An improved technique for the determination of organic phosphorus in sediments and soils by 31P nuclear magnetic resonance spectroscopy 总被引:1,自引:0,他引:1
Knowledge of organic phosphorus (P) species quantity and distribution in sediments and soils is needed to determine the potential for aquatic and terrestrial organism growth. This can be achieved with 31P nuclear magnetic resonance spectroscopy (NMR), but resolving peaks within spectra can be problematic because of broadening via paramagnetics, which can cause peaks to overlap. We compared 31P NMR spectra of NaOH-EDTA extracts of three sediments and three soils to those that had first been pre-treated with Ca-EDTA-dithionite to remove Fe and Mn, paramagnetics that cause broadening of peaks, but leave organic P alone. Broadening of peaks in Ca-EDTA-dithionite pre-treated samples decreased by 46% and revealed peaks that were hidden compared to untreated samples. The spectrum of one pre-treated soil was similar if not better than the same soil that had received pre-treatment with Chelex resin (also to remove paramagnetics). Therefore, pre-treatment with Ca-EDTA-dithionite is recommended as a simple and cost-effective method for improving organic P idenTIFication and determination in subsequent NaOH-EDTA extracts of sediments and soils rich in Fe and Mn. 相似文献

The influence of gonadal maturation on seawater adaptability at the metabolic level was assessed in the euryhaline teleost Sparus auratus by treatment of immature fish with 17ß-estradiol (E2) followed by acclimation to different environmental salinities. Fish were injected with coconut oil alone (sham) or containing E2 (10 µg g–1 body weight) and maintained in seawater (40). After 5 days, fish from oil- and E2-implanted groups were sampled. At the same time, fish of both groups were transferred to brackish water (5, hypoosmotic test), seawater (40, transfer test), and hypersaline water (55, hyperosmotic test); 4 days after transfer (9 days post-implant) all groups were sampled. Data obtained from liver suggest that E2 treatment produced effects comparable to those already reported in literature, including decreased glycogen levels, increased glycolytic potential, and decreased gluconeogenic potential. This, together with the fact that many changes displayed in shams among salinities disappeared in E2-treated fish allow us to suggest that the response of liver carbohydrate metabolism to osmotic acclimation is exceeded by the response elicited by E2 treatment. In gills, E2 treatment produced increased lactate levels, decreased capacity for use of exogenous glucose, and decreased the potential of the pentose phosphate pathway. These findings suggest that the energy demand occurring in gills during osmotic acclimation should be increasingly fuelled by substrates other than exogenous glucose. Finally, data obtained in brain of E2-treated fish suggest a lower necessity of exogenous glucose, increased lactate levels, and decreased glycolytic potential.Communicated by S.A. Fifty-two samples of surface soils were taken in the urban area of Seville, to assess the possible influence of different
land uses on their metal contents and their relationship with several soil properties. The samples corresponded to five categories
or land uses: agricultural, parks, ornamental gardens, riverbanks, and roadsides. Sequential extraction of metal according
to the procedure proposed by the former Community Bureau of Reference (BCR) was carried out, and pseudo-total (aqua regia soluble) metal contents were determined. Lower organic C, total N and available P and K contents were found in riverbank
samples, probably due to the lack of manuring of those sites, left in a natural status. In contrast, significantly higher
electrical conductivity was found in those sites, due to the tidal influence of the nearby Atlantic Ocean. Other land uses
did not show significant differences in the general properties. Concentrations of Cu, Pb and Zn, both aqua-regia soluble and sequentially extracted, were clearly higher in soils from ornamental gardens, whereas the concentrations in the
riverbank samples were slightly lower than the other categories. In contrast, other metals (Cd, Cr, Fe, Mn, Ni) were uniformly
distributed throughout all land uses. A strong statistical association is found among the concentrations of Cu, Pb, Zn and
organic C, suggesting that the larger contents of these metals in ornamental gardens are partly due to organic amendments
added to those sites more frequently than to other kinds of sites. Considering the conclusions of previous studies, heavy
traffic can also contribute to those `urban’ metals in urban soils. Periodic monitoring of the concentrations of urban metals
in busy city centres and of the quality of amendments added to soils of recreational areas are recommended. 相似文献

