INTENTION, GOAL, SCOPE, BACKGROUND: Cyanobacteria have the natural ability to degrade moderate amounts of organic pollutants. However, when pollutant concentration exceeds the level of tolerance, bleaching of the cells and death occur within 24 hours. Under stress conditions, cyanobacterial response includes the short-term adaptation of the photosynthetic apparatus to light quality, named state transitions. Moreover, prolonged stresses produce changes in the functional organization of phycobilisomes and in the core-complexes of both photosystems, which can result in large changes in the PS II fluorescence yield. The localization of ferredoxin-NADP+ reductase (FNR) at the ends of some peripheral rods of the cyanobacterial phycobilisomes, makes this protein a useful marker to check phycobilisome integrity. OBJECTIVE: The goal of this work is to improve the knowledge of the mechanism of action of a very potent pesticide, lindane (gamma-hexaclorociclohexane), in the cyanobacterium Anabaena sp., which can be considered a potential candidate for bioremediation of pesticides. We have studied the effect of lindane on the photosynthetic apparatus of Anabaena using fluorescence induction studies. As ferredoxin-NADP+ reductase plays a key role in the response to oxidative stress in several systems, changes in synthesis, degradation and activity of FNR were analyzed. Immunolocalization of this enzyme was used as a marker of phycobilisome integrity. The knowledge of the changes caused by lindane in the photosynthetic apparatus is essential for rational further design of genetically-modified cyanobacteria with improved biorremediation abilities. METHODS: Polyphasic chlorophyll a fluorescence rise measurements (OJIP) have been used to evaluate the vitality and stress adaptation of the nitrogen-fixing cyanobacterium Anabaena PCC 7119 in the presence of increasing concentrations of lindane. Effects of the pesticide on the ultrastructure have been investigated by electron microscopy, and FNR has been used as a marker of phycobilisome integrity. RESULTS AND DISCUSSION: Cultures of Anabaena sp. treated with moderate amounts of lindane showed a decrease in growth rate followed by a recovery after 72 hours of pesticide treatment. Concentrations of lindane below 5 ppm increased the photosynthetic performance and activity of the cells. Higher amounts of pesticide caused a decrease in these activities which seems to be due to a non-competitive inhibition of PS II. Active PS II units are converted into non-QA reducing, so called heat sink centers. Specific activity and amount of FNR in lindane-treated cells were similar to the values measured in control cultures. Release of FNR from the thylakoid after 48 hours of exposure to 5 ppm of lindane towards the cytoplasm was detected by immunogold labeling and electron microscopy. CONCLUSIONS: From these results, we conclude that the photosynthetic performance and activity of the cells are slightly increased in the presence of lindane up to 5 ppm. Moreover, in those conditions, lindane did not produce significant changes in the synthesis, degradation or activity of FNR. The high capability of Anabaena to tolerate lindane makes this cyanobacterium a good candidate for phytoremediation of polluted areas. RECOMMENDATION AND OUTLOOK: The results of this study show that cultures of Anabaena PCC 7119 tolerate lindane up to 5 ppm, without significant changes in the photosynthetic vitality index of the cells. However, a slight increase in phycobiliprotein synthesis is observed, which is related to total protein content. This change might be due to degradation of proteins less stable than phycobiliproteins. An identification of the proteins with altered expression pattern in the presence of the pesticide remains the subject of further work and will provide valuable information for the preparation of strains which are highly tolerant to lindane.  相似文献

The biotransformation and mineralization of a mixture of two polycyclic aromatic hydrocarbons (PAHs), anthracene and pyrene, which are known contaminants of soil and groundwater, by an enrichment culture in the presence or absence of 100 mg l(-1) Tergitol NP-10, a non-ionic surfactant, and at temperatures of 10 degrees C and 25 degrees C were investigated. The overall biotransformation of 2 mg l(-1) total PAHs with free cell suspensions in batch culture was greater than 97.2% at both examined temperatures. At 25 degrees C, the overall mineralization of anthracene was 48.8% and that of pyrene was 66.1%. However, the decrease of temperature to 10 degrees C had a negative effect on the mineralization of PAHs and reduced it to 18.5% and 61.5% for anthracene and pyrene, respectively. Using a higher PAHs concentration of 20 mg l(-1) at 25 degrees C, the overall biotransformation of anthracene was 80.7% and that of pyrene was 100%, where only 17.3% anthracene and 7.6% pyrene were mineralized to carbon dioxide and water. The addition of surfactant at 25 degrees C increased the overall mineralization of anthracene and pyrene to 33.0% and 27.6%, respectively. However, the addition of surfactant at 10 degrees C had a negative impact on the overall biotransformation of anthracene and pyrene, reducing them to 20.6% and 14.0%, respectively. These results have significant implications in the bioremediation of PAHs-contaminated sites.  相似文献

This article summarizes the current status of known contamination arising from abandoned uranium mines (AUMs) within the Navajo Nation and examines the options for addressing the elevated risks to health that AUM waste poses to the people of the Navajo Nation. More specifically, this article provides the following:
  1. An overview of past uranium mining conducted on the Navajo Nation, the current status of AUM waste within the Navajo Nation, and the human health risks associated with uranium exposure.
  2. A discussion of Navajo Fundamental Law, the Diné (the Navajo People) Uranium Remediation Advisory Commission, and the Navajo Nation Department of Justice's position regarding institutional controls and cleanup of AUMs.
  3. A summary of cleanup actions taken to date to address AUM waste and the lessons learned from such actions.
  4. Options for cleanup of AUM waste consistent with Diné Fundamental Law, the Diné Natural Resources Protection Act of 2005, and the Comprehensive Environmental Response, Compensation, and Liability Act of 1980 (CERCLA).

