During screening for poly-β-hydroxybutyrate (PHB) producing bacteria from distillery effluent sample, six out of 30 isolates comprising of three strains of Alcaligenes sp., two strains of Bacillus sp., and one strain of Pseudomonas sp. were found to accumulate varying levels of intracellular PHB. Amongst the various isolates, Alcaligenes sp. RZS4 was found as the potent PHB-producing organism, accumulating higher amounts of PHB. PHB productivity was further enhanced in the presence of oxygen, nitrogen-limiting conditions, and cloning of PHB synthesizing genes of Alcaligenes sp. RZS 4 into Escherichia coli. A twofold increase in PHB yield was obtained from recombinant E. coli vis-à-vis Alcaligenes sp.; the recombinant E. coli accumulated more PHB in NDMM, produced good amount of PHB in a single-stage cultivation process under both nutrient-rich and nutrient-deficient conditions. Extraction of PHB with acetone–alcohol (1:1) was found as suitable method for optimum extraction of PHB as this mixture selectively extracted PHB without affecting the non-PHB cell mass. PHB extract from recombinant E. coli showed the presence of C–H, =O stretching, =C–H deformation, =C–H, =CH, and =C–O functional groups characteristic of PHB.  相似文献

The effects of solution pH on adsorption of trace metals to different types of natural aquatic solid materials have been studied extensively, but few studies have been carried out to investigate the effect of pH at which the solid materials were formed on the adsorption. The purpose of present study is to examine this effect of culture pH on metal adsorption to natural freshwater biofilms. The adsorption of Pb and Cd to biofilms which were developed at different culture pH values (ranging from 6.5 to 9.0) was measured at the same adsorption pH value (6.5). The culture pH had considerable effects on both composition and metal adsorption ability of the biofilms. Higher culture pH usually promoted the accumulation of organic material and Fe oxides in the biofilms. The culture pH also affected the quantity and species of algae in the biofilms. The adsorption of Pb and Cd to the biofilms generally increased with the increase of culture pH. This increase was minor at lower pH range and significant at higher pH range and was more remarkable for Cd adsorption than for Pb adsorption. The notable contribution of organic material to the adsorption at higher culture pH values was also observed. The profound impacts of culture pH on adsorption behavior of biofilms mainly resulted from the variation of total contents of the biofilm components and were also affected by the alteration of composition and properties of the components.  相似文献

The objective of this study was to develop a bioremediation strategy for cadmium (Cd) and carbendazim co-contaminated soil using a hyperaccumulator plant (Sedum alfredii) combined with carbendazim-degrading bacterial strains (Bacillus subtilis, Paracoccus sp., Flavobacterium and Pseudomonas sp.). A pot experiment was conducted under greenhouse conditions for 180 days with S. alfredii and/or carbendazim-degrading strains grown in soil artificially polluted with two levels of contaminants (low level, 1 mg kg?1 Cd and 21 mg kg?1 carbendazim; high level, 6 mg kg?1 Cd and 117 mg kg?1 carbendazim). Cd removal efficiencies were 32.3–35.1 % and 7.8–8.2 % for the low and high contaminant level, respectively. Inoculation with carbendazim-degrading bacterial strains significantly (P?<?0.05) increased Cd removal efficiencies at the low level. The carbendazim removal efficiencies increased by 32.1–42.5 % by the association of S. alfredii with carbendazim-degrading bacterial strains, as compared to control, regardless of contaminant level. Cultivation with S. alfredii and inoculation of carbendazim-degrading bacterial strains increased soil microbial biomass, dehydrogenase activities and microbial diversities by 46.2–121.3 %, 64.2–143.4 %, and 2.4–24.7 %, respectively. Polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) analysis revealed that S. alfredii stimulated the activities of Flavobacteria and Bradyrhizobiaceae. The association of S. alfredii with carbendazim-degrading bacterial strains enhanced the degradation of carbendazim by changing microbial activity and community structure in the soil. The results demonstrated that association of S. alfredii with carbendazim-degrading bacterial strains is promising for remediation of Cd and carbendazim co-contaminated soil.  相似文献

