Karasu River, which is the only river in the Erzurum plain, is the source of the Euphrates River (Eastern Anatolia of Turkey). The river is in a serious environmental situation as a result of pollution by agricultural and industrial sewage and domestic discharges. The present study aims to evaluate genotoxic effects of toxic metals in chub, Leuciscus cephalus, and transcaucasian barb, Capoeta capoeta, collected from contaminated site of the Karasu River, in comparison with fish from an unpolluted reference site. Heavy metal concentrations in surface water of the river were determined. The condition factor (CF) was taken as a general biomarker of the health of the fish, and genotoxicity assays such as micronucleus (MN) and other nuclear abnormalities (NA) were carried out on the fish species studied. MN and NA such as kidney-shaped nucleus, notched nucleus, binucleated, lobed nucleus, and blebbed nucleus were assessed in peripheral blood erythrocytes, gill epithelial cells, and liver cells of the fish. A significant decrease in CF values associated with a significant elevation in MN and NA frequencies was observed in fish collected from the polluted sites compared with those from the reference site. Results of the current study show the significance of integrating a set of biomarkers to identify the effects of anthropogenic pollution. High concentrations of heavy metals have a potential genotoxic effects, and the toxicity is possibly related to industrial, agricultural, and domestic activities.  相似文献

The present study investigates the response of three hepatic biomarkers in adult sea bass (Dicentrarchus labrax, Linnaeus 1758) caged at a wastewater outlet of an oil refinery with fish caged at a pristine site used as controls. The biomarkers that were investigated were the hepatosomatic index (HSI), 7-ethoxyresorufin-O-deethylase (EROD) activity and glutathione-S-transferase (GST) activity. In addition, we have measured the levels of polycyclic aromatic hydrocarbons (PAHs) and selected heavy metals (lead, cadmium, mercury, copper and zinc) in sediment samples at the polluted site. Although the polluted site had high environmental levels of PAHs and heavy metals, there was no difference in hepatic EROD activity and HSI between fish caged at the polluted site and controls. On the other hand, GST activity was significantly lower in fish caged at the polluted site compared to controls. Our results point out that the studied biomarkers have limited use in environmental risk assessment studies, at least when caged adult sea bass is used as the sentinel species and complex toxicant mixtures are involved.  相似文献

The objective of this study was to investigate the impact of anthropogenic activities on soil quality using the land snail Helix aspersa as a bioindicator. Soil samples and snails were collected from several sites in Northeast Algeria during the summer and winter of 2010. All of the sites were chosen due to their proximity to industrial factories—a potential source of soil pollution via heavy metal contamination. The concentration of heavy metals (Pb, Cd, Mn, and Fe) in soil samples was analyzed using atomic absorption spectrophotometry. Activity levels of glutathione S-transferase (GST) and acetylcholinesterase (AChE), indicators of oxidative stress and neurotoxicity, respectively, were measured in snails collected from each site. GST and AChE activity were found to vary between sites and by season. The highest levels of GST activity were registered during the summer at sites closest to potential sources of pollution. AChE activity levels also peaked during the summer with the highest values recorded at the site in El Hadjar. These increased levels of bioindicative stress response correlated with increasing metal concentration in soil samples collected at each site.  相似文献

The aim of this study is to summarise the determination of concentrations of microcystins (MCs) in muscle and liver of freshwater fish species caught in stagnant waters of the Czech Republic. Within the years 2007–2009, 351 muscle samples and 291 liver samples of 16 freshwater fish species derived from four fishponds, and four water reservoirs were analysed. MCs were detected in 53 liver samples. The highest concentrations of microcystins were determined in liver samples of carnivorous fish species; 50.3 ng/g of fresh weight (FW) in perch (Perca fluviatilis) and 22.7 ng/g FW in pikeperch (Sander lucioperca). MCs in liver were detected in other five fish species; asp (Aspius aspius), pike (Esox lucius), common carp (Cyprinus carpio), grass carp (Ctenopharyngodon idella) and European eel (Anguilla anguilla). Concentrations of MCs in liver of nine fish species (European bream, whitefish, tench, silver carp, European catfish, roach, chub, crucian carp and rudd) were below the detection limit of 1.2–5.4 ng/g FW for different MC congeners. However, the concentrations of MCs in all muscle samples were below the detection limit. The assessment of MCs concentrations might be influenced by the detection method used. Due to the concentrations of MCs being below the detection limit in muscle samples of all fish species analysed, it seems that there might be a low potential threat for human health in case of fish muscle consumption.  相似文献

