Although information is limited, it is evident that prolonged persistence of infectious Hepatitis A virus (HAV) is a factor in the transmission of the virus via fresh produce. Consequently, data on persistence of the virus on produce, and in environments relevant to production, such as soils, water and surfaces, are required to fully understand the dynamics of transmission of HAV via foods. Furthermore, information on effective disinfection procedures is necessary to implement effective post-harvest control measures. This review summarises current information on HAV persistence in fresh produce and on relevant disinfection procedures. On vegetables, HAV can remain infectious for several days; on frozen berries, it can persist for several months. HAV can remain infectious on surfaces for months, depending on temperature and relative humidity, and can survive desiccation. It can survive for several hours on hands. Washing hands can remove the virus, but further data are required on the appropriate procedure. Chlorination is effective in water, but not when HAV is associated with foodstuffs. Bleach and other sodium hypochlorite disinfectants at high concentrations can reduce HAV on surfaces, but are not suitable for use on fresh produce. There is only limited information on the effects of heating regimes used in the food industry on HAV. HAV is resistant to mild pasteurisation. Some food components, e.g. fats and sugars, can increase the virus’ resistance to higher temperatures. HAV is completely eliminated by boiling. Quantitative prevalence data are needed to allow the setting of appropriate disinfection log reduction targets for fresh produce.  相似文献

Nutrient sufficiency of zooxanthellae in the sea anemone Aiptasia pallida cultured in low nutrient seawater depends on the availability of particulate food to the host. Zooxanthellae in anemones unfed for 20 to 30 d exhibited the following characteristics of nutrient deficiency: cell division rates decreased; chlorophyll a content gradually decreased from 2 to <1 pg cell–1; and C:N ratios increased from 7.5 to 16. Over a 3-mo period, algal populations in unfed anemones gradually decreased, indicating that zooxanthellae were lost faster than they were replaced by division. The mitotic index of zooxanthellae in unfed anemones was stimulated either by feeding the host or by the addition of inorganic N and P to the medium. Whether algae are nutrient-limited in hosts under field conditions has not been examined fully; however, C:N ratios in zooxanthellae from field-collected hosts are slightly higher (9.4 vs 7.5) than in hosts fed to repletion in laboratory cultures. This study examined the associations between dissolved organic matter(DOM) characteristics and potential nitrification occurrence in the presence of chloramine along a drinking water distribution system. High-performance size exclusion chromatography(HPSEC) coupled with a multiple wavelength detector(200–280 nm) was employed to characterise DOM by molecular weight distribution, bacterial activity was analysed using flow cytometry, and a package of simple analytical tools, such as dissolved organic carbon, absorbance at 254 nm, nitrate,nitrite, ammonia and total disinfectant residual were also applied and their applicability to indicate water quality changes in distribution systems were also evaluated. Results showed that multi-wavelength HPSEC analysis was useful to provide information about DOM character while changes in molecule weight profiles at wavelengths less than 230 nm were also able to be related to other water quality parameters. Correct selection of the UV wavelengths can be an important factor for providing appropriate indicators associated with different DOM compositions. DOM molecular weight in the range of 0.2–0.5 k Da measured at210 nm correlated positively with oxidised nitrogen concentration(r = 0.99), and the concentrations of active bacterial cells in the distribution system(r = 0.85). Our study also showed that the changes of DOM character and bacterial cells were significant in those sampling points that had decreases in total disinfectant residual. HPSEC-UV measured at210 nm and flow cytometry can detect the changes of low molecular weight of DOM and bacterial levels, respectively, when nitrification occurred within the chloraminated distribution system.  相似文献

Due to the difficulties in testing for specific pathogens, water samples are tested for the presence of nonpathogenic indicator organisms to determine whether a water supply has been contaminated by fecal material. An implicit assumption in this approach is that where pathogenic microorganisms are present fecal indicator organisms are present as well; yet surprisingly few studies have been conducted that directly compare the transport of indicator organisms with pathogenic organisms in ground water environments. In this study we compared the cell properties and transport of Escherichia coli, a commonly used indicator organism, and Campylobacter jejuni, an important enteropathogen commonly found in agricultural wastes, through saturated porous media. Differences in cell properties were determined by measuring cell geometry, hydrophobicity, and electrophoretic mobility. Transport differences were determined by conducting miscible displacement experiments in laboratory columns. Under the experimental conditions tested, C. jejuni was much more negatively charged and more hydrophobic than E. coli. In addition, C. jejuni cells were slightly longer, narrower, and less spherical than E. coli. The variations in cell properties, primarily surface charge, resulted in significant differences in transport between these two microorganisms, with the transport of C. jejuni exceeding that of E. coli when conditions favored low attachment rates, thus calling into question the usefulness of using E. coli as an indicator organism for this important pathogen.  相似文献