The acyl carrier protein (ACP) phosphodiesterase gene (SO 4396) of Shewanella oneidensis MR-1 which was analyzed to have azoreductase activity was heterologously expressed in Escherichia coli. The ACP phosphodiesterase was found to reach maximum enzyme velocity 220.59 U/mg, named azoreductase in this study. The azoreductase had highest specific activity (153.16 U/mg) at pH 6.5, which showed a preference for nicotinamide adenine dinucleotide (NADH) as electron donor. The phylogenetic tree analysis indicated that the azoreductase had preference for NADH and dependence for flavin mononucleotide (FMN). However, the azoreductase from S. oneidensis MR-1 still had high enzyme activity in the absence of FMN. The Mg2+ had a positive influence on the enzyme activity with 25 mM concentration, whereas Cr3+, Cd2+ usually had significantly negative effect on enzyme activity. The purified azoreductase retained nearly 100 % activity after incubating in 30 % dimethyl sulfoxide (DMSO), 30 % acetone, 30 % methanol, 20 % ethanol, 20 % isopropanol, and 10 % propanol.  相似文献

Pterocarya stenoptera is a native deciduous tree species and a candidate for reforestation in the riparian zones of the Three Gorges Reservoir Region of Yangtze River in China. Water treatments of continuous flooding (CF) and periodic flooding–drought (PF) were applied to examine the growth dynamics of 4-month-old P. stenoptera seedlings and its effects on soil chemical properties. Results showed that P. stenoptera seedlings in both CF and PF significantly decreased leaf biomass accumulation and the height and diameter growth as compared to that in control (CK; treatment with well-watered, well-drained soil), respectively. There was no significant difference in stem biomass among the three groups, but root biomass in PF showed severe reduction compared to that in both CK and CF. Total biomass in PF was significantly decreased compared to that in CK, but comparable to that in CF. Furthermore, no significant difference was found between CF and CK in total biomass. Water treatments in the unplanted soil pots significantly influenced soil pH, soil organic matter (OM), total nitrogen (TN), and alkali hydrolysable nitrogen (AN) contents, in contrast to no significant effects in total phosphorus (TP), total potassium (TK), available phosphorus (AP), and available potassium (AK) contents. In P. stenoptera soils, there were significant effects by water treatment, time, and treatment × time in the eight tested soil chemical properties, except treatment in TK and time effect in OM content. Compared to unplanted soils, the growth of P. stenoptera seedlings significantly increased soil pH value and OM, TN, TP, and TK contents, while decreasing AN, AP, and AK contents in CK group, augmented the mean value of each of the tested soil chemical properties with an exception of AK content in CF group, and increased soil pH value and TN, AN, TP, and AP contents with no significant differences in OM, TK, and AK contents in PF group. Given the fact that TN and TP contents significantly increased in P. stenoptera soils as compared to those in unplanted soils, growth of P. stenoptera seedlings should be a successful candidate for restoration within the highly dynamic hydrologic zone of the riparian zones of the Three Gorges Reservoir Region.  相似文献

One strategy to combat nitrate (NO3-N) contamination in rivers is to understand its sources. NO3-N sources in the East Tiaoxi River of the Yangtze Delta Region were investigated by applying a 15N–18O dual isotope approach. Water samples were collected from the main channel and from the tributaries. Results show that high total N and NO3-N are present in both the main channel and the major tributaries, and NO3-N was one of the most important N forms in water. Analysis of isotopic compositions (δ 18O, δD) of water suggests that the river water mainly originated from three tributaries during the sampling period. There was a wide range of δ 15N-NO3 (?1.4 to 12.4 ‰) and a narrow range of δ 18O-NO3 (3.7 to 9.0 ‰) in the main channel waters. The δ 15N and δ 18O-NO3 values in the upper, middle, and lower channels along the river were shifted as 8.2, 3.5, and 9.5 ‰, and 9.0, 4.2, and 6.0 ‰, respectively. In the tributary South Tiao, the δ 15N and δ 18O-NO3 values were as high as 9.5 and 7.0 ‰, while in the tributaries Mid Tiao and North Tiao, NO3-N in most of the samples had relatively low δ 15N and δ 18O-NO3 values from 2.3 to 7.5 ‰ and 4.7 to 7.0 ‰, separately. Our results also suggest that the dual isotope approach can help us develop the best management practice for relieving NO3-N pollution in the rivers at the tributary scale.  相似文献