The present study reports the first analysis of water pollutants in Sri Lankan waters using a suite of biomarkers in Nile tilapia (Oreochromis niloticus) residing in Bolgoda Lake which receives urban, industrial and domestic wastes from multiple sources. The fish were collected from the lake in the dry period (April 2005) and wet periods (September 2005, October 2006) and the levels of biomarkers viz. hepatic ethoxyresorufin O-deethylase (EROD), glutathione S-transferase (GST), metallothioneins, biliary fluorescent aromatic compounds, brain and muscle cholinesterases (ChE) were compared with those of the laboratory reared control fish and the fish obtained from a less polluted water body, Bathalagoda reservoir (reference site). The results revealed that biomarker levels of the fish collected from the reference site were not significantly different from the controls. Hepatic EROD and GST activities in fish from Bolgoda Lake were induced 4.2-16.6 folds and 1.4-3.3 folds respectively compared with the control fish. Analysis of bile in the lake fish revealed recent uptake of naphthalene, pyrene and benzo(a)pyrene type polycyclic aromatic hydrocarbons (PAHs). The induction of EROD activities in feral fish reflects the exposure of fish to aryl hydrocarbon receptor agonists including PAHs present as pollutants in the Bolgoda Lake. Cholinesterase activity in the fish inhabiting one sampling site of Bolgoda Lake was lower (22-40% inhibition) than the activity measured in the control fish indicating the presence of anticholinesterase pollutants in the area. Hepatic metallothionein levels in the lake fish were higher (1.9-3.2 folds) in comparison to the controls indicating metal exposure. The results support the potential use of these biomarkers in Nile tilapia in assessing pollution in tropical water bodies.  相似文献

Populations of invasive fishes quickly reach extremely high biomass. Before control methods can be applied, however, an understanding of the contaminant loads of these invaders carry is needed. We investigated differences in concentrations of selected elements in two invasive carp species as a function of sampling site, fish species, length and trophic differences using stable isotopes (delta (15)N, delta (13)C). Fish were collected from three different sites, the Illinois River near Havana, Illinois, and two sites in the Mississippi River, upstream and downstream of the Illinois River confluence. Five bighead carp (Hypophthalmichthys nobilis) and five silver carp (Hypophthalmichthys molitrix) from each site were collected for muscle tissue analyses. Freshwater mussels (Amblema plicata) previously collected in the same areas were used as an isotopic baseline to standardize fish results among sites. Total fish length, trophic position, and corrected (13)C, were significantly related to concentrations of metals in muscle. Fish length explained the most variation in metal concentrations, with most of that variation related to mercury levels. This result was not unexpected because larger fish are older, giving them a higher probability of exposure and accumulation of contaminants. There was a significant difference in stable isotope profiles between the two species. Bighead carp occupied a higher trophic position and had higher levels of corrected (13)C than silver carp. Additionally bighead carp had significantly lower concentrations of arsenic and selenium than silver carp. Stable isotope ratios of nitrogen in Asian carp were at levels that are more commonly associated with higher-level predators, or from organisms in areas containing high loads of wastewater effluent.  相似文献

