Muramic acid (Mur) is found in bacterial peptidoglycan (PG) whereas 3-hydroxy fatty acids (3-OH FAs) are found in Gram-negative bacterial lipopolysaccharide (LPS). Thus Mur and 3-OH FAs serve as markers to assess bacterial levels in indoor air. An initial survey, in a school, demonstrated that the levels of dust, PG and LPS (pmol m(-3)) were each much higher in occupied rooms than in the same rooms when unoccupied. In each instance, the Mur content of dust was increased and the hydroxy fatty acid distribution changed similarly suggesting an alteration in the bacterial population. Here, findings are compared with results from two additional schools. Follow-up aerosol monitoring by particle size was also performed for the first time for all 3 schools. The particle size distribution was shown to be quite different in occupied versus unoccupied schoolrooms. Within individual classrooms, concentrations of airborne particles [greater-than-or-equal]0.8 [micro sign]m in diameter, and CO(2) were correlated. This suggests that the increased levels of larger particles are responsible for elevation of bacterial markers during occupation. Release of culturable and non-culturable bacteria or bacterial aggregates from children (e.g. from flaking skin) might explain this phenomenon.  相似文献

Previously it was demonstrated that the levels of large particles (>2 micron) and associated bacterial cell envelope markers increase greatly on occupation in schools; it was hypothesized that the source of both was shed human skin. In the current work to test this hypothesis, room air cleaners were used to collect airborne dust (>50-100 mg) from occupied and unoccupied school rooms which was then subjected to proteomic analysis. Proteins were extracted from the dust and separated using two dimensional gel electrophoresis (2D GE). In situ digestion of protein spots with trypsin released peptides, which were subsequently analyzed by matrix assisted laser desorption/deionization, time-of-flight mass spectrometry (MALDI-TOF-MS) and tandem mass spectrometry (MALDI-TOF-MS-MS). In Coomassie blue stained gels, a single spot generally dominated the 2D gels; this protein was identified by tandem mass spectrometry as K10 epithelial keratin. The results experimentally confirm previous anecdotal reports that human skin is readily shed into air and suggest that increased levels of microbial markers and large particles observed in occupied rooms are also derived from skin.  相似文献

The interrelationships between ventilation rate, indoor air quality, and energy consumption in operation rooms at rest are yet to be understood. We investigate the effect of ventilation rate on indoor air quality indices and energy consumption in ORs at rest. The study investigates the air temperature, relative humidity, concentrations of carbon dioxide, particulate matter (PM), and airborne bacteria at different ventilation rates in operation rooms at rest of a medical center. The energy consumption and cost analysis of the heating, ventilating, and air conditioning (HVAC) system in the operation rooms at rest were also evaluated for all ventilation rates. No air-conditioned operation rooms had very highest PM and airborne bacterial concentrations in the operation areas. The bacterial concentration in the operation areas with 6–30 air changes per hour (ACH) was below the suggested level set by the United Kingdom (UK) for an empty operation room. A 70% of reduction in annual energy cost by reducing the ventilation rate from 30 to 6 ACH was found in the operation rooms at rest. Maintenance of operation rooms at ventilation rate of 6 ACH could save considerable amounts of energy and achieve the goal of air cleanliness.  相似文献

Gas chromatography-mass spectrometry (GC-MS) using a quadrupole instrument and GC-tandem MS (GC-MS-MS) using an ion trap instrument were applied to determine 3-hydroxy fatty acids (3-OH FAs) with 10-18 carbon chain lengths, specific components of the endotoxin (lipopolysaccharide, LPS) of Gram-negative bacteria, in 30 house dust samples. The two methods provided similar detection sensitivity for methyl ester/trimethylsilyl derivatives of the 3-OH FAs and allowed these acids to be distinguished from co-eluting 2-OH FA derivatives. The correlation coefficients between endotoxin activity (Limulus test) and the combined amounts of 3-OH C10, 3-OH C12, and 3-OH C14 were 0.60 and 0.61 when using GC-MS and GC-MS-MS, respectively. The superior selectivity of GC-MS-MS was illustrated in analyses of sub-milligram amounts of dust, where the chromatograms achieved by GC-MS were difficult to interpret due to a high background and several closely eluting compounds. GC-MS-MS is therefore preferable to GC-MS for determining 3-OH FAs in minute (sub-milligram) amounts of dust.  相似文献