Chromite ore processing residues (COPR) is the source of the Cr(VI) contamination in the environment. Pannonibacter phragmitetus BB was used to treat two different types of COPRs in this research. The water-soluble Cr(VI) of COPR A and B is 3,982.9 and 1,181.4 mg/kg, respectively. In the column biotreatment process, P. phragmitetus BB can reduce Cr(VI) in the leachate to an undetectable level at the flow rate of 1 and 2 ml/min. In the direct biotreatment process, Cr(VI) in the liquid supernatant of COPR A and B decreased from 265 and 200 mg/l to 145 and 40 mg/kg after 240 h of incubation. In one-step and two-step biotreatment processes, Cr(VI) in the liquid supernatant of both COPRs can be reduced to an undetectable level. The toxicity characteristic leaching procedure results indicate that the Cr(VI) concentration of treated COPR A (3.48 mg/l) is lower than the identification standards for hazardous wastes of China (5 mg/l) (GB 5085.6-2007). The information obtained in this study has significance for the application of P. phragmitetus BB to remediate COPR contamination.  相似文献

The concentrations, compositional profiles, and possible sources of polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/PCDF) in water column and surface sediment samples from the lower reaches of the Yangtze River were investigated, and the potential risks posed by these chemicals were evaluated. Concentrations of the 17 2,3,7,8-substituted PCDD/PCDF in the water and sediment samples ranged from 0.45 to 10.0 pg TEQ/L (mean 2.34 pg TEQ/L) and from 0.31 to 51 pg TEQ/g dw (mean 5.25 pg TEQ/g dw), respectively. Water and sediments from the Nanjing–Nantong sections of the Yangtze River were found to have elevated PCDD/PCDF levels, and the PCDD/PCDF TEQ concentrations in 13 sediment samples exceeded the probable effect levels (PEL), above which adverse biological effects are likely. Multivariate statistical analysis indicated that the principal PCDD/PCDF contamination source for the water and sediment was the production and use of pentachlorophenol (PCP) or sodium pentachlorophenate (PCP-Na). PCDD/PCDF concentrations were stable over time near the Yangtze River Estuary but had increased sharply in recent decades in the Nanjing–Nantong section, which will have been related to the growth in industrial activities and other anthropogenic PCDD/PCDF sources. Total organic carbon (TOC) and PCDD/PCDF concentrations correlated significantly (R?=?0.474, P?<?0.01), suggesting that TOC plays an important role in the transport and re-distribution of PCDD/PCDF in the Yangtze River basin.  相似文献

Release of domestic sewage leads to accumulation of xenoestrogens in holding waters, especially in closed or semi-enclosed waters such as lakes. In the study, the occurrence, distribution, estrogenic activity and risk of eight xenoestreogens were evaluated in Lake Donghu, China. Nonylphenol (NP), octylphenol (OP), and bisphenol A (BPA) were identified as the main xenoestrogens ranging from tens of ng L−1 (in the surface water) or ng g−1 dw (in the suspended particles and sediment) to tens of μg L−1 or μg g−1 dw. The sum of 17β-estradiol equivalents (∑EEQs) ranged from 0.32 to 45.02 ng L−1 in the surface water, 0.53 to 71.86 ng g−1 dw in the suspended particles, and 0.09 to 24.73 ng g−1 dw in the sediment. Diethylstilbestrol (DES) was determined as the main contributor to ∑EEQs followed by NP. The risk assessment showed a higher risk in the surface water than in the suspended particles and sediment in such domestic sewage-holding lake.  相似文献