Nuclear bud (NB) formation was investigated in blood erythrocytes of 1892 flounder Platichthys flesus, herring Clupea harengus, and eelpout Zoarces viviparus specimens that were collected at 82 sites representing different regions of the Baltic Sea in 2009–2011. This is the first attempt to evaluate the baseline levels of NB and rank the genotoxicity risk for native fish species. NB levels were compared to the previously published micronuclei (MN) data from the same individual fish specimens in order to compare the two methods of genotoxicity assessment and investigate the relationship between MN as the cytogenetic measure of genotoxicity and the DNA damage reflecting NB. In 2009–2011, elevated NB levels in 89.4 % of flounder sampling groups indicated high and extremely high genotoxicity risk levels. Herring and eelpout sampling groups showed elevated levels of NB, 74.6 and 45.7 %, respectively. In general, herring and eelpout NB measure was more sensitive as the genotoxicity biomarker than MN.  相似文献

The Yellow River, the second largest river in China, is the most important resource of water supply in North China. In the last 40 years, even in the upper Yellow River, with the development of industry and agriculture, more and more contaminants have been discharged into this river and greatly polluted the water. Although a routine chemical component analysis has been performed, little is known about the real toxic effects of the polluted water on organisms at environmental level. To explore whether the pollutants induced oxidative stress and damage to aquatic organisms, malondialdehyde (MDA) level and activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione S-transferase (GST) in hepatopancreas, kidney and intestine of the field-collected carp Cyprinus carpio from a mixed polluted (Lanzhou Region, LZR) and a relatively unpolluted (Liujiaxia Region, LJXR) sites of the upper Yellow River were measured. The results showed that when the values of LZR compared with those of LJXR, SOD and GST activities increased and GPx activity decreased significantly in all the three organs (P < 0.05–0.01); CAT activity decreased but MDA level increased significantly (P < 0.05–0.01) only in kidney and intestine. In conclusion, the results of this study suggest that the pollutants can induce obvious oxidative damage in the carp, and the SOD, GST and GPx might be better indicators for the oxidative damage in aquatic organisms.  相似文献

以松花江流域的野生鱼类为研究对象,在松花江流域选取松林、大顶子山、佳木斯3个具有代表性的点位,在2014—2016年的每年5月底6月初采集鲤鱼和鲫鱼样品,3年内共制备45个鲤鱼单样,44个鲤鱼混样,30个鲫鱼混样,共计119个。采用电感耦合等离子体-质谱法对鱼类肌肉组织中残留的As、Cd、Cr、Pb、Hg 5种重金属进行检测。检测结果显示,As、Cd、Cr、Pb、Hg 5种重金属全部检出,检测结果全部达标,达标率为100%。根据检测结果采用单因子指数法和金属污染指数法等评价了松花江流域鱼体重金属残留状况及食用安全性,评价结果为松花江流域鱼类重金属残留量相对较少,鱼体健康,食用安全。  相似文献

Tropical coastal lagoons are highly productive environments exhibiting high biodiversity. However, the use of these ecosystems by local communities is of concern, since this generally leads to environmental degradation. The Imboassica coastal lagoon, located in Macaé city, in Northern Rio de Janeiro, is an important ecosystem in the state, however, already displaying signs of anthropogenic impacts. Carnivorous fish Hoplias malabaricus specimens were sampled from this impacted site, as well as from a reference area. Fish from Imboassica Lagoon presented lower condition factor, lower cholinesterase activity, and higher percentage of erythrocyte micronuclei when compared to fish from the reference site. Metals in fish from Imboassica Lagoon were always higher than Encantada Lagoon, with some seasonal differences, where some metals were higher in the rainy season compared to the dry season in muscle tissue, with the exception of Cu, Fe, Sr, and Zn; and in the liver, except for Ba, Cd, Cr, Ni, and Sr. Cr and Mn in the edible muscle portion of the fish were higher than the limits established by Brazilian and International legislations as permissible for human consumption, thus leading to concerns regarding public health risks for the local population that use fish as their main protein source.  相似文献

