The aim of this study was to investigate the cytotoxic effects of different concentrations of Aluminium (Al) and Cobalt (Co) heavy metal ions on Phaseolus vulgaris L. cv Barbunia (Fabaceae) root tips. We used the germination percentage (GP), root length (RL), weight gain (WG) and micronucleus (MN) frequency as indicators of cytotoxicity and correlated these data with statistical parameters. Additionally to the cytogenetic analysis, lipid peroxidation and DNA analyses were performed in root tips of barbunia seeds treated with Al and Co metals. The seeds were divided into five groups as control, Al and Co treatment groups. They were treated with 25 and 50 ppm doses of Al and Co during 7 days. The results indicated that there was an alteration in the GP, RL, WG and MN frequency depending on the treatment dose in the seeds exposed to Al and Co metal ions when compared with the controls. Al and Co metal ions at both the doses significantly reduced the GP, RL and WG in seeds of all the treatment groups. The highest GP was observed in seeds of the control group (in proportion as 96%). 25 and 50 ppm doses of Co and Al caused 30, 50 and 42, 64% decrease of seed germination, respectively. In the control group, the final weights of all the seeds increased about 1.31 g when compared to initial weight. The mean RL of control seeds were measured as 3.71 cm at the end of experimental period. In Co andAl groups, the final weights of seeds increased about 0.34 g and 0.19 g according to initial weight at 50 ppm dose, respectively But, Al and Co ions caused a dose-dependent increase in the frequency of MN. The highest frequency of MN was observed at 50 ppm dose of Al and least frequency of MN was observed at 25 ppm dose of Co. Besides, 25 and 50 ppm concentrations of Al and Co significantly enhanced the lipid peroxidation and caused an increase in malondialdehyde (MDA) levels at both the doses. In roots treated with 25 and 50 ppm doses of Al, the increase of MDA was about 62 and 136% according to control, respectively In Co-treated roots, the increase of MDA was about 31 and 91% according to the control at 25 and 50 ppm doses, respectively The investigated parameters (except MN and MDA) were higher in the seeds exposed to Co than the seeds treated with Al. Moreover, it was observed that the yields of DNA in the seeds treated with Al andCo metals were lower than recorded in the controls. Hence, DNA yields exposed to Al and Co were run ahead on agarose gel according to the control group. The results of the present study indicate that Al and Co metal ions have toxic effects on barbunia root tip cells, and the selected parameters such as the GP, RL, WG, MN and MDA are very sensitive and useful biomarkers for biomonitoring these effects.  相似文献

One of the most significant diseases related to environmental asbestos exposure is malignant mesothelioma (MM). Sivas province is located in the Central Anatolia where asbestos exposure is common. We aimed to study clinical, demographical and epidemiologic features of the patients with MM in Sivas, along with the history of asbestos exposure. In total, 219 patients with MM who were diagnosed in our hospital between 1993 and 2010 were retrospectively analyzed in terms of demographical and clinical features. Rock, soil and house plaster samples were taken from the habitats of those patients and were evaluated with optical microscopy and X-ray diffraction methods. The age of the patients ranged between 18 and 85 years. The male-to-female ratio was 1.4:1. Most of the patients confirmed an asbestos exposure history. The most frequent symptoms of the patients were chest pain (60 %) and dyspnea (50 %). The gap between the start of first symptoms and the diagnosis date was approximately 4 months in average. The plaster materials used in most of the houses were made up of mainly carbonate and silicate minerals and some chrysotile. Ophiolitic units contained fibrous minerals such as serpentine (clino + orthochrysotile) chiefly and pectolite, brucite, hydrotalcite and tremolite/actinolite in smaller amounts. MM is not primarily related to occupational asbestos exposure in our region, and hence, environmental asbestos exposure may be indicted. Yet, single or combined roles and/or interactions of other fibrous and non-fibrous minerals in the etiology of MM are not yet fully understood and remain to be investigated.  相似文献

In this study, the levels of some heavy metals (Al, Cd, Co, Cr, Cu, Fe, Li, Mn, Ni, Pb and Zn) in muscle, gill, and liver of Carassius carassius and in the water samples from Eber Lake (Afyonkarahisar, Turkey) have been investigated. Additionally, one of the lipid peroxidation markers, malondialdehide (MDA) and glutathion (GSH) levels were investigated. All the metal analysis was performed by using ICP-AES. According to results obtained, it was observed that heavy metals were accummulated in liver in the highest degree and lowest one in the muscle tissues. MDA and GSH levels varied in the seasons but their winter levels were found to be statistically meaningful when compared with other seasons. The obtained data of metals from the study were compared with the acceptable levels of Turkish governmental regulation and they were found not to be harmful for human health. On the other hand, it was suggested that oxidative stress markers should be checked regularly in order to get important data for continuous life of aquatic organisms.  相似文献

Abstract

Organochlorine pesticides (OCPs) were analyzed in three different ages (half-, 1.5-, 2.5-year-old) for needles and semi permeable membrane devices (SPMDs) at three deployment periods from sea level to 1881 meter above sea level. Individual HCHs concentrations ranged between 1.4 and 129?pg/g fw depending on the age and sampling season while 2.5-year-old needles showed higher HCHs levels compared to half and 1.5- year-old. Correlation between elevation and HCH concentration in SPMDs was found but not in needle samples. Concentrations of HCHs in SPMDs indicated clearly cold condensation effect on accumulation in winter period and increased with altitude. Concentrations of DDTs in half and 1.5-year-old needles were lower than 2.5-year-old needles. The highest total concentration of DDTs was detected in 1-year-period SPMD. Higher concentrations were found in 2.5-year-old needles for other OCPs. Seasonal and altitude-dependent changes were not observed for other OCPs in SMPDs. Total accumulation of OCPs in SPMDs were found higher than in needles. On the contrary, an increased accumulation rate was observed for HCHs in SPMD. In general, Total concentrations of DDTs and HCHs were similar to total of other OCPs in all altitudes when dominating endosulfan wasnot taken into account in the computation of total concentration of other OCPs.  相似文献

