The purpose of this study was to demonstrate that shellfish can be used to detect enteric viruses in marine waters where they are present at very low concentrations. Aqua-cultured mussels were placed in the sea just off the mouth of a drainage channel affected by human and animal faecal contamination. Samples were taken from the channel, the sea and the mussels at intervals over two 4-week periods. The samples were tested to verify the presence of both rotaviruses and E. coli. Rotaviruses were detected by Real Time-PCR, typed by multiplex PCR and subsequently sequenced. E. coli was enumerated in water matrices by a filtering method and in mussels by the MPN method. The presence of E. coli in the examined matrices demonstrates contamination of faecal origin throughout the studied environments. Rotaviruses were recorded in channel waters, but not in sea water. In both experiments, rotaviruses were detected in mussels 21 and 28 days after being placed in the sea water off the channel mouth. The use of mussels thus enabled the detection of rotaviruses in waters where the high dilution rendered direct investigation impossible. This study indicates that mussels can be used in marine virological surveillance programs.  相似文献

In the last decade, various molecular methods (e.g., fluorescent hybridization assay, sandwich hybridization assay, automatized biosensor detection, real-time PCR assay) have been developed and implemented for accurate and specific identification and estimation of marine toxic microalgal species. This review focuses on the recent quantitative real-time PCR (qrt-PCR) technology developed for the control and monitoring of the most important taxonomic phytoplankton groups producing biotoxins with relevant negative impact on human health, the marine environment, and related economic activities. The high specificity and sensitivity of the qrt-PCR methods determined by the adequate choice of the genomic target gene, nucleic acid purification protocol, quantification through the standard curve, and type of chemical detection method make them highly efficient and therefore applicable to harmful algal bloom phenomena. Recent development of qrt-PCR-based assays using the target gene of toxins, such as saxitoxin compounds, has allowed more precise quantification of toxigenic species (i.e., Alexandrium catenella) abundance. These studies focus only on toxin-producing species in the marine environment. Therefore, qrt-PCR technology seems to offer the advantages of understanding the ecology of harmful algal bloom species and facilitating the management of their outbreaks.  相似文献

If Europe is serious about reaching its target to keep global mean temperature increase below 2 °C, it must strive for a 100% renewable electricity system by 2050. The SuperSmart Grid approach combines what is often perceived as two exclusive alternatives: wide area power generation and decentralised power generation. We argue that by combining these, in fact, complementary measures, it is possible to address the crucial issue of renewable generation—fluctuating supply—in a comprehensive as well as in a technologically and economically viable manner. Thus, the SuperSmart Grid simultaneously can contribute to energy security, climate security, social security, and national security.  相似文献

Variation of the amplification effect of burrows of the leptodactylid frog Eupsophus emiliopugini on conspecific calls generated externally was investigated. Advertisement calls broadcast through a loudspeaker placed in the vicinity of a burrow were monitored with small microphones positioned inside and outside the cavity. For 150 presentations of calls of 15 individuals in 12 burrows, 134 were amplified and 16 were attenuated (range –6–13 dB). The fundamental resonant frequency of burrows, measured with broadcast noise and pure tones, averaged 814 Hz (range 302–1361 Hz) and covaried with burrow length. The dominant frequency of the calls of burrow occupants (average 1062 Hz, range 636–1459 Hz) was not correlated with the fundamental resonant frequency of these cavities. In burrows with low resonant frequencies, externally broadcast calls with high dominant frequencies were attenuated, or amplified to a lower extent than calls with lower dominant frequencies. The dominant frequencies of the calls experienced shifts towards the burrows’ fundamental resonant frequencies. The amplification of calls inside burrows of E. emiliopugini exhibits manifest variability, with considerable potential for facilitating acoustic interactions in this species.
