Several ABC transporters have been characterized from many aquatic organisms, but no information is yet available for Antarctic fish. The aim of this work was to identify the expression of genes for ABC proteins in Trematomus bernacchii, a bioindicator species of the Southern Ocean. Partial cDNA sequences of ABCB1, ABCC1, ABCC2, ABCC4 and ABCC9 were cloned from liver. Using RACE technology, 3.5 and 2.2 kb contigs were obtained for ABCB1 and ABCC2. Considering the elevated natural bioavailability of cadmium at Terra Nova Bay, responsiveness of ABCB1 and ABCC2 to this element was investigated under laboratory conditions. ABCB1 and ABCC2 mRNA levels were approximately four-fold higher in Cd-exposed fish compared to the controls. Induction of ABCB1 protein was also found by western blot. This study provides the first identification of five ABC genes in the liver of an Antarctic key species, some of which may be involved in cellular detoxification. 相似文献

Aquatic ecosystems around the world, lake, estuaries and coastal areas are increasingly impacted by anthropogenic pollutants through different sources such as agricultural, industrial and urban discharges, atmospheric deposition and terrestrial drainage. Lake Victoria is the second largest lake in the world and the largest tropical lake. Bordered by Tanzania, Uganda, and Kenya, it provides a livelihood for millions of Africans in the region. However, the lake is under threat from eutrophication, a huge decline in the number of native fish species caused by several factors including loss of biodiversity, over fishing and pollution has been recently documented. Increasing usage of pesticides and insecticides in the adjacent agricultural areas as well as mercury contamination from processing of gold ore on the southern shores are currently considered among the most emergent phenomena of chemical contamination in the lake. By the application of globally consistent and comprehensive geospatial data-sets based on remote sensing integrated with information on heavy metals accumulation and insecticides exposure in native and alien fish populations, the present study aims at assessing the environmental risk associated to the contamination of the Lake Victoria water body on fish health, land cover distribution, biodiversity and the agricultural area surrounding the lake. By the elaboration of Landsat 7 TM data of November 2002 and Landsat 7 TM 1986 we have calculated the agriculture area which borders the Lake Victoria bay, which is an upland plain. This process has greatly enhanced nutrient loading to the soil, which is subsequently transported to the lake by rain or as dry fall. All the data has been insert in the Geographical information System (ARCGIS) to be upgraded and consulted. Heavy metals in fish fillets showed concentrations rather low except for mercury being higher than others as already described in previous investigations. In the same tissue, cholinesterases activity (ChE) as an indicator of insecticides exposure showed significant differences among fish species in both activity and sensitivity of selected inhibitor insecticides. This integrated approach aims at identifying and quantifying selected aquatic environmental issues which integrated with monitoring techniques such as contaminant concentrations and biological responses to insecticides exposure in fish populations will provide a scientific basis for aquatic zones management and assist in policy formulations at the national and international levels. 相似文献

Exposure to lead (Pb) may affect adversely human health. Mapping soil Pb contents is essential to obtain a quantitative estimate of potential risk of Pb contamination. The main aim of this paper was to determine the soil Pb concentrations in the urban and peri-urban area of Cosenza–Rende to map their spatial distribution and assess the probability that soil Pb concentration exceeds a critical threshold that might cause concern for human health. Samples were collected at 149 locations from residual and non-residual topsoil in gardens, parks, flower-beds, and agricultural fields. Fine earth fraction of soil samples was analyzed by X-ray Fluorescence spectrometry. Stochastic images generated by the sequential Gaussian simulation were jointly combined to calculate the probability of exceeding the critical threshold that could be used to delineate the potentially risky areas. Results showed areas in which Pb concentration values were higher to the Italian regulatory values. These polluted areas were quite large and likely, they could create a significant health risk for human beings and vegetation in the near future. The results demonstrated that the proposed approach can be used to study soil contamination to produce geochemical maps, and identify hot-spot areas for soil Pb concentration. 相似文献

Using a dynamic model of Lake Chozas developed by Marchi et al. (2011), we tested three hypotheses about recovery of the indigenous community and water quality after radical changes caused by introduction of an invasive allochthonous crayfish, Procambarus clarkii:

- Can the lake resist the pressure of an invasive species, like P. clarkii, by adaptation?
- 2.
- Can the ecosystem recover when all the crayfish are removed and low phosphorus concentrations persist in inflow water?
- 3.
- Does the simulated recovery of submerged vegetation occur at a total phosphorus concentration below 100 mg TP m−3, as estimated by Scheffer et al. (1993), Scheffer (1997), Jeppesen et al. (1998) and Zhang et al. (2003)?
- 1.
- Lake Chozas can at least partly resist by adaptation. A combination of possible parameter changes could lead to a significant increase in eco-exergy.
- 2.
- Removal of the phosphorus represented by crayfish (by harvesting) implies complete recovery of the lake and its eco-exergy, albeit not necessarily with the same organisms having the same properties.
- 3.
- The expected hysteresis created by introduction and harvesting of crayfish is observed under the following conditions: phytoplankton dominance at total phosphorus ≥ about 200-250 mg TP m−3 and submerged vegetation returns at total phosphorus < 100 mg TP m−3.

