Organotin compounds are widely used as biocides in marine and terrestrial environments. Several currently used techniques allow either the measurement of the chemicals or their effects on living organisms. Our current research focuses on the development of a complementary method based on a bacterial bioluminescence-based bioassay for the specific detection of organotin compounds. The performance of the bioassay was assessed. The Escherichia coli bacterial strain used in this study is specific for TBT and DBT (with Cl, Br or I as the halogen group) with the central tin atom important for light production. The assay is conducted after overnight culture of the bacterial strain, followed by 60 min of contact time with the organotin compound for significant light production. The detection limits were found to be 0.08 microM for TBT (26 microgl(-1)) and 0.0001 microM for DBT (0.03 microgl(-1)) with a linear range of one logarithm. The repeatability of the bioassay is 8% and the reproducibility for TBT and DBT was approximately 14%. Lyophilization of the strains did not significantly modify the detection limit as well as the range of detection. Applications of the bioassay to environmental samples are discussed.  相似文献

Terrestrial invertebrates are becoming widely established as tools to assess heavy metal pollution at contaminated sites. A practical and time saving method to sample terrestrial invertebrates consist of pitfall traps, often filled with a 4% formaldehyde solution and some detergent. The reliability of metal concentrations based on organisms captured and stored in this solution might however be questioned and we therefore tested the effect of formaldehyde on Zn, Cu, Cd and Pb concentration experimentally in three isopod species. Our results showed that in many cases, significant decreases in Cu concentrations compared to animals stored in a freezer were observed that could be as high as 40%, while Zn, Cd and Pb concentrations increased. A regression analysis of individual dry weight on individual size revealed that formaldehyde decreases the dry weight substantially and in that way causes increased measurements of Zn, Cd and Pb concentrations. We conclude that pitfall traps with formaldehyde should better not be used to collect animals in which concentrations of heavy metals or other toxic substances will be determined.  相似文献

Emission factors for selected volatile organic compounds (VOCs) and particulate emissions were developed while processing eight commercial grades of polycarbonate (PC) and one grade of a PC/acrylonitrile-butadiene-styrene (ABS) blend. A small commercial-type extruder was used, and the extrusion temperature was held constant at 304 degrees C. An emission factor was calculated for each substance measured and is reported as pounds released to the atmosphere/million pounds of polymer resin processed [ppm (wt/wt)]. Scaled to production volumes, these emission factors can be used by processors to estimate emission quantities from similar PC processing operations.  相似文献

Temporal and spatial variability in wetland water-quality variables were examined for twenty-one wetlands in the Minneapolis/St. Paul metropolitan area and eighteen wetlands in adjacent Wright County. Wetland water quality was significantly affected by contact with the sediment (surface water vs. groundwater), season, degree of hydrologic isolation, wetland class, and predominant land-use in the surrounding watershed (p<0.05). Between years, only nitrate and particulate nitrogen concentrations varied significantly in Wright County wetland surface waters. For eight water-quality variables, the power of a paired before-and-after comparison design was greater than the power of a completely randomized design. The reverse was true for four other water-quality variables. The power of statistical tests for different classes of water-quality variables could be ranked according to the predominant factors influencing these: climate factors>edaphic factors>detritivory>land-use factors>biotic-redox or other multiple factors.For two wetlands sampled intensively, soluble reactive phosphate and total dissolved phosphorus were the most spatially variable (c.v.=76–249%), while temperature, color, dissolved organic carbon, and DO were least variable (c.v.=6–43%). Geostatistical analyses demonstrated that the average distance across which water-quality variables were spatially correlated (variogram range) was 61–112% of the mean radius of each wetland. Within the shallower of the two wetlands, nitrogen speciation was explained as a function of dissolved oxygen, while deeper marsh water-quality variables were explained as a function of water depth or distance from the wetland edge. Compositing water-quality samples produced unbiased estimates of individual sample means for all water quality variables examined except for ammonium.  相似文献

