The peroxynitrite/porphyrin biomimetic system was established to investigate the effects of peroxynitrite on benzo[a]pyrene (B[a]P) metabolism. Three model systems consisting of different iron porphyrins were compared, and the results showed that the peroxynitrite/T(p-Cl)PPFeCl system was the highest catalytic efficiency in the metabolism of B[a]P. We analyzed the B[a]P metabolites produced from this system by RP-HPLC method and firstly identified the formation of nitrobenzo[a]pyrenes which are the special metabolites of B[a]P induced by peroxynitrite.  相似文献

研究了6株真菌对土壤中芘和苯并芘(BaP)的降解动态,用Michaelis-Menton和Monod动力学模型对结果进行拟合.结果表明,6株真菌对芘和BaP的降解速率有显著性差异,降解率相差不大.产黄青霉(Penicillium chrysogenum,SF04),在42d内对BaP的降解能力最强,可达71.31%,对芘的降解能力相对最弱.镰刀菌(Fusariumsp.,SF11),黑曲霉(Aspergillusniger,SF05),木霉(Trichodermasp.,SF02)和毛霉(Mucorsp.,SF06)42d对芘的降解率分别为86.22%,86.18%,85.41%,85.04%,对BaP的降解率分别为71.11%,69.44%,69.05%,69.72%.木霉(Trichodermasp.,SF02)和毛霉(Mucorsp.,SF06)对芘和BaP的降解速率均很快.  相似文献

蚯蚓细胞色素P450酶系对土壤中芘或苯并[a]芘的响应   总被引:1,自引:0,他引:1  
以赤子爱胜蚓(Eisenia fetida)为受试生物,通过人工污染草甸棕壤微宇宙试验方法,研究了暴露于不同环境浓度芘或苯并[a]芘(添加量分别为0.12、0.24、0.48、0.96 mg·kg~(-1))污染的土壤中14 d后,蚯蚓体内细胞色素P450(CYPs)酶系,如CYPs总量、CYP1A1及CYP2C9活性的变化.结果显示,芘或苯并[a]芘暴露导致蚯蚓CYPs总量、CYP1A1及CYP2C9活性的变化不同.其中,CYPs总量与CYP1A1活性对芘的响应趋势相似:试验3 d后,0.96 mg·kg~(-1)芘导致二者都显著高于对照水平;14 d后,都显著低于对照水平;CYP1A1活性对芘的响应更为敏感.CYPs总量与CYP2C9活性对苯并[a]芘的响应趋势相似:暴露初期,苯并[a]芘最高暴露剂量(0.96 mg·kg~(-1))导致二者都显著低于对照水平;试验结束后,苯并[a]芘最低暴露剂量(0.12 mg·kg~(-1))导致二者都显著高于对照水平,最高剂量暴露导致二者都显著低于对照水平,CYP2C9活性对苯并[a]芘的响应更为敏感.因此,推断CYPs亚酶对污染物的响应具有选择性且亚酶的响应敏感度高于CYPs总量.将CYPs总量与CYP1A1活性结合,或与CYP2C9活性结合分别诊断土壤芘污染或并[a]芘污染,较为准确有效.  相似文献

从被石油污染的土壤样品中,以苯并[a]芘为唯一碳源反复驯化,分离筛选出1株高效降解苯并[a]芘的菌株JL 14 .经形态及生理生化特征分析,初步鉴定属于氮单胞菌属(Azomonas) .菌株JL 14在苯并[a]芘浓度为5mg·L- 1 ,2 8℃振荡培养10d ,苯并[a]芘的降解率达到48. 9% .培养基初始pH值分别为4、6、8、10时,2 8℃振荡培养10d ,苯并[a]芘的降解率分别为7 1% ,2 4 8% ,49 6%和2 5 9% .JL 14在苯并[a]芘浓度分别为10mg·L- 1 与2 0mg·L- 1 的条件下,2 8℃振荡培养10d ,苯并[a]芘的降解率分别为3 8. 2 %与2 7. 6% .Zn2 + (2 0 0mg·L- 1 )、Cd2 + (5 0mg·L- 1 )与Pb2 + (2 0 0mg·L- 1 )不影响JL 14对苯并[a]芘的降解作用,但Cu2 + (5 0mg·L- 1 )、Cr2 + (5 0mg·L- 1 )对JL 14有毒性;菲和蔗糖均可促进菌株JL 14对苯并[a]芘的降解作用.  相似文献

