In this study we evaluated the hyperglycemic and hyperlipidemic effects of chlorpyrifos (CPF) after an acute exposure in rats. The mechanisms involved in hyperglycemia induced by CPF were studied. A single dose of CPF (50 mg kg−1, subcutaneous, s.c.) was administered to overnight-fasted rats. Glucose and corticosterone levels, lipid status and paraoxonase (PON1) activity were determined in plasma of rats. Cardiovascular risk factors and the atherogenic index were calculated. Glycogen levels, tyrosine aminotransferase (TAT) and glucose-6-phosphatase (G6Pase) activities were determined in livers of rats. Cerebral acetylcholinesterase (AChE) activity was also determined. CPF caused an increase in glucose and glycogen levels as well as in TAT and G6Pase activities. The CPF exposure caused an increase in corticosterone levels, an inhibition of AChE activity and a reduction of PON1 activity. Regarding the lipid status, CPF induced an increase in triglycerides (TG) and low-density lipoprotein-cholesterol (LDL) levels and a decrease in high-density lipoprotein (HDL) levels associated with an increase of cardiovascular risk factors and the atherogenic index. The present study demonstrated that a single CPF administration caused hyperglycemia and hyperlipidemia in rats. The activation of the gluconeogenesis pathway, probably elicited by hypercorticosteronemia, is involved in the hyperglycemic effect of CPF in rats. 相似文献
In this study, we investigated the effect of diphenyl diselenide [(PhSe)2] on chlorpyrifos (CPF)-induced hepatic and hematologic toxicity in rats.
Methods
Rats were pre-treated with (PhSe)2 (5?mg/kg) via the oral route (oral gavage) once a day for 7?days. On the eighth and ninth days, rats were treated with (PhSe)2 (5?mg/kg) 30?min prior to CPF (50?mg/kg, by subcutaneous route). The aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase activities were determined in plasma of rats. Lipid peroxidation, protein carbonyl, and non-protein thiol levels as well as catalase, superoxide dismutase, glutathione peroxidase, glutathione reductase, and gluthatione S-transferase activities were determined in livers of rats. Hematological parameters were also determined.
Results
The results showed that CPF caused hepatic oxidative damage, as demonstrated by an increase in lipid peroxidation and protein carbonyl levels which was associated with a decrease in antioxidant defenses. CPF exposure caused a reduction in the leukocyte, indicating hematologic toxicity. (PhSe)2 was effective in attenuating these toxic effects caused by CPF exposure in rats.
Conclusions
The results indicated that (PhSe)2 was effective in protecting the hepatic and hematologic toxicity induced by acute CPF exposure in rats. 相似文献
A rapid, sensitive, accurate and reliable multiresidue method for the identification and quantification of 210 relevant pesticides in four representative fruit and vegetable commodities (tomato, potato, spring onion and orange) has been developed and validated by gas chromatography in tandem with triple quadrupole mass spectrometry. The method has been fully validated and applied to 292 samples from different countries. Prior to instrumental analysis, an extraction procedure based on a sample extraction of multiclass analytes, using the ethyl acetate method was employed. Mass spectrometric conditions were individually optimized for each compound in the selected reaction monitoring (SRM) mode to achieve maximum sensitivity. The pesticides were separated in less than 25 min. This was followed by an exhaustive control of the retention times. The Retention Time Locking Method was applied, working at a constant pressure throughout the analysis. System maintenance was reduced by using a purged capillary flow device that provided backflush capabilities by reversing column flow immediately after elution of the last compound of interest. We used data on number of carcasses of wildlife species sold in 79 bushmeat markets in a region of Nigeria and Cameroon to assess whether species composition of a market could be explained by anthropogenic pressures and environmental variables around each market. More than 45 mammal species from 9 orders were traded across all markets; mostly ungulates and rodents. For each market, we determined median body mass, species diversity (game diversity), and taxa that were principal contributors to the total number of carcasses for sale (game dominance). Human population density in surrounding areas was significantly and negatively related to the percentage ungulates and primates sold in markets and significantly and positively related to the proportion of rodents. The proportion of carnivores sold was higher in markets with high human population densities. Proportion of small‐bodied mammals (<1 kg) sold in markets increased as human population density increased, but proportion of large‐bodied mammals (>10 kg) decreased as human population density increased. We calculated an index of game depletion (GDI) for each market from the sum of the total number of carcasses traded per annum and species, weighted by the intrinsic rate of natural increase (rmax) of each species, divided by individuals traded in a market. The GDI of a market increased as the proportion of fast‐reproducing species (highest rmax) increased and as the representation of species with lowest rmax (slow‐reproducing) decreased. The best explanatory factor for a market's GDI was anthropogenic pressure—road density, human settlements with >3000 inhabitants, and nonforest vegetation. High and low GDI were significantly differentiated by human density and human settlements with >3000 inhabitants. Our results provided empirical evidence that human activity is correlated with more depleted bushmeat faunas and can be used as a proxy to determine areas in need of conservation action. 相似文献