In order to be able to make a decision, as to whether a room or building has a health-endangering pentachlorophenol (PCP) concentration, usually the PCP concentrations in air and settled dust are measured. The variability of the PCP concentration in indoor air and dust was studied. Air and dust samples were taken from 75 rooms in 30 buildings with suspicion of application of PCP-containing wood preservatives. Sampling was repeated four times within 18 months. Thirty-six rooms were reconstructed within the study; 39 rooms had unchanged contamination status during the study. The four times repeated measurements of PCP concentrations in air and dust in these rooms showed large variations of the measured values. The variability of the results is to a large extent in the same range as the measured values. The observed relative standard deviation of the PCP concentrations in air and dust does not depend on the average PCP concentration detected in the individual rooms.  相似文献

The main objective of this work was to quantify and characterize the major indoor air contaminants present in different stages of a municipal WWTP, including microorganisms (bacteria and fungi), carbon dioxide, carbon monoxide, hydrogen sulfide ammonia, formaldehyde, and volatile organic compounds (VOCs). In general, the total bacteria concentration was found to vary from 60 to >52,560 colony-forming units (CFU)/m3, and the total fungi concentration ranged from 369 to 14,068 CFU/m3. Generally, Gram-positive bacteria were observed in higher number than Gram-negative bacteria. CO2 concentration ranged from 251 to 9,710 ppm, and CO concentration was either not detected or presented a level of 1 ppm. H2S concentration ranged from 0.1 to 6.0 ppm. NH3 concentration was <2 ppm in most samples. Formaldehyde was <0.01 ppm at all sampling sites. The total VOC concentration ranged from 36 to 1,724 μg/m3. Among the VOCs, toluene presented the highest concentration. Results point to indoor/outdoor ratios higher than one. In general, the highest levels of airborne contaminants were detected at the primary treatment (SEDIPAC 3D), secondary sedimentation, and sludge dehydration. At most sampling sites, the concentrations of airborne contaminants were below the occupational exposure limits (OELs) for all the campaigns. However, a few contaminants were above OELs in some sampling sites.  相似文献

A GC-MS method for the quantification of bacterial lipopolysaccharides (LPS, endotoxin) is presented. After hydrolytic cleavage of 3-hydroxy fatty acids (3-OH FAs) from the lipid A region of LPS, derivatisation of both the hydroxyl and the carboxyl group was performed in one step with a mixture of methyl-bis(trifluoracetamide)(MBTFA) and N-methyl-N-(tert-butyldimethylsilyl)trifluoracetamide (MTBSTFA). Using GC-MS in the EI mode with selected ion monitoring (SIM) for analysis, baseline separation of 3-OH FAs (and of possibly interfering 2-OH FAs) was achieved. The sensitivity of the method (LOD 7-50 pg/injection for the different 3-OH FAs investigated) allows for the efficient quantification of LPS in occupational and environmental samples. Degradation of 3-OH FAs as well as of their derivatives during sample preparation and GC-MS separation as a possible source of errors in analytical methods based on 3-OH FA determination is reported for the first time. Thermal elimination of water from the underivatised 3-OH FAs and of trifluoroacetic acid from the derivatives was identified as the cause of degradation. The resulting alpha,beta-unsaturated compounds showing the same mass spectra as the 3-OH FA derivatives were detected as more or less prominent satellite peaks. By using alkaline instead of acidic hydrolysis and cool on-column instead of split/splitless injection, elimination was reduced to an acceptable level.  相似文献

During periods of two weeks in February and June 2010 the performance of portable air treatment units (PATUs) was evaluated in a primary school classroom using indicators of indoor air quality. Air samples were collected in an undisturbed setting on weekend days and in an occupied setting during teaching hours. In the first week PATUs were turned off and in the second week they were turned on. On weekend days PATUs reduced indoor levels of PM-10 by 87% in February and by 70% in June compared to weekend days when PATUs were turned off. On schooldays, indoor PM-10 was increased by 6% in February and reduced by 42% in June. For PM-2.5 reductions on weekend days were 89% in February and 80% in June. On school days PM-2.5 was increased by 15% in February and reduced by 83% in June. Turning on the PATUs reduced total VOC by 80% on weekend days and by 57% on school days (but not in June). No influence on formaldehyde, NO(2), O(3) and molds was observed. PATUs appeared to be less effective in removal of air pollutants when used in an occupied classroom compared to an unoccupied setting. Our study suggests that such devices should be tested in real-life settings to evaluate their influence on indoor air quality.  相似文献