Tributyltin(TBT), an organometal used as an antifouling biocide, has been reported to induce masculinization of fish. Benzo [a]pyrene (BaP), a widespread carcinogenic polycyclic aromatic hydrocarbon, has been reported that its microsomal metabolites can produce an estrogenic response when tested in vitro. This study was therefore designed to examine the potential in vivo influence of TBT, BaP and their mixture on sex hormone levels in serum of Sebastiscus marmoratus, which were given 2 separate intraperitoneally (ip) injections(a single injection every 7 d) of TBT(0.5, 1, 5 and 10 mg/kg), BaP(0.5, 1, 5 and 10 mg/kg), or both in combination(0.5, 1, 5 and 10 mg/kg); control fish received olive oil vehicle only. Six days after the 2nd injection, serum samples were collected and analyzed for sex hormone levels and alkali labile protein phosphorus (ALPP), which is related to the yolk precursor protein vitellogenin. The pollutants at all doses significantly reduced serum testosterone, estradiol and ALPP content after 2 injections compared with the corresponding controls. The reduction of the estradiol levels should be response for the decrease of the vitellogenin levels. The results in the present study suggested that aromatase seems not the major target acted by TBT and BaP in fish. This study demonstrated that TBT or BaP exposure both inhibit the reproductive potential in female Sebastiscus marmoratus. Combined effect of TBT and BaP on the serum testosterone, estradiol and ALPP was not antagonism from the anticipation.  相似文献

一株苯并[a]芘高效降解真菌的筛选与降解特性   总被引:2,自引:0,他引:2  
从长期受多环芳烃(PAHs)污染的土壤中分离出一株能够降解苯并[a]芘(B[a]P)的真菌,经鉴定为绿色木霉(Trichoderma viride)(命名为BF-1),并对其以B[a]P为唯一碳源进行反复驯化,考察了B[a]P浓度、不同重金属和培养基对其降解能力的影响.结果表明,菌株BF-1在B[a]P浓度为5mg.L-1,32℃振荡培养约6d的条件下,降解速度最快,B[a]P的降解率达68.28%.BF-1在B[a]P浓度分别为10与25mg.L-1,32℃振荡培养6d的条件下,B[a]P的降解率分别为73.29%与87.36%.Cu2+(50mg.L-1)基本不影响BF-1对B[a]P的降解率;Cd2+(100mg.L-1)、Pb2+(300mg.L-1)对BF-1降解B[a]P有一定影响,但仍表现出较高的耐受能力;而Zn2+(200mg.L-1)对BF-1有明显的抑制作用.选用含5mg.L-1B[a]P的土豆葡萄糖液体培养基,6d后B[a]P的降解率为71.31%.对比前述实验结果表明,培养基对B[a]P降解率的影响并不明显.因此,BF-1的应用价值较高.  相似文献

研究了苯并[a]芘暴露对鲫鱼(Carassius auratus)的几种生物标志物的影响.在鲫鱼接受3个不同质量分数苯并[a]芘(BaP)的腹腔注射14 d后,分析了其肝脏7-乙氧基-3-异吩噁唑酮-脱乙基酶(EROD)活性,以及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)等抗氧化酶活性和红细胞核异常(NA)率变化.结果显示:w(BaP)为1 mg/kg处理组的EROD活性显著升高(P<0.001).w(BaP)为0.1和1 mg/kg处理组SOD活性显著低于对照组(P<0.001),在暴露结束时,w(BaP)为1 mg/h处理组CAT活性显著低于对照组(P<0.01),而w(BaP)为0.1和1 mg/kg处理组GPx活性被显著抑制(P<0.001).表明BaP对EROD活性有诱导作用,而对SOD,CAT和GPx等抗氧化酶活性的抑制作用明显.所有处理组NA率与对照组均无显著差异.因此,鲫鱼EROD,SOD,CAT和GPx等酶活性的改变适于作为生物标志物,可以用来显示暴露于多环芳烃有机污染物鱼类的生活状态.   相似文献