Downstream flow in rivers is repeatedly delayed by hydrologic exchange with off‐channel storage zones where biogeochemical processing occurs. We present a dimensionless metric that quantifies river connectivity as the balance between downstream flow and the exchange of water with the bed, banks, and floodplains. The degree of connectivity directly influences downstream water quality — too little connectivity limits the amount of river water exchanged and leads to biogeochemically inactive water storage, while too much connectivity limits the contact time with sediments for reactions to proceed. Using a metric of reaction significance based on river connectivity, we provide evidence that intermediate levels of connectivity, rather than the highest or lowest levels, are the most efficient in removing nitrogen from Northeastern United States’ rivers. Intermediate connectivity balances the frequency, residence time, and contact volume with reactive sediments, which can maximize the reactive processing of dissolved contaminants and the protection of downstream water quality. Our simulations suggest denitrification dominantly occurs in riverbed hyporheic zones of streams and small rivers, whereas vertical turbulent mixing in contact with sediments dominates in mid‐size to large rivers. The metrics of connectivity and reaction significance presented here can facilitate scientifically based prioritizations of river management strategies to protect the values and functions of river corridors. 相似文献
Total mercury (T-Hg) and methylmercury (MeHg) concentrations have been measured in the muscle tissue of 16 fish species consumed
in the Mojana region of Colombia. T-Hg analysis was performed by cold-vapor atomic-absorption spectroscopy (CV-ASS) and MeHg
analysis by gas chromatography with electron-capture detection. Higher T-Hg and MeHg concentrations were detected in carnivorous
species (T-Hg = 0.371 ± 0.172 (μg g−1 fresh wt, MeHg = 0.346 ± 0.171 μg g−1 fresh wt) than in non-carnivorous fish (T-Hg = 0.155 ± 0.108 μg g−1 fresh wt, MeHg = 0.146 ± 0.102 μg g−1 fresh wt). In the different species mercury was present almost completely as the methylated form, with percentages between
80.5 and 98.1% (mean 92.0 ± 3.4%). In 13.5% of fish-tissue samples T-Hg concentrations exceeded the maximum level recommended
by the World Health Organization for human consumption (Hg = 0.5 μg g−1 fresh wt). Although mean T-Hg concentrations in all fish samples (0.269 ± 0.181 μg g−1 fresh wt) did not exceed this limit, risk assessment suggested that the consumption of 0.12 kg fish day−1 could increase the risk of mercury poisoning of the inhabitants of this region. 相似文献
In this study, we evaluated, in a pioneering way, the influence of wavelengths from the decomposition of white light on the production and physicochemical properties of silver nanoparticles (AgNPs). Bearing in mind a process of green synthesis, an extract of the bracts of Bougainvillea glabra Choisy (BgC) was used, a species native to tropical and subtropical regions and frequently used in ornamentation, possessing in its photochemical composition, biomolecules capable of acting as reducing agents for convert Ag+ to Ag0. We used light-emitting diodes (LED) to obtain the desired wavelengths (violet, blue, green, yellow, orange, and red) in the test called rainbow, and we evaluated the obtaining of AgNPs compared to white LED light, nature, and absence of light. In the rainbow assay, we obtained a gradual increase in the intensity of the plasmonic band resonance from the red wavelength (0.124 ± 0.067 a.u.) to violet (0.680 ± 0.199 a.u.), indicating a higher reaction yield in obtaining AgNPs. Smaller hydrodynamic sizes (approximately 150 nm) at more energetic wavelengths (violet, blue, and green) about less energetic wavelengths (yellow, orange, and red) (approximately 400 nm) were obtained. Analysis by SEM microscopy, FTIR spectroscopy, and X-ray diffraction indicates the presence of silver nanoparticles in all LED colors used together with white LED light and Laboratory light (natural light). Due to the high environmental demand to remove pollutants from water sources, including textile dyes, we applied AgNPs/BgC to remove methylene blue (MB) dye from an aqueous solution. A minimum removal percentage greater than 65%, with emphasis on formulations synthesized by the colors of violet LED (84.27 ± 2.65%) and orange LED (85.91 ± 1.95%), was obtained.
