Polycyclic aromatic hydrocarbon (PAH) exposure and genetic susceptibility were conductive to genotoxic effects including gene damage, which can increase mutational probability. We aimed to explore the dose-effect associations of PAH exposure with damage of exons of epidermal growth factor receptor (EGFR) and breast cancer susceptibility gene 1 (BRCA1), as well as their associations whether modified by Flap endonuclease 1 (FEN1) genotype. Two hundred eighty-eight coke oven male workers were recruited, and we detected the concentration of 1-hydroxypyrene (1-OH-pyr) as PAH exposure biomarker in urine and examined base modification in exons of EGFR and BRCA1 respectively, and genotyped FEN1 rs174538 polymorphism in plasma. We found that the damage indexes of exon 19 and 21 of EGFR (EGFR-19 and EGFR-21) were both significantly associated with increased urinary 1-OH-pyr (both Ptrend < 0.001). The levels of urinary 1-OH-pyr were both significantly associated with increased EGFR-19 and EGFR-21 in both smokers and nonsmokers (both P < 0.001). Additionally, we observed that the urinary 1-OH-pyr concentrations were linearly associated with both EGFR-19 and EGFR-21 only in rs174538 GA+AA genotype carriers (both P < 0.001). Moreover, FEN1rs rs174538 showed modifying effects on the associations of urinary 1-OH-pyr with EGFR-19 and EGFR-21 (both Pinteraction < 0.05). Our findings revealed the linear dose-effect association between exon damage of EGFR and PAH exposure and highlight differences in genetic contributions to exon damage and have the potential to identify at-risk subpopulations who are susceptible to adverse health effects induced by PAH exposure.
Adsorbable organic halogens(AOX) are a general indicator for the total amount of compounds containing organically bonded halogens. AOX concentrations and components were investigated along the wastewater treatment process in four large-scale pharmaceutical factories of China, and genotoxicity based on the SOS/umu test was also evaluated. The results showed that AOX concentrations in wastewater of four factories ranged from 4.6 to 619.4 mg/L, which were high but greatly different owing to differences in the raw materials and products. The wastewater treatment process removed 50.0%–89.9% of AOX, leaving 1.3–302.5 mg/L AOX in the effluents. Genotoxicity levels ranged between 2.1 and 68.0 μg 4-NQO/L in the raw wastewater and decreased to 1.2–41.2 μg 4-NQO/L in the effluents of the wastewater treatment plants(WWTPs). One of the main products of factory I, ciprofloxacin, was identified as the predominant contributor to its genotoxicity. We used aerated systems to assess the influence of the bacterioplankton community on cyanobacterial blooms in algae/post-bloom of Lake Taihu, China. Bacterioplankton community diversity was evaluated by polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE) fingerprinting. Chemical analysis and nitrogen dynamic changes illustrated that NH4+-N was nitrified to NO2-N and NO3-N by bacterioplankton. Finally, NH4+-N was exhausted and NO3-N was denitrified to NO2-N, while the accumulation of NO2-N indicated that bacterioplankton with completely aerobic denitrification ability were lacking in the water samples collected from Lake Taihu. We suggested that adding completely aerobic denitrification bacteria(to denitrify NO2-N to N2)would improve the water quality. PCR-DGGE and sequencing results showed that more than 1/3 of the bacterial species were associated with the removal of nitrogen, and Acidovorax temperans was the dominant one. PCR-DGGE, variation of nitrogen, removal efciencies of chlorophyll-a and canonical correspondence analysis indicated that the bacterioplankton significantly influenced the physiological and biochemical changes of cyanobacteria. Additionally, the unweighted pair-group method with arithmetic means revealed there was no obvious harm to the microecosystem from aeration. The present study demonstrated that bacterioplankton can play crucial roles in aerated ecosystems, which could control the impact of cyanobacterial blooms in eutrophicated fresh water systems. 相似文献
