The relationship between immune defense mechanisms and environmental pollutants has been a focus of intensive research during the last decade. In animal experiments, diesel exhaust particles (DEP) have been shown to exert adjuvant effects on the IgE response against aeroantigens and to compromise broncho-pulmonary immune defense. Important target cells are monocytes/alveolar macrophages which display the important functions of phagocytosis, antigenpresentation and modulation of inflammatory processes. To further evaluate the influence of DEP upon these cells, we investigated whether exposure to suspended DEP would change the phagocytic capacity and the tumor necrosis factor alpha (TNF-α) release of human peripheral monocytes/alveolar macrophages. Our results demonstrated a dose-dependent reduction of phagocytosis (p < 0.001) and an additional increase of an ongoing TNF-α response (p < 0.005). These findings may help to link exposure to environmental pollutants to such biologic effects as increased susceptibility to broncho-pulmonary disease or facilitated sensitization against aeroallergens.  相似文献

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) exerts its acute toxicity by inducing a gradually increasing voluntary feed refusal. However, this seems not to be caused by a direct effect on the central nervous system, as far higher concentrations of TCDD were found in the brain after intracerebroventricular (i.c.v.) than after lethal intravenous (i.v.) injections, but were not accompanied by a wasting syndrome. TCDD causes inhibition of several key enzymes of gluconeogenesis, with phosphoenolpyruvate carboxykinase (PEPCK) responding earliest and strongest to the insult. Responses of pyruvate carboxylase (PC) and glucose-6-phosphatase (G-6-Pase) are less pronounced and begin at later time points. Blood and brain levels of tryptophan increase following TCDD treatment with a lag period of about three days, shortly after the decrease of PEPCK activity becomes apparent. Since thi samino acid is the precursor of the appetite suppressant neurotransmitter serotonin, and since it is normally degraded via gluconeogenesis, a series of events can be suggested to explain the TCDD-induced wasting syndrome. By an as yet unrevealed mechanism TCDD decreases the activity of PEPCK to about 40 percent of normal, leading to a back-up of gluconeogenic substrates, among them tryptophan, which in turn can further inhibit PEPCK activity in vivo. This causes an increase in serotonin turnover in brain and possibly in other tissues. Increased serotonergic activity in turn is likely to play an important role in the increasing feed refusal of TCDD-treated rats which eventually leads to death.  相似文献

The optimised biodegradability test system "O2/CO2 Headspace Test with GC-TCD" is used for the assessment of synthetic ester lubricants. The effects of both additives and usage on biodegradability are examined and discussed. Ester based cutting fluids and hydraulic fluids with and without additives are used under defined conditions at machine tools and hydraulic and plain bearing test benches. The lubricants are characterised additionally with respect to kinematic viscosity, acidity and elemental composition. Furthermore, a formulated mineral oil is characterised before and after usage at an hydraulic test bench. The results clearly show that the mineral oil is far less biodegradable than the ester oils and that their biodegradability is not affected by usage. Biodegradability of the ester oils is mainly depending on the characteristics of the base fluids and not affected by the additives. Antioxidants are influencing stability respectively biodegradability indirectly, since they prevent oxopolymerisation effects. Other effects of usage on biodegradation are not detected. In this context, the antioxidants ensure ready biodegradability and have a positive effect on the environmental fate of synthetic ester lubricants.  相似文献

In the context of a monitoring program, persistent organic pollutants (POPs) were quantified in the blood of 10 year old children at four different demographic regions in Baden-Wuerttemberg, a highly industrialised federal state in South West Germany. DDE, HCB, PCB 138, PCB 153 and PCB 180 were measured in 1996/1997, 1998/1999, 2000/2001 and 2002/2003 in individual samples of about 400 children per year. PCDD/PCDFs and some relevant coplanar PCBs were determined in pooled samples from children in seven cycles from 1993 to 2003. Blood concentrations of the investigated compounds decreased in that time period by a factor of 2-4 with the exception of most PCDFs. The concentrations of POPs in the blood of the children were distinctly lower than the concentrations reported for adults. Breast feeding was associated with about 30% higher median concentrations of DDE, HCB, PCBs and a 30% increase for mean PCDD/PCDF concentrations. Concerning demographic differences, significant lower concentrations of HCB, PCBs and PCDD/PCDFs could be seen in children from Mannheim compared to the region of Aulendorf. About 10-20% higher concentrations were found in boys compared to girls for HCB, indicator PCBs and PCDD/PCDFs. The pattern of non-ortho and mono-ortho PCBs in the blood of children was similar to the pattern reported for mother's milk, and PCB 126 and PCB 156 contributed about 70% to the toxicity of dioxin-like PCBs and about one-third to total TEQ including PCDD/PCDFs.  相似文献

