Mitochondrial DNA (mtDNA) and allozyme variation were analysed in samples of mussels collected in 1984 and 1985 from four localities in South West England and one locality in South Wales, a region of Britain where the common mussel (Mytilus edulis) occurs sympatrically and hybridises with the Mediterranean mussel (M. galloprovincialis). Significant differences in mtDNA genotype frequencies for three restriction enzymes (BstEII, XbaI, and EcoRI) were observed between mussels from M. galloprovincialis populations (Padstow and Bude) and those from an M. edulis population (Swansea). Some mtDNA genotypes at high-frequency in M. galloprovincialis were not observed in M. edulis, although there was no indication that mtDNA variation provides greater overall diagnostic power than allozyme variation in distinguishing between the two forms of mussel. Construction of a phylogenetic tree of multiple mtDNA genotypes revealed small mutational distances between the genotypes characterising M. edulis and M. galloprovincialis. The results were consistent with predominant mtDNA flow from M. edulis to M. galloprovincialis. This can be explained by the dispersal of larvae to South West England from M. edulis regions to the north and east, but little dispersal in the opposite directions. Samples from two hybrid populations (Whitsand and Croyde) were analysed. mtDNA genotype frequencies at Croyde were in line with predictions made on the basis of two partially diagnostic allozyme loci (Est-D and Odh), mtDNA frequencies at Whitsand were not. Frequencies of some mtDNA genotypes at Whitsand were characteristic of M. edulis, others of M. galloprovincialis. Differential selective mortality or flow of different mtDNA genotypes and allozyme variation are proposed as possible causes of these results.  相似文献

Taxonomic identification and genetic analysis of larval marine invertebrates have been vexing problems. We describe a polyacrylamide mini-gel electrophoresis technique for resolving proteins from individual larval bivalves (shell length 250 to 350 m) and apply this technique to three species of laboratory-cultured larval oysters [Ostrea edulis L., 1758, Crassostrea gigas (Thunberg, 1793) and c. virginica (Gmelin, 1791)] reared during summer 1989. Electrophoretic patterns of proteins clearly discriminate among the three species and allow genetic analysis of a polymorphic allozyme locus (Pgi) in field-collected larvae and juveniles of C. virginica. This technique provides an economical tool for largescale taxonomic, ecologic, and genetic studies of meroplanktonic stages of various species.  相似文献

Growth comparisons were made involving mussels (Mytilus spp.) collected from five different localities in Britain in 1980–1981. Two of the localities, Mumbles, South Wales, and Bude, Southwest England, have pure populations of M. edulis and M. galloprovincialis, respectively. The other three, Whitsand Bay, Southwest England, Croyde Bay, Southwest England and Robin Hood's Bay, Northeast England, have hybrid populations with both M. edulis and M. galloprovincialis ancestry. To make growth comparisons, mussels from different populations were mixed in oyster nets and transplanted to three different localities. After periods of transplantation varying between several months and one year, growth was assessed by measuring increase in shell length or dry body weight. Starch-gel electrophoresis was used to assay variation in the transplanted mussels at three allozyme loci partially diagnostic for M. edulis and M. galloprovincialis. The results provide evidence of small growth differences between populations and between allozyme genotypes within populations. These differences accounted for no more than a few percent of the total variation in growth between mussels. Statistically significant results were obtained, but were frequently found not to be reproducible. There is no clear evidence of a growth difference between M. edulis and M. galloprovincialis. Allozyme heterozygotes appear to have growth rates intermediate between allozyme homozygotes; this study thus fails to provide evidence for overdominance with respect to growth rate.  相似文献

