INTENTION, GOAL, SCOPE, BACKGROUND: Cyanobacteria have the natural ability to degrade moderate amounts of organic pollutants. However, when pollutant concentration exceeds the level of tolerance, bleaching of the cells and death occur within 24 hours. Under stress conditions, cyanobacterial response includes the short-term adaptation of the photosynthetic apparatus to light quality, named state transitions. Moreover, prolonged stresses produce changes in the functional organization of phycobilisomes and in the core-complexes of both photosystems, which can result in large changes in the PS II fluorescence yield. The localization of ferredoxin-NADP+ reductase (FNR) at the ends of some peripheral rods of the cyanobacterial phycobilisomes, makes this protein a useful marker to check phycobilisome integrity. OBJECTIVE: The goal of this work is to improve the knowledge of the mechanism of action of a very potent pesticide, lindane (gamma-hexaclorociclohexane), in the cyanobacterium Anabaena sp., which can be considered a potential candidate for bioremediation of pesticides. We have studied the effect of lindane on the photosynthetic apparatus of Anabaena using fluorescence induction studies. As ferredoxin-NADP+ reductase plays a key role in the response to oxidative stress in several systems, changes in synthesis, degradation and activity of FNR were analyzed. Immunolocalization of this enzyme was used as a marker of phycobilisome integrity. The knowledge of the changes caused by lindane in the photosynthetic apparatus is essential for rational further design of genetically-modified cyanobacteria with improved biorremediation abilities. METHODS: Polyphasic chlorophyll a fluorescence rise measurements (OJIP) have been used to evaluate the vitality and stress adaptation of the nitrogen-fixing cyanobacterium Anabaena PCC 7119 in the presence of increasing concentrations of lindane. Effects of the pesticide on the ultrastructure have been investigated by electron microscopy, and FNR has been used as a marker of phycobilisome integrity. RESULTS AND DISCUSSION: Cultures of Anabaena sp. treated with moderate amounts of lindane showed a decrease in growth rate followed by a recovery after 72 hours of pesticide treatment. Concentrations of lindane below 5 ppm increased the photosynthetic performance and activity of the cells. Higher amounts of pesticide caused a decrease in these activities which seems to be due to a non-competitive inhibition of PS II. Active PS II units are converted into non-QA reducing, so called heat sink centers. Specific activity and amount of FNR in lindane-treated cells were similar to the values measured in control cultures. Release of FNR from the thylakoid after 48 hours of exposure to 5 ppm of lindane towards the cytoplasm was detected by immunogold labeling and electron microscopy. CONCLUSIONS: From these results, we conclude that the photosynthetic performance and activity of the cells are slightly increased in the presence of lindane up to 5 ppm. Moreover, in those conditions, lindane did not produce significant changes in the synthesis, degradation or activity of FNR. The high capability of Anabaena to tolerate lindane makes this cyanobacterium a good candidate for phytoremediation of polluted areas. RECOMMENDATION AND OUTLOOK: The results of this study show that cultures of Anabaena PCC 7119 tolerate lindane up to 5 ppm, without significant changes in the photosynthetic vitality index of the cells. However, a slight increase in phycobiliprotein synthesis is observed, which is related to total protein content. This change might be due to degradation of proteins less stable than phycobiliproteins. An identification of the proteins with altered expression pattern in the presence of the pesticide remains the subject of further work and will provide valuable information for the preparation of strains which are highly tolerant to lindane. 相似文献
Many oil industry related sites have become contaminated due to the activities characteristic of this industry, such as oil exploration and production, refining, and petro-chemistry. In Mexico, reported hydrocarbon spills for the year 2000 amounted to 185 203, equivalent to 6252 tons (PEMEX, 2000). The first step for the remediation of these polluted sites is to assess the size and intensity of the oil contamination affecting the subsoil and groundwater, followed by a health risk assessment to establish clean up levels. The aim of this work was to characterize the soil and water in a north-central Mexico Oil Storage and Distribution Station (ODSS), in terms of TPHs, gasoline and diesel fractions, BTEX, PAHs, MTBE, and some metals. Besides, measurements of the explosivity index along the ODSS were made and we describe and discuss the risk health assessment analysis performed at the ODSS, as well as the recommendations arising from it. Considering soils with TPH concentrations higher than 2000 mg kg−1, the contaminated areas corresponding to the railway zone is about 12 776.5 m2, to the south of the storage tanks is about 6558 m2, and to the south of the filling tanks is about 783 m2. Total area to be treated is about 20 107 m2 (volume of 20 107 m3), considering 1 m depth. 相似文献
The entire northwestern coast of Portugal is undergoing severe erosion and there are several areas at high risk of erosion.
Commonly considered as a problem—because it jeopardizes human development along the coast—erosion is indeed a natural process
of sediment redistribution. This paper presents a brief analysis of erosion driving forces and the subsequent state of vulnerability
that coastal segments between the mouth of the River Douro and Cape Mondego are facing. The paper also discusses erosion risk
levels, low or high, and the subsequent questions whether there are populations, economical assets or natural habitats at
risk and/or areas prone to coastal flooding. Main challenges and future trends along the study area are identified in the
light of understanding the underlying causes of conflicts and what realistically can be achieved given the morphodynamics
and hydrodynamic processes, human development established along this coastal segment and the existing policies. 相似文献
Planktonic patches are defined as areas where the abundance of plankters is above a threshold value τ. The estimation of patch
size and shape can be approached using spatial statistical tools, using truncated random fields or indicator random fields
as classifiers. In all cases there is the risk of false positive and false negative errors. In this paper we present the results
of a comparative study on the performance of four commonly used methods: conditional simulation and kriging, both in the original
measurement units of the data and under an indicator transform. We used a misclassification cost function to compare the four
methods. Our results show that conditional simulation in the original measurement units attains the lowest misclassification
cost. We also illustrate how the point at which this minimum is attained can be used to chose an optimal cut-off value for
binary classification.