We conducted kinetic and equilibrium sorption experiments on removal of Zn(II) from aqueous solutions by scoria (a vesicular pyroclastic rock with basaltic composition) from Jeju Island, Korea, in order to examine its potential use as an efficient sorbent. The batch-type kinetic sorption tests under variable conditions indicated that the percentage of Zn(II) removal by scoria increases with decreasing initial Zn(II) concentration, particle size, and sorbate/sorbent ratio. However, the sorption capacity decreases with the decrease of the initial Zn(II) concentration and sorbate/sorbent ratio. Equilibrium sorption tests show that Jeju scoria has a larger capacity and affinity for Zn(II) sorption than commercial powdered activated carbon (PAC); at initial Zn(II) concentrations of more than 10mM, the sorption capacity of Jeju scoria is about 1.5 times higher than that of PAC. The acquired sorption data are better fitted to the Langmuir isotherm than the Freundlich isotherm. Careful examination of ionic concentrations in sorption batches suggests that the sorption behavior is mainly controlled by cation exchange and typically displays characteristics of 'cation sorption'. The Zn(II) removal capacity decreases when solution pH decreases because of the competition with hydrogen ions for sorption sites, while the Zn(II) removal capacity increases under higher pH conditions, likely due to hydroxide precipitation. At an initial Zn(II) concentration of 5.0mM, the removal increases from 70% to 96% with the increase of initial pH from 3.0 to 7.0. We recommend Jeju scoria as an economic and efficient sorbent for Zn(II) in contaminated water.  相似文献

Bioremedial treatment to remove low level organic contamination to regulatory standards has met with limited success. In this study source water from a contaminated surficial aquifer at a former wood treatment facility was used to evaluate the potential for indigenous microorganisms to degrade low level (< 1.0 mg) pentachlorophenol (PCP) to a regulatory drinking water standard of 0.001 mg/L. PCP degradation was evaluated in series of batch reactors in a two phase study to (a) determine the rate and extent of PCP removal and (b) evaluate the impact of nutrient amendment (N and P) on removal rate. All reactors with the exception of the abiotic control demonstrated PCP removal to a level < 0.002 mg/L within a maximum period of 32 d with and without nutrient amendment. A regression analysis of reactive phosphate (ortho-P) concentration versus removal rate produced an R2 of 0.94 (p = 0.006) indicating a significant correlation between the level of available phosphate and PCP degradation rate. Selective bacterial enumeration (for PCP degrading bacteria) revealed PCP-degrading bacteria increased in abundance prior to and in conjunction with the degradation phase to a density of between 10(3) to 10(4) CFU/ml. Isolates were also analyzed for total fatty acids using Fatty Acid Methyl Ester (FAME) methodology and the results indicated that PCP degrading bacteria were present in the aquifer and consisted of predominately fluorescent, oxidase positive Pseudomonas species. Overall, data indicate that autochthonous microbes are capable of removing low level PCP (< 1.0 mg/L) to approach if not reach the regulatory standard of 0.001 mg/L with the addition of oxygen, with or without nutrient amendment. Results of this research can be applied to full-scale implementation of in-situ or ex-situ bioremediation of groundwater at former wood treatment facilities.  相似文献

Goal, Scope, and Background

Soil organisms play a crucial role in terrestrial ecosystems. Plant Protection Products (PPPs) are known to affect these organisms and might have negative impacts on soil functions influenced by these organisms. Little research has been done to day on the impact of PPPs on tropical ecosystems. Therefore, in this study it was investigated whether the effects of pesticides differ between tropical and temperate regions and whether data generated under temperate conditions can be used for the Environmental Risk Assessment (ERA) in tropical regions.

Methods

The effects of one fungicide (Carbendazim) and one insecticide (lambda-Cyhalothrin) on soil invertebrates (earthworms and arthropods) and functional parameters (organic matter (OM) breakdown, feeding rate) were evaluated in semi-field tests using Terrestrial Model Ecosystems (TMEs) and in field tests using litter-bags. The soil for the TMEs was extracted from the same site near Manaus (Amazonas, Brazil) where the field test was conducted (see Part 1, Römbke et al. 2005 〈DOI: http://dx.doi.org/10.1065/uswf2004.12.088.1〉). The soil was acid clay, typical for tropical regions. In the TMEs the standard test species as well as native tropical species were introduced. Several application scenarios, selected according to their relevance for practical conditions, were realized in TME-tests and field (duration: 150 and 365 days, respectively). Finally, an Environmental Risk Assessment (ERA) for the two chemicals was performed.

