Understanding the formation mechanisms of secondary air pollution is very important for the formulation of air pollution control countermeasures in China. Thus, a large-scale outdoor atmospheric simulation smog chamber was constructed at Chinese Research Academy of Environmental Sciences (the CRAES Chamber), which was designed for simulating the atmospheric photochemical processes under the conditions close to the real atmospheric environment. The chamber consisted of a 56-m3 fluorinated ethylene propylene (FEP) Teflon film reactor, an electrically-driven stainless steel alloy shield, an auxiliary system, and multiple detection instrumentations. By performing a series of characterization experiments, we obtained basic parameters of the CRAES chamber, such as the mixing ability, the background reactivity, and the wall loss rates of gaseous compounds (propene, NO, NO2, ozone) and aerosols (ammonium sulfate). Oxidation experiments were also performed to study the formation of ozone and secondary organic aerosol (SOA), including α-pinene ozonolysis, propene and 1,3,5-trimethylbenzene photooxidation. Temperature and seed effects on the vapor wall loss and SOA yields were obtained in this work: higher temperature and the presence of seed could reduce the vapor wall loss; SOA yield was found to depend inversely on temperature, and the presence of seed could increase SOA yield. The seed was suggested to be used in the chamber to reduce the interaction between the gas phase and chamber walls. Colony is a key to Microcystis becoming a dominant population and forming blooms. To find the mechanism of colony formation, we investigated cell wall structures of colonial and unicellular strains. Results showed that colonial strains had significant surface layer protein (S-layer) on the surface of cells than unicellular strains by transmission electron microscopy. Western blot showed colonial strains had more S-layer than the unicellular strains. When the S-layer gene (GenBank accession number CAO89090.1) of Microcystis aeruginosa PCC7806 was expressed in Synechocystis sp. PCC6803, PCC6803 aggregated into colonial morphology. The results indicated that the S-layer could promote colony formation in Microcystis. Based on the S-layer sequences of PCC6803 and PCC7806, nine S-layer genes in other Microcystis strains were screened from the GenBank. Sequence comparing showed that the S-layers conserved regions were all located in N-terminal. The S-layers contain repeats-in-toxin (RTX) sequences with Ca2+-binding site, and their amino acid composition, hydrophobicity, isoelectric point, etc. were consistent with the characteristics of RTX-type S-layer in bacteria.
The aim of this study was to investigate in vitro effects of fenitrothion (FNT) on mouse splenic lymphocytes. Here, naïve mice had their spleens harvested and splenocytes isolated. After exposure to FNT for 48 hr: splenocyte viability was measured using a tetrazolium dye assay; cell phenotypes, i.e., B-cells (CD19+), T-cells (CD3+), and T-cell subsets (CD4+ and CD8+), were quantified by flow cytometry; and, production of cytokines/granzyme-B was assessed via enzyme-linked immunosorbent assay. The ability for FNT to induce oxidative stress in the cells was evaluated by measuring hydroxyl radical (·OH) and malondialdehyde (MDA) production and changes in glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) activity. The results showed that FNT significantly inhibited splenocyte proliferation, and decreased production of interleukin (IL)-2, interferon gamma, IL-4, and granzyme B, but had no impact on IL-6 production. FNT also selectively decreased splenic T-cell levels but did not induce changes in CD19+ B-cells. Further, within the T-cell populations, percentages of CD3+, CD4+, and CD8+ T-cells (particularly CD8+ T-cells) were reduced. Lastly, FNT selectively increased MDA and ·OH production and inhibited SOD and GSH-Px activities in the splenic lymphocytes. These findings suggest that, due to oxidative damage, FNT selectively inhibits splenic T-lymphocyte survival and cytokine/granzyme production in vitro. 相似文献
Surface sediments from estuarine and coast of CanGio wetland (CGW) of Hochiminh City, Vietnam, were investigated to identify the spatial and temporal variations of polycyclic aromatic hydrocarbons (PAHs). The total PAHs showed wide variation but similar to patterns observed (F?=?0.901, p?=?0.46) in LongTau (31?±?77 ng/g dry weight, n?=?13), SoaiRap (53?±?81, n?=?4), ThiVai (60?±?62, n?=?10) estuaries, and coastal areas (112?±?211, n?=?9). A decreasing trend in the wet season (F?=?8.8, p?=?0.01) reflected that inland sources such as wastewater discharged and atmospherically transported contaminants contributed to PAHs in sediments. The risk posed by the PAHs in the coastal and estuarine areas of CGW is still negligible. The present study provides baseline data, which can be used for regular monitoring and future strategy of environmental protection for the study area.