The availability of solid food (Artemia nauplii) and dissolved inorganic nutrients (ammonium, nitrate, phosphate) to the shallow-water marine hydroid Myrionema amboinense was manipulated for 1-8 days in order to investigate their role in the growth of intracellular symbiotic dinoflagellates (zooxanthellae) of the genus Symbiodinium. Symbionts from hydroids collected from the field or maintained under laboratory conditions (25°C, 12 h:12 h light:dark cycle, 80 µE m-2 s-1 fluorescent lighting) always exhibited a single peak in mitotic index (MI) at dawn. Symbionts in freshly collected field animals had an MI peak of about 15%. Symbiotic dinoflagellates in hydroids fed Artemia nauplii twice daily in the laboratory maintained this dawn peak of MI between 10% and 15%, but in the absence of feeding or added inorganic nutrients, this peak declined to less than 1% within 2-4 days. In contrast, when hydroids were placed in solutions containing ammonium (20 µM NH4Cl), nitrate (10 µM NaNO3), and a combination of ammonium and phosphate (2 µM Na2HPO4) immediately after collection, the algal MI remained between 5% and 15% for 4-7 days; the addition of 2 µM phosphate did not increase MI relative to unfed rates. When unfed animals were placed in dissolved nitrogen or fed Artemia, the symbiont MI increased from <1% to 10-17% within 2-3 days; P alone had no effect. However, the increase resulting from added inorganic nutrients was temporary, lasting only 5-7 days. These observations suggest that algal division in the host is maintained indefinitely in the field or by feeding particulate foods twice daily in the laboratory, but the addition of inorganic nutrients alone (ammonium, nitrate and ammonium/phosphate) appeared to support the completion of a maximum of one additional round of cell division. Nutrients required for continued growth and division of symbiotic dinoflagellates are linked to host feeding and host growth; without external food, neither host nor symbiont continue to grow. The same phenomenon is seen in zooxanthellate anemones, clams and corals, where total numbers of symbionts appear to be linked to changes in host-tissue biomass (protein), achieving relatively stable densities in M. amboinense, corals and other cnidarian symbioses, depending on their local environmental conditions. The results of the present study help explain the cellular responses of algal symbionts in reef-dwelling invertebrates to additions of dissolved inorganic nutrients to coral-reef ecosystems.  相似文献

Management effectiveness evaluation has been recognized as an important mechanism for both reporting on and improving protected area management. The Convention on Biological Diversity’s program of work on protected areas calls on all countries to implement such systems. In 2004, the first whole of system assessment of park management effectiveness, based on the IUCN-WCPA Management Effectiveness Evaluation Framework, was undertaken in New South Wales, Australia as part of a State of the Parks reporting requirement. This article describes the development of the State of the Parks assessment tool, its elements, and how it addresses the management effectiveness difficulties associated with assessments conducted across an extensive and diverse range of park types. The importance of engaging staff, at all levels, throughout the process is highlighted, as well as the adjustments made to the assessment tool based on staff feedback. While some results are presented, the main purpose of the article is to identify and discuss important procedural and methodological considerations. These include balancing quantitative and qualitative assessment approaches, achieving a comprehensive understanding of the management processes, and responding to any problems associated with assessments.  相似文献

Abstract

Commercially prepared ceralure‐B1, an attractant for the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), was evaluated in trapping tests by comparison with commercial ceralure and trimedlure. Jackson traps were used for medfly captures. The residual ceralure‐B1, ceralure, and trimedlure contents of aged dispensers were determined. The results of this study show that male medfly captures from traps with ceralure‐B1 were as high or higher than male medfly captures from traps with twice the quantity of ceralure or trimedlure. The use of regioselectively synthesized ceralure‐B1, enriched in B1, as a source of a more potent attractant than commercial ceralure and as a more persistent male medfly attractant than trimedlure is reported.  相似文献

Abstract

For the first time, velocities were measured inside a fieldscale rotary kiln incinerator. Combustion gas velocities and temperatures were measured at multiple points across a quadrant of the kiln near its exit using a bidirectional pressure probe and suction pyrometer. To accommodate the new bidirectional probe and gain access to the upper portion of the kiln, a lighter and stiffer positioning boom was designed. The kiln was directly fired using natural gas in a steady state mode. Results indicate strong vertical stratification of both velocity and temperature, with the highest values corresponding to the top of the kiln. Access restraints prevented the lower region of the kiln from being mapped. Horizontal variations in both temperature and velocity were insignificant. Operating conditions were varied by adjusting the amount of ambient air added to the front of the kiln. Increasing the flow of ambient air into the front of the kiln reduced the measured temperatures as expected, but did hot have as significant an effect on measured velocities. The quality of the results is examined by performing mass balances across the incinerator and by comparison to an existing numerical model. Both methods indicate that the experimental results are reasonable.  相似文献

Visual Determination of the Opacity of Emissions from Stationary Sources (Method 9) is a reference method established by U.S. Environmental Protection Agency (EPA) to quantify plume opacity. However, Method 9 relies on observations from humans, which introduces subjectivity. In addition, it is expensive to teach and certify personnel to evaluate plume opacity on a semiannual basis. In this study, field tests were completed during a "smoke school" and a 4-month monitoring program of plumes emitted from stationary sources with a Method 9 qualified observer to evaluate the use of digital photography and two computer algorithms as an alternative to Method 9. This Digital Optical Method (DOM) improves objectivity, costs less to implement than Method 9, and provides archival photographic records of the plumes. Results from "smoke school" tests indicate that DOM passed six of eight tests when the sun was located in the 140 degrees sector behind one of the three cameras, with the individual opacity errors of 15% or less and average opacity errors of 7.5% or less. DOM also passed seven of the eight tests when the sun was located in the 216 degrees sector behind another camera. However, DOM passed only one of the eight tests when the sun was located in the 116 degrees sector in front of the third camera. Certification to read plume opacity by a "smoke reader" for 6 months requires that the "smoke reader" pass one of the smoke school tests during smoke school. The average opacity errors and percentage of observations with individual opacity errors above 15% for the results obtained with DOM were lower than those obtained by the smoke school trainees with the sun was located behind the camera, whereas they were higher than the smoke school trainee results with the sun located in front of the camera. In addition, the difference between plume opacity values obtained by DOM and a Method 9 qualified observer, as measured in the field for two industrial sources, were 2.2%. These encouraging results demonstrate that DOM is able to meet Method 9 requirements under a wide variety of field conditions and, therefore, has potential to be used as an alternative to Method 9.  相似文献