The purpose of this study was to evaluate the level and composition of bacteria and fungi in the indoor air of an Italian office building equipped with a heating, ventilation and air conditioning (HVAC) system. Airborne bacteria and fungi were collected in three open-space offices during different seasons. The microbial levels in the outdoor air, supply air diffusers, fan coil air flow and air treatment unit humidification water tank were used to evaluate the influence of the HVAC system on indoor air quality (IAQ). A medium–low level of bacterial contamination (50–500 CFU/m3) was found in indoor air. Staphylococcus and Micrococcus were the most commonly found genera, probably due to human presence. A high fungal concentration was measured due to a flood that occurred during the winter. The indoor seasonal distribution of fungal genera was related to the fungal outdoor distribution. Significant seasonal and daily variation in airborne microorganisms was found, underlining a relationship with the frequency of HVAC system switching on/off. The results of this monitoring highlight the role of the HVAC system on IAQ and could be useful to better characterise bacterial and fungal population in the indoor air of office buildings.  相似文献

In order to characterize the status of indoor air pollution in some important facilities, a list of key criteria pollutants [particulate matter (PM(10)), carbon dioxide (CO(2)), carbon monoxide (CO), formaldehyde (HCHO), and bioaerosol] was measured from a total of 91 randomly selected sites in 18 different cities, Korea (February 2006 to December 2009). The target facilities include 43 child care facilities, 38 medical facilities, 6 elementary schools, and 4 postnatal care centers. The results showed that some air pollutants (e.g., CO and HCHO) did not exceed the recommended guideline [e.g., the Korean indoor air standard (KIAS) values of 10 ppm and 100 ppb, respectively]. However, concentration of PM(10), CO(2), and bioaerosol occasionally exceeded their respective guidelines (e.g., seven, three, and two cases). Discrete seasonalities were observed from indoor pollutants because of varying ventilation practice (e.g., summer time dominance of PM(10), HCHO, and bioaerosol or winter dominance of CO(2) and CO). However, as the concentrations of the indoor pollutants were scarcely above the recommended guideline level, more diversified approaches are desirable to diagnose the status of indoor pollution and to provide a realistic strategy for the improvement of IAQ.  相似文献

The aim of this study was to estimate mite antigen exposure in farming environments by passive sampling of airborne dust. Antigen concentrations were measured with enzyme immunoassays specific for three storage mites (SM): Acarus siro, Lepidoglyphus destructor, Tyrophagus putrescentiae and the house dust mite (HDM) Dermatophagoides pteronyssinus. Dust samples were collected with electrostatic dust fall collectors (EDCs) in three different areas of cattle farms. EDCs were placed in cow stables (working area), in changing rooms (transit area) and in different rooms of farmer dwellings (living area). Mite concentrations in the living area of farm homes were compared to those of urban homes. In dust samples from stables, antigens of all four mite species could be detected. The highest exposure level was to L. destructor (median 56.7 μg/m(2)), the lowest to A. siro (median 14.4 μg/m(2)). Mite concentrations of different species showed no correlation within the cow stables. In comparison to stables, the median mite concentrations in farm homes were significantly lower, ranging from below the detection limit to 1.5 μg/m(2). Antigens of SM were predominantly found in changing rooms and kitchens, and HDM antigens were mainly detected in bedrooms. Antigens of all mites were measured the least often in living rooms. T. putrescentiae was the most prevalent mite in all room types, and the exposure levels correlated strongly between different rooms. The number of SM positive samples in farm homes was considerably higher than in urban homes, while the percentage of HDM positive samples did not differ significantly.  相似文献

