Phytoplankton is an important water quality indicator because of its high species differentiation, growth rates and responsiveness to environmental actuators. The new European Water Framework Directive calls for assessment of the duration, intensity and succession of phytoplankton blooms to determine the ecological status of various types of waters. For common phytoplankton growth rates basic signal processing theory yields a minimum monitoring frequency of once per day, which is much more than applied in standard practice. To assess the nature of this discrepancy we followed the behaviour of about 40 groups of organisms/particles found in the Oude Rijn river by a two-week daily cytometric analysis. Particle counts of the 20 most abundant groups are shown. Their variation rate and magnitude confirm that daily sampling is needed to follow such ecosystems in detail. It is shown that limiting the monitoring to the "coarse line" does not allow a correspondingly decreased sampling frequency. Automated systems may fill the gaps between the microscopical examinations by gathering highly frequent information. The information depth of bulk measurements is poor however, and not used as such. The data shown here demonstrate that modern scanning flow cytometry (SFC) offers an information depth close to the taxonomic level. In the past decade, acquisition and operation costs of these systems have come down considerably, whereas operation is hands free, even in situ and submerged, and data analysis has become more efficient. SFC is used most efficiently complementary to microscopical analyses for mutual validation. In these cases it presents a realistic solution to generate the essential high frequency observations required to assess ecosystem variability.  相似文献

The ability to detect low levels of 2,4,6-trinitrotoluene (TNT) in aqueous samples is important due to the toxicity of both TNT and its breakdown products. We have been characterizing recombinant anti-TNT antibodies isolated from the Griffin library of phage displayed scFvs by selection for binders to the TNT-surrogate 2,4,6-trinitrobenzene (TNB) coupled to the protein bovine serum albumin. Two candidate antibody fragments, TNB1 and TNB2, were isolated and evaluated by ELISA for their ability to bind to TNB coupled to the protein ovalbumin. Competition ELISA was then used to demonstrate antibody fragment binding to TNT in solution and to examine cross-reactivity towards several TNT-related compounds and other explosives. Both recombinant antibody fragments were incorporated into a continuous flow assay for the detection of TNT. TNB2, the best single chain antibody, showed a limit of detection of 1 ng ml(-1), comparable to a commercially available anti-TNT antibody in the same assay format.  相似文献

Our objectives were to determine if concentrations of environmental pollutants and microbial contamination in nonviable eggs of the endangered Mississippi sandhill crane (Grus canadensis pulla) contributed to egg failure. Six eggs collected in 1990 and four in 1991 contained only background levels of organochlorine pesticides and polychlorinated biphenyls (PCBs), and tests for microbial contamination were all negative. Two eggs contained late dead embryos, but neither revealed obvious abnormalities. Three eggs contained potentially harmful concentrations (23, 39, 146 pg/g, wet mass) of polychlorinated dibenzo-p-dioxins (PCDDs) and dibenzofurans (PCDFs), based on 2,3,7,8-tetrachlorodibenzo-p-dioxin equivalents (TCDD-EQ) for combined compounds. Because of the scarcity of material suitable for laboratory examination and the endangered status of the crane, we recommend that nonviable eggs continue to be monitored for toxic pollutants.  相似文献

Epidemiological studies have demonstrated the exacerbation of respiratory diseases following sandstorm-derived particulate matter(PM) exposure.The presence of anthropogenic and biological agents on the sandstorm PM and the escalation of PM 2.5 μm(PM2.5)pollution in China have led to serious concerns regarding the health effects of PM2.5during Asian sandstorms.We investigated how changes in PM2.5composition,as the weather transitioned towards a sandstorm,affected human airway epithelial cells.Six PM2.5samples covering two sandstorm events and their respective background and transition periods were collected in Baotou,an industrial city near the Gobi Desert in China.PM samples from all three periods had mild cytotoxicity in human bronchial epithelial cell line BEAS-2B,which was positively correlated with the contents of polycyclic aromatic hydrocarbons and several metals.All PM samples potently increased the release of interleukin-6(IL-6) and interleukin-8(IL-8).Endotoxin in all samples contributed significantly to the IL-6 response,with only a minor effect on IL-8.Cr was positively correlated with both IL-6 and IL-8 release,while Si was only associated with the increase of IL-6.Our study suggests that local agricultural and industrial surroundings in addition to the sandstorm play important roles in the respiratory effects of sandstorm-derived PM.  相似文献

Estimation of Nitrous Oxide Emissions from US Grasslands   总被引:2,自引:0,他引:2  
2 O) emissions from temperate grasslands are poorly quantified and may be an important part of the atmospheric N2O budget. In this study N2O emissions were simulated for 1052 grassland sites in the United States using the NGAS model of Parton and others (1996) coupled with an organic matter decomposition model. N2O flux was calculated for each site using soil and land use data obtained from the National Resource Inventory (NRI) database and weather data obtained from NASA. The estimates were regionalized based upon temperature and moisture isotherms. Annual N2O emissions for each region were based on the grassland area of each region and the mean estimated annual N2O flux from NRI grassland sites in the region. The regional fluxes ranged from 0.18 to 1.02 kg N2O N/ha/yr with the mean flux for all regions being 0.28 kg N2O N/ha/yr. Even though fluxes from the western regions were relatively low, these regions made the largest contribution to total emissions due to their large grassland area. Total US grassland N2O emissions were estimated to be about 67 Gg N2O N/yr. Emissions from the Great Plains states, which contain the largest expanse of natural grassland in the United States, were estimated to average 0.24 kg N2O N/ha/yr. Using the annual flux estimate for the temperate Great Plains, we estimate that temperate grasslands worldwide may potentially produce 0.27 Tg N2O N/yr. Even though our estimate for global temperate grassland N2O emissions is less than published estimates for other major temperate and tropical biomes, our results indicate that temperate grasslands are a significant part of both United States and global atmospheric N2O budgets. This study demonstrates the utility of models for regional N2O flux estimation although additional data from carefully designed field studies is needed to further validate model results.  相似文献

