A low-cost, high throughput bioanalytical screening method was developed for monitoring cis/trans-permethrin in dust and soil samples. The method consisted of a simple sample preparation procedure [sonication with dichloromethane followed by a solvent exchange into methanol:water (1:1)] with bioanalytical detection using a magnetic particle enzyme-linked immunosorbent assay (ELISA). Quantitative recoveries (83–126 %) of cis/trans-permethrin were obtained for spiked soil and dust samples. The percent difference of duplicate ELISA analyses was within ± 20 % for standards and ± 35 % for samples. Similar sample preparation procedures were used for the conventional gas chromatography/mass spectrometry (GC/MS) analysis except that additional cleanup steps were required. Recoveries of cis/trans-permethrin ranged from 81 to 108 % for spiked soil and dust samples by GC/MS. The ELISA-derived permethrin concentrations were highly correlated with the GC/MS-derived sum of cis/trans-permethrin concentrations with a correlation coefficient (r) of 0.986. The ELISA method provided a rapid qualitative screen for cis/trans-permethrin in soil and dust while providing a higher sample throughput with a lower cost as compared to the GC/MS method. The ELISA can be applied as a complementary, low-cost screening tool to prioritize and rank samples prior to instrumental analysis for exposure studies. 相似文献

The years 2012 and beyond seem likely to record major changes in energy use and power generation. The Japanese tsunami has resulted in large countries either scaling back or abolishing the future use of nuclear energy. The discovery of what seems like vast amounts of economically deliverable natural gas has many forecasting a rapid switch from coal- to gas-fired generating plants. On the other hand, environmentalists have strong objections to the production of natural gas and of petroleum by hydraulic fracturing from shale, or by extraction of heavy oil. They believe that global warming from the use of fossil fuels is now established beyond question. There has been rapid progress in the development of alternative energy supplies, particularly from on-shore and off-shore wind. Progress toward a viable future energy mix has been slowed by a U.S. energy policy that seems to many to be driven by politics. The author will review the history of power and energy to put all of the above in context and will look at possible future developments. He will propose what he believes to be an idealized energy policy that could result in an optimum system that would be arrived at democratically. 相似文献

The development of state implementation plans (SIPs) for attainment of criteria pollutant standards is an integral component of air quality management in the United States. However, the content and efficacy of SIPs have rarely been examined systematically. Here, 20 SIPs developed in response to the 1997 8-hr ozone standard are reviewed as case studies of attainment efforts at the state level. Comparison of observed and model predicted ozone concentrations shows the US Environmental Protection Agency (EPA) recommended modeled attainment test to be a somewhat conservative predictor of attainment. Among 12 SIPs for regions that sought attainment by 2009, the test correctly predicted attainment and nonattainment in four and five regions, respectively; in the other three regions, attainment was observed despite predictions of nonattainment. However weight-of-evidence determinations and deviations from the recommended modeled attainment test methodology led five of these SIPs to predict attainment that was not in fact observed by 2009; three of those regions achieved attainment in 2010. Ozone and NO2 concentrations declined across much of the United States during the period covered by the SIPs, with rates of improvement strongly correlated with the initial pollution levels and hence greatest in nonattainment regions. However at monitors with mid-range levels of ozone initially, rates of reduction were largely independent of the initial attainment status of the region. This is consistent with thefact that apart from California, the majority of ozone precursor reductions documented by SIPs resulted from federal measures rather than from state or local controls specific to the nonattainment regions. 相似文献