Micronuclei tests is a system of mutagenicity testing used for determining the pollution and chemicals causing changes in DNA fragments such as micronuclei in the cytoplasm of interphase cells. Damage caused on the DNA by genotoxic pollutants is the first consequence occurring in the aquatic organisms. Thus, it was attempted to determine whether pollution affected the erythrocytes and gills of fish Gobius niger and haemolymph and gills of mussels Mytilus galloprovincialis living in Izmir Bay at the level of DNA by the means of micronuclei (MN) test. Organisms used in the MN test were collected from seven locations (Alsancak, Alaybey Shipyard, Karsiyaka, Bostanli, Göztepe, Konak and Pasaport) which are known as the most polluted part of inner Bay of Izmir (Western Coast of Turkey). According to the results of the present study, frequency of MN was found at high level in Alaybey Shipyard and Pasaport where wastes from existing dockyard contributed to high level of pollution. In conclusion, this study indicates that the micronuclei test gives sensitive results in monitoring the pollution, especially the pollution of harbor, and thus it might be used as standard method in regular monitoring of pollution of coastal ecosystem.  相似文献

Activities of glutathione S-transferases (GST) and cholinesterase (ChE) from Paracentrotus lividus were investigated as possible biomarkers of environmental contamination in the coastal zone. In the first phase of the study, the activity of both enzymes was determined in various tissues in order to select the most appropriate ones to be used in the following assays. In the second phase, the ChEs present in ambulacra were characterized using different substrates and selective inhibitors. In the next phase, laboratory bioassays were performed with dilutions of water-accommodated fraction of #4 fuel-oil (WAF) and benzo[a]pyrene (BaP) to determine the response of those enzymes to these pollutants and, finally, the activity of both enzymes was determined during a year in indigenous specimens from six sites on the Northwest coast of Portugal, with different pollution levels, to determine basal values and seasonal variations of ChE and GST activities. Among the several tissues tested, ambulacra and the anterior portion of the intestine were selected for ChE and GST assays, respectively. The determination of ChE in ambulacra tissue may be performed in a non-destructive way. Ambulacra ChE hydrolysed acetylthiocholine preferentially to propionylthiocholine and butyrylthiocholine and, inhibition by excess of substrate was observed. Enzymatic activity was almost fully inhibited by eserine sulfate (>98%) at concentrations equal or higher than 6.25 microM. Sensitivity to both BW284C51 (reaching 98% at 200 microM) and iso-OMPA (73% at 8 mM) was found. In laboratory bioassays, GSTs activity was inhibited by WAF and induced by BaP, whereas ChE activity was not affected by any of these environmental contaminants. Seasonal variations in enzymatic activities were found. For example, in a reference site, ChE values changed from 52.2 +/- 9.3 U mg(-1) protein in autumn to 71.8 +/- 13.3 U mg(-1) protein in spring, while GST activity changed from 129.9 +/- 29.8 U mg(-1) protein in winter to 279.0 +/- 48.0 U mg(-1) protein in autumn. Sea-urchins from reference sites presented significantly higher values of both ChE and GST than animals from contaminated sites in all seasons. In conclusion, the results of this study indicate that (i) ambulacra and the anterior portion of intestine are the most suitable tissues to measure ChE and GST activities, respectively; (ii) only one form of ChE seems to be present in ambulacra, showing properties of both typical acetylcholinesterase (AChE) and pseudocholinesterase (PChE); (iii)P. lividus GST is sensitive to both WAF and BaP even after acute exposures while ChE is not, and (iv) in spite of the significant seasonal variations observed in both enzymes in the field, P. lividus ChE and GST were capable of discriminate sites with different contamination levels and, thus, they are suitable for use as biomarkers in biomonitoring studies in the coastal zone.  相似文献