We analyzed polychlorobiphenyls (PCBs), perfluorooctanesulfonate (PFOS) and perfluorooctanoate (PFOA) in the edible part of farmed sea bass reared in two fish farms in Liguria (Northern Italy). The aim was to determine the contamination levels and human exposure to these chemicals through fish consumption. Concentrations of “dioxin-like” PCBs (DL-PCBs) ranged from 0.033 to 0.759 pg ΣTEQ-PCB g−1 whole weight (w.w.) in fish farm 1 and from 0.032 to 1.60 pg ΣTEQ-PCB g−1 w.w. in fish farm 2, and the six indicators of “non-dioxin-like” (NDL-PCBs) from 0.538 to 9.33 ng Σ6PCB g−1 w.w. and from 1.62 to 27.6 ng Σ6PCB g−1 w.w. Concentrations were generally lower in farm 1 than in farm 2. One reason for this difference might be the proximity of farm 2 to the seaport of La Spezia, which could be a punctual source of pollutants influencing the contamination of the water in the farm. Principal component analysis (PCA) showed differences also in the congeners profiles for the two sites, with higher-chlorinated PCBs more abundant in farm 1, and lower-chlorinated PCBs were more abundant in farm 2. Most of the concentrations of PFOS and PFOA were below the limit of detection (LOD 0.05 ng g−1 w.w.). Only about 10% of the samples analyzed had levels slightly higher than the LOD. Assessments of exposure using these data showed that consumption of farmed fish may contribute significantly to PCBs through the diet, whereas the contribution of PFOS and PFOA seems to be low. 相似文献

Gene expression experiments were targeted in order to monitor the ABC efflux transporters, which is potentially involved in cellular detoxification/defense. Changes in expression levels of different ABC genes in kidney of Oncorhynchus mykiss fed with melamine and melamine + cyanuric acid enriched diets were recorded in both treated groups by mRNA ΔΔCT relative quantification method. Expression profiles of eight different ABC genes basically showed low alterations in melamine group and more consistent changes in melamine + cyanuric acid treated fish, compared with own control. In the last group ABCC2 gene over expression was the more evident alteration. These results suggest that ABC efflux system could be involved in mobilization of hydrophilic molecules in the forcing condition of chronic exposure. 相似文献

Achieving ethically responsible decisions is crucial for the success of biodiversity conservation projects. We adapted the ethical matrix, decision tree, and Bateson's cube to assist in the ethical analysis of complex conservation scenarios by structuring these tools so that they can implement the different value dimensions (environmental, social, and animal welfare) involved in conservation ethics. We then applied them to a case study relative to the decision-making process regarding whether or not to continue collecting biomaterial on the oldest of the two remaining northern white rhinoceroses (Ceratotherium simum cottoni), a functionally extinct subspecies of the white rhinoceros. We used the ethical matrix to gather ethical pros and cons and as a starting point for a participatory approach to ethical decision-making. We used decision trees to compare the different options at stake on the basis of a set of ethical desiderata. We used Bateson's cube to establish a threshold of ethical acceptability and model the results of a simple survey. The application of these tools proved to be pivotal in structuring the decision-making process and in helping reach a shared, reasoned, and transparent decision on the best option from an ethical point of view among those available. 相似文献

The characterization of thermal decomposition of cumene hydroperoxide (CHP) in cumene from kinetic point of view is of great interest for industrial applications. However, the efforts done so far by the researchers lead only to the availability of single kinetic laws (first order, autocatalytic, etc.) whose usefulness is strongly limited by the presence in the starting solutions of some impurities. A detailed model aiming at simulating both, thermal and kinetic behavior of the system, is now presented. In other words, the proposed model tries to simulate CHP thermal decomposition process at varying reaction conditions (different initial temperature, in the range 120–160 °C, different initial CHP concentrations, addition of α-methylstyrene to CHP solutions in cumene) and during isothermal, scanning and adiabatic runs. A good capability of the model is observed to simulate the system behavior by using data collected under isothermal and adiabatic conditions and during scanning calorimetric runs. It is important to stress that the use of the model proposed in the present investigation does not require the adoption of adjustable parameters. 相似文献

Bivalve mollusks as filter-feeders concentrate in their digestive tissue microorganisms likely present in the harvesting water, thus becoming risky food especially if consumed raw or poorly cooked. To eliminate bacteria and viruses eventually accumulated, they must undergo a depuration process which efficacy on viruses is on debate. To better clarify the worth of the depuration process on virus elimination from mussels, in this study we investigated rotavirus kinetics of accumulation and depuration in Mytilus galloprovincialis experimentally contaminated. Depuration process was monitored for 9 days and virus residual presence and infectivity were evaluated by real time quantitative polymerase chain reaction, cell culture and electron microscopy at days 1, 2, 3, 5, 7, 9 of depuration. Variables like presence of ozone and of microalgae feeding were also analyzed as possible depuration enhancers. Results showed a two-phase virus removal kinetic with a high decrease in the first 24 h of depuration and 5 days necessary to completely remove rotavirus. 相似文献
