The rate and extent of biodegradation of benzene, toluene, ethylbenzene and xylenes (BTEX) in ground-water was studied in samples from a contaminated site which contained total BTEX concentrations of up to 20 000 microg litre(-1). All compounds were rapidly degraded under natural aerobic conditions. Elevation of incubation temperature, supply of organic nutrients or addition of inorganic fertiliser did not increase the rate or extent of biodegradation and it appeared that oxygen supply was the factor limiting BTEX degradation at this site. Attempts to increase the dissolved oxygen concentration in the ground-water by the addition of hydrogen peroxide to give a final concentration of 200 mg litre(-1) resulted in the complete inhibition of biodegradation. No biodegradation occurred under anaerobic conditions except when nitrate was provided as a terminal electron acceptor for microbial respiration. Under denitrifying conditions there was apparent biodegradation of benzene, toluene, ethyl-benzene, m-xylene and p-xylene but o-xylene was not degraded. Degradation under denitrifying conditions occurred at a much slower rate than under oxygenated conditions.  相似文献

The remediation of nitroaromatic contaminated groundwater is sometimes difficult because nitroaromatic compounds are resistant to biodegradation and, when they do transform, the degradation of the products may also be incomplete. A simple nitroaromatic compound, nitrobenzene, was chosen to assess the feasibility of an in situ multi-zone treatment system at the laboratory scale. The proposed treatment system consists of a zero valent granular iron zone to reduce nitrobenzene to aniline, followed by a passive oxygen release zone for the aerobic biodegradation of the aniline daughter product using pristine aquifer material from Canadian Forces Base (CFB) Borden, Ontario, as an initial microbial source. In laboratory batch experiments, nitrobenzene was found to reduce quickly in the presence of granular iron forming aniline, which was not further degraded but remained partially sorbed onto the granular iron surface. Aniline was found to be readily biodegraded with little metabolic lag under aerobic conditions using the pristine aquifer material. A sequential column experiment, containing a granular iron reducing zone and an aerobic biodegradation zone, successively degraded nitrobenzene and then aniline to below detection limits (0.5 microM) without any noticeable reduction in hydraulic conductivity from biofouling, or through the formation of precipitates.  相似文献

The objective of this investigation was to evaluate the anaerobic biodegradability of benzene, toluene, ethylbenzene, ortho-, meta- and para-xylene (BTEX) and trichloroethylene (TCE) in aquifer sediment down gradient of an unlined landfill. The major organic contaminants identified in the shallow unconfined aquifer are cis-dichloroethylene (c-DCE) and toluene. The biodegradative potential of the contaminated aquifer was measured in three sets of microcosms constructed using anaerobic aquifer sediment from three boreholes down gradient of the landfill. The degradability of BTEX and TCE was examined under ambient and amended conditions. TCE was degraded in microcosms with aquifer material from all three boreholes. Toluene biodegradation was inconsistent, exhibiting biodegradation with no lag in one set of microcosms but more limited biodegradation in two additional sets of microcosms. TCE exhibited an inhibitory effect on toluene degradation at one location. The addition of calcium carbonate stimulated TCE biodegradation which was not further stimulated by nutrient addition. TCE was converted to ethylene, a harmless byproduct, in all tests. Benzene, ethylbenzene and xylene isomers were recalcitrant in both ambient and amendment experiments. Biodegradation occurred under methanogenic conditions as methane was produced in all experiments. Bromoethane sulfonic acid (BES), a methanogenic inhibitor, inhibited methane and ethylene production and TCE biodegradation. The results indicate the potential for intrinsic bioremediation of TCE and toluene down gradient of the Wilder's Grove, North Carolina, landfill. The low concentrations of TCE in monitoring wells was consistent with its biodegradation in laboratory microcosms.  相似文献

This study examined the role of denitrifying and sulfate-reducing bacteria in biodegradation of pentaerythritol tetranitrate (PETN). Microbial inocula were obtained from a PETN-contaminated soil. PETN degradation was evaluated using nitrate and/or sulfate as electron acceptors and acetate as a carbon source. Results showed that under different electron acceptor conditions tested, PETN was sequentially reduced to pentaerythritol via the intermediary formation of tri-, di- and mononitrate pentaerythritol (PETriN, PEDN and PEMN). The addition of nitrate enhanced the degradation rate of PETN by stimulating greater microbial activity and growth of nitrite reducing bacteria that were responsible for degrading PETN. However, a high concentration of nitrite (350 mg L−1) accumulated from nitrate reduction, consequently caused self-inhibition and temporarily delayed PETN biodegradation. In contrast, PETN degraded at very similar rates in the presence and absence of sulfate, while PETN inhibited sulfate reduction. It is apparent that denitrifying bacteria possessing nitrite reductase were capable of using PETN and its intermediates as terminal electron acceptors in a preferential utilization sequence of PETN, PETriN, PEDN and PEMN, while sulfate-reducing bacteria were not involved in PETN biodegradation. This study demonstrated that under anaerobic conditions and with sufficient carbon source, PETN can be effectively biotransformed by indigenous denitrifying bacteria, providing a viable means of treatment for PETN-containing wastewaters and PETN-contaminated soils.  相似文献

