The present study analyzed the bioactivity of whole body extracts from six solitary and eight colonial ascidian taxa against 20 sympatric bacterial isolates and one sympatric diatom species from the Western Antarctic Peninsula. Ascidians had crude lipophilic and hydrophilic extracts assayed against 20 bacterial strains. The lipophilic extract of one ascidian caused growth inhibition in all bacterial isolates at 3× tissue-level concentrations. The lipophilic and hydrophilic extracts were fractionated into seawater-soluble and insoluble fractions and assayed at three concentrations against a sympatric diatom species. Significant diatom mortality was detected at 3× and 1× concentrations in all but one ascidian taxon. Lipophilic fractions caused higher diatom mortality than hydrophilic extracts. The specificity of secondary metabolites against diatom fouling and the lack of activity against bacteria suggest high selective pressure for chemical defenses against diatom fouling or the potential that bacterial pathogens are controlled by the ascidian immune system.  相似文献

The General Motors Research Laboratories and the Sloan-Kettering Institue for Cancer Research are collaborating to determine the contribution by automotive vehicles to the polynuclear aromatic hydrocarbons in city air. Sampling of particulate matter at the rate of 140 M3/min (5000 cfm) was carried out at two heavily-trafficked sites in Detroit and one suburban site in Warren, Michigan. Carbon monoxide was determined continuously, and particulate matter was analyzed for “tar,” polynuclear aromatic hydrocarbons, lead, vanadium, and sulfates. Polynuclear aromatic hydrocarbons in automobile exhaust gas are assumed to be dispersed in air along with carbon monoxide or lead from automobiles. It is further assumed that automobiles are the sole source of carbon monoxide and lead in the atmosphere. Concentrations of carbon monoxide and lead in exhaust gas and in the air are utilized to estimate the percentage of polynuclear aromatic hydrocarbons in the air attributable to automobiles. The mean automobile contributions to benzo(a)pyrene in the air, based on lead concentrations, were 18% at a Freeway Interchange, 5% in a downtown commercial area, and 42% in suburban Warren. The average concentrations of benzo(a)pyrene at the sites were 6 μg/103 M3, 7 μg/103 M3 and 1 μg/103 M3, respectively. Mean contributions based on carbon monoxide concentrations were approximately twice the levels based on lead concentrations. Benzo(a)pyrene and benz(a)anthracene in air were not statistically related to carbon monoxide or lead in air, but were higher in winter than in summer, probably because of the higher levels of these materials emitted in space heating combustion in winter.  相似文献

The attenuation and fate of erythromycin-resistance-methylase (erm) and extended-spectrum beta-lactamse (bla) genes were quantified over time in aquatic systems by adding 20-L swine waste to 11,300-L outdoor mesocosms that simulated receiving water conditions below intensive agricultural operations. The units were prepared with two different light-exposure scenarios and included artificial substrates to assess gene movement into biofilms. Of eleven genes tested, only erm(B), erm(F), blaSHV and blaTEM were found in sufficient quantity for monitoring. The genes disappeared rapidly from the water column and first-order water-column disappearance coefficients were calculated. However, detected gene levels became elevated in the biofilms within 2 days, but then disappeared over time. Differences were observed between sunlight and dark treatments and among individual genes, suggesting that ecological and gene-specific factors play roles in the fate of these genes after release into the environment. Ultimately, this information will aid in generating better predictive models for gene fate.  相似文献

Alterations in development and reproduction as a consequence of exposure to endocrine-disrupting chemicals (EDCs) have been demonstrated in many wildlife species. Animals living in, or closely associated with, the aquatic environment are particularly vulnerable to endocrine disruption because thousands of chemicals are actively disposed into rivers, estuaries and seas. Fish have thus been a focus in endocrine disruption studies, and some of the most comprehensive studies on the disruption of sexual development and function are on the roach (Rutilus rutilus). This paper provides a critical analysis of the roach as a sentinel for studies into endocrine disruption. The paper starts by describing what is known on the basic reproductive biology of the roach, information essential for interpreting chemical effect measures on sexual development and function. We then analyze where and how the roach has been applied to improve our understanding of the estrogenic nature of discharges from wastewater treatment works (WWTWs) and describe the phenomenon of feminized male roach in UK rivers. In this paper, the causation of these effects and issues of relative susceptibility and sensitivity of the roach to the effects of estrogenic EDCs are addressed. The paper then describes the ongoing work on the development of genetic and genomic resources for roach and analyses how these are being applied in studies to understand the mechanisms of disruption of sexual development. Finally, the paper addresses the biological significance of sexual disruption and intersex for the individual and discusses the possible implications for wild populations.  相似文献