Results and Conclusions

The results from the TME-tests show that both test chemicals (depending on their concentration) had toxic effects on the tested organisms and organic matter breakdown, but not on the feeding rates. In the field organic matter breakdown was affected for some time too, but effects on single species were, with one exception (the native earthworm Andiorrhinus amazonicus) not observed; a result probably influenced by the small size of the test plots. Observed differences in OM breakdown between control and treated TMEs were often significant, while due to the higher variability even with differences >10% between means this was not the case in the field.

Recommendation and Perspective

According to the preliminary results of the Environmental Risk, Assessment presented here, the use of the three model chemicals in the tropics (e.g. in Brazil) can cause a damage to the environmental compartment soil. Therefore, a more detailed investigation of these PPPs is recommended, in particular a better evaluation of the exposure (residue analysis). In general, the work performed in Amazonia can be summarised as follows: (1) Depending on the properties of the PPP, the effects of pesticides in tropical resp, temperate regions can differ. (2) If a PPP is going to be used in the tropics, data from tests modified for these conditions should be performed and their results should be incorporated in an Environmental Risk Assessment (ERA) for tropical regions.  相似文献

ABSTRACT: A single-family residence in Tucson, Arizona, was retrofitted with water-conserving fixtures, rainwater harvesting, and graywater reuse systems. During a four-year study, efficient use of water was shown to significantly decrease demand for domestic water at the house without reducing the residents' quality of life. The use of municipal water was reduced by 66 percent to 148 gallons per day (gpd) and total household use was reduced by 27 percent to 245 gpd. Graywater reuse averaged approximately 77 gpd or 32 percent of the total household water use. Evaporative cooling required about 15 gpd. Water use for toilet flushing was only 9 gallons per capita per day (gpcd) or 14 percent of interior water use.  相似文献

The elimination half-lives (t1/2) in Sprague-Dawley rats for 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), 1,2, 3,7,8-pentachlorodibenzo-p-dioxin (PeCDD), 1,2,3,4,7,8-hexachlorodibenzo-p-dioxin (HxCDD), 1,2,3,4,6,7,8-heptachlorodibenzo-p-dioxin (HpCDD) and 1,2,3,4,6,7,8,9-octachlorodibenzo-p-dioxin (OCDD) were estimated in long-term studies by Schlatter, Poiger and others. Furthermore, there are some published half-lives of TCDD in adult humans. The average half-life of TCDD in adult humans is approximately 2840 days, while in Sprague-Dawley rats the average t1/2 of TCDD is 19 days. The t1/2 of TCDD in humans is about 150 times that of rats. This factor was used to calculate the t1/2 values of the other polychlorinated dibenzo-p-dioxins (PCDDs) in humans from the rat data. Furthermore, the terminal t1/2 values of PCDDs in adult humans were calculated from the regression equation: logt1/2H = 1.34 logt1/2R + 1.25 which was recently established for 50 xenobiotics (t1/2H = terminal half-lives in days for humans, t1/2R = terminal half-lives in days for rats). The following terminal half-lives in adult humans were obtained: 12.6 years for 1,2,3,7,8-PeCDD, 26-45 years for 1,2,3,4,7,8-HxCDD, 80-102 years for 1,2,3,4,6,7,8-HpCDD and ca. 112-132 years for OCDD. These half-lives of PCDDs are critically compared with measured t1/2 values of PCDDs and other persistent organic pollutants in rats, monkeys and humans.  相似文献

GIS overlay techniques were used to provide a quantitative historic documentation of deforestation and land-use dynamics in the Middle Mountains of Nepal between 1947 and 1990. Deforestation was most critical in the 1960s, but active afforestation programs in the 1980s have reversed the process. In spite of these trends, the degradation problem is more complex. The GIS evaluation showed that 86% of the recently afforested land is now under pine plantations located primarily at lower elevations and moderately steep slopes. In contrast, rainfed agricultural expansion is most pronounced on acidic soils and steeper, upper elevation sites, suggesting marginalization of agriculture. Agricultural expansion coupled with major losses of grazing land to pine forests are the key processes pointing towards major animal feed deficits. An alternative animal feed source is suggested through GIS using a topographically based microclimatic classification to generate a tree-planting map where the optimum ecological conditions for selective native fodder tree species are identified.  相似文献
