Synthetic multiple-target RNA and DNA oligonucleotides were constructed for use as quantification standards for nucleic acid amplification assays for human norovirus genogroup I and II, hepatitis E virus, murine norovirus, human adenovirus, porcine adenovirus and bovine polyomavirus. This approach overcomes the problems related to the difficulty of obtaining practical quantities of viral RNA and DNA from these viruses. The quantification capacity of assays using the standards was excellent in each case (R 2 > 0.998 and PCR efficiency > 0.89). The copy numbers of the standards were equivalent to the genome equivalents of representative viruses (murine norovirus and human adenovirus), ensuring an accurate determination of virus presence. The availability of these standards should facilitate the implementation of nucleic acid amplification-based methods for quantitative virus detection.  相似文献

The aim of this study was to determine the impact of specific environmental contaminants on the activities of antioxidant enzymes in terrestrial, free-living birds. To investigate this, several biomarkers and the levels of lead, mercury, and arsenic were determined in blood of white stork (Ciconia ciconia) nestlings from two different colonies located in the west of Spain that were exposed to different environmental conditions and human activities. Different biomarker and contaminant patterns were observed between the two colonies, suggesting different levels of environmental contamination. In comparison to nestlings from a natural reference area, the levels of the three hazardous elements Pb, As, and Hg were significantly higher in the colony nearby a landfill. Analysis of antioxidant biomarkers revealed significantly higher levels of glutathione and glutathione S-transferase activity in birds from the colony close to the landfill site than were found in those from the “control” area. In contrast, the levels of malondialdehyde did not significantly differ between the two sampling sites. In addition, a positive correlation between glutathione and both Hg and As has been observed. Taken together, exposure to the three hazardous elements is reflected by enhanced activity of glutathione S-transferases, and elevated levels of glutathione and malondialdehyde. White storks can therefore be considered as a suitable model species for environmental monitoring programs.  相似文献

Abandoned metal mines in the Sierra de Guadarrama, Madrid, Spain, are often located in areas of high ecological value. This is true of an abandoned barium mine situated in the heart of a bird sanctuary. Today the area sustains grasslands, interspersed with oakwood formations of Quercus ilex and heywood scrub (Retama sphaerocarpa L.), used by cattle, sheep and wild animals. Our study was designed to establish a relationship between the plant biodiversity of these grasslands and the bioavailability of heavy metals in the topsoil layer of this abandoned mine. We conducted soil chemical analyses and performed a greenhouse evaluation of the effects of different soil heavy metal concentrations on biodiversity. The greenhouse bioassays were run for 6 months using soil samples obtained from the mine polluted with heavy metals (Cu, Zn, Pb and Cd) and from a control pasture. Soil heavy metal and Na concentrations, along with the pH, had intense negative effects on plant biodiversity, as determined through changes in the Shannon index and species richness. Numbers of grasses, legumes, and composites were reduced, whilst other species (including ruderals) were affected to a lesser extent. Zinc had the greatest effect on biodiversity, followed by Cd and Cu. When we compared the sensitivity of the biodiversity indicators to the different metal content variables, pseudototal metal concentrations determined by X-ray fluorescence (XRF) were the most sensitive, followed by available and soluble metal contents. Worse correlations between biodiversity variables and metal variables were shown by pseudototal contents obtained by plasma emission spectroscopy (ICP-OES). Our results highlight the importance of using as many different indicators as possible to reliably assess the response shown by plants to heavy metal soil pollution.  相似文献

In this work, a series of reactive copolymers of glycidyl methacrylate (G), styrene (S) and acrylonitrile (AN), were synthesized through reversible addition-fragmentation chain transfer polymerization and evaluated as macromolecular chain extenders in reactive extrusion of recycled poly(ethylene terephthalate) (rPET). The results obtained indicate that the addition of the reactive copolymers as chain extenders modifies the chain conformation in rPET causing low crystallization rate and low crystallinity. The physical and rheological properties (melt flow and intrinsic viscosity) of chain-extended rPET improved, rendering better processability. rPET modified with polymeric chain extenders shows improved rheological properties (complex viscosity, storage and loss modulus) and also displays higher elongation at break and impact properties as the GMA content in the chain extenders increase.  相似文献