A series of miscible blends consisting of cellulose acetate propionate (CAP) and poly(ethylene glutarate) (PEG) or poly(tetramethylene glutarate) (PTG) were evaluated in a static bench-scale simulated municipal compost environment. Samples were removed from the compost at different intervals, and the weight loss was determined before evaluation by gel permeation chromatography, scanning electron microscopy, and1H NMR. The type of polyester (PEG versus PTG) in the blend made no difference in composting rates. At fixed CAP degree of substitution (DS), when the content of polyester in the blend was increased, the rate of composting and the weight loss due to composting increased. When the CAP was highly substituted, little degradation was observed within 30 days and almost all of the weight loss was ascribed to loss of polyester. Although the polyester was still observed to degrade faster, when the CAP DS was below approximately 2.0, both components are observed to degrade. The data suggests that initial degradation of the polyester is by chemical hydrolysis and the rate of this hydrolysis is very dependent upon the temperature profile of the compost and upon the DS of the CAP.  相似文献

This report presents results of a review of available methods for control of environmental hazards applied to indoor air pollutants. Indoor air pollution originates from transport of ambient outdoor air contaminants into occupied spaces by natural infiltration ventilation, or by mechanical ventilation using outdoor makeup air, plus contributions from indoor emission sources. When air exchange with the external ambient environment is reduced to conserve energy, contributions from indoor emission sources may dominate indoor air pollutant levels. This paper identifies alternative methods available to control indoor air pollutant exposures. The performance characteristics of ventilation systems and of air cleaning devices used in mixed modes for ventilation of occupied spaces are described. Models for predicting effectiveness of several alternative modes are reviewed, with field trial validation results cited where available. Results of previous confined-space studies are briefly reviewed as points of departure for consideration of necessary air quality, ventilation, and air cleaning. Understanding of indoor air contaminant generation and controls is aided by examination of earlier studies of indoor air quality, using modern perspectives on occupational environmental health and hygiene.  相似文献

We report our experience of 14 preimplantation genetic diagnosis (PGD) cycles in eight couples carrying five different single gene disorders, during the last 18 months. Diagnoses were performed for myotonic dystrophy (DM), cystic fibrosis (CF) [ΔF508 and exon 4 (621+1 G>T)], fragile X and CF simultaneously, and two disorders for which PGD had not been previously attempted, namely neurofibromatosis type 2 (NF2) and Crouzon syndrome. Diagnoses for single gene disorders were carried out on ideally two blastomeres biopsied from Day 3 embryos. A highly polymorphic marker was included in each diagnosis to control against contamination. For the dominant disorders, where possible, linked polymorphisms provided an additional means of determining the genotype of the embryo hence reducing the risk of misdiagnosis due to allele dropout (ADO). Multiplex fluorescent polymerase chain reaction (F-PCR) was used in all cases, followed by fragment analysis and/or single-stranded conformation polymorphism (SSCP) for genotyping. Embryo transfer was performed in 13 cycles resulting in one biochemical pregnancy for CF, three normal deliveries (a twin and a singleton) and one early miscarriage for DM and a singleton for Crouzon syndrome. In each case the untransferred embryos were used to confirm the diagnoses performed on the biopsied cells. The results were concordant in all cases. The inclusion of a polymorphic marker allowed the detection of extraneous DNA contamination in two cells from one case. Knowing the genotype of the contaminating DNA allowed its origin to be traced. All five pregnancies were obtained from embryos in which two blastomeres were biopsied for the diagnosis. Our data demonstrate the successful strategy of using multiplex PCR to simultaneously amplify the mutation site and a polymorphic locus, fluorescent PCR technology to achieve greater sensitivity, and two-cell biopsy to increase the efficiency and success of diagnoses. ABSTRACT: Sequential water reuse by various users, i.e., municipal, industrial, agricultural, is common practice in all river systems. In the arid and semiarid western United States where streams are not large and water demands are high relative to the virgin water supply, the practice causes a reduction in stream flow and an increase in stream salinity. This paper outlines a quick and easy application of the materials balance principle to approximate the effects of increasing the level of sequential reuse on residual stream flow and salinity.  相似文献
