为揭示SigL及其增强子结合蛋白(EBPs)在苏云金芽胞杆菌(Bt)中的调控功能,在全基因组测序的基础上,采用生物信息学方法,对YBT-1520菌株的SigL及其EBPs的结构和功能进行深入分析.结果表明,YBT-1520基因组中存在1个SigL和6个EBPs,而且EBPs在结构上具有丰富的多样性,包含了EBPs的所有可能的结构域组织类型.SigL所调控的基因涉及11个假定的COG代谢途径,其中包括能量代谢、氨基酸代谢、翻译与细胞周期等.根据EBPs在基因组的位置推测,YBT-1520的EBPs参与γ-氨基丁酸代谢途径、精氨酸代谢途径、支链脂肪酸降解途径、多糖分解代谢等代谢途径的调控.本研究将为揭示Bt杀虫晶体蛋白大量表达的调控机制提供新的思路. 相似文献

有机磷农药的构效关系及其对浮游生物的毒性效应 总被引:2,自引:0,他引:2
以定量构效关系理论和实验室内急性毒性试验相结合研究了六种有机磷农药对四尾栅藻(S.quadricanda)和多刺裸腹溞(Moina macrocopa)的生态毒性。有机磷农药的毒性取决于磷原子的电正性,各取代基种类和构象对电荷分布作用显著。根据构效关系原理,磷氧双键(P=O)被磷硫双键(P=S)取代后毒性降低;羟基(-OH)被甲、乙、丙氧基(-CH3O、-CH3CH2O、-CH3CH2CH2O)取代后有机磷农药毒性依次增强;磷氧单键(P-O)毒性高于磷碳单键(P-C)。该实验中毒死蜱、辛硫磷(-CH3CH2O)毒性高于其它四种(-CH3O),敌敌畏和敌百虫(P=O)毒性高于乐果(P=S),敌敌畏(P-O)又高于敌百虫(P-C),甘氨酸异丙胺盐取代羟基(-OH)的草甘膦毒性最低。毒性试验结果表明,毒死蜱、辛硫磷、敌百虫、敌敌畏、乐果、草甘膦异丙胺盐对四尾栅藻的96 h EC50分别为6.34、6.62、59.53、82.12、141.37和7.25 mg.L-1。它们对多刺裸腹溞的48 h LC50分别为0.20、0.12、0.28、0.17、1.12和5.03 mg.L-1。六种有机磷农药对多刺裸腹溞的毒性强度结果与QSAR分析具有较高的一致性,即表征有机磷农药毒性特征的取代基团作用的毒性强弱顺序为:-OH〉=O〉-O;对四尾栅藻毒性试验结果中,由于草甘膦异丙胺盐作用于植物类靶位点的特殊性,导致其对四尾栅藻毒性显著增强。综合以上研究,以构效关系理论和浮游生物毒性试验相结合进行有机磷农药的生态风险评价将更加便捷和准确。 相似文献

考察了pH值对"Fe0-厌氧微生物"体系降解2,4,6,-三氯酚(2,4,6-TCP)效果的影响,结果表明:pH值是影响"Fe0-厌氧微生物"体系降解2,4,6-TCP效果的重要参数,初始pH值直接影响微生物活性和铁腐蚀,进而影响过程pH值变化,反过来又影响铁腐蚀和微生物活性,pH 7.0~9.0的中性偏碱范围较适于厌氧微生物生长。Fe0与微生物对目标污染物的降解具有协同促进作用,其协同促进机制表现在3方面:Fe0与微生物对体系过程pH值具有互补调节作用,可将体系的pH值调节值适于微生物生长的中性范围;Fe0腐蚀产生的Fe2+和H2可为微生物代谢提供电子对和营养物质,从而促进生物还原脱氯的进行;Fe0的腐蚀过程直接对氯代有机物还原脱氯,而微生物又可促进Fe0腐蚀。 相似文献

Secondary organic aerosols(SOAs) are an important component of particulates, but whether biogenic SOAs(BSOAs) or anthropogenic SOAs(ASOAs) are the dominant contributors to haze pollution remains poorly characterized. In this study, particulate samples were collected from September 2014 to August 2015 at an urban site in Jinan, which is the capital of Shandong Province and a typical city in the North China Plain. The PM2.5 samples were analyzed for BSOA(isoprene(SOAI) and monoterpenes(SOAM)) and ASOA(aromatic(SOAA)) tracers. The concentrations of the SOAAtracer(1.1 ± 1.0 ng/m3) were lowest, and those of SOAItracers(41.8 ± 86.2 ng/m3) were highest, with the concentrations of SOAMtracers(19.4 ± 9.9 ng/m3) being intermediate. The SOAItracers were more abundant in the summer and less abundant in the winter. Both SOAIand SOAMincreased with increasing ozone level but decreased with increasing NOx level. Correlation analysis revealed a good correlation between 2,3-dihydroxy-4-oxopentanoic acid and levoglucosan levels in three seasons. These results suggested that biomass burning activities occurring in the NCP can enhance the emissions of aromatics and should be controlled, especially in the autumn and winter. SOA tracers were classified according to pollution degree, and the results showed that as pollution increases, the contributions of SOAAincrease. These results indicate that reducing anthropogenic emissions is necessary to prevent SOA pollution, especially during heavy pollution episodes. 相似文献