Mussel samples were collected at 4 to 6 wk intervals throughout 1987 from two hybridMytilus edulis/M. galloprovincialis populations, at Croyde Bay and Whitsand Bay, in southwest England. These were analyzed at two polymorphic loci which are diagnostic for allozyme differences which typifyM. edulis andM. galloprovincialis. Dry mantle weight as a function of shell length was determined for all individuals of each sample. Size-frequency data for the two populations was obtained in September 1987 and March 1988. For all genotypes at both sites, fecundity was a function of shell length, and in both populations the frequency ofM. galloprovincialis alleles was positively correlated with shell length. At both sites, allozyme genotype explained a significant amount of variation in mantle weight either when assessed as a main effect or when assessed as an interaction with shell length or time of collection. At Croyde,M. galloprovincialis mussels had greater estimated fecundity per unit length than theM. edulis mussels. Differences in the timing of spawning activity between theM. edulis and theM. galloprovincialis mussels were inferred, and these differences might act to reduce the amount of interbreeding at Croyde. At Whitsand, a reduced level of variability in the timing of spawning activity and fecundity between the genotypes was observed and explained by a higher degree of genetic mixing. Because theM. galloprovincialis mussels had (1) a greater estimated fecundity at any length, and (2) a greater mean length than theM. edulis mussels, the mean genotypic annual fecundity perM. galloprovincialis mussel was 2.8 times greater than an individualM. edulis mussel at Croyde, and 2.2 times greater than an individualM. edulis mussel at Whitsand. This evidence thatM. galloprovincialis mussels have an advantage in fecundity, and thus perhaps in fertility, taken together with the evidence thatM. galloprovincialis also has a higher viability, indicates directional selection in favour of theM. galloprovincialis phenotype. Because of the observed temporal stability of the population it seems likely that this selection is counterbalanced by a massive imigration ofM. edulis spat from neighbouring populations.  相似文献

Mussel samples were collected from a hybrid mussel (Mytilus galloprovincialis and M. edulis) Population at Croyde, southwest England, in January, March and May 1990. The strength of attachment of each mussel to the substrate was measured with a spring balance. A number of diagnostic characters were also recorded. These are shell lengh, width and height, mantle colour and genotype at two allozyme loci, esterase-D and octopine dehydrogenase. Multiple-regression analysis was used to assess the effect of the diagnostic characters on strength of attachment as dependent variable. Mussels possessing the relatively high shells and darker mantle colouration characteristic of M. galloprovincialis had higher values, on average, for strength of attachment than mussels resembling M. edulis. Phenotypically intermediate mussels had intermediate values for strength of attachment. The results suggest an adaptive difference which can account for reports of differential mortality acting in favour of M. galloprovincialiis.  相似文献

We examined natural selection within a population of marine mussels, sampled in southwestern England in June 1991, containing a high frequency of hybrids between Mytilus edulis L. and M. galloprovincialis Lmk. This system is particularly tractable for the assessment of natural selection because hybridization is common and individual mussels can be aged, allowing changes in the frequency of hybrid genotypes among age classes to be determined. We show that strong viability selection occurs among hybrid genotypes which results in the virtual elimination of M. edulis–like genotypes from the population over a period of 3 years. Recombinant hybrid genotypes are intermediate in fitness, with M. edulis–like genotypes having a lower survival rate and M. galloprovincialis–like genotypes having a higher survival rate than genotypes of mixed ancestry. Since intermediate fitness for hybrid genotypes is inconsistent with endogenous selection models we conclude that the structure and position of this hybrid zone is probably generated by exogenous selection. This pattern of selection is a recurring feature of this hybrid population and likely occurs elsewhere in the hybrid zone. Selection against M. edulis–like genotypes appears to be offset by extensive immigration of larvae dispersed from pure populations of M. edulis. Two South Australian rocky intertidal platforms were sampled in April 1981 and December 1982 (Marino Rocks) and in December 1982 (Lady Bay), respectively. Three snail species, Nerita atramentosa Reeve, 1855, Bembicium nanum (Lamarck, 1822) and Austrocochlea concamerata (Wood, 1828), exhibited shore-level size-gradients, with smaller individuals occupying the higher intertidal levels. The reef crab (Ozius truncatus M.-Edwards, 1834), whose distribution overlaps that of the gastropods, is an important predator of these gastropods. The shore-level distribution of shell sizes can be explained by differential selection against smaller individuals by reef crabs. The decreased density of small snails at lower intertidal levels may be the result of either their consumption by reef crabs and/or their movement to higher shore levels where crab predation is less intense.  相似文献

In the coastal waters of Florida (USA) tadpole larvae of the colonial ascidian Ecteinascidia turbinata contain chemicals which make them unpalatable to planktivorous juvenile pinfish Lagodon rhomboides. Experiments demonstrate that the bright organe color of E. turbinata tadpoles is aposematic. Fish that have recently tasted larvae of E. turbinata will not attack the palatable tadpoles of Clavelina oblonga when the latter are dyed organe to resemble larvae of E. turbinata. Tadpoles of E. turbinata that have been mouthed and rejected by fish generally survive to complete a normal metamorphosis. Individual selection explains the evolution of aposematic coloration in E. turbinata better than kin selection. The identity of the defensive chemical is unknown. The unpalatable substance in larvae of E. turbinata is removed by dialysis, indicating that it has a molecular weight less than 14000 d. Larvae are not acidic, nor is the active substance denatured by doiling.  相似文献