To better understand the affect of different levels of EMF on one’s brain activity, the Hodgkin–Huxley model has been used to describe the generation of an active potential in the nerve cell membrane. The theoretical calculations have shown that by the simultaneous affect of both passing an electrical potential and external very low frequency AC, one’s excitation pulses may be suppressed beginning with a certain level of external current. The level of this current depends on the frequency. It is postulated that to eliminate this suppression, the brain increases its normal level relatively to the lower limit. It is also postulated that the brain “tunes up” in the presence of external AC, by increasing the amplitude of its own excitation signals so that suppression does not occur. It is possible that this “tuning” process lasts some time after removing the external EMF as the brain adapts. This has been observed experimentally.  相似文献

Increased sulphate (SO4) export from wetlands following summer droughts in central Ontario, Canada has been associated with the delayed chemical recovery of downstream surface waters following decreased sulphur (S) emissions. Prolonged summer droughts result in a decrease or cessation of stream flow, declines in wetland water table level and oxidation of reduced S compounds to SO4, which is subsequently flushed into drainage streams when stream flow resumes. Sulphate input-output budget calculations (1983–1995 and 1999–2001) at a conifer Sphagnum swamp in the Plastic Lake catchment, indicate that SO4 is retained in most years but is exported on a net basis following particularly severe summer droughts that result in the cessation of stream flow for more than 54 days (95% CI: 41–72 days). Hindcast calculations using long-term (1916–2000) stream discharge records from a nearby station indicate that while droughts occurred frequently in south-central Ontario over the past 85 years, sufficiently dry conditions to cause net SO4 export occurred in only18 of the past 85 years, and indicate a cumulative positive SO4 balance for the swamp (i.e. net SO4 retention). Furthermore, the S pool at the Plastic Lake swamp has been estimated to be ∼1500 kg S/ha in the upper 40 cm peat layer, which is large compared to the amount of net SO4 export that occurs even in years with particularly dry summers (e.g. −43 kg S/ha in 1987/88). Together, these data suggest that the wetland S pool at Plastic Lake has not been depleted by previous droughts and will continue to sustain episodic drought-related SO4 export for the foreseeable future.  相似文献

Short-chain chlorinated paraffins are a class of organic compounds widely used in many industrial applications, extensively diffused into the environment, persistent, bioaccumulative, and toxic towards aquatic organisms. However, their study and monitoring in the environment are still limited. Because of the enormous number of positional isomers that characterise their mixtures, the analysis of this class of pollutants is very difficult to perform. Beside this, the lack of certified reference materials poses a problem for the assessment of the quality assurance/quality control of any analytical procedure. At present, the scientific community does not agree on any analytical reference method, although the monitoring of short-chain chlorinated paraffins has already started in order to comply with the Water Framework Directive of the European Union on water quality. In this paper the regulatory framework, in which chlorinated paraffins are included, and the status concerning their determination are summarized. The main analytical difficulties still existing are discussed, and the definition of a method-defined parameter as well as the development of a standardised method are suggested as a way to obtain comparable monitoring data.  相似文献

A combination of bioenergetics and biochemical biomarkers in mussels was applied to assess possible pollution impacts in a protected semi-enclosed estuary (Amvrakikos Gulf, NW Greece) that receives pesticide discharges through riverine transport. Scope for growth, a physiological condition index representing the energy budget of the organism, was applied to detect general stress effects on the health status of mussels. The low energy budgets of mussels revealed stress conditions and provided early warning signals of possible consequences at higher levels of biological organization. Biochemical markers of exposure confirmed a risk of pesticide contamination. Decreased acetylcholinesterase activities indicated exposure to organophosphate and carbamate pesticides. Responses of the antioxidant enzyme glutathione peroxidase suggested the presence of contaminants capable of reactive oxygen species production that could be related to organochlorine pesticide contamination in the area. On the other hand, metallothionein levels implied low metal contamination.  相似文献

Few data are available to quantify the transfer of both natural and anthropogenic radionuclides to detritivorous invertebrates to facilitate estimation of the internal dose to such biota in models used to assess radiation exposure. To enhance the available data, activity concentrations of 137Cs, 40K, 90Sr, 239?+?240Pu, 241Am, 235U and 238U were measured in ants (Formicidae) and corresponding undisturbed soil collected from the Zlatibor mountain in Serbia and ant/soil concentration ratios (CR) calculated. The 241Am concentration ratios for ants were fourfold higher than those calculated for ants in a previous study whereas they are similar to the more numerous data previously reported for a range of detritivorous invertebrates in other studies. CR values for 137Cs in ants were similar to the few other reported values and slightly lower than those for a range of detritivorous invertebrates. Those for 239?+?240Pu were slightly higher than those for ants in two other studies but they were close to upper limit of a range of data reported for detritivorous invertebrates. All the CR values will be included in a future revision of the ERICA Tool database and will particularly improve the information available for uranium.  相似文献