ABSTRACT: As population growth occurs in regions of the eastern United States that do not have abundant water supplies, new transbasin diversions may be required. Such diversions are exceptionally politically divisive and are at the core of most of the interstate water conflicts that presently exist in the eastern states. This study examines alternative means by which these conflicts might be resolved. The strengths and weaknesses of these alternatives are examined. The study concludes that the federal-interstate compact should be the preferred alternative by which interstate water conflicts in the eastern United States are resolved.  相似文献

ABSTRACT: A UNIX-based windows application was developed to integrate a phosphorus transport model with the Geographic Resources Analysis Support System (GRASS). The system prioritizes potential phosphorus loading from fields or cells in a watershed and can evaluate the effects of alternate management practices on phosphorus yield. The model simulates phosphorus loading by using a daily mass balance on a unit-area basis and incorporates the effects from rainfall, topography, soil properties, fertilizer and animal waste application, and management. Model predictions include dissolved and sediment-bound phosphorus yield, runoff volume, and sediment yield. Within the integrated GRASS-modeling system, the user can obtain model input data, execute the model using various options, rank model input and out. put data, and display them as GRASS-based maps or data tables. All functions are menu driven, developed using C language and X-window tools to run on a SUN workstation platform. The system provides a powerful and efficient tool for prioritizing phosphorus loading from nonpoint sources.  相似文献

Grassland to cropland conversion in the northern prairie of the United States has been a topic of recent land use change studies. Within this region more corn and soybeans are grown now (2017) than in the past, but most studies to date have not examined multi-decadal trends and the synergistic web of socio-ecological driving forces involved, opting instead for short-term analyses and easily targeted agents of change. This paper examines the coalescing of biophysical and socioeconomic driving forces that have brought change to the agricultural landscape of this region between 1980 and 2013. While land conversion has occurred, most of the region’s cropland in 2013 had been previously cropped by the early 1980s. Furthermore, the agricultural conditions in which crops were grown during those three decades have changed considerably because of non-biophysical alterations to production practices and changing agricultural markets. Findings revealed that human drivers played more of a role in crop change than biophysical changes, that blending quantitative and qualitative methods to tell a more complete story of crop change in this region was difficult because of the synergistic characteristics of the drivers involved, and that more research is needed to understand how farmers make crop choice decisions.  相似文献

This Impacts article proposes strategies for mitigating negative impacts of urbanization in rural locations in the United States. Issues addressed include impacts of population growth and development, loss of agricultural lands, and impacts of climate change on agriculture and rural communities. Conclusions are supported by stakeholder survey data, geographic information systems-based data, and desktop reviews of research journal publications. We propose a sustainable, diversified approach that supports mitigation of issues, including increasing demand on food production and decline of rural communities. A key issue that we address is where we will find suitable landscapes to reduce enough food for 9.6 billion people living in 2050.

Urban and rural development planners are grappling with solutions to escalating impacts global populations, stresses on food production, and effects of climate change. Solutions are identified, including strengthening rural and urban contexts by establishing connected and interdependent links that support diversification of rural and urban contexts as viable solutions to these issues.

Diversified rural-to-urban sustainable agriculture production is a promising approach to addressing climate change impacts. Organic agriculture principles exhibit strong diversity and are accredited by United States Department of Agriculture as the only federally certified sustainable agriculture practice in the United States. Sustainable agriculture practices are evolving into profitable diversified alternative food sources. We offer substantiated alternative solutions for remediating impacts of urbanization on rural agriculture and communities. Collectively, these solutions can strengthen symbiotic relationships between sustainable agriculture and rural communities, addressing our growing population issues and preserving our dwindling farmlands and rural communities.  相似文献

Bacterioplankton community composition was compared across 10 lakes and 14 streams within the catchment of Toolik Lake, a tundra lake in Arctic Alaska, during seven surveys conducted over three years using denaturing gradient gel electrophoresis (DGGE) of PCR-amplified rDNA. Bacterioplankton communities in streams draining tundra were very different than those in streams draining lakes. Communities in streams draining lakes were similar to communities in lakes. In a connected series of lakes and streams, the stream communities changed with distance from the upstream lake and with changes in water chemistry, suggesting inoculation and dilution with bacteria from soil waters or hyporheic zones. In the same system, lakes shared similar bacterioplankton communities (78% similar) that shifted gradually down the catchment. In contrast, unconnected lakes contained somewhat different communities (67% similar). We found evidence that dispersal influences bacterioplankton communities via advection and dilution (mass effects) in streams, and via inoculation and subsequent growth in lakes. The spatial pattern of bacterioplankton community composition was strongly influenced by interactions among soil water, stream, and lake environments. Our results reveal large differences in lake-specific and stream-specific bacterial community composition over restricted spatial scales (<10 km) and suggest that geographic distance and connectivity influence the distribution of bacterioplankton communities across a landscape.  相似文献