The spring waters of Tuzla–Icmeler are on the Marmara Sea coast in Tuzla town of Istanbul city. The springs discharge a natural sodium chloride mineral water that consumed for ages for therapeutic purposes attributed to their chemical properties. Development of springs commenced during the Ottoman times and a surface collection structure was built at the discharge point of the main spring. Two deep wells were drilled to tap mineral water within the past decades. The bottled water of these springs is also sold for a couple of years and its consumption as a beverage is increasing. The geochemical properties of these springs were investigated by several researchers in the past. This study comprises geochemical and geophysical measurements performed between July 2001 and July 2002 in order to construct a conceptual hydrogeological model for environmental and land use planning purposes. The seasonal evaluation of Tuzla–Icmeler (mineral spring) shows that the chemical properties fluctuate from the beginning of summer until the beginning of winter. This indicates that the overdraft of water during the summer season causes the movement and mix of normal groundwater with the mineralized groundwater. As a result, mixing of less mineralized groundwater decreases the salinity of mineralized groundwater. Using the site-specific hydrogeological, geochemical and geophysical data, zones of protection areas were delineated in order to prevent a possible pollution access to the springs and surroundings from nearby dockyards, dwellings and vehicle traffic. For this purpose, a new land use plan was proposed using the existing settlement sustainability plans.  相似文献

In this study, it was aimed to determine organochlorine pesticides (OCPs) in three types of milk (cow's, buffalo's, and sheep's milk) produced in Afyonkarahisar province of Turkey. The results indicated that these milk specimens were found to be contaminated by 21 different pesticides. Sixteen OCP residues were detected in sheep's milk and it was followed with 14 pesticides in buffalo's milk and 11 pesticides in cow's milk. Dominant pesticides in all samples examined were beta-HCH in buffalo's, cow's, and sheep's milk in the concentrations of 63.36, 91.32, and 122.98 ng/ml, respectively. Total OCP levels were found to be 243.81 ng/ml in sheep's milk, 151.02 ng/ml in cow's milk, and 133.38 ng/ml in buffalo's milk. Some of the pesticides detected were found to be in the excess amount of the acceptable level regarding the EU regulations.  相似文献

In this study, the potential re-use of waste foundry sand in high-strength concrete production was investigated. The natural fine sand is replaced with waste foundry sand (0%, 5%, 10%, and 15%). The findings from a series of test program has shown reduction in compressive and tensile strengths, and the elasticity modulus which is directly related to waste foundry inclusion in concrete. Nevertheless the concrete with 10% waste foundry sand exhibits almost similar results to that of the control one. The slump and the workability of the fresh concrete decreases with the increase of the waste foundry sand ratio. Although the freezing and thawing significantly reduces the mechanical and physical properties of the concrete. The obtained results satisfies the acceptable limits set by the American Concrete Institute (ACI).  相似文献

The Melet is one of Turkey's economically important rivers. Most of the petroleum plants are located at fairly nearby of the river This situation is considered as main source of heavy metal pollution in the river. The present study was designed to evaluate the protective role of Ginkgo biloba (GB) on cytotoxicity induced by petroleum wastewater in Vicia faba root tip cells. For this aim, we used the germination percentage, root length, weight gain and micronucleus (MN) frequency as indicators of cytotoxicity. Additionally to the cytological analysis, lipid peroxidation analyses were also performed in V. faba roots. Heavy metal concentrations in wastewater were measured by atomic absorption spectrophotometer (AAS). The V. faba seeds were divided into six groups. They were treated with petroleum wastewater and 10, 20 and 30 microM doses of GB. As a result, the mean concentrations of heavy metals in wastewater were observed in the order: Pb > A > Ni > Cr > Fe > Cu > Zn > Cd. The highest germination percentage was observed in the seeds of the control and positive control groups (in proportion as 98 and 96%, respectively). Wastewater treatment caused a significant decrease in the germination percentage of Group III (in proportion as 44%). The highest root length and weight gain were observed in the seeds of the control and positive control groups at the end of the experimental period. The least root length and weight gain were observed in the seeds of Group III treated with wastewater alone. In the control group, the final weights of all the seeds increased about 4.08 g according to initial weight. The root lengths of the control seeds were measured as 6.80 cm at the end of the experimental period. The final weights of the seeds exposed to wastewater alone increased about 0.90 g according to initial weight. Besides, there was a significantly increase in the MDA levels of the roots exposed to wastewater. Heavy metals in wastewater significantly affected the MDA production indicating lipid peroxidation. But, GB-treatment caused amelioration in indices of the germination percentage, root length, weight gain, MN frequency and lipid peroxidation when compared with group III. Each dose of GB provided protection against wastewater toxicity and its strongest protective effect observed at dose of 30 microM. In vivo results showed that GB is a potential protector against toxicity induced by petroleum wastewater and its protective role is dose-dependent.  相似文献