采用水培试验,研究了镉胁迫下喷施细胞分裂素类物质6-苄氨基嘌呤(6-BA)和激动素(KT)对玉米幼苗生长、叶片叶绿素含量、脯氨酸含量、植株镉含量和活性氧清除酶活性的影响.结果表明,与单纯镉胁迫相比,喷施6-BA和KT提高了玉米幼苗的生物量、根和地上部镉含量、叶绿素和脯氨酸含量,降低了叶片丙二醛(MDA)含量,促进了叶片过氧化物酶(POD)和过氧化氢酶(CAT)活性的升高,但对超氧化物歧化酶(SOD)活性的影响不明显.说明细胞分裂素类物质6-BA和KT可缓解镉胁迫对玉米幼苗的伤害,其机制可能与细胞分裂素物质能维持叶绿素稳定、刺激细胞内H2O2清除酶类活性及促进脯氨酸积累有关.当6-BA浓度在0~20mg·L-1之间时,其对玉米幼苗镉胁迫的缓解效应随喷施浓度的提高而增强,而KT的最适喷施浓度为70mg·L-1. 相似文献

Distribution of polybrominated diphenyl ethers in the surface sediments of the Taihu Lake, China 总被引:5,自引:0,他引:5
Twenty-eight sediment samples were collected from the Taihu Lake and analyzed to acquire information about the levels, distribution, possible sources, time trend and inventory of polybrominated diphenyl ethers (PBDEs) in the Taihu Lake. Our results showed that the most abundant BDE congeners in surface sediments were BDE-47, 99 and 209, with a median value of 0.124, 0.279, and 22.72 ng(gdw)(-1), respectively. The levels of BDE-209 in our samples were much higher relative to those of the other BDE congeners and made up more than 80% of the PBDEs levels in almost all samples. Disregarding BDE-209, among the seven PBDEs (BDE-28, 47, 99, 100, 153, 154, 183) more concerned, the most abundant ones were BDE-47 and 99, which contributed 44.65% (ranged from 29.02% to 73.11%), 24.24% (ranged from 2.27% to 39.66%) to ∑(7)PBDEs, respectively. The correlation between PBDE and TOC was poor, indicating that PBDEs concentrations in sediments of the Taihu Lake were controlled not only by TOC contents, but also by a combined effect of transport, mixing, depositional mechanisms associated with PBDEs, uncontaminated sediments, or fresh input of PBDEs. As expected, the time trend of PBDEs concentration in surface sediment from Taihu Lake were increasing, and the inventory of ∑(25)PBDEs and BDE-209 were estimated to be 3668 kg and 26296 kg. 相似文献

2018年11月5—7日,韩国首尔出现了一次PM_(2.5)污染过程。利用拉格朗日轨迹分析(HYSPLIT)模型分析了首尔峰值浓度气团的来源,结合污染物监测和气象资料,定性分析了中国对韩国浓度高值可能的影响及其程度。利用嵌套网格空气质量预报模式(NAQPMS)及其耦合的在线污染来源追踪模块进行了污染来源解析和敏感性测试,分别计算了同一时期中韩两国相互间的PM_(2.5)传输贡献。结果显示:2018年11月5—7日,中国对韩国首尔污染过程的日均贡献不足10%;此次污染过程后期,首尔的污染气团对上海PM_(2.5)浓度峰值产生了影响。 相似文献

In this study, 74 soil samples collected from the Pearl River Delta were analyzed for polycyclic aromatic hydrocarbons (PAHs). The PAH mixture in the soils is mainly of low molecular weight compounds, with naphthalene (21.4%) and phenanthrene (21.8%) being dominant. Soil PAH levels from the Pearl River Delta are relatively low (28?C711 ng/g, averaged 192 ng/g) compared to those from urban soils in temperate regions. The mean concentration of ??PAHs generally decrease with increasing distance from the city center, with ??PAHs of paddy soils > crop soil > natural soil. PAHs in the air were measured during a year-round sampling campaign using semipermeable membrane devices, and the transfer of chemicals between the soil and air compartments were estimated. Soil?Cair fugacity quotient calculations showed a highly uncertain equilibrium position of PAHs, with net volatilization of naphthalene and fluorene, whereas net deposition of phenanthrene, fluoranthene, and pyrene, indicating a capacity for the air to supply the soil with more substances. 相似文献