Mainly due to intrinsic biodegradation, monitored natural attenuation can be an effective and inexpensive remediation strategy at petroleum release sites. However, gasoline additives such as methyl tert-butyl ether (MTBE) can jeopardize this strategy because these compounds often degrade, if at all, at a slower rate than the collectively benzene, toluene, ethylbenzene and the xylene (BTEX) compounds. Investigation of whether a compound degrades under certain conditions, and at what rate, is therefore important to the assessment of the intrinsic remediation potential of aquifers. A natural gradient experiment with dissolved MTBE-containing gasoline in the shallow, aerobic sand aquifer at Canadian Forces Base (CFB) Borden (Ontario, Canada) from 1988 to 1996 suggested that biodegradation was the main cause of attenuation for MTBE within the aquifer. This laboratory study demonstrates biologically catalyzed MTBE degradation in Borden aquifer-like environments, and so supports the idea that attenuation due to biodegradation may have occurred in the natural gradient experiment. In an experiment with batch microcosms of aquifer material, three of the microcosms ultimately degraded MTBE to below detection, although this required more than 189 days (or >300 days in one case). Failure to detect the daughter product tert-butyl alcohol (TBA) in the field and the batch experiments could be because TBA was more readily degradable than MTBE under Borden conditions.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are one of the main classes of contaminants in the terrestrial environment. Concentrations of biphenyl, fluorene, phenanthrene and pyrene were added to soil samples in order to investigate the anaerobic degradation potential of PAHs under denitrifying conditions. A mixed population of microorganisms obtained from a paddy soil was incubated for 20 days in anaerobic conditions in the presence of soil alone or with nitrate, adding, as electron donors, PAHs and, in some samples, glucose or acetate. At regular time intervals oxidation-reduction potential, PAHs concentration, microbial ATP and nitrate concentration into the solution were measured. Degradation trends for each hydrocarbon are similar under all conditions, indicating that the molecular conformation prevails over other parameters in controlling the degradation. Poor degradation results were obtained when PAHs were the only organic matter available for the inoculum, thus confirming the recalcitrance to degradation of these compounds. Biodegradation was influenced by the addition of other carbon sources. As better degradation results were generally obtained when acetate or glucose were added, the hypothesis of a co-metabolic enhancement of PAH biodegradation seems likely. Thus, anaerobic biodegradation of PAHs studied, biphenyl, fluorene, phenanthrene and pyrene, seems to be possible both through fermentative and respiratory metabolism, provided that low molecular weight co-metabolites and suitable electron acceptors (nitrate) are present.  相似文献

The present study was conducted to investigate the anaerobic biodegradation potential of biostimulation by nitrate (KNO3) and methyl-β-cyclodextrin (MCD) addition on an aged organochlorine pesticide (OCP)-contaminated paddy soil. After 180 days of incubation, total OCP biodegradation was highest in soil receiving the addition of nitrate and MCD simultaneously and then followed by nitrate addition, MCD addition, and control. The highest biodegradation of chlordanes, hexachlorocyclohexanes, endosulfans, and total OCPs was 74.3, 63.5, 51.2, and 65.1 %, respectively. Meanwhile, MCD addition significantly increased OCP bioaccessibility (p?<?0.05) evaluated by Tenax TA extraction and a three-compartment model method. Moreover, the addition of nitrate and MCD also obtained the highest values of soil microbial activities, including soil microbial biomass carbon and nitrogen, ATP production, denitrifying bacteria count, and nitrate reductase activity. Such similar trend between OCP biodegradation and soil-denitrifying activities suggests a close relationship between OCP biodegradation and N cycling and the indirect/direct involvement of soil microorganisms, especially denitrifying microorganisms in the anaerobic biodegradation of OCPs.  相似文献