We assessed the effects on fish of lead (Pb) released to streamsby smelters located in Trail, BC (Canada), E. Helena, MT, Herculaneum, MO, and Glover, MO. Fish were collected by electrofishing from sites located downstream of smelters and from reference sites. Blood from each fish was analyzed for -aminolevulinic acid dehydratase (ALAD) activity and hemoglobin (Hb), and samples of blood, liver, or carcass were analyzed for Pb, zinc (Zn), or both. Fish collected downstreamof all four smelters sites had elevated Pb concentrations, decreased ALAD activity, or both relative to their respectivereference sites. At E. Helena, fish from the downstream site also had lower Hb concentrations than fish from upstream. Differences among taxa were also apparent. Consistent with previous studies, ALAD activity in catostomids (Pisces: Catostomidae-northern hog sucker, Hypentelium nigricans;river carpsucker, Carpiodes carpio; largescale sucker, Catostomus macrocheilus; and mountain sucker, C. platyrhynchus) seemed more sensitive to Pb-induced ALAD inhibition than the salmonids (Pisces: Salmonidae-rainbow trout,Oncorhynchus mykiss; brook trout, Salvelinus fontinalis) or common carp (Cyprinus carpio). Some of these differences may have resulted from differential accumulation of Zn, which was not measured at all sites. We detected no ALAD activity in channel catfish (Ictaluruspunctatus) from either site on the Mississippi River at Herculaneum, MO. Our findings confirmed that Pb is releasedto aquatic ecosystems by smelters and accumulated by fish, andwe documented potentially adverse effects of Pb in fish. We recommend that Zn be measured along with Pb when ALAD activityis used as a biomarker and the collection of at least 10 fish ofa species at each site to facilitate statistical analysis.  相似文献

We analyzed water samples from the confluence of three municipal sewage treatment effluent streams, surface water, and whole carp (Cyprinus carpio) for synthetic musks for a period of 7-12 months. The lipid content of each fish was determined and compared with the concentration of musks in the whole fish tissue. Enhanced methods were used for water sampling and musk extraction. The data presented here provide insight as to the relationship between concentrations of synthetic musks in the municipal effluent and associated biota. This study confirmed the presence of polycyclic and nitro musk compounds in sewage effluent, Lake Mead water, and carp. The concentrations were found to be considerably lower than previous studies conducted in other countries. This study also provides data for polycyclic and nitro musk compounds, as well as some of the nitro musk metabolites in sewage treatment plant effluent, lake water, and carp.  相似文献

Considering that diesel oil is one of the most common aquatic contaminants, we compare the oxidative stress between two species of fish with different habitats (Pterygoplichthys anisitsi, benthic and Oreochromis niloticus, nektonic) exposed to diesel oil. Malondialdehyde concentrations (MDA) and the activities of ethoxyresorufin-O-deethylase (EROD), glutathione S-transferase (GST), catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase were all analyzed in the fishes' livers and gills after 2 and 7 days of exposure to different concentrations of diesel (0.1 and 0.5 mL/L). In the tilapia, MDA levels and the activities of EROD and GST activity in the liver, as well as MDA levels and the activities of GST and SOD in the gill had statistically significant differences between the treatments and between the times of exposure. For the catfish, the same occurred in the case of MDA, EROD, and SOD in the liver and in CAT and SOD in the gills. There were significant differences in the enzyme activity and lipid peroxidation between the species. Although the activity of most enzymes seemed to be more expressive and responsive to diesel in O. niloticus, diesel oil also caused significant effects on oxidative stress parameters in P. anisitsi, even though this species is benthic and thus has less access to insoluble fractions of diesel oil. Therefore, both species can be used as sentinel organisms in environmental biomonitoring of diesel contamination.  相似文献

This study assessed metals in water and different tissues of Labeo rohita and the impact of these metals on DNA and proteins as biomarkers of gills and muscles of these fish from three different polluted sites (reference or low = KW, medium = CH and high = SK) of the Indus River, Pakistan. The Mn, Pb, Cu, Zn, Hg, and Cr levels in water, gills, liver, muscles, and skin of these fish were compared with the international permissible levels. All metals except Pb and Hg in water were within the acceptable limits of drinking water. In contrast, the Mn, Hg, and Cr levels in the fish tissues were higher than their permissible limits for fish as a human food. Here, the gills contained higher metals than the other tissues. Different patterns of biomarkers were found in fish from these sites. While the gills did not show four protein bands (55, 30, 18.4, and 16.4 kDa), the muscles showed four new protein bands (100, 85, 45, and 20 kDa) for fish from the medium and high polluted sites as compared to the reference or low polluted site. The fish from the CH and SK sites of the Indus River contained low molecular weight DNA in their gills but high molecular weight DNA in their muscles when compared with the KW site. This study suggests that the proteins and DNA profiles of L. rohita could be used as biomarkers to assess the impact of potential environmental stressors such as metals on the freshwater systems.  相似文献