Many marine organisms inherently express the presence and function of a new defence mechanism, termed multixenobiotic resistance mechanism (MXRM) because of its similarity to multidrug resistance (MDR) found in tumor cell lines resistant to chemotherapeutic drugs. However, previously no information was available on a possible induction of the activity of MXRM in organisms living at polluted sites. The purpose of the present study was to demonstrate the inducibility of this defence mechanism in the marine snail Monodonta turbinata, an organism known to inhabit a wide range of environmental conditions existing along a pollution gradient. Specimens of M. turbinata used in the present study were collected from March 1993 through March 1994 near Rovinj, Croatia. The accumulation of generally labelled 3H-vincristine (3H-VCR) in the gills of snails living at an unpolluted site was 67% higher and very sensitive to verapamil, an inhibitor of P-glycoprotein activity in comparison to snails living at a polluted site. The accumulation of vincristine (VCR) in snails from a polluted site was reduced and insensitive to verapamil due to the induced state of P-glycoprotein, as was demonstrated immunochemically by the enhanced concentration of a 140 kDa protein cross-reacting with the polyclonal antibodies raised against mammalian P-glycoprotein. MXRM could be induced in snails from the unpolluted site to the level found in snails living at a polluted site either by transplantation for 3 d to a polluted site, or by exposure for 3 d to sea water polluted with Diesel-2 oil (hydrocarbon concentration equivalent to 50 ppb of Kuwait oil). Discovery of the inducibility of this new defence mechanism in aquatic organisms may have important implications in ecotoxicology, as was demonstrated by a 104% enhanced accumulation of VCR in the presence of the MXRM-inhibitor, the so-called chemosensitizer. Besides, a striking difference in the levels of MXRM-elements (the sensitivity of the accumulation rate of xenobiotics to verapamil, immunochemical expression of P-glycoprotein, and the sensitivity of the binding of xenobiotics on membrane vesicles to verapamil) found between specimens living at the polluted and unpolluted sites offers a new molecular biomarker for exposure to pollutants.  相似文献

This paper reports data on 28 allozyme loci in wild and artificially reared sea bass (Dicentrarchus labrax) samples, originating from either coastal lagoon or marine sites in the Mediterranean Sea. F ST analysis (θ estimator) indicated strong genetic structuring among populations; around 34% of the overall genetic variation is due to interpopulation variation. Pairwise θ estimates showed that, on average, the degree of genetic structuring was much higher between marine populations than between samples from lagoons. Six polymorphic loci showed differences in allele frequencies between marine and lagoon samples. Multivariate analyses of individual allozymic profiles and of allele frequencies suggested that different arrays of genotypes prevail in lagoons compared to marine samples, particularly at those loci that, on the basis of previous acclimation experiments, had been implicated in adaptation to freshwater. On the other hand, variation at “neutral” allozyme loci reflects to a greater extent the geographic location of populations. Allozyme differentiation was also studied in a D. labrax population from the Portuguese coast. Average genetic distance between this population and the Mediterranean populations was quite high (Nei's D = 0.236) and calls into question the taxonomic status of the Portuguese population. Finally, genetic relationships between D. labrax and D. punctatus were evaluated. Average Nei's D was 0.648, revealing high genetic differentiation between the two species, even for two sympatric populations of these species in Egypt; thus gene flow was not indicated between species. Electrophoresis was used to survey a number of enzymes for loci of potential value as stock markers in Chlamys opercularis (L.). Two loci, Pgi and Pgd, were then studied in larger samples from each of two localities around the Isle of Man. One Pgi allele was found to be restricted to a particular year-class in both populations. This was confirmed by subsequent studies of three further populations, in each of which the same allele occurred only in the one year-class. Otherwise, no significant differences in allele frequencies were found between year-classes or between populations in any of the samples used. From the results it is concluded that there is temporal variation in the sources of recruitment of C. opercularis stocks around the Isle of Man. Possible origins of larvae contributing to these stocks are discussed in the light of what is known of current circulation patterns in the northern Irish Sea.  相似文献

Multilocus allozyme heterozygosity (MLH) has been positively correlated with growth in some marine bivalves and was suggested to facilitate swimming activity in pectinids. Using two highly mobile scallops, Placopecten magellanicus and Argopecten purpuratus, we examined escape response performance and morphometric characteristics as a function of allelic variability at metabolic loci. Ten allozyme systems were used for A. purpuratus and 7 for P. magellanicus. In each species, the morphometric characteristics and escape response parameters were analyzed separately using principal components analysis (PCA) and the scores of the major principal components were related to allozyme heterozygosity. In both P. magellanicus and A. purpuratus, positive correlations were found between MLH and morphometric parameters, but escape response parameters were only positively linked to MLH in P. magellanicus, and then weakly. The hypothesis that MLH improves fitness of pectinids by increasing the capacity to escape predators is not supported.  相似文献