Changes in the chemical and chemical-structural composition of the organic matter of two different sewage sludges (aerobic and anaerobic) mixed with sawdust (1:1 and 1:3, v/v) during composting were determined by monitoring chemical and microbiological parameters as well as by pyrolysis-gas chromatography. Composting was carried out in periodically turned outdoor piles, which were sampled for analysis 1, 30, 60 and 90 days after the beginning of the composting process. Both volatile organic matter and the water soluble C fraction decreased during composting, indicating that the more labile C fractions are mineralized during the process. Microbial activity as measured by microbial respiration (CO(2) evolved from compost samples during incubation) also decreased with composting, reflecting the more stable character of the resulting compost. No major differences were observed between the four composts studied as regards their chemical-structural characteristics. The acetonitrile, acetic acid and phenol pyrolytic fragment tended to increase with composting. Although the final composts were more aromatic in nature than the starting materials, a low degree of humification was observed in all four composts studied, as determined by their high proportion of polysaccharides and alkyl compounds. For this reason, the relationship between pyrolytic fragments, such as benzene/toluene or benzene+toluene/pyrrol+phenols, which are used as indices of humification for soil organic matter, are not of use for such poorly evolved sludge composts; instead, ratios that involve carbohydrate derivatives and aromatic compounds, such as furfural+acetic/benzene+toluene or acetic/toluene, are more sensitive indices for reflecting the transformations of these materials during composting. Both the chemical and microbiological parameters and pyrolytic analysis provided valuable information concerning the nature of the compost's organic matter and its changes during the composting process.  相似文献

The present study evaluates the capacity of shrimp (Farfantepenaeus aztecus) head to remove toxic Ni(II) ions from aqueous solutions. Relevant parameters that could affect the biosorption process, such as shrimp head pretreatment, solution pH level, contact time and initial Ni(II) concentration, were studied in batch systems. An increase in Ni(II) biosorption capacity and a reduction in the time required to reach Ni(II) biosorption equilibrium was manifested by shrimp head biomass pretreated by boiling in 0.5 N NaOH for 15 min; this biomass was thereafter denominated APSH. The optimum biosorption level of Ni(II) ions onto APSH was observed at pH 7.0. Biosorption increased significantly with rising initial Ni(II) concentration. In terms of biosorption dynamics, the pseudo-second-order kinetic model described Ni(II) biosorption onto APSH best. The equilibrium data adequately fitted the Langmuir isotherm model within the studied Ni(II) ion concentration range. According to this isotherm model, the maximum Ni(II) biosorption capacity of APSH was 104.22 mg/g. Results indicate that APSH could be used as a low-cost, environmentally friendly, and promising biosorbent with high biosorption capacity to remove Ni(II) from aqueous solutions.  相似文献

The scaling of metabolic rates with body mass is one of the best known and most studied characteristics of aquatic animals. Herein, we studied how size is related to oxygen consumption, ammonia excretion, and ingestion rates in tropical (Octopus maya) and cold-water (Enteroctopus megalocyathus) cephalopod species in an attempt to understand how size affects their metabolism. We also looked at how cephalopod metabolisms are modulated by temperature by constructing the relationship between metabolism and temperature for some benthic octopod species. Finally, we estimated the energy balance for O. maya and E. megalocyathus in order to validate the use of this information for aquaculture or fisheries management. In both species, oxygen consumption and ammonia excretion increased allometrically with increasing body weight (BW) expressed as Y = aBW b . For oxygen consumption, b was 0.71 and 0.69 for E. megalocyathus and O. maya, respectively, and for ammonia excretion it was 0.37 and 0.43. Both species had low O/N ratios, indicating an apparent dependence on protein energy. The mean ingestion rates for E. megalocyathus (3.1 ± 0.2% its BW day−1) and O. maya (2.9 ± 0.5% its BW day−1) indicate that voracity, which is characteristic of cephalopods, could be independent of species. The scope for growth (P = I − (H + U + R) estimated for E. megalocyathus was 28% higher than that observed in O. maya (320 vs. 249 kJ day−1 kg−1). Thus, cold-water cephalopod species could be more efficient than tropical species. The protein and respiratory metabolisms of O. maya, E. megalocyathus, and other octopod species are directly dependent on temperature. Our results offer complementary evidence that, as Clarke (2004) stated, the metabolic response (R and U) cannot be determined mechanistically by temperature, as previously proposed (Gillooly et al. 2002).  相似文献

A series of porous γ-Al2O3 materials was prepared by solution-combustion and ball-milling processes. The as-prepared powders were physicochemically characterized by x-ray diffraction (XRD), scanning electron microscopy (SEM), and N2 physisorption measurements and their performances in CO2 adsorption at different pressures (0.5 to 1.5 MPa) and temperatures (40 to 60ºC) were investigated. It was found that γ-Al2O3 synthesized by the solution-combustion process and ball milled at 10 hr exhibited the best CO2 adsorption performance at 60ºC and 1.5 MPa, achieving a maximum of 1.94 mmol/g compared to the four studied materials, as a result of their interesting microstructure and surface properties (i.e., nanocrystallinity, specific surface area, narrow pore size distribution, and large total pore volume). Our study shows that the γ-Al2O3 prepared by solution combustion followed by ball milling presents a fairly good potential adsorbent for efficient CO2 capture.

Implications: In this work, γ-Al2O3 materials were successfully obtained by solution combustion and modified via ball milling. These improved materials were systematically investigated as solid adsorbents of accessible surface areas, large pore volumes, and narrow pore size distribution for the CO2 capture. These studied solid adsorbents can provide an additional contribution and effort to develop an efficient CO2 capture method as means of alleviating the serious global warning problem.  相似文献
