Aromatic sulfonates (R-SO(3)(-)) can be used as sulfur sources by sulfate-starved bacteria in laboratory cultures and the corresponding phenols are excreted from the cells. The present study was conducted to demonstrate whether such desulfonation reactions also occur in sulfate-leached agricultural soil, where desulfonation of organic sulfur compounds may have agronomic importance as a S source for plants. Xenobiotic linear alkylbenzene sulfonates (LAS) were added to nominal concentrations of 0, 10 and 100 mgkg(-1) dry weight in a sandy soil that was depleted in sulfate by leaching the soil with water (sulfate depletion, approximately 75%). The soil was incubated at 20 degrees C in duplicate 3-dm(3) mesocosms for 8 weeks. Primary degradation of LAS was rapid with half-lives of 1-4 days. Sulfophenylcarboxylates were identified and quantified as intermediates, whereas linear alkylphenols (the expected primary desulfonation products) were not detected by high-pressure liquid chromatography coupled with both fluorescence and electrospray ionization-mass spectrometry. Thus, LAS was used by the bacteria as a source of energy and carbon, rather than as a source of sulfur. Measurements of soil pH, fluorescein diacetate (FDA) hydrolysis and arylsulfatase activity showed that stable microbial conditions prevailed in the soil mesocosms. FDA hydrolysis (a measure of total microbial activity) was transiently inhibited at the highest LAS concentrations. Arylsulfatase activity (i.e., hydrolysis of aromatic sulfate esters) was not significantly affected by the soil incubation, although arylsulfatases may be upregulated in sulfate-starved bacteria. However, an increased production of arylsulfatase may be difficult to detect due to the background of extracellular arylsulfatases stabilised in the soil. Therefore, the present data does not exclude a regulatory response to sulfate depletion by the soil microorganisms. However, the importance of desulfonation reactions in natural environments still needs to be demonstrated.  相似文献

This work describes the development of an instrument for measuring free and total copper in seawater by continuous flow analysis (CFA) with an Orion copper (II) ion selective electrode (CuISE). Sample analysis times are reduced considerably by using an extrapolation technique based on the fitting of an empirical mathematical expression to the electrode time-response curve enabling a prediction of the final equilibrium potential. CuISE measurements in seawater samples containing nanomolar levels of total copper can be very time consuming, and this predictive approach significantly reduces sample analysis time, and improves sample throughput. The time taken to measure pCu in seawater to a precision of +/- 0.1, using conventional potentiometry, varies considerably depending on the condition of the electrode membrane but can be reduced by a factor of 3-6 (typically from 60 to 10 min) by using the extrapolation technique in conjunction with CFA. Details are given of the protocols used for preconditioning the CuISE. The system can be used as a portable instrument for field measurements or for shipboard measurements of free copper in seawater. Extrapolated equilibrium potentials are within +/- 0.5 mV of true steady state values.  相似文献

The temporal occurrence of parasites in Sagitta setosa J. Müller, caught one to three times a month during 1982 and 1983 off Plymouth, England, was investigated. S. setosa was infected by eight parasitic species: one nematode, five trematodes, one cestode and, possibly, one protozoon. Infection by a single parasite, normally found in the body coelom, was the rule. The nematode Hysterothylacium aduncum, as III-stage larva, was by far the most common parasite (56% of all parasites found), followed by the metacercariae of three non-encysted trematodes: Derogenes varicus, a didymozoid species and a Lecithochirium species. Two trematode species new to Chaetognatha were found. The percentage of infected S. setosa specimens ranged from 0 to 7%. Absence or low numbers of parasites from June to November were correlated to the disappearance of large S. setosa and the appearance of a new S. setosa generation. Trematodes may show great annual and seasonal differences in occurrence in S. setosa. No injuries due to parasites were seen. The reproduction of the S. setosa populations off Plymouth did not seem to be affected by parasite infections during 1982–1983.  相似文献