Inter-and intraspecific allozyme differentiation in the mysid crustacean genus Mysis in the North Atlantic region was studied in order to evaluate earlier concepts of evolutionary and systematic relationships and to assess patterns of subdivision within widespread taxa. The results support a relatively ancient divergence of the marine and non-marine species of the genus, and are generally in line with the current subgeneric tridivision into Mysis s.str., Michteimysis and Auricomysis. However, the North American littoral species M. gaspensis should be returned to subgenus Mysis s.str. from its present position in Michteimysis with M. mixta. The closest observed affinities within Mysis s.str. were between M. gaspensis and the freshwater M. relicta group, and between M. oculata and M. litoralis. Intraspecific differentiation among North European coastal populations of M. oculata and M. litoralis was moderately strong (F ST0.1), suggesting population bottlenecks and limited dispersal in the post-glacial time. On the other hand, ransoceanic differences were not essentially greater, indicating the systematic homogeneity and long-term dispersal capacity in the marine species. This contrasts with the strong genetic and systematic fragmentation earlier found within the circumboreal M. relicta species group.  相似文献

In-situ feeding habits of the copepods Temora turbinata and T. stylifera were investigated by scanning electron microscope examination of fecal pellets, the contents of which reflected copepod gut contents upon capture. Pellet contents were compared with assemblages of available phytoplankton in the water column at the times of zooplankton sampling. Samples were collected in continental shelf and slope waters of the Gulf of Mexico near the mouth of the Mississippi River. Both species ingested a wide size range and taxonomic array of phytoplankters, and to a lesser extent, other crustaceans. Fecal pellets contained primarily the remains of the phytoplankters that were most abundant in the water at times of collection. There was considerable overlap in the food items ingested by adult females of both copepod species, or two stages of T. turbinata copepodites. Thus, T. turbinata and T. stylifera are omnivores, but primarily opportunistic herbivores.  相似文献

In Ireland, mussels on exposed rocky shores constitute an interbreeding mixture of two forms of mussels, the blue mussel, Mytilus edulis, and the Mediterranean mussel, M. galloprovincialis. Results from an Irish study in the 1980s, using partially diagnostic allozyme markers, indicated that mussels higher up the shore were more galloprovincialis-like than those lower down. In this study we set out to test two hypotheses: (a) recruits arriving on the shore are composed of genetically distinct cohorts that settle preferentially at different levels on the shore, and maintain genetic distinctiveness into adulthood; (b) recruits are genetically homogeneous, but once settled they diverge genetically over time, due to within-habitat site specific-selection. The diagnostic Me 15/16 DNA marker was used to analyse the genetic composition of newly-settled spat recruiting to artificial substrates, which were placed at two-week intervals from May–October 2002, on the mid- and low shore areas of two exposed sites in Galway Bay. Adult mussels were also collected on each sampling date. Results did not support the preferential settlement hypothesis, i.e., the genetic composition of primary settlers (≤ 500 μm) was similar between tidal heights and shores. Neither was there evidence of post settlement selective mortality, as adults were genetically similar to settling spat. In spat and adults the frequency of the M. galloprovincialis allele was high (0.56–0.80), due to high frequencies of M. galloprovincialis (> 37%) and hybrid (> 33%) genotypes, and correspondingly low frequencies of the M. edulis genotype (< 11%). Adult mussels from a nearby sheltered estuarine site, while significantly different to exposed shore mussels, still had low frequencies of the M. edulis genotype (< 17%), indicating no apparent advantage for the genotype in this environment. There are indications that the genetic composition of mussels may be changing on the Atlantic coasts of Ireland.  相似文献

Clonal reproduction, a common life history strategy among sessile marine invertebrates, can lead to high local abundances of one to a few genotypes in a population. Analysis of the clonal structure of such populations can provide insight into the ecological and evolutionary history of the population, but requires markers that can identify individual genets. Forensic and demographic studies have demonstrated that DNA fingerprinting can provide markers that are unique for an individual genotype. We have generated DNA fingerprints for over 70 colonies of the clonal gorgonian, Plexaura A (Plexaura sp. A) collected from June 1990 through July 1991 in the San Blas Islands, Panama. DNA fingerprints within a singic individual were identical and fingerprinting resolved multiple genotypes within and among reefs. On one reef in the San Blas Islands, Panama, 59% of the colonies sampled were of one genotype and this genotype was not found on any other sampled reefs. A previous study using tissue grafts identified 13 putative clones on these reefs, while DNA fingerprints of the same colonies differentiated 17 genotypes. The present study demonstrates the utility of DNA fingerprinting for distinguishing clones and for identifying clonal structure of marine invertebrate populations.  相似文献

