an increase of PA by one unit (1%Heart rate max) was found to reduce the immediate negative effects of particulate matter (PM) upon PEF (PM2.5, 0.02 L/min, p = 0.047; PM10, 0.02 L/min p = 0.02; PMcoarse, 0.03 L/min, p = 0.02) and the several hours delayed negative effects of PM upon FVC (PMcoarse, 0.11 mL, p = 0.02). The negative impact of exposure to TRAP constituents on FEV1/FVC and PEF was attenuated in those participants with higher TRAP pre-exposure levels.ConclusionsOur results suggest that associations between various pollutant exposures and respiratory measures are modified by the level of PA during exposure and TRAP pre-exposure of participants. 相似文献

基于海洋经济发展进程的生态环境演化模型,建立了环渤海地区海洋经济与生态环境发展水平评价指标体系,通过构建响应度模型定量分析2004至2014年环渤海地区海洋经济发展的海洋生态环境响应变化及影响因素,得出以下结论:(1)通过构建海洋经济与生态环境发展水平评价指标体系及综合评价模型进行测算发现,环渤海地区海洋经济发展综合评价值由2004年的0.135提升至2014年的0.850,表明环渤海地区海洋经济发展水平不断提高,并呈现出良好的发展势头;海洋生态环境发展综合评价值先由2004年的0.647降至2012年的0.335,后又缓慢回升至2014年的0.374,表明环渤海地区海洋生态环境水平虽在2012至2014年间略有好转,但总体仍呈波动下降趋势。(2)通过构建海洋生态环境响应度模型进行测算发现,环渤海地区海洋经济发展的生态环境响应指数由2004年的-0.073下降至2012年的-0.576,而后又增大至2014年的-0.297,表明环渤海地区海洋经济发展对生态环境演变产生了"胁迫"影响,总体呈现出由"弱胁迫"向"强胁迫"变化的态势,但胁迫程度在2012至2014年间有所缓和。(3)通过多元回归分析发现,与海洋生态环境响应指数的偏回归系数较高的3个自变量分别为海洋科技课题成果应用数量、海域利用效率、单位面积工业废水排放达标量,且海域利用效率的回归系数最高为0.749。表明海域利用效率是影响环渤海地区海洋生态环境响应演变的主要因素,此外,海洋科技发展水平的提高以及海洋污染治理力度加强等因素也对胁迫程度的减小具有一定的推动作用。 相似文献

It is essential to investigate hydrologic responses to climate change and human activities across different physiographic regions so as to formulate sound strategies for water resource management. Mann–Kendall, wavelet and geospatial analyses were coupled in this study, associated with ENSO indicators, flashiness index and baseflow index, in order to explore the hydrologic sensitivity to climate change and human activities in the Jiulong River Basin (JRB), a subtropical coastal watershed of southeast China. The results showed that the average annual precipitation presented an increasing trend (Z = 2.263, p = 0.024) and that this tendency has become weaker from estuary to inland in the JRB over the past 50 years. The annual frequency of rainstorm events increased from 3.4 to 5.2 days in the estuary and from 5.1 to 5.6 days in the West River, whereas it decreased from 6.0 to 5.5 days in the North River from 1954 to 2010. The 10-year average streamflow during 2001–2010 in the North River and West River decreased by 9.2 and 6.7 %, respectively, compared to the average annual streamflow during 1967–2000. Annual fluctuations were the most representative signals in streamflow variability for the North River and West River over the period 1967–2010. Human activities including dam construction, land change and socioeconomic development posed increasing influences on hydrologic conditions in the JRB. Seasonal variability of streamflow and sediment discharge changed significantly between the two periods divided by the jumping point (1992), identified when dams were constructed extensively in the North River and West River. This research provided important insights into the hydrologic consequences of climate change and human activities in a subtropical coastal watershed of southeast China. 相似文献

Periphyton communities can be used as monitors of ecosystem health and as indicators of contamination in lotic systems. Measures of biomass, community structure, and genetic diversity were used to investigate impacts of uranium (U) exposure on periphyton. Laboratory exposures of periphyton in river water amended with 238U were performed for 5 days, followed by 2 days of U depuration in unamended river water. Productivity as measured by biomass was not affected by concentrations up to 100 microg238U L(-1). Phospholipid fatty acid (PLFA) profiles and denaturing gradient gel electrophoresis (DGGE) banding patterns revealed no changes in community or genetic structure related to U exposure. We suggest that the periphyton community as a whole was not significantly impacted by exposures of 238U up to a concentration of 100 microgL(-1). These findings have significance for the assessment and prediction of U impacts on aquatic ecosystems. 相似文献

As a guide to the possible effects of a CO2-induced warming on the cryosphere, we review the effects of three warm periods in the past, and out theoretical understanding of fluctuations in mountain glaciers, the Greenland and Antarctic ice sheets, ground ice, sea ice and seasonal snow cover. Between 1890 and 1940 A.D. the glaciated area in Switzerland was reduced by over 25%. In the Hypsithermal, at about 6000 BP, ground ice in Eurasia retreated northward by several hundred kilometres. In the interglacial Stage 5e, at about 120 000 BP, global sea-level rose by over 6 m. Fluctuations of mountain glaciers depend on mesoscale “weather” and on their mechanical response to it. Any melting of the Greenland ice sheet is likely to be slow in human terms. The West Antarctic ice sheet (its base below sea-level) is susceptible to an ungrounding, and such an event may have been the cause of the sea-level rise above. The East Antarctic ice sheet is susceptible to mechanical “surges”, which might be triggered by a warming at its margin. Both an ungrounding and a surge might occupy less than 100 yr, and are potetially the most important ice changes in human terms. Modelling studies suggest that a 5°C warming would remove the Arctic pack ice in summer, and this may be the most significant effect for further climatic change. 相似文献