We examined the effects of a commonly used preservation technique on mercury concentration in fish tissue. After fixing fish muscle tissue in formalin followed by preservation in isopropanol, we found that mercury concentration in fish muscle tissue increased by 18%, reaching an asymptote after 40 days. We used formalin–isopropanol-preserved longear sunfish (Lepomis megalotis) from the Sam Noble Oklahoma Museum of Natural History to examine historical changes and predict current mercury concentrations in fish from two rivers in southeastern Oklahoma. Glover River was free-flowing, while Mountain Fork River was impounded in 1970 and a coldwater trout fishery was established upstream from the collection site in 1989. Mercury concentrations in longear sunfish from Glover River showed no historical changes from 1963 to 2001. Mercury concentrations in longear sunfish from Mountain Fork River showed no change from 1925 to 1993 but declined significantly from 1993 to 2003. We also compared mercury concentrations of the most recently collected longear sunfish in the museum to mercury concentrations of unpreserved fish collected from the rivers in 2006. Concentrations of mercury in museum fish were not significantly different from mercury concentrations in unpreserved fish we collected from the rivers. Our study indicates that preserved museum fish specimens can be used to evaluate historical changes and predict current levels of mercury contamination in fish.  相似文献

Total mercury (Hg) concentrations were determined by CV-AAS in selected tissues (liver, kidney, and muscle) of the Great Cormorant and some fish species (herring, ruffe, European smelt, tench, roach, Crucian carp, and Prussian carp) from the Vistula Lagoon ecosystem (Poland). Significant correlations between Hg concentrations in the kidneys and muscle of cormorants (U test, p?<?0.05) were found; levels of the metal were highest in the birds?? liver and kidneys. Total Hg concentrations in the fish decreased in the following order: roach > Prussian carp > Crucian carp > tench > European smelt > ruffe > herring. The biomagnification factor of Hg for the cormorant relative to the fish decreased in the following sequence: herring > ruffe > European smelt > tench > Crucian carp > roach = Prussian carp. It was significantly greater than unity, especially for the cormorant?Cherring trophic relationship. This implies that Hg is biomagnified in the cormorant in relation to its prey.  相似文献

The river Ravi, while passing through Lahore, the second largest city of Pakistan, gets highly polluted owning heavy loads of untreated municipal sewage and industrial effluents of diverse kinds. The fish, Catla catla sampled in two different seasons from three downstream polluted sites were compared with the samples of the same fish from an upstream, a less polluted site, for their physico-chemical parameters. The data were statistically analysed to study the effect of sites, seasons and their interaction on the physico-chemical parameters of waters and mineral uptake in fish muscles. Significant differences (P?<?0.001) among the sampling sites and seasons were observed. The river appeared to be polluted as indicated by the high values of total suspended solids (909 mg/l) and sulphate (964 mg/l) in comparison to the respective values of 150 and 600 mg/l being suggested as the safer values of drinking water of the National Environmental Quality Standards. Most trace and macro elements in fish muscles were increased with the increasing pollution loads from the upstream to the downstream sites of this river. The remarkable increases in the levels of all the investigated minerals in fish muscles from the polluted sites raise concerns about the long-term health of the river Ravi ecosystem and consequently the fish and its consumer’s health. The results contradict the opinion of the local population that the riverine fish are natural, more health-promoting and precious than the pond fish. Therefore, we strongly argue for the utilization of an effect-based monitoring approach to alleviate the detrimental effects of anthropogenic activities on fish and the fish consumers’ health.  相似文献