In a study of consumptiveness among Swedish and Norwegian anglers in Engerdal, eastern Norway, three segments of low-, mid-, and high-consumptive anglers were identified by replicating a methodology of an earlier study of North American anglers. The Engerdal anglers were somewhat more catch-oriented than the North American anglers. Anglers in Norway fish to satisfy a complex set of experience preferences. Low-consumptive anglers rate nature experiences higher than mid- and high-consumptive anglers. High-consumptive anglers have a somewhat narrower range of experience preferences and are more oriented towards competitive aspects of fishing. All anglers generally support management efforts directed toward restricting other types of use of the waters, such as reducing pollution and reducing local fishermen's leisure gill-net fishing. They also support stocking of fish, but oppose actions restricting angling. However, low-consumptive anglers do not oppose angling restrictions to the same degree as mid- and high-consumptive groups. Low-consumptive fishermen show a higher degree of specialization and report higher overall satisfaction with their fishing trips than the other two groups.  相似文献

The necessity to provide information about radionuclide concentrations in Arctic marine species has been heightened in recent years due to a number of accidents in Arctic regions involving nuclear vessels and the presence of a large number of potential radioactive contamination sources. The provision for such information is largely dependent on the use of radionuclide uptake and transfer models. The uptake of radionuclides in Arctic seal species in this study has been modelled using a probabilistic biokinetic approach. In this paper, model results are compared with empirical data from relevant samples taken within the Arctic region. Results indicate that the model performs well when estimating concentrations of (137)Cs in two seal species for both median values and reproduction of the distribution of data values, but not as well for a third seal species. Likely factors affecting the results are the probability density functions used for the input parameters.  相似文献

On-line determination of respiration rates at steady-state dissolved oxygen tensions was performed in a thermostatted measuring chamber equipped with a galvanic oxygen electrode. The measuring chamber is designed to completely exclude gas bubbles from the liquid phase and is named a mono-phase oxystat. Additions of oxygenated water to maintain steady-state dissolved oxygen tensions are controlled by a computer, and the respiration rate is determined from the rate of these additions. An external loop with a flow cell in a spectrophotometer makes it possible to determine and control the concentration of algal cells in the mono-phase oxystat and allowed simultaneous determination of respiration and filtration rates of filter-feeding invertebrates. Because the system maintains steady-state conditions the experiments do not suffer from time limitations. Within the scope of a bilateral cooperation between Svensk K?rnbr?nslehantering (SKB) and Forschungszentrum Karlsruhe, Institut für Nukleare Entsorgung (FZK-INE), an actinide migration experiment is currently being performed at the Asp? Hard Rock Laboratory (HRL) in Sweden. This paper covers laboratory and in situ investigations on actinide migration in single-fractured granite core samples. For the in situ experiment, the CHEMLAB 2 probe developed by SKB was used. The experimental setup as well as the breakthrough of inert tracers and of the actinides Am, Np and Pu are presented. The breakthrough curves of inert tracers were analyzed to determine hydraulic properties of the fractured samples. Postmortem analyses of the solid samples were performed to characterize the flow path and the sorbed actinides. After cutting the cores, the abraded material was analyzed with respect to sorbed actinides. The slices were scanned optically to visualize the flow path. Effective volumes and inner surface areas were measured. In the experiments, only breakthrough of Np(V) was observed. In each experiment, the recovery of Np(V) was < or = 40%. Breakthrough of Am(III) and Pu(IV) as well as of Np(IV) was not observed.  相似文献

Using a human security perspective, we investigate how the history of land tenure changes has driven sustainability in southern African savannas. The paper examines four countries—Botswana, Mozambique, South Africa and Zimbabwe. Two research questions are addressed: first, how has colonial land tenure and distribution affected people's options and capacity to end, mitigate or adapt to risks to their human, environmental and social rights, and what have been the related effects on land uses and degradation? Second, to what extent have post-independence land reforms addressed threats to human security and political causes of land degradation? The inequitable distribution of land and colonial legacy of dual or pluralistic systems of tenure are found to be at the root of many agrarian and environmental problems. Post-independence land reforms have largely failed to address these fundamental issues, sometimes even reinforcing threats to social, economic and environmental sustainability. The skewed distribution of land and resources, insecure rights, and the marginalisation and restriction of savanna livelihood systems have persisted, undermining human security and environmental integrity in the region as well as leading to mounting conflict and insecurity.  相似文献