Biological and abiotic approaches for treating co-mingled perchlorate, nitrate, and nitramine explosives in groundwater were compared in microcosm and column studies. In microcosms, microscale zero-valent iron (mZVI), nanoscale zero-valent iron (nZVI), and nickel catalyzed the reduction of RDX and HMX from initial concentrations of 9 and 1 mg/L, respectively, to below detection (0.02 mg/L), within 2 h. The mZVI and nZVI also degraded nitrate (3 mg/L) to below 0.4 mg/L, but none of the metal catalysts were observed to appreciably reduce perchlorate ( approximately 5 mg/L) in microcosms. Perchlorate losses were observed after approximately 2 months in columns of aquifer solids treated with mZVI, but this decline appears to be the result of biodegradation rather than abiotic reduction. An emulsified vegetable oil substrate was observed to effectively promote the biological reduction of nitrate, RDX and perchlorate in microcosms, and all four target contaminants in the flow-through columns. Nitrate and perchlorate were biodegraded most rapidly, followed by RDX and then HMX, although the rates of biological reduction for the nitramine explosives were appreciably slower than observed for mZVI or nickel. A model was developed to compare contaminant degradation mechanisms and rates between the biotic and abiotic treatments.  相似文献

The addition of ferric hydroxide to sludge from a municipal anaerobic digester stimulated the rate of methanogenesis from canola oil when the initial oil concentration was high (4600 mg/L; P < 0.002), but not when it was low (920 mg/L; P > 0.05). Similar trends were observed when oleic acid, a fatty acid that is a major component of canola oil triglycerides, was provided, but the effects were statistically significant only when the initial concentration of ferric hydroxide was also high (18 g/L; P = 0.015). Iron reduction occurred when ferric hydroxide was added to microcosms containing anaerobic digester sludge, but the extent of ferrous iron production was much less in acetate-amended microcosms than in those that were provided with canola oil or oleic acid. Methanogenesis and acetate consumption were completely inhibited when the initial acetate concentration was approximately 5000 mg/L, regardless of the initial ferric hydroxide concentration. The main effect of ferric hydroxide in this system appears to have been a result of stimulation of the rate of fatty acid oxidation.  相似文献

Two natural-gradient pulse tracer tests were conducted in a petroleum-contaminated aquifer to evaluate the potential for benzene, toluene, ethylbenzene, and xylenes (BTEX) biodegradation under enhanced nitrate-reducing conditions. Addition of nitrate resulted in loss of toluene, ethylbenzene, and m,p-xylenes (TEX) after an initial lag period of approximately 9 days. Losses of benzene were not observed over the 60-day monitoring period. Tracer breakthrough curves (BTCs) were analyzed to derive transport and biodegradation parameters, including advective velocities, retardation factors, dispersion coefficients, biodegradation rate constants, and nitrate utilization ratios. Using the parameters derived from the BTC analysis, numerical simulations of one of the tracer experiments were conducted using BIONAPL/3D [Molson, J., BIONAPL/3D User Guide, A 3D Coupled Flow and Multi-Component Reactive transport model. University of Waterloo, Waterloo, Ontario, Canada]. Simulations using the BTC-derived transport and biodegradation parameters successfully reproduced benzene, TEX, and nitrate concentrations measured during the tracer experiment. Comparisons of observed and simulated nitrate concentrations indicate that the mass ratio of nitrate-N utilized to TEX degraded increased over time during the experiment, reaching values many times that expected based on stoichiometry of TEX oxidation coupled to nitrate reduction. Excess nitrate loss is likely due to oxidation of other organics in addition to TEX.  相似文献

Hexahydro-1,3,5-trinitro-1,3,5-triazine (RDX), octahydro-1,3,5,7-tetranitro-1,3,5,7-tetrazocine (HMX), and 2,4,6-trinitrotoluene (TNT) are explosives that are frequently found as environmental contaminants on military installations. Hydrogen has been shown to support the anaerobic transformation of these explosives. We investigated ethanol and propylene glycol as electron donors for providing syntrophically produced H2 for stimulating the anaerobic biodegradation of explosives in contaminated soil. The study was conducted using anoxic microcosms constructed with slurries of the contaminated soil and groundwater. The addition of 5mM ethanol and propylene glycol enhanced the biodegradation of RDX and HMX relative to the control bottles. Ethanol was depleted within about 20 days, resulting in the transient formation of hydrogen, acetate, and methane. The hydrogen headspace concentration increased from 8 ppm to 1838 ppm before decreasing to background concentrations. Propylene glycol was completely degraded after 15 days, forming hydrogen, propionate, and acetate as end-products. The hydrogen headspace concentrations increased from 56 ppm to 628 ppm before decreasing to background concentrations. No methane formation was observed during the incubation period of 48 days. Our findings indicate the addition of ethanol and propylene to the aquifer slurries increased the hydrogen concentrations and enhanced the biotransformation of RDX and HMX in the explosive-contaminated soil.  相似文献

