Decimal reduction times (DRT or D-value) of Campylobacter jejuni, Salmonella enterica (formerly Salmonella choleraesuis) serovar Senftenberg, Escherichia coli, and Listeria monocytogenes were determined in two different matrices, diluted fermenter sludge (DFS) and 0.95% NaCl-solution (NaCl) at 50 degrees C, both under aerobic and anaerobic conditions. Depending on aeration, matrix composition, and the respective organism, the D-values varied between 10min and more than 15h. Generally the viability of bacteria decreased faster in DFS compared to NaCl-solution and under aerobic compared to anaerobic conditions. After 24h no viable cells could be detected in DFS, both under aerobic as well as under anaerobic conditions, whereas viable cells were still found in NaCl solutions. In both matrices the detection limits determined by means of PCR-based and classical microbiological methods were compared and pointed to lower detection limits of the latter methods. Results of the present investigation show that test organisms were far from surviving several days in DFS whereas hydraulic retention times normally used for thermophilic anaerobic digestion are in the range of 2 weeks. However, an underestimation of survival rates of the test organisms seems probable when applying aerobic standard methods.  相似文献

Human health risk assessments for dioxins and dioxin-like PCBs (with the exception of the one by US-EPA) recommend health based exposure limits within the range of 1-4 pg WHO-TEQ/kg bw per day. As all humans are exposed to measurable levels of dioxins and related substances, the determination of the tolerated daily intake is a very significant decision and may influence limit values guiding risk reduction measures and target levels. The proposed TDI has to protect all human subpopulations. In the case of dioxin this is particularly important as the exposure of infants through breast-feeding may exceed the exposure of adults by one or two orders of magnitude. An overview of recently recommended limit values (WHO, SCF, JECFA) for PCDDs, PCDFs and dioxin-like PCBs using WHO-TEFs shows the common feature that the values were derived only from non carcinogenic endpoints. In November 2000 the Scientific Committee on Food of the European Commission published an 'Opinion of the SCF on the Risk Assessment of Dioxins and Dioxin-like PCBs in Food' [SCF, Scientific Committee on Food 2000. On the basis of this extensive review of data and experimental results the Committee recommended a temporary tolerable weekly intake (t-TWI) of 7 pg WHO-TEQ/kg bw. Only six months later the SCF carried out a re-evaluation of its t-TWI from November 2000. The reconsideration of 'pivotal studies' led to the situation that the re-assessment is now based only on rat studies which investigated only reproductive effects only on male offspring and, in addition, three of these studies are single dose studies at gestational day 15. Applying an overall uncertainty factor of 10 to the LOAEL derived estimated human daily intakes (EHDI) the SCF concluded that 14 pg/kg bw per week should be considered as a tolerable intake for 2,3,7,8-TCDD. The SCF stated that on a body weight basis, the dioxin intake of breast-fed infants has been estimated to be one to two orders of magnitude higher than the average adult intake. Recent German data suggest that the body burden of formerly breast-fed children aged 9-11 is still about 30% higher than those of their formula-fed age-mates. As breast-feeding has measurable benefits for neurological and immunological development, formula feeding cannot be recommended as an alternative to lower dioxin intake. So the only remaining way to lower the dioxin uptake is to drastically reduce the background exposure of the general population. It is acknowledged that any recommendation of a precise number for a TDI is flawed by uncertainties and the possibility of different weight being given to the studies of relevance. The determination of the TDI has influence on all regulatory limit values that are based on the TDI value. A higher TDI lowers the level of protection for humans. It is proposed by the German Federal Environmental Agency that the TDI should be reassessed in a process transparent to the public and on the basis of all relevant endpoints from animal experiments and human epidemiology, including the assessment of cancer risks.  相似文献

