This work describes TCE biotic removal in a single-pulse bioreactor under aerobic conditions. Activated sludge from a wastewater-treatment plant was used for inoculation of the cultivator. The experiment focused on a more detailed verification of microbial composition of mixed heterotrophic culture during pulsed phenol dosage. Attention was given to suppressing eucaryotic organisms, particularly yeasts and fungi, by the addition of cycloheximide. The TCE-removal capacity of the heterotrophic culture, described by kinetic tests, was dependent on pulsed phenol injection and on cyclic addition of phenol and TCE. Maximum TCE degradation was determined in a batch test. It was found that the addition of cycloheximide (an antibiotic against propagation and growth of fungi and yeast) increased the TCE degradation activity of the mixed microbial suspension. A certain residual amount of TCE remained in some of the experiments.  相似文献

The common tube-building polychaete Lanice conchilega is known as a habitat structuring species and can form dense aggregations. The effects of L. conchilega on the surrounding benthic community have received little attention, especially in subtidal areas. Therefore, the presence of L. conchilega in different habitats in the North Sea and its effect on the abundance, species richness, diversity and community structure in these habitats are evaluated in the present paper, based on data from the ICES North Sea Benthos Survey of 2000. Lanice conchilega has a wide geographical distribution and a low habitat specialization, but optimally occurs in shallow fine sands. In the present study, the presence of L. conchilega resulted in a density increase and a significant (positive) correlation of the benthos density with the density of L. conchilega. Furthermore, the species richness (number of species) increased with increasing density of L. conchilega. This trend was, however, not consistent: the number of species reached more or less an asymptotic value or even decreased after reaching a critical density of L. conchilega (>500–1,000 ind/m2), as observed in shallow fine sands. The same overall pattern was detected concerning the expected number of species. The N 1-diversity index showed similar or slightly higher values in L. conchilega patches compared to patches without L. conchilega. From the results of the community analysis, it can be concluded that the species, which were responsible for the increase of the diversity, belonged to the overall species-pool of that habitat. The effects on density and diversity differed between the four discerned habitats (shallow muddy sand, shallow fine sand, shallow medium sand and deep fine sand), and were most pronounced in shallow fine sands. These patterns can be attributed to the habitat structuring capacity of L. conchilega. The mechanisms responsible for the increase of the habitat quality in patches of L. conchilega can be summarized as (1) changes in the hydrodynamics, (2) increases of the habitat stability and oxygen supply, and (3) a creation of habitat heterogeneity in a uniform environment. In this way, L. conchilega alters the habitat characteristics and affects other organisms, and can therefore even be considered as an ecosystem engineer. In other words, L. conchilega patches are responsible for an increased habitat quality in an otherwise uniform habitat, which results in a higher survival of the surrounding benthic species.  相似文献

Attachment of pathogenic bacteria to food contact surfaces and the subsequent biofilm formation represent a serious threat for the food industry, since these bacteria are more resistant to antimicrobials or possess more virulence factors. The main aim of this study was to investigate the correlation between antibiotic resistance against 13 antibiotics, distribution of 10 virulence factors and biofilm formation in 105 Escherichia coli strains according to their origin. The high prevalence of antibiotic resistance that we have found in wildlife isolates could be acquired by horizontal transfer of resistance genes from human or domestic or farm animals. Consequently, these commensal bacteria might serve as indicator of antimicrobial usage for human and veterinary purposes in the Czech Republic. Further, 46 out of 66 resistant isolates (70%) were able to form biofilm and we found out statistically significant correlation between prevalence of antibiotic resistance and biofilm formation ability. The highest prevalence of antibiotic resistance was observed in weak biofilm producers. Biofilm formation was not statistically associated with any virulence determinant. However, we confirmed the correlation between prevalence of virulence factors and host origin. Chicken isolates possessed more virulence factors (66%), than isolates from wildlife (37%). We can conclude that the potential spread of antibiotic resistance pattern via the food chain is of high concern for public health. Even more, alarming is that E. coli isolates remain pathogenic potential with ability to form biofilm and these bacteria may persist during food processing and consequently lead to greater risks of food contamination.  相似文献