An inexpensive scheme to reclaim groundwaters contaminated by volatile organics at the Gloucester landfill site, near Ottawa, Canada, involved pumping of contaminated water into an conducted to evaluate the possible enhancement of biodegradation to improve this remedial measure.Batch experiments revealed that toluene (0.8–1.5 mg L−1) was rapidly degraded aerobically while chloroform (1.2–1.0 mg L−1) was recalcitrant under aerobic or anaerobic (denitrifying) conditions. Dynamic column experiments confirmed these findings. In column experiments, both chloroform and toluene were retarded relative to the pore water velocity, but to a far lesser extent than predicted by hydrophobic sorption theory. This discrepancy is attributed to the large particulate nature of the sorbent (sawdust) which prevented rapidly migrating organics from attaining an equilibrium partitioning with the bulk of this organic matter. Addition of acetate and nitrate at 20 mg L−1 stimulated aerobic activity, but was inhibitory to toluene degradation. However, when nutrient addition ceased, enhanced toluene degradation was produced by the larger microbial population.In the field experiment, addition of acetate and nitrate to groundwater influent into the sawdust/sand pit may have stimulated benzene biodegradation, but had no apparent influence on volatile chlorinated hydrocarbon biodegradation. As in the column experiments, sorptive retardation was considerably less than predicted, due to a lack of equilibrium partitioning of organic solutes within the sawdust.Enhancing biodegradation in this remedial scheme was unsuccessful for a broad range of volatile organic compound types. Periodic nutrient addition to create variable environments for microbes appears to hold more promise for remediation than the continuous nutrient input scheme. This experience demonstrates the need for realistic field-scale experiments before translating laboratory studies into full-scale remedial operations.  相似文献

The feasibility of biodegradation of the fuel oxygenate methyl tert-butyl ether (MTBE) under iron-reducing conditions was explored in batch and continuous-flow systems. A porous pot completely-mixed reactor was seeded with diverse cultures and operated under iron-reducing conditions. For batch studies, culture from the reactor was transferred anaerobically to serum bottles containing either MTBE alone or MTBE with ethanol (EtOH) and excess electron acceptor. In the continuous-flow reactor, MTBE conversion to tert-butyl alcohol (TBA) was observed after 181 days of operation, and stable removal was achieved throughout the remainder of the study. Simultaneously, both the MTBE only and the MTBE and EtOH iron-reducing batch serum bottles also began to degrade MTBE. Bottles were respiked and the degradation rate was determined to be 2.36 +/- 0.10 x 10(-4) mmol MTBE/min-kgVSS. The EtOH present with MTBE degraded faster (7.76 +/- 0.08 x 10(-3) mmol EtOH/min-kg VSS) but did not have a noticeable effect on the rate of MTBE degradation. No evidence of TBA degradation was observed by the iron-reducing cultures. Stoichiometry of iron utilization was determined from the iron balance of the continuous-flow reactor, and it was found that the bulk of the electron acceptor was required for energy and maintenance with little remaining for cell synthesis. This is consistent with a yield coefficient of less than 0.1. Molecular analysis of the iron-reducing culture by denaturing gradient gel electrophoresis indicated that uncultured strains of delta-Proteobacteria were dominant in the reactor.  相似文献

Biomass from a prototype reactor was used to investigate the kinetics of chemoheterotrophic reduction of solutions of ferric ethylenediaminetetraacetic acid (EDTA) and solutions containing the nitrosyl adduct of ferrous EDTA using ethanol as the primary electron donor and carbon source. A series of batch experiments were conducted using biomass extracted from the scrubber solution treatment and regeneration stage of a prototype iron EDTA-based unit process for the absorption of nitric oxide with subsequent biological treatment. Using a linear-sweep voltammetric method for analysis of the ferric EDTA concentration, iron-reducing bacteria were found to behave according to the Monod kinetic model, at initial concentrations up to 2.16 g chemical oxygen demand (COD) as ethanol per liter, with a half-velocity constant of 0.532 g COD as ethanol/L and a maximum specific utilization rate of 0.127 mol/L of ferric ethylenediamine-tetraacetic acid [Fe(III)EDTA]*(g volatile suspended solids [VSS]/L)d(-1). Based on batch analyses, biomass yield and endogenous decay values of iron-reducing bacteria were estimated to be 0.055 g VSS/g COD and 0.017 L/d, respectively. An average of 1.64 times the theoretical (stoichiometric) demand of ethanol was used to complete reduction reactions. Kinetics of the reduction of the nitrosyl adduct of ferrous EDTA are summarized by the following kinetic constants: half-velocity constant (Ks) of 0.39 g COD/L, maximum specific utilization rate (k) of 0.2 mol/L [NO x Fe(II)EDTA(2-)](g VSS/L)d(-1), and inhibition constant (K(I)) of 0.33 g COD/L, as applied to the modified Monod kinetic expression described herein. Based on batch analyses, the biomass yield of nitrosyl-adduct-reducing bacteria was estimated to be 0.259 g VSS/g COD, endogenous decay was experimentally determined to be 0.0569 L/d, and an average of 1.26 times the stoichiometric demand of ethanol was used to complete reduction reactions.  相似文献