Dwellings showing a presence of moulds are considered to be unhealthy both by the inhabitants and by sanitary authorities. Although the thresholds of pathogenicity have not yet been established, the toxic, allergic and infectious risk of indoor moulds is better understood today. A study on indoor fungi contamination for 128 dwellings was done between October and May in France. It concerned 69 dwellings, the occupants of which either complained to the sanitary authorities about problems of moulds and humidity or consulted a doctor who related their symptoms to housing conditions. Fifty-nine other dwellings, the occupants of which were healthy, constituted the control group. We present the statistical analysis of questionnaires, which aimed to clarify characteristics of dwellings associated with high concentrations of airborne moulds. Air samples were taken with an impactor in 500 rooms. On visiting dwellings, investigators obtained answers to 25 questions concerning characteristics of inhabitants and living space, as well as the presence of mould indicators. Indoor and outdoor temperature and indoor relative humidity of air measurements were taken. The total concentration of fungi in the air was significantly higher in ground floor apartments versus those on other floors (p = 0.047), in small and highly occupied dwellings (p = 0.03 and 0.003), in dwellings with electric heating (p = 0.04), without a ventilation system (p = 0.003), with water damage (p = 0.003), and finally, in those where the investigator noted an odour of moisture or visible moulds (p < 0.001). The efficacy of the latter criteria in the evaluation of insalubrity is discussed.  相似文献

Airborne dust bioaerosols, ammonia and formaldehyde levels were determined inside two different (ventilated and unventilated) wood working shops. Airborne dust was found at mean values of 4.3 and 3.01 mg m(-3). These levels were higher than that recommended by Egyptian environmental law [1 mg m(-3) indoor maximum allowable concentration (MAC) for hard wood]. The highest frequency of aerodynamic size distribution of airborne wood dust was detected at a diametre of 4.9 microm which was recorded during a machining operation. Total viable bacteria were recorded at a mean value of 10(4) colony-forming units (cfu) m(-3), whereas Gram-negative bacteria were found at very low counts (10(1) cfu m(-3)). Fungi levels were recorded at mean values of 10(3) and 10(2) cfu m(-3) in ventilated and unventilated shops, respectively. Penicillium, Aspergillus, Cladosporium and yeast species were dominant isolates. Moreover, actinomycetes were found at a mean value of 10(3) cfu m(-3) at both workshops. Ammonia was detected in relatively low concentrations (mean values of 457 and 623 microg m(-3)), whereas formaldehyde was found in relatively moderate concentrations (mean values of 0.42 and 0.64 ppm).  相似文献

The aim of this study was to evaluate the indoor (I) and outdoor (O) levels of NO?, speciated volatile organic compounds (VOCs) and carbonyls at fourteen primary schools in Lisbon (Portugal) during spring, autumn and winter. Three of these schools were also selected to be monitored for comfort parameters, such as temperature and relative humidity, carbon dioxide (CO?), carbon monoxide (CO), total VOCs, and both bacterial and fungal colony-forming units per cubic metre. The concentration of CO? and bioaerosols greatly exceeded the acceptable maximum values of 1800 mg m?3 and 500 CFU m?3, respectively, in all seasons. Most of the assessed VOCs and carbonyls occurred at I/O ratios above unity in all seasons, thus showing the importance of indoor sources and building conditions in indoor air quality. However, it has been observed that higher indoor VOC concentrations occurred more often in the colder months, while carbonyl concentrations were higher in the warm months. In general, the I/O NO? ratios ranged between 0.35 and 1, never exceeding the unity. Some actions are suggested to improve the indoor air quality in Lisbon primary schools.  相似文献

Elevated concentrations of endotoxin in indoor air due to cigarette smoking   总被引:1,自引:0,他引:1  
Exposure to environmental tobacco smoke (ETS) is an important worldwide public health issue. The present study demonstrates that cigarette smoke can be a major source of endotoxin (lipopolysaccharide, LPS) in indoor environments. Gas-chromatography/mass-spectrometry was used to determine 3-hydroxy fatty acids as markers of endotoxin in air-borne house dust in homes of smokers and non-smokers. Air concentrations of endotoxin were 4-63 times higher in rooms of smoking students than in identical rooms of non-smoking students. The fact that cigarette smoke contains large amounts of endotoxin may partly explain the high prevalence of respiratory disorders among smokers and may also draw attention to a hitherto neglected risk factor of ETS.  相似文献