The distribution and enrichment of selected trace metals (Cd, Cr, Cu, Ni, Pb, Sn, Zn) in benthic sediments of the Southport
Broadwater, a semi-enclosed coastal body of water adjacent to the Gold Coast city, south-eastern Queensland, Australia, was
studied with the objective of assessing the extent and degree of sediment contamination. Sediment samples from the 0–10 cm
and 10–20 cm depth intervals of 32 sites within the Southport Broadwater and surrounding residential canals were analysed
for particle size distribution, pH, organic C and ‘near-total’ major (Al, Ca, Fe, Mn) and trace (Cd, Cr, Cu, Ni, Pb, Sn, Zn)
metal contents. Sediment contamination for each trace metal was assessed by (1) comparison with Australian sediment quality
guidelines, (2) calculation of the index of geoaccumulation based on regional background values, and (3) geochemical normalisation
against Al (i.e. the abundance of alumino-silicate clay minerals). Based on this approach, the results indicate that submerged
sediments in the study area are not presently enriched with Cd, Cr or Ni, with the spatial distribution of these metals being
very well explained by the abundance of alumino-silicate clay minerals. However, several sites were strongly enriched with
Cu, Pb, Sn and Zn, arising from sources related to either urban runoff or vessel maintenance activities. The study indicates
that several varying approaches are needed for a satisfactory assessment of contaminant enrichment in estuarine sediments. 相似文献

Caste theory predicts that social insect colonies are organized into stable groups of workers specialized on particular task sets. Alternative concepts of organization of work suggest that colonies are composed of extremely flexible workers able to perform any task as demand necessitates. I explored the flexibility of workers in temporal castes of the honey bee Apis mellifera by determining the ability of colonies to reorganize labor after a major demographic disturbance. I evaluated the flexibility of temporal castes by comparing the foraging rates of colonies having just lost their foragers with colonies having also lost their foragers but having been given a week to reorganize. The population sizes and contents of the colonies in each group were equalized and foraging rates were recorded for one week. Colonies given a weeks initial recovery time after the loss of their foragers were found to forage at significantly higher rates than those colonies given no initial recovery time. This result was consistent for nectar and pollen foraging. These results suggest that honeybee workers lack sufficient flexibility to reorganize labor without compromising foraging. This finding is consistent with the caste concept model of organization of work in insect societies. 相似文献