Large-scale contaminated sites with multiple contaminants in the groundwater present a challenge to risk assessment and remediation. Attenuation reactions take place in the subsurface and act to contain contaminants, but must be thoroughly investigated on a site-specific basis. Field data from monitoring wells at a contaminated industrial site in Bitterfeld, Germany, are presented and analyzed for evidence of the prevalent biodegradation reactions. The groundwater in the Tertiary aquifer is contaminated with large quantities of chlorinated aliphatic compounds, in addition to chlorobenzenes and BTEX. In this strictly anaerobic environment, geochemical indications for several microbial processes were found, including methanogenesis, sulfate and iron reduction as well as reductive dechlorination of the chlorinated hydrocarbons. Direct evidence for the latter degradation reaction was observed along the flowpath due to the appearance of intermediates and an increase in the degree of dechlorination.  相似文献

When present in the vadose zone, potentially toxic nitrate and perchlorate anions can be persistent sources of groundwater contamination. Gaseous electron donor injection technology (GEDIT), an anaerobic variation of petroleum hydrocarbon bioventing, involves injecting electron donor gases, such as hydrogen or ethyl acetate, into the vadose zone, to stimulate biodegradation of nitrate and perchlorate. Laboratory microcosm studies demonstrated that hydrogen and ethanol promoted nitrate and perchlorate reduction in vadose zone soil and that moisture content was an important factor. Column studies demonstrated that transport of particular electron donors varied significantly; ethyl acetate and butyraldehyde were transported more rapidly than butyl acetate and ethanol. Nitrate removal in the column studies, up to 100%, was best promoted by ethyl acetate. Up to 39% perchlorate removal was achieved with ethanol and was limited by insufficient incubation time. The results demonstrate that GEDIT is a promising remediation technology warranting further validation.  相似文献

Co-produced water from the oil and gas industry accounts for a significant waste stream in the United States. This "produced water" is characterized by saline water containing a variety of pollutants, including water soluble and immiscible organics and many inorganic species. To reuse produced water, removal of both the inorganic dissolved solids and organic compounds is necessary. In this research, the effectiveness of a pretreatment system consisting of surfactant modified zeolite (SMZ) adsorption followed by a membrane bioreactor (MBR) was evaluated for simultaneous removal of carboxylates and hazardous substances, such as benzene, toluene, ethylbenzene, and xylenes (BTEX) from saline-produced water. A laboratory-scale MBR, operated at a 9.6-hour hydraulic residence time, degraded 92% of the carboxylates present in synthetic produced water. When BTEX was introduced simultaneously to the MBR system with the carboxylates, the system achieved 80 to 95% removal of BTEX via biodegradation. These results suggest that simultaneous biodegradation of both BTEX and carboxylate constituents found in produced water is possible. A field test conducted at a produced water disposal facility in Farmington, New Mexico confirmed the laboratory-scale results for the MBR and demonstrated enhanced removal of BTEX using a treatment train consisting of SMZ columns followed by the MBR. While most of the BTEX constituents of the produced water adsorbed onto the SMZ adsorption system, approximately 95% of the BTEX that penetrated the SMZ and entered the MBR was biodegraded in the MBR. Removal rates of acetate (influent concentrations of 120 to 170 mg/L) ranged from 91 to 100%, and total organic carbon (influent concentrations as high as 580 mg/L) ranged from 74 to 92%, respectively. Organic removal in the MBR was accomplished at a low biomass concentration of 1 g/L throughout the field trial. While the transmembrane pressure during the laboratory-scale tests was well-controlled, it rose substantially during the field test, where no pH control was implemented. The results suggest that pretreatment with an SMZ/MBR system can provide substantial removal of organic compounds present in produced water, a necessary first step for many water-reuse applications.  相似文献