The European Union Water Framework Directive (WFD) requires Member States to set water quality objectives and identify cost-effective mitigation measures to achieve "good status" in all waters. However, costs and effectiveness of measures vary both within and between catchments, depending on factors such as land use and topography. The aim of this study was to develop a cost-effectiveness analysis framework for integrating estimates of phosphorus (P) losses from land-based sources, potential abatement using riparian buffers, and the economic implications of buffers. Estimates of field-by-field P exports and routing were based on crop risk and field slope classes. Buffer P trapping efficiencies were based on literature metadata analysis. Costs of placing buffers were based on foregone farm gross margins. An integrated optimization model of cost minimization was developed and solved for different P reduction targets to the Rescobie Loch catchment in eastern Scotland. A target mean annual P load reduction of 376 kg to the loch to achieve good status was identified. Assuming all the riparian fields initially have the 2-m buffer strip required by the General Binding Rules (part of the WFD in Scotland), the model gave good predictions of P loads (345-481 kg P). The modeling results show that riparian buffers alone cannot achieve the required P load reduction (up to 54% P can be removed). In the medium P input scenario, average costs vary from £38 to £176 kg P at 10% and 54% P reduction, respectively. The inorganic nitrogen transformations occurring at a municipal waste leachate treatment facility were investigated. The treatment
facility consisted of a collection well and an artificial wetland between two aeration ponds. The first aeration pond showed
a decrease in ammonium (from 3480 (± 120) to 630(± 90) mg ⋅ L−1), a reduction in inorganic nitrogen load (3480 to 1680 mg N ⋅ L−1), and an accumulation of nitrite (< 1.3 mg-N ⋅ L−1 in the collection well, to 1030 mg-N ⋅ L−1). Incomplete ammonium oxidation was presumably the result of the low concentration of carbonate alkalinity (∼2 mg ⋅ L−1), which may cause a limitation in the ammonium oxidation rate of nitrifiers. Low carbonate alkalinity levels may have been
the result of stripping of CO2 from the first aeration pond at the high aeration rates and low pH. Various chemodenitrification mechanisms are discussed
as the reason for the reduction in the inorganic nitrogen load, including; the reduction of nitrite by iron (II) (producing
various forms of gaseous nitrogen); and reactions involving nitrous acid. It is suggested that the accumulation of nitrite
may be the result of inhibition of nitrite oxidizers by nitrous acid and low temperatures. Relative to the first aeration
pond, the speciation and concentration of inorganic nitrogen was stable in the wetlands and 2nd aeration pond. The limited

Despite the widespread and successful use of luminescence-based bioassays in water testing, their applications to soils and sediments is less proven. In part this is because such bioassays have mainly been carried out in an aqueous-based medium and, as such, favour contaminants that are readily water-soluble. In this study, aqueous solutions and soils contaminated with heavy metals (HM), polar organic contaminants and hydrophobic organic contaminants (HOCs) were tested using a range of luminescence-based bioassays (Vibrio fischeri, Escherichia coli HB101 pUCD607 and Pseudomonas fluorescens 10586r pUCD607). For the first two chemical groups, the assays were highly reproducible when optimised extraction procedures were employed but for HOCs the bioassay response was poor. Quantitative structure-activity relationships (QSARs) obtained from aqueous solutions had a linear response although correlation for the chemicals tested using bacterial bioassays was significantly less sensitive than that of sublethal tests for Tetrahymena pyriformis. Bacterial and Dendrobaena veneta bioassay responses to extracts from HM amended soils showed that a clear relationship between trophic levels could be obtained. There is no doubt that the wide range of bioluminescent-based bioassays offers complementary applications to traditional testing techniques but there is a significant need to justify and optimise the extraction protocol prior to application. 相似文献

The Escambia Wood Treating Company (ETC) Superfund site, Pensacola, FL, is contaminated with polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans (PCDD/F), benzo(a)pyrene, lead and arsenic from pentachlorophenol (PCP), creosote, and other compounds used to treat utility poles and foundation pilings. Although ETC's operations ceased in 1982, soils in the areas surrounding the facility continue to exhibit elevated levels of contaminants attributable to ETC operations. In July 2000, individuals who may have been affected by contamination from the ETC site, including current and former residents and former workers and their household members were invited to participate in a study, which included a health and exposure history and routine blood analysis. We also conducted a toxicological health evaluation of a subset of these eligible workers/residents by analyzing serum levels of 17 PCDD/F congeners. Members of the ETC cohort exhibited elevated serum PCDD/F relative to the general population, and congener profiles in members of the cohort reflected patterns commonly observed in persons exposed to PCP. Hypertension prevalence in the cohort was found to correlate with PCDD/F levels, although no other significant relationships were identified with monitored health indices. 相似文献

The use of molecular tools, principally qPCR, versus traditional culture-based methods for quantifying microbial parameters (e.g., Fecal Indicator Organisms) in bathing waters generates considerable ongoing debate at the science–policy interface. Advances in science have allowed the development and application of molecular biological methods for rapid (~2 h) quantification of microbial pollution in bathing and recreational waters. In contrast, culture-based methods can take between 18 and 96 h for sample processing. Thus, molecular tools offer an opportunity to provide a more meaningful statement of microbial risk to water-users by providing near-real-time information enabling potentially more informed decision-making with regard to water-based activities. However, complementary studies concerning the potential costs and benefits of adopting rapid methods as a regulatory tool are in short supply. We report on findings from an international Working Group that examined the breadth of social impacts, challenges, and research opportunities associated with the application of molecular tools to bathing water regulations. 相似文献