BaP和Cd单一复合对BaP蚯蚓亚细胞分配的影响   总被引:1,自引:0,他引:1  
选用钙离子通道阻断剂氯化镧(LaCl3)和巯基蛋白阻断剂N-乙马来酰亚胺(NEM)对赤子爱胜蚓进行预暴露,然后构建BaP单一或Cd-BaP复合污染,研究不同阻断剂对BaP在蚯蚓不同亚细胞组分(Fraction C:细胞溶质组分;Fraction D:固体颗粒组分;Fraction E:细胞碎片组分)中分配积累特征的影响.结果表明,无论单一或复合污染,BaP主要分布于细胞碎片组分中(占55.42%~69.96%),其次为固体颗粒组分(占27.91%~32.90%),在细胞溶质组分中的浓度最低(占2.13%~11.67%).单一BaP污染下,两种阻断剂对BaP的作用相近,即LaCl3和NEM的加入均能不同程度地促进3个亚细胞组分中BaP的积累.而在Cd和BaP复合污染下,两种阻断剂对BaP的分布积累的影响略有不同,LaCl3能够促进复合污染下BaP在3个亚细胞组分中的积累,而NEM促进了复合污染下BaP在固体颗粒组分和细胞碎片组分中的积累,但抑制了BaP在细胞溶质组分中的积累,浓度从原来的0.99mg/kg降低至0.59mg/kg.因此,钙离子通道和巯基蛋白可能参与BaP在蚯蚓亚细胞的分配积累,相比单一污染,Cd的复合污会进一步改变BaP的积累分配特征.  相似文献

研究了模拟生物泥浆法修复多环芳烃污染土壤.选择了几种从石油污染土壤中分离出来的真菌,研究它们对土壤内苯并[a]芘的降解,并研究了土壤内共存底物:菲、芘、邻苯二甲酸对苯并[a]芘降解的影响,及其之间的共代谢过程.结果表明,芘可以促进镰刀菌、毛霉对苯并[a]芘的降解,并认为这是共代谢作用的结果;菲也可以促进镰刀菌对苯并[a]芘的降解,但抑制了毛霉和青霉对苯并[a]芘的降解;而邻苯二甲酸对青霉和毛霉降解苯并[a]芘的过程均有抑制作用.   相似文献

通过静态吸附实验,研究了3种不同类型的表面活性剂(阳离子∶十六烷基三甲基溴化铵,CTAB;阴离子∶十二烷基苯磺酸钠,SDBS∶非离子,曲拉通100,TX100)对苯并[a]芘(BaP)在黑炭表面吸附解吸的影响.结果表明,3种表面活性剂的存在均会增加BaP在黑炭表面吸附的非线性程度.CTAB的存在可以促进BaP在黑炭表面的吸附同时抑制了BaP的解吸,然而SDBS的存在降低了BaP在黑炭表面的吸附并增加了吸附过程的可逆性.与CTAB和SDBS不同,TX100对BaP在黑炭表面吸附解吸的影响取决于其添加浓度.低浓度的TX100(50 mg.L-1)能够促进BaP在黑炭上的吸附并抑制BaP的解吸,吸附能力参数Kd值(在ce=50μg.L-1下)从188 062 mL.g-1增大到264 179 mL.g-1,解吸滞后指数HI从0.44降低到0.39.随着TX100浓度增加到150 mg.L-1和200 mg.L-1,TX100对BaP的吸附表现出抑制作用并能够促进BaP的解吸,Kd值(在ce=50μg.L-1下)从188 062 mL.g-1分别降低到182 751 mL.g-1和178 730 mL.g-1,解吸滞后指数从0.44分别升高到0.65和0.70.本研究为预测多环芳烃在表面活性剂污染的环境中的分布特征和最终归趋提供了理论依据.  相似文献

The effects of interspecific fungal interactions between Trametes versicolor and Phanerochaete chrysosporium on laccase activity and enzymatic oxidation of polycyclic aromatic hydrocarbons (PAHs) were investigated. A deadlock between the two mycelia rather than replacement of one fungus by another was observed on an agar medium. The laccase activity in crude enzyme extracts from interaction zones reached a maximum after a 5-day incubation, which was significantly higher than that from regions of T. versicolor or P. chrysosporium alone. The enhanced induction of laccase activity lasted longer in half nutrition than in normal nutrition. A higher potential to oxidize benzo[a]pyrene by a crude enzyme preparation extracted from the interaction zones was demonstrated. After a 48 hr incubation period, the oxidation of benzo[a]pyrene by crude enzyme extracts from interaction zones reached 26.2%, while only 9.5% of benzo[a]pyrene was oxidized by crude extracts from T. versicolor. The oxidation was promoted by the co-oxidant 2,2'-azinobis-3-ethylbenzthiazoline-6-sulphonate diammonium salt (ABTS). These findings indicate that the application of co-culturing of white-rot fungi in bioremediation is a potential ameliorating technique for the restoration of PAH-contaminated soil.  相似文献