Social insect colonies often have one or a few queens. How these queens maintain their reproductive monopoly, when other
colony members could gain by sharing in the reproduction, is not generally known. DNA microsatellite genotyping is used to
determine reproductive interests of various classes of colony members in the paper wasp, Polistes annularis. The relatedness estimates show that the best outcome for most individuals is to be the reproductive egg-layer. For workers,
this depends on the sex of offspring: they should prefer to lay their own male eggs, but are indifferent if the queen lays
the female eggs. The next-best choice is usually to support the current queen. As a rule, subordinates and workers should
prefer the current queen to reproduce over other candidates (though subordinates have no strong preference for the queen over
other subordinates, and workers may prefer other workers as a source of male eggs). This result supports the theory that reproductive
monopoly stems from the collective preferences of non-reproductives, who suppress each other in favor of the queen. However,
we reject the general hypothesis of collective worker control in this species because its predictions about who should succeed
after the death of the present queen are not upheld. The first successor is a subordinate foundress even though workers should
generally prefer a worker successor. If all foundresses have died, an older worker succeeds as queen, in spite of a collective
worker preference for a young worker. The results support the previous suggestion that age serves as a conventional cue serving
to reduce conflict over queen succession.

In order to study the bioaccumulation of Pb, Cr, Ni, and Zn and the stress response, the floating aquatic plant Limnobium laevigatum was exposed to increasing concentrations of a mixture of these metals for 28 days, and its potential use in the treatment of wastewater was evaluated. The metal concentrations of the treatment 1 (T1) were Pb 1 μg L−1, Cr 4 μg L−1, Ni 25 μg L−1, and Zn 30 μg L−1; of treatment 2 (T2) were Pb 70 μg L−1, Cr 70 μg L−1, Ni 70 μg L−1, and Zn 70 μg L−1; and of treatment 3 (T3) were Pb 1000 μg L−1, Cr 1000 μg L−1, Ni 500 μg L−1, and Zn 100 μg L−1, and there was also a control group (without added metal). The accumulation of Pb, Cr, Ni, and Zn in roots was higher than in leaves of L. laevigatum, and the bioconcentration factor revealed that the concentrations of Ni and Zn in the leaf and root exceeded by over a thousand times the concentrations of those in the culture medium (2000 in leaf and 6800 in root for Ni; 3300 in leaf and 11,500 in root for Zn). Thus, this species can be considered as a hyperaccumulator of these metals. In general, the changes observed in the morphological and physiological parameters and the formation of products of lipid peroxidation of membranes during the exposure to moderate concentrations (T2) of the mixture of metals did not cause harmful effects to the survival of the species within the first 14 days of exposure. Taking into account the accumulation capacity and tolerance to heavy metals, L. laevigatum is suitable for phytoremediation in aquatic environments contaminated with moderated concentrations of Cr, Ni, Pb, and Zn in the early stages of exposure.
Volatile organic compounds (VOCs) from several different municipal solid wastes’ treatment plants in Mallorca (Spain) have been analysed by thermal desorption-gas chromatography-mass spectrometry (TD-GC-MS). Ambient (immission) air was collected during February and March 2011 by active sampling onto sorbents Tenax™ TA and Carboxen™ 1000. The study presents the chemical characterisation of 93 volatile organic compounds (VOCs) from an overall set of 84 immission air samples. 70 VOCs were positively identified. The linear fit for all 93 external standard calibration, from 10 mg L−1 to 150 mg L−1 (n = 4), was within the range 0.974 < r2 < 0.998. Limits of detection of the method (LOD) for all the standards were within the range 1.1-4,213 pg, as the absolute standard amount spiked into sorbent tubes in 1 μL standard mixture (dissolved in methanol).Overall results stated systematic correlation between waste’s nature and VOCs’ air composition. Organic wastes show main contribution of terpenes, waste water sludge residues’ of reduced sulphured compounds (RSCs) and municipal solid wastes show contribution of a wide sort of VOCs. The use of a chemometric approach for variable’s reduction to 12 principal components enables evaluation of similarities and dissimilarities between facilities. PCA clearly related samples to its corresponding facility on the basis of their VOCs composition and the ambient temperature. 相似文献
Myelocystocele is a rare spinal cord disorder and has not been described prenatally. We report a case in which prenatal ultrasound and magnetic resonance imaging (MRI) demonstrated a posterior cervical mass which was initially thought to be a meningocele or an atypical cystic hygroma. Surgery performed at 1 day of age showed this to be a myelocystocele. Therefore, the differential diagnosis of an extracranial cystic mass in the posterior cervical region should be expanded to include myelocystoceles. 相似文献
A simplified dynamic model for the activated sludge process with high-strength wastewaters is presented. The model is based
on activated sludge models and predicts effluent chemical oxygen demand concentration using only four parameters. Experimental
data for testing the model were obtained in an activated sludge plant using flax retting wastewater. The proposal of the model
is described: mass balance to the system, selection of variables and behaviour of the theoretical values versus experimental
results. 相似文献