Muramic acid (MA) is a unique amino sugar that is a constituent of the peptidoglycan (PG) present in prokaryotic cell walls. MA can serve as a marker for quantifying bacterial load, e.g. in indoor environments, by using gas chromatography-tandem mass spectrometry (GC-MS-MS). We demonstrated recently that the methyl ester O-methyl acetate (MMA) derivative can be used to detect MA in house dust by ion-trap GC-MS-MS. However, since the MMA derivative is not formed from free MA quantification was not optimal. Here we report 1) significant improvements in sample preparation of the MMA derivative and 2) an evaluation of the performance of derivative, using for comparison the alditol acetate derivative, the gold standard in quantitative trace analysis of MA in complex matrices. The MMA derivative was analysed using an MS instrument with internal ionization and the alditol acetate derivative was analysed using an instrument with external ionization. 13C-labelled cyanobacteria, containing MA in their PG, were used as the internal standard. A linear relationship was found between the two methods in studies on 27 parallel samples of airborne dust from school classes collected on filters. Although the analytical sensitivity of the MMA derivatives was somewhat slightly lower than of the alditol acetate derivative, this may be due to differences in yield of derivative, sample clean-up efficiency, or different performance of the GC columns or MS instruments. However preparation of the MMA derivative is quick and compatible with preparation of methyl esters of 3-hydroxy fatty acids (used as markers of Gram negative endotoxin) allowing the levels of both markers to be determined in the same dust sample. In conclusion, the MMA procedure can be used to determine MA in environmental samples with good reproducibility provided the concentration of the 13C-labelled MA internal standard in the cyanobacteria is first determined with an alternative method.  相似文献

2-Ethyl-1-hexanol is a possibly causative chemical in sick building symptoms, although 2-ethyl-1-hexanol has received little attention as a hazardous substance in studies on indoor air pollution. Airborne 2-ethyl-1-hexanol concentrations were measured from 2002 to 2004 in 99 rooms of 42 non-domestic buildings in Nagoya, Japan. The diffusive sampling method is effective for the measurement of a low level of 2-ethyl-1-hexanol in indoor air. The geometric mean (geometric standard deviation) of 2-ethyl-1-hexanol concentrations was 16.5 (5.4) microg m(-3) in indoor air and 1.9 (2.2) microg m(-3) in outdoor air. The maximum concentration of 2-ethyl-1-hexanol in indoor air and outdoor air was 2709 microg m(-3) and 12.4 microg m(-3), respectively. Fewer rooms in a small number of new buildings showed high concentrations of 2-ethyl-1-hexanol, while low concentrations were observed in many rooms of these buildings as well as the other new buildings. The room-to-room concentrations of 2-ethyl-1-hexanol in each building exhibited a wide variation. The geometric mean of the 2-ethyl-1-hexanol concentrations was significantly higher for indoor air than for outdoor air (p < 0.01). The correlation of the 2-ethyl-1-hexanol concentrations between indoor and outdoor air was not significant. Mechanical ventilation was effective in the temporary reduction of indoor 2-ethyl-1-hexanol level. These results suggest that the predominant source of 2-ethyl-1-hexanol was indoor areas.  相似文献

We examined the selectivity of 53 sets of primers for environmental monitoring of indoor air quality. Thirty-six fungal strains, representing 26 species from 14 genera of commonly occurring fungi, and 16 different bacterial strains, representing both gram-negative and gram-positive species, were included in the experiment. We verified the specificity of 28 of the 53 sets of primers, which were classified as universal fungal, universal bacterial, group or species specific. The PCR conditions required for optimal specificity were also determined. These results can serve as a guide for the step-wise PCR-based detection and identification of airborne fungi commonly found in indoor environments.  相似文献

Measureable levels of chlorpyrifos were seen in air and on horizontal and vertical surfaces over an 84-day sampling period following application by two different methods. Pressurized aerosol applications had the highest airborne levels over the 84-day sampling period, and movement into adjacent, nontreated rooms was seen 7 days after application. Highest surface residues found were located at floor/wall interfaces and were due probably as a result of splash or overspray around treated areas. Residue levels from desk sides were very low and all surface residues were highly variable. One could not predict what surface levels would be based upon airborne concentrations.  相似文献
