Abstract

Investigations have shown that the physical state of the DNA strongly influences the pattern of methylation observed when DNA or a substance containing DNA is treated with the fumigant methyl bromide. 1‐Methyl‐adenine and 7‐methylguanine were identified, after hydrolysis, as the major methylated bases of DNA which had been treated in the solid state. 3‐Methylcytosine, 3‐methyladenine and 7‐methyladenine were found as minor products. The overall methylation pattern was similar to that observed earlier for DNA of maize and wheat which had been fumigated. In contrast, when buffer solutions of DNA were treated with methyl bromide, the N‐7 position of guanine was the major site and the N‐3 position of adenine was the second most important site of methylation. This result corresponds to that previously observed in similar studies with buffer solutions of DNA and other methylating agents.  相似文献

The U.S. Environmental Protection Agency (EPA), in cooperation with the Toronto Harbour Commissioners (THC), conducted a Superfund Innovative Technology Evaluation (SITE) demonstration of the THC Soil Recycle Treatment Train. The treatment train consists of three technologies operated in sequence: a soil wash process, a metals removal process, and a biological treatment process. The THC conducted an extensive demonstration of the treatment train at a 55 tons per day pilot plant in order to evaluate an approach for remediation of industrial/commercial sites that are situated in the Toronto Port Industrial District (PID). Three soils were processed during the THC demonstration. The EPA SITE demonstration project examined, in detail, soil processing from one of the sites being evaluated as part of the overall THC project. Contaminants included organic compounds and heavy metals. It has been estimated by THC that as much as 2,200,000 tons of soil from locations within the PID may require some form of treatment due to heavy metal and/or organic contamination that resulted from various industrial processing operations. The objective of the SITE demonstration was to evaluate the technical effectiveness of the process in relation to THC’s target criteria.

Gravel and sand that met the THC target criteria for medium to fine soil suitable for industrial/commercial sites was produced. The fine soil from the biological treatment process did not meet the target level of 2.4 ppm for benzo(a)pyrene. However, there was a significant reduction in polynuclear aromatic hydrocarbon (PAH) compounds. The metals removal process achieved reductions of greater than seventy percent for copper, lead, nickel, and zinc.  相似文献

Copper (Cu) metabolism is altered in rats fed diets high in molybdenum (Mo) and low in Cu. This 10-week study was carried out to examine the effects of supplemental Mo (7.5–240 μg/g diet) on male Sprague–Dawley rats fed diets adequate in Cu (5 μg/g diet) and to determine the susceptibility of Mo-treated animals to the environmental pollutant 3,3′,4,4′-tetrabromobiphenyl (TBB). After 7 weeks of dietary treatment, half of the rats in each group received a single IP injection of TBB (150 μM/kg bw), while the other half received the corn oil vehicle. Rats sacrificed at 10 weeks showed no effects of Mo on growth, feed efficiency, or selected organ or tissue weights. Dose-dependent effects on plasma Mo (0–5.1 μg/mL), plasma Cu (0.95–0.20 μg/mL), and bone Cu (3.4–10 μg/g) in control through the high dose were found. Cu sequestration in the bone of Mo-treated rats is a new finding. TBB treatment resulted in dramatic weight loss and loss of absolute organ mass. Relative organ weights were increased, except for the thymus. TBB altered the concentrations of certain amino acids. Compared to control rats, this polybrominated biphenyl congener significantly decreased plasma Cu and ceruloplasmin at higher concentrations of dietary Mo and promoted the process of plasma Cu decrease by Mo, suggesting a combined effect.  相似文献

A novel microbial source tracking (MST) method based on the detection of human and non-human markers was developed and applied to track the origin of fecal pollution in water systems. Mitochondrial DNA sequences were used to develop new quantitative real-time polymerase chain reaction (qPCR) assays for dog, poultry, and gull. The targets were included as part of a toolbox including human, cow, pig, and sheep assays. A primer and probe set for the detection of the human-specific nifH gene of Methanobrevibacter smithii was also designed as an indicator of human fecal contamination. The assays were tested for specificity and applied to fecal-spiked surface waters and environmental samples collected from two river catchments impacted by sources of human and non-human fecal contamination. The MST methods described were applicable to both spiked waters and environmental samples, and using the two approaches the origin of fecal pollution could be successfully determined in mixed source fecally polluted waters.  相似文献