The marine copepod Calanopia americana Dahl undergoes twilight diel vertical migration (DVM) in the Newport River estuary, North Carolina, USA, in synchrony with the light:dark cycle. Copepods ascend to the surface at sunset, descend to the bottom around midnight, and make a second ascent and descent before sunrise. Behavioral assays with C. americana in the laboratory during fall 2002/2003 and summer 2004 investigated aspects of three hypotheses for the proximate role of light in DVM: (1) preferendum hypothesis (absolute irradiance), (2) rate of change hypothesis (relative rates of irradiance change), and (3) endogenous rhythm hypothesis. Results suggest that C. americana responds to exogenous light cues consistent with its DVM pattern; changes in absolute irradiance evoked swimming responses that would result in an ascent at sunset and descent at sunrise, while relative rates of irradiance decrease at sunset (–0.0046 s–1) evoked an ascent response, and relative rates of irradiance increase at sunrise (0.0042 s–1) evoked a descent response. Furthermore, C. americana expressed an endogenous rhythm in vertical migration that was positively correlated with field observations of twilight DVM. Collectively, these results indicate that both exogenous light cues and endogenous rhythms play a proximate role in twilight DVM of C. americana, providing redundancy in the causes of its vertical migration.Communicated by J.P. The date when a landbird migrant arrives on its breeding grounds may have reproductive consequences. Generally, early arriving individuals begin breeding earlier and consequently experience greater seasonal reproductive performance. Here, we describe relationships between arrival timing and seasonal reproductive performance in the American redstart (Setophaga ruticilla), a long-distance passerine migrant, arriving at northerly breeding grounds in Michigans eastern Upper Peninsula. Evidence suggests that both males and females benefited from early arrival at the breeding grounds. Early males appeared to settle on higher quality territories and hatched nestlings sooner than later arrivals. Early females began their clutches early, produced heavier nestlings and possibly laid more eggs than later arrivals. Larger clutches and heavier offspring increase the likelihood of offspring recruiting into the breeding population. The findings of this study point to fitness consequences arising from when a bird arrives at its breeding grounds. These results also have implications for understanding how events occurring during spring migration influence reproductive performance as migratory delays likely influence arrival timing.Communicated by: W. The tarpon (Megalops atlanticus) is a highly valued game fish and occasional food fish in the eastern and western Atlantic Ocean. Tarpon have a high capacity for dispersal, but some regional biological differences have been reported. In this study we used two molecular genetic techniques—protein electrophoresis of nuclear DNA loci, and restriction fragment length polymorphism analysis of the mitochondrial DNA (mtDNA)—to assess this species population genetic structure in the eastern (coastal waters off Gabon and Sierra Leone, Africa) and western (coastal waters off Florida, Caribbean Sea) Atlantic Ocean north of the equator. Genetic differentiation was observed between tarpon from Africa and tarpon from the western Atlantic Ocean. A unique allele and haplotype, significant differences in allozyme allele and mtDNA haplotype frequencies between the African and western Atlantic samples, and significant FST analyses suggest that levels of gene flow between tarpon from these two regions is low. Among the western Atlantic Ocean collections, genetic diversity values and allele and haplotype frequencies were similar. AMOVA analyses also showed a degree of genetic relatedness among most of the western Atlantic Ocean collections: however, some significant population structuring was detected in the allozyme data. A regional jackknifed FST analysis indicated the distinction of the Costa Rica population from the other western Atlantic populations and, in pairwise analyses, FST values tended to be higher (i.e., genetic relatedness was lower) when the Costa Rican sample was paired with any of the other western Atlantic samples. These data suggest that Costa Rican tarpon could be partially isolated from other western Atlantic tarpon populations. This study presents the concentrations of about 50 metals and ions in 33 different brands of bottled waters on the Swedish market. Ten of the brands showed calcium (Ca) concentrations ≤10 mg L−1 and magnesium (Mg) levels <3 mg L−1, implying very soft waters. Three of these waters had in addition low concentrations of sodium (Na; <7 mg L−1), potassium (K; <3 mg L−1) and bicarbonate (HCO3 ≤31 mg L−1). These brands were collected from barren districts. Nine of the brands were collected from limestone regions. They showed increased Ca-levels exceeding 50 mg L−1 with a maximum of 289 mg L−1. Corresponding Mg-levels were also raised in two brands exceeding 90 mg L−1. Two soft and carbonated waters were supplemented with Na2CO3 and NaCl, resulting in high concentrations of Na (644 and 648 mg L−1) and chloride (Cl; 204 and 219 mg L−1). Such waters may make a substantial contribution to the daily intake of NaCl in high water consumers. The storage of carbonated drinking water in aluminum (Al) cans increased the Al-concentration to about 70 μg L−1. Conclusion As there was a large variation in the material as regards concentrations of macro-elements such as Ca, Mg, Na, K and Cl. Supplementation with salts, e.g., Na2CO3, K2 CO3 and NaCl, can lead to increased concentrations of Na, K and Cl, as well as decreased ratios of Ca/Na and larger ratios of Na/K. Water with high concentrations of e.g., Ca and Mg, may make a substantial contribution to the daily intake of these elements in high water consumers. Al cans are less suited for storage of carbonated waters, as the lowered pH-values may dissolve Al. The levels of potentially toxic metals in the studied brands were generally low. 相似文献

Misuse of alcohol is a significant public health problem, potentially resulting in unintentional injuries, motor vehicle crashes,
drownings, and, perhaps of greatest concern, serious acts of violence, including assaults, rapes, suicides, and homicides.
Although previous research establishes a link between alcohol consumption increased levels of violence, studies relating the
density of alcohol outlets (e.g., restaurants, bars, liquor stores) and the likelihood of violent crime have been less common.
In this paper we test for such a relationship at the small area level, using data from 79 neighborhoods in the city of Minneapolis,
Minnesota. We adopt a fully Bayesian point of view using Markov chain Monte Carlo (MCMC) computational methods as available
in the popular and freely available WinBUGS language. Our models control for important covariates (e.g., neighborhood racial
heterogeneity, age heterogeneity) and also account for spatial association in unexplained variability using conditionally
autoregressive (CAR) random effects. Our results indicate a significant positive relationship between alcohol outlet density
and violent crime, while also permitting easy mapping of neighborhood-level predicted and residual values, the former useful
for intervention in the most at-risk neighborhoods and the latter potentially useful in identifying covariates still missing
from the fixed effects portion of the model. 相似文献