The redox-active quinalphos main metabolite, 2-hydroxyquinoxaline, is particularly effective under excitation by light. We have studied the photocatalytic destruction of melatonin and its precursors, because the cytoprotective indoleamine has been detected in high quantities in mammalian skin. In photooxidation reactions, in which melatonin, N-acetylserotonin and serotonin are destroyed by 2-hydroxyquinoxaline, the photocatalyst is virtually not consumed. Rates of melatonin and serotonin destruction are not changed by the singlet oxygen quencher 1,4-diazabicyclo-(2,2,2)-octane, indicating that this oxygen species is not involved in the primary reactions, so that the persistence of 2-hydroxyquinoxaline has to be explained by redox cycling. This should imply formation of an organic radical, presumably the quinoxaline-2-oxyl radical, from which 2-hydroxyquinoxaline is regenerated by electron abstraction from indolic radical scavengers. Electron donation by 2-hydroxyquinoxaline is demonstrated by reduction of the 2,2'-azino-bis-(3-ethylbenzthiazolinyl-6-sulfonic acid) cation radical under ultrasound excitation. The compound 2-hydroxyquinoxaline interacts with the specific superoxide anion scavenger Tiron. Formation of oligomeric products from melatonin and serotonin is strongly inhibited by sodium dithionite. Products from photocatalytic indolamine conversion are predominantly dimers and oligomers. No kynuramines were detected in the case of serotonin oxidation, and melatonin's otherwise prevailing oxidation product N(1)-acetyl-N(2)-formyl-5-methoxykynuramine, another cytoprotective metabolite, is only formed in relatively small quantities. The proportion between products from melatonin is changed by 1,4-diazabicyclo-(2,2,2)-octane: singlet oxygen, also formed under the influence of excited 2-hydroxyquinoxaline, only affects secondary reactions.  相似文献

The establishment of high quality long-term monitoring programmes is essential if the implementation of the Water Framework Directive (WFD) is to be effective. It is recognised that monitoring can be very expensive and so guidance is needed to establish cost-effective, risk-based and targeted groundwater monitoring across Europe that enables WFD objectives to be met. In this context, the Groundwater Working Group (WGC) of the Common Implementation Strategy (CIS) of the WFD has developed recommendations aiming to implement consistent groundwater monitoring across Europe. This has been published on the internet in the form of a non-legally binding guidance document, which provides useful elements for the development and maintenance of networks at high standards and thereby provide the necessary information to assess (ground)water status, identify trends in pollutant concentrations, support establishment and assessment of programmes of measures and the effective targeting of economic resources. Phthalates are found in numerous consumer products, including interior materials like polyvinyl chloride (PVC). Several studies have identified phthalates in indoor air. A recent case-control study demonstrated associations between allergic symptoms in children and the concentration of phthalates in dust collected from their homes. Here we have analyzed the content of selected phthalates in particulate matter (PM): PM(10) and PM(2.5) filter samples collected in 14 different indoor environments. The results showed the presence of the phthalates di-n-butyl phthalate (DBP), butyl benzyl phthalate (BBP), dicyclohexyl phthalate (DCHP) and diethyl hexyl phthalate (DEHP) in the samples. The dominating phthalate in both PM(10) and PM(2.5) samples from all locations was DBP. More than a 10-fold variation in the mean concentration of total phthalates between sampling sites was observed. The highest levels of total phthalates were detected in one children's room, one kindergarten, in two primary schools, and in a computer room. The relative contribution of total phthalates in PM(10) and PM(2.5) was 1.1 +/- 0.3% for both size fractions. The contribution of total phthalates in PM(2.5) to total phthalates in PM(10) ranged from 23-81%, suggesting different sources. Of the phthalates that were analyzed in the PM material, DBP was found to be the major phthalate in rubber from car tyres. However, our analyses indicate that tyre wear was of minor importance for indoor levels of both DBP as well as total phthalates. Overall, these results support the notion that inhalation of indoor PM contributes to the total phthalate exposure.  相似文献

A first medium term monitoring of atmospheric transport and distribution for persistent organic pollutants (POPs) in Bj?rn?ya (Bear island) air samples has been performed in the period between week 51/1999 and week 28/2003. A total of 50 single compounds consisting of polychlorinated biphenyls (33 congeners), hexachlorobenzene (HCB), hexachlorocyclohexane isomers (alpha-, beta-, gamma-HCH), alpha-endosulfan, cyclodiene pesticides (chlordanes, nonachlor-isomers, oxy-chlordane, heptachlor and chlordane) as well as dichlorodiphenyltrichloroethane (DDT) derivatives were analysed and quantified. Atmospheric transport of POPs was identified as an important contamination source for the island. PCBs, HCB and HCH isomers were the predominant POP groups, contributing with 70-90% to the overall POP burden quantified in the Bj?rn?ya air samples. The highest concentration levels for a single compound were found for HCB (25-35 pg m(-3)). However, the sum of 33 PCB congeners was found to be in the same concentration range (annual means between 15 and 30 pg m(-3)). Cyclodiene pesticides, DDT derivatives and alpha-endosulfan were identified as minor contaminants. Several atmospheric long-range transport episodes were identified and characterised. Indications for industrial emissions as well as agricultural sources were found for the respective atmospheric transport episodes. A first simple statistical correlation assessment showed that for long-range transport of pollution, the local meteorological situation is not as important as the air mass properties integrated over the time period of the transport event. The local weather situation, on the other hand, is important when investigating deposition rates and up-take/accumulation properties in the local ecosystem. Based upon chemical data interpretation, valuable information about the influence of primary and secondary sources on the air mass contamination with chlorinated insecticides (e.g., HCHs) was found and discussed. The interdisciplinary interpretation of contaminant data using statistical methods, chemical analysis, meteorological modelling and classical meteorological information for a comprehensive evaluation of atmospheric long range transport into the European Arctic (Bj?rn?ya) has proven to be a highly versatile tool not only for atmospheric scientists but also with strong potential for regulatory purposes.  相似文献