Near surface disposal facility design and management are examined and compared using a systems approach that defines facility performance as a function of three components (or subsystems): the disposal facility design (cover systems and bottom liners); the properties of the waste (waste composition, waste form and waste package); and the site‐specific environmental features (climate, geology, and hydrology). We report an evaluation of five DOE near surface disposal facility case studies, selected to provide a “representative” sample that included disposal sites with a range of waste and environmental characteristics across the DOE. The facilities selected were the Savannah River E‐Area Engineered Trenches, Hanford Integrated Disposal Facility, Idaho Radioactive Waste Management Complex, Oak Ridge Environmental Management Waste Management Facility, and Nevada National Security Site Area 5. ©2015 Wiley Periodicals, Inc.  相似文献

Extra-pair copulations create a potential for sexual conflict in pair-bonding birds. Here we report an experimental study of the bluethroat, Luscinia s. svecica, in which the throat ornament of males was blackened with Nyanzol D in order to reduce their sexual attractiveness and thus increase the sexual conflict over fertilizations. In an earlier study, we showed that males blackened before pairing had a lower success in attracting social mates than controls, whereas males blackened after pairing guarded their mates more intensely and sang less than controls. Here we add behavioural data from one more year on males blackened after pairing and corroborate our previous finding that the manipulation caused males to guard their mates more intensely and advertise less for additional mates. Blackened males did not suffer more intrusions from neighbouring males than did controls. Paternity analyses of the combined data set, using multilocus DNA fingerprinting and microsatellite typing, revealed that blackened males lost significantly more paternity than controls. There was also a tendency for blackened males to show a lower success in achieving extra-pair fertilizations. These results indicate that females have the upper hand in the sexual conflict over fertilizations, as females paired with unattractive males can achieve more extra-pair paternity despite the greater constraint posed by the intensified mate guarding. Still, within the blackened group, there were some indications that males guarding more intensely and singing less had higher paternity than males guarding less and singing more, suggesting a marginal positive effect of guarding for unattractive males. BACKGROUND, AIM, AND SCOPE: The subsurface at the Oak Ridge Field Research Center represents an extreme and diverse geochemical environment that places different stresses on the endogenous microbial communities, including low pH, elevated nitrate concentrations, and the occurrence of heavy metals and radionuclides, including hexavalent uranium [U(VI)]. The in situ immobilization of U(VI) in the aquifer can be achieved through microbial reduction to relatively insoluble U(IV). However, a high redox potential due to the presence of nitrate and the toxicity of heavy metals will impede this process. Our aim is to test biostimulation of the endogenous microbial communities to improve nitrate reduction and subsequent U(VI) reduction under conditions of elevated heavy metals. MATERIALS AND METHODS: Column experiments were used to test the possibility of using biostimulation via the addition of ethanol as a carbon source to improve nitrate reduction in the presence of elevated aqueous nickel. We subsequently analyzed the composition of the microbial communities that became established and their potential for U(VI) reduction and its in situ immobilization. RESULTS: Phylogenetic analysis revealed that the microbial population changed from heavy metal sensitive members of the actinobacteria, alpha- and gamma-proteobacteria to a community dominated by heavy metal resistant (nickel, cadmium, zinc, and cobalt resistant), nitrate reducing beta- and gamma-proteobacteria, and sulfate reducing Clostridiaceae. Coincidentally, synchrotron X-ray absorption spectroscopy analyses indicated that the resulting redox conditions favored U(VI) reduction transformation to insoluble U(IV) species associated with soil minerals and biomass. DISCUSSION: This study shows that the necessary genetic information to adapt to the implemented nickel stress resides in the endogenous microbial population present at the Oak Ridge FRC site, which changed from a community generally found under oligotrophic conditions to a community able to withstand the stress imposed by heavy metals, while efficiently reducing nitrate as electron donor. Once nitrate was reduced efficient reduction and in situ immobilization of uranium was observed. CONCLUSIONS: This study provides evidence that stimulating the metabolism of the endogenous bacterial population at the Oak Ridge FRC site by adding ethanol, a suitable carbon source, results in efficient nitrate reduction under conditions of elevated nickel, and a decrease of the redox potential such that sulfate and iron reducing bacteria are able to thrive and create conditions favorable for the reduction and in situ immobilization of uranium. Since we have found that the remediation potential resides within the endogenous microbial community, we believe it will be feasible to conduct field tests. RECOMMENDATIONS AND PERSPECTIVES: Biostimulation of endogenous bacteria provides an efficient tool for the successful in situ remediation of mixed-waste sites, particularly those co-contaminated with heavy metals, nitrate and radionuclides, as found in the United States and other countries as environmental legacies of the nuclear age.  相似文献