This study is planned to perform a sanitary survey of the largest sewage treatment plant in Riyadh, KSA, fortnightly for 6 months to examine its effluent quality as an example for the growing dependence on reuse of treated municipal wastewater in agricultural irrigation purposes to cope with increasing water shortage. The biological and physico-chemical parameters of 12 wastewater samples from the plant were examined using standard methods. The physico-chemical analysis indicated that the surveyed municipal wastewater treatment plant contained some of the studied parameters, such as turbidity, total suspended solids, biochemical oxygen demand, chemical oxygen demand and residual chlorine above the maximum permissible wastewater limits set by the Saudi Standards. However, heavy metal concentrations in all samples were lower than the recommended standards. Total and faecal coliform counts were above the permissible limits indicating poor sanitation level. Fifty percent of all wastewater samples were contaminated with faecal coliforms but, surprisingly, Escherichia coli were only detected in 8.3 % of the samples. Regular monitoring and enhancement of microbial and physico-chemical parameters of the wastewater quality served by different wastewater treatment plants for reuse in agricultural irrigation is recommended to preserve the environment and public health.  相似文献

1H NMR metabolomics can be used to assess the sub-lethal toxicity of contaminants to earthworms by identifying alterations in the metabolic profiles of contaminant- exposed earthworms in contrast to those of healthy (control) individuals. In support of this method this study sought to better characterize the baseline metabolic profile of healthy, mature earthworms of the species, Eisenia fetida, which is recommended for both acute and sub-lethal toxicity testing for soil contaminants. Profiles of D2O-buffer extracted metabolites were determined using 1H NMR spectroscopy and both inter-individual metabolic variability and pair-wise metabolic correlations were assessed. The control earthworm extracts exhibited low overall inter-individual metabolic variability, with a spectrum-wide median relative standard deviation (%RSD = standard deviation/mean × 100) of 14%, which suggests that the metabolic profile of E. fetida earthworms is well controlled in laboratory conditions and supports further use of this organism in environmental metabolomics research. In addition, strong positive correlations were detected between the levels of maltose, betaine, glycine, and glutamate as well as between the levels of lactate, valine, leucine, alanine, lysine, tyrosine, and phenylalanine which had not previously been reported. Since comparison of pair-wise metabolic correlations between control and treated organisms can reveal changes in the underlying pattern of biochemical relationships between the metabolites, identification of these significant metabolic correlations in control earthworms provides an additional characteristic that may be applied to delineate between control and treated earthworms in future NMR-based metabolomic studies.  相似文献

A significant emerging environmental problem is the disturbance and oxidation of soils with high levels of iron sulphide minerals resulting in acidification and causing the mobilization of metals into groundwater. This process is occurring in many parts of the world. In Western Australia, impacted groundwater is extracted by residents for domestic use. Packaging materials are one of the largest contributors to municipal solid waste production. This paper evaluates the material impacts packaging policy in The Netherlands in the period 1986-2007. Five different voluntary agreements were implemented over this period to reduce the environmental impact of packaging. The analysis shows that among the investigated indicators, population statistically is the most robust indicator to estimate the packaging demand. A baseline is developed on the basis of population growth in The Netherlands, and is used as reference to evaluate policy impacts. The policy periods are evaluated on the basis of overall effectiveness compared to the baseline and target achievement. Dutch packaging policies have been effective to reduce the total packaging volume until 1999. After 2000, packaging consumption increased more rapidly than the baseline, suggesting that policy measures have not been effective. The largest increase in packaging recycling rates was achieved during the first policy period. More clear and consistent packaging policy measures and targets could improve the effectiveness of policies.  相似文献