Cyanide phytoremediation by water hyacinths (Eichhornia crassipes)   总被引:1,自引:0,他引:1  
Although cyanide is highly toxic, it is economically attractive for extracting gold from ore bodies containing only a few grams per 1000 kg. Most of the cyanide used in industrial mining is handled without observable devastating consequences, but in informal, small-scale mining, the use is poorly regulated and the waste treatment is insufficient. Cyanide in the effluents from the latter mines could possibly be removed by the water hyacinth Eichhornia crassipes because of its high biomass production, wide distribution, and tolerance to cyanide (CN) and metals. We determined the sodium cyanide phytotoxicity and removal capacity of E. crassipes. Toxicity to 5-50 mg CN L(-1) was quantified by measuring the mean relative transpiration over 96 h. At 5 mgCNL(-1), only a slight reduction in transpiration but no morphological changes were observed. The EC(50) value was calculated by probit analysis to be 13 mgCNL(-1). Spectrophotometric analysis indicated that cyanide at 5.8 and 10 mgL(-1) was completely eliminated after 23-32 h. Metabolism of K(14)CN was measured in batch systems with leaf and root cuttings. Leaf cuttings removed about 40% of the radioactivity from solution after 28 h and 10% was converted to (14)CO(2); root cuttings converted 25% into (14)CO(2) after 48 h but only absorbed 12% in their tissues. The calculated K(m) of the leaf cuttings was 12 mgCNL(-1), and the V(max) was 35 mg CN(kg fresh weight)(-1)h(-1). Our results indicate that E. crassipes could be useful in treating cyanide effluents from small-scale gold mines.  相似文献

Imprinting Disorders (ImpDis) are a group of congenital syndromes associated with up to four different types of molecular disturbances affecting the monoallelic and parent-of-origin specific expression of genomically imprinted genes. Though each ImpDis is characterized by aberrations at a distinct genetic site and a specific set of postnatal clinical signs, there is a broad overlap between several of them. In particular, the prenatal features of ImpDis are non-specific. Therefore, the decision on the appropriate molecular testing strategy is difficult. A further molecular characteristic of ImpDis is (epi)genetic mosaicism, which makes prenatal testing for ImpDis challenging. Accordingly, sampling and diagnostic workup has to consider the methodological limitations. Furthermore, the prediction of the clinical outcome of a pregnancy can be difficult. False-negative results can occur, and therefore fetal imaging should be the diagnostic tool on which decisions on the management of the pregnancy should be based. In summary, the decision for molecular prenatal testing for ImpDis should be based on close exchanges between clinicians, geneticists, and the families before the initiation of the test. These discussions should weigh the chances and challenges of the prenatal test, with focus on the need of the family.  相似文献

This study examined the effects of transhumance pressure on total abavoe-ground biomass and forage availability on rangelands in Benin. We also investigated the implications of land cover transitions on rangelands over a 31-year period. Our work was carried out in three regions of Benin representing distinct phytogeographic regimes: Ketou, Tchaourou, and Sinende. Ground-truthing and biomass sampling of the herbaceous and phanaerophyte strata were carried out between the 2016 peak vegetation period and the onset of the 2017 rainy season. Herbaceous biomass was determined by destructive sampling, and biomass of shrub and trees was estimated using non-destructive sampling and allometric equations. Historical and present-day Landsat data allowed an analysis of land cover change for the 1986–2002 and 2002–2017 periods. Land cover analyses yielded evidence of significant expansion of agricultural areas, especially in the latter period. The data also revealed progressive landscape fragmentation and transformations to a land cover of reduced total phytomass. There were no long-term effects of transhumance on trees, but likely on herbaceous biomass. Land cover changes in the study regions seem primarily the result of population pressure, infrastructural changes, persisting norms, and traditions regarding environmental management and the increasing popularity of livestock keeping as an insurance strategy. Rangeland transformations had negative impacts on transhumant herds’ mobility and forage availability. As rangeland stability and consent between agricultural and pastoral land users are at a tipping point, informed policies, and land use planning that foster compromises among all stakeholders are needed.