A one-dimensional flow and transport model was developed to describe the movement of two fluid phases, gas and water, within a porous medium and the transport of 226Ra and 222Rn within and between these two phases. Included in this model is the vegetative uptake of water and aqueous 226Ra and 222Rn that can be extracted from the soil via the transpiration stream. The mathematical model is formulated through a set of phase balance equations and a set of species balance equations. Mass exchange, sink terms and the dependence of physical properties upon phase composition couple the two sets of equations. Numerical solution of each set, with iteration between the sets, is carried out leading to a set-iterative compositional model. The Petrov-Galerkin finite element approach is used to allow for upstream weighting if required for a given simulation. Mass lumping improves solution convergence and stability behavior. The resulting numerical model was applied to four problems and was found to produce accurate, mass conservative solutions when compared to published experimental and numerical results and theoretical column experiments. Preliminary results suggest that the model can be used as an investigative tool to determine the feasibility of phytoremediating radium and radon-contaminated soil.  相似文献

Wildland firefighters work in unfavourable environments involving both heat and moisture. Moisture in clothing systems worn by wildland firefighters may increase or decrease heat transfer, depending on its source and location in the clothing system, location on the body, timing of application and degree of sorption. In this experiment, 4 outerwear/underwear combinations were exposed to 1 of 5 different conditions varying on amount and location of moisture. The fabric systems were then exposed to either a high-heat-flux flame exposure (83 kW/m(2)) or a low-heat-flux radiant exposure (10 kW/m(2)). Under high-heat-flux flame exposures, external moisture tended to decrease heat transfer through the fabric systems, while internal moisture tended to increase heat transfer. Under low-heat-flux radiant exposures, internal moisture decreased heat transfer through the fabric systems. The nature and extent of such differences was fabric dependent. Implications for test protocol development are discussed.  相似文献

ABSTRACT: This paper compares approaches to quantifying land cover/land use (LCLU) in riparian corridors of 23 watersheds in Oregon's Willamette Valley using color infrared aerial photography (AP) and Thematic Mapper (TM) imagery. For each imagery source, LCLU adjacent to stream networks were quantified across a range of lateral and longitudinal scales. Single‐date AP data had higher LCLU class accuracies than the multi‐date TM data, except for row crops. Correlations among LCLU classes for the two imagery sources increased with increased spatial extent. In general, LCLU proportions for AP and TM differed, but lateral/longitudinal patterns were similar. An aggregated vegetation class comprised of forest, shrub/scrub, and grass/forb was strongly associated with a fish index of biotic integrity (IBI) for both AP and TM data, although AP correlations were higher. Highest fish IBI correlations for both data sources were with the aggregated vegetation class close to the stream and for the longest longitudinal scales. The row crop class was strongly associated with stream nitrate for both data sources, although correlations for the TM data were higher. Stream nitrate correlations were strongest for the widest lateral and longest longitudinal scales. Overall, both single‐date AP and multi‐date TM imagery appear to have potential for use in estimating indicators of stream ecological condition.  相似文献

ABSTRACT: Various temporal sampling strategies are used to monitor water quality in small streams. To determine how various strategies influence the estimated water quality, frequently collected water quality data from eight small streams (14 to 110 km2) in Wisconsin were systematically subsampled to simulate typically used strategies. These subsets of data were then used to estimate mean, median, and maximum concentrations, and with continuous daily flows used to estimate annual loads (using the regression method) and volumetrically weighted mean concentrations. For each strategy, accuracy and precision in each summary statistic were evaluated by comparison with concentrations and loads of total phosphorus and suspended sediment estimated from all available data. The most effective sampling strategy depends on the statistic of interest and study duration. For mean and median concentrations, the most frequent fixed period sampling economically feasible is best. For maximum concentrations, any strategy with samples at or prior to peak flow is best. The best sampling strategy to estimate loads depends on the study duration. For one‐year studies, fixed period monthly sampling supplemented with storm chasing was best, even though loads were overestimated by 25 to 50 percent. For two to three‐year load studies and estimating volumetrically weighted mean concentrations, fixed period semimonthly sampling was best.  相似文献