The development of monitoring strategies for the early warning of seawater pollution, with particular reference to faecal and hydrocarbon contamination, has been the specific goal of research carried out within the Cluster 10-SAM (Advanced Systems for Coastal Marine Monitoring) Project, funded by the Italian Ministry for University and Scientific Research. Advanced analytical approaches have been designed and applied to detect bacterial species that have been selected as potential indicators of pollution in seawater samples. We report the results obtained using the fluorescent antibody and enzymatic assays for the detection of Escherichia coli, and a real-time PCR protocol for monitoring marine areas for hydrocarbon pollution. Immunofluorescence and enzymatic methods revealed the occurrence of different faecal pollution levels, reaching 105 E. coli cells 100 ml?1 in the Straits of Messina. Real-time PCR results corresponded to the different degree of oil pollution of the analysed samples. The specificity and speed make these methods promising for the detection and evaluation of marine pollution.  相似文献

The genetic relationships between morphologically indistinguishable marine and brackish-water populations of Monocelis lineata (O.F. Müller, 1774) (Proseriata: Monocelididae) were analysed by means of allozyme electrophoresis. Fifteen samples of M. lineata (13 from the Mediterranean and two from the Atlantic) from coastal marine and brackish-water habitats were examined for variation at 18 loci. Eleven loci were polymorphic in at least one population of M. lineata. Low levels of within-population genetic variability were found, with average observed and expected heterozygosity values ranging from Ho=0.015±0.015 to 0.113±0.044, and from He=0.028±0.028 to 0.138±0.054, respectively. The occurrence of a number of private alleles indicated a marked genetic divergence among populations of M. lineata, with Rogers genetic distances ranging from DR=0.003 to 0.676 and a highly significant FST value (0.918±0.012, P<0.001). UPGMA (unweighted pair-group method using arithmetic average) cluster analysis and multidimensional scaling showed a clear genetic divergence between marine and brackish-water populations. Moreover, Atlantic and Mediterranean populations were sharply separated. Our results suggest that M. lineata is a complex of sibling species.Communicated by R. Cattaneo-Vietti, Genova  相似文献

Investigations into several potential fitness parameters in collections of the estuarine bivalve Geukensia demissa (Dillwyn), (from Newport Bay and Alamitos Bay in southern California, conducted from 1981 to 1984) under both field and laboratory conditions, revealed differences in growth rate, tissue weight, and mortality which were correlated with leucine aminopeptidase-1 (LAP-1) genotype. The results of these fitness studies are consistent among themselves and with the results of previous population genetic studies on G. demissa. As a whole, these studies suggest that differential fitness among LAP-1 genotypes in conjunction with a variable environment act to maintain the LAP-1 polymorphism in natural populations of this species.  相似文献

Calanoid copepods typically exhibit escape reactions to hydrodynamic stimuli such as those generated by the approach of a predator. During the summers of 2000, 2001 and 2004, two small calanoid species, Temora turbinata Dana, 1849 and Paracalanus parvus Claus, 1863 were exposed to a visual predatory fish, the blenny Acanthemblemaria spinosa Metzelaar, 1919, and their predator–prey interactions were recorded using both high-speed and standard videographic techniques. Copepod escape reaction components, including swimming pattern, reactive distance, turning rate, and jump kinetics, were quantified from individual predation events using motion analysis techniques. Among the observed escape reaction components, differences were noted between the species’ swimming patterns prior to attack and their response latencies. Temora turbinata was a continuous cruiser and P. parvus exhibited a hop-and-sink swimming pattern. During periods of sinking, P. parvus stopped beating its appendages, which presumably reduced any self-generated hydrodynamic signals and increased perceptual abilities to detect an approaching predator. Response latency was determined for each copepod species using a hydrodynamic stimulus produced by a 1 ms acoustic signal. Response latencies of T. turbinata were significantly longer than those of P. parvus. Despite some apparent perceptual advantages of P. parvus, the blenny successfully captured both species by modifying its attack behavior for the targeted prey.  相似文献