A bacterial strain BAP5 with a relatively high degradation ability of benzo[a]pyrene (BaP) was isolated from marine sediments of Xiamen Western Sea, China and identified as Ochrobactrum sp. according to 16S rRNA gene sequence as well as Biolog microbial identification system. Strain BAP5 could grow in mineral salt medium with 50 mg/L of BaP and degrade about 20% BaP after 30 d of incubation. Ochrobactrum sp. BAP5 was able to utilize other polycyclic aromatic hydrocarbons (PAHs) (such as phenanthrene, pyrene and fluoranthene) as the sole carbon source and energy source, suggesting its potential application in PAHs bioremediation. The profile of total soluble protein from Ochrobactrum sp. BAP5 was also investigated. Some over- and special-expressed proteins of strain BAP5 when incubated with the presence of BaP were detected by two-dimensional polyacrylamide gel electrophoresis, and found to be related with PAHs metabolism, DNA translation, and energy production based on peptide fingerprint analysis through matrix-assisted laser desorption/ionization-time of flight mass spectrometry.  相似文献

用RBCA和CLEA模型推导土壤中苯并[a]芘的标准值   总被引:4,自引:1,他引:3  
我国现行土壤环境质量标准中尚未包含苯并[a]芘(B[a]P). 分别应用2种国际上认可的RBCA和CLEA模型,获取了土壤中B[a]P的标准值,并对2种模型的计算结果和取值进行了比较. 结果表明,在目标风险水平设定为10-5时,RBCA和CLEA模型计算的住宅用地土壤B[a]P标准值分别为0.83和1.21 mg/kg;工业用地标准值分别为4.10和26.30 mg/kg. 2种模型住宅用地的标准差异不大,且与其他国家的标准水平基本接近;工业用地的标准差异较大,其中RBCA模型的结果更接近于其他国家标准,而CLEA模型的结果则明显偏高. 造成2种模型计算结果差异的原因主要来自于两方面,即模型的方法原理不同导致的暴露量计算差异以及采用的毒理学数据来源不同导致的数据水平差异. 2种模型暴露途径解析的结果均表明,经口摄入和皮肤接触是土壤污染物B[a]P人体暴露的主要途径,而口鼻吸入导致的暴露量很小.   相似文献

观察两种致癌性多环芳烃化合物二甲基苯蒽(DMBA)和苯并(a)芘(BaP)对小鼠肝脏脂质过氧化产物和金属硫蛋白含量的影响。选用雄性C57BL/OLA129小鼠,观察DMBA和BaP各50mg/kg一次腹腔注射染毒后24、48、72和144h后的动物肝脏重量、金属硫蛋白(MT)含量和脂质过氧化程度的变化。MT测定采用镉-血红蛋白亲和分析法;脂质过氧化指标为硫代巴比妥酸测定丙二醛法。结果表明,DMBA和BaP均能引起小鼠肝脏重量增加、MT含量增加和MDA含量增加。随着DMBA和BaP染毒后的时间延长小鼠肝脏的重量不断增加,呈现时间效应关系。小鼠肝脏MT的含量在染毒后24和48h后明显增加1倍。肝脏MDA的含量增加在染毒后24h后达到高峰。本研究观察到DMBA和Bap可以MT增加。MT的增加与MDA的增加呈现一定平行的关。提示MT诱导可能与多环芳烃化合物所致的氧化损伤有关。  相似文献

Two indigenous microorganisms, Bacillus sp. SB02 and Mucor sp. SF06, capable of degrading polycyclic aromatic hydrocarbons (PAHs) were co-immobilized on vermiculite by physical adsorption and used to degrade benzo[a] pyrene (BaP). The characteristics of BaP degradation by both free and co-immobilized microorganism were then investigated and compared. The removal rate using the immobilized bacterial-fungal mixed consortium was higher than that of the freely mobile mixed consortium. 95.3% of BaP was degraded using the co-immobilized system within 42 d, which was remarkably higher than the removal rate of that by the free strains. The optimal amount of inoculated co-immobilized system for BaP degradation was 2%. The immobilized bacterial-fungal mixed consortium also showed better water stability than the free strains. Kinetics of BaP biodegradation by co-immobilized SF06 and SB02 were also studied. The results demonstrated that BaP degradation could be well described by a zero-order reaction rate equation when the initial BaP concentration was in the range of 10--200 mg/kg. The scanning electronic microscope (SEM) analysis showed that the co-immobilized microstructure was suitable for the growth of SF06 and SB02. The mass transmission process of co-immobilized system in soil is discussed. The results demonstrate the potential for employing the bacterial-fungal mixed consortium, co-immobilized on vermiculite, for in situ bioremediation of BaP.  相似文献
