Fecal bacteria have traditionally been used as indicator organisms to monitor the quality of recreational waters. Recent work has questioned the robustness of traditional indicators, particularly at seawater bathing beaches. For example, a study of Florida beaches found unexpectedly high abundances of Escherichia coli, fecal coliforms, and enterococci in beach sand. The aim of the present study was to explain these abundances by assessing the survival of E. coli and enterococci in beach sand relative to seawater. We used a combination of quantitative laboratory mesocosm experiments and field observations. Results suggested that E. coli and enterococci exhibited increased survivability and growth in sand relative to seawater. Because fecal bacteria are capable of replicating in sand, at least under controlled laboratory conditions, the results suggest that sand may be an important reservoir of metabolically active fecal organisms. Experiments with "natural" mesocosms (i.e., unsterilized sand or water rich in micropredators and native bacteria) failed to show the same increases in fecal indicators as was found in sterile sand. It is postulated that this was due to predation and competition with indigenous bacteria in these "natural" systems. Nonetheless, high populations of indicators were maintained and recovered from sand over the duration of the experiment as opposed to the die-off noted in water. Indicator bacteria may wash out of sand into shoreline waters during weather and tidal events, thereby decreasing the effectiveness of these indicators as predictors of health risk and complicating the interpretations for water quality managers.  相似文献

Polychlorinated dibenzo-p-dioxins (PCDDs) are toxic and widespread persistent organic pollutants (POPs). Cost-effective technologies for destroying or detoxifying PCDDs are in high demand. The overall purpose of this study was to develop a zero-valent zinc based technology for transforming toxic PCDDs to less- or non-toxic forms. We measured the dechlorination rates of 1,2,3,4-tetrachlorodibenzo-p-dioxin (1,2,3,4-TCDD) in the presence of zero-valent zinc under aqueous conditions, identified the daughter compounds of the reaction, and constructed possible pathways for the reactions. The reaction rates of daughter compounds with zero-valent zinc were also measured independently. Our results showed that the zero-valent zinc is a suitable candidate for reducing PCDDs. Reductive dechlorination of 1,2,3,4-TCDD was stepwise and complete to dibenzo-p-dioxin (DD) mainly via 1,2,4-trichlorodibenzo-p-dioxin (1,2,4-TrCDD), 1,3-dichlorodibenzo-p-dioxin (1,3-DCDD), 1-chlorodibenzo-p-dioxin (1-MCDD) to DD and via 1,2,4-TrCDD, 2,3-dichlorodibenzo-p-dioxin (2,3-DCDD), 2-chlorodibenzo-p-dioxin (2-MCDD) to DD. In each separate system, the observed half-lives of 1,2,3,4-TCDD, 1,2,3-TrCDD, 1,2,4-TrCDD, 1,2-DCDD, 1,3-DCDD, 1,4-DCDD and 2,3-DCDD are 0.56, 2.62, 5.71, 24.93, 41.53, 93.67 and 169.06 h respectively. The tendency of rate constant follows TCDD>TrCDD>DCDD. Our results suggest that zero-valent zinc is a suitable candidate for rapidly reducing highly chlorinated PCDDs to less or non-chlorinated daughter products.  相似文献

A series of lab and greenhouse studies were undertaken to understand how Cu and Zn toxicity influences Bromus carinatus (Hook and Arn.) growth, to what degree an organic amendment (yard waste compost) may reduce Cu and Zn bioavailability in Cu-Zn minespoil and promote plant growth in combination with fertilizer, and how the vertical distribution of compost in the minespoil influences rooting depth. Root Cu and Zn toxicity thresholds were determined to be 1 mgL(-1) and 10 mgL(-1) in solution, respectively. The compost amendment had exceptionally high Cu and Zn binding capacities (0.17 and 0.08 g metal g C(-1), for Cu and Zn, respectively) that were attributed to high compost humic and fulvic acid concentrations. Maximum plant biomass was achieved when minespoil was amended with compost and fertilizer in combination. Fertilizer alone had no effect on plant growth. Mixing compost into the minespoil was essential to promote adequate rooting depth.  相似文献

Visibility impairment in the Columbia River Gorge National Scenic Area is an area of concern. A field study conducted from July 2003 to February 2005 was followed by data analysis and receptor modeling to better understand the temporal and spatial patterns of haze and the sources contributing to the haze in the Columbia River Gorge in the states of Washington and Oregon. The nephelometer light scattering and surface meteorological data at eight sites along the gorge showed five distinct wind patterns, each with its characteristic diurnal and spatial patterns in light scattering by particles (bsp). In summer, winds were nearly always from west to east (upgorge) and showed decreasing bsp with distance into the gorge and a pronounced effect of the Portland, OR, metropolitan area on haze, especially in the western portions of the gorge. Winter often had winds from the east with very high levels of bsp, especially at the eastern gorge sites, with sources east of the gorge responsible for much of the haze. The major chemical components responsible for haze were organic carbon, sulfate, and nitrate. Positive matrix factorization (PMF) using chemically speciated Interagency Monitoring of Protected Visual Environments data indicated seven source factors in the western gorge and five factors in the eastern gorge. Organic mass is a large contributor to haze in the gorge in all seasons, with a peak in fall. The PMF analysis suggests that approximately half of the organic mass is biomass smoke, with mobile sources as the second largest contributor. PMF analysis showed nitrates (important in fall and winter) mainly attributed to a generic secondary nitrate factor, with the next largest contributor being oil combustion at Mt. Zion, WA and mobile sources at Wishram, WA. Sulfate is a significant contributor in all seasons, with peak sulfate concentrations in summer.  相似文献

Waterbirds are known to disperse invertebrate propagules that survive gut passage, but there is very little information about how the probability of dispersal changes at different times of the annual cycle when birds move in different directions, or how it is affected by changes in diet. We studied internal transport of brine shrimp Artemia cysts by migratory waders in the Odiel saltworks in south-west Spain. Viable cysts of parthenogenetic Artemia were abundant in the faeces and regurgitated pellets of redshank Tringa totanus, pellets of spotted redshank T. erythropus, and faeces of black-tailed godwit Limosa limosa during spring and/or autumn migrations in 2001–2002, but were not recorded during winter. Godwits did not produce pellets, and spotted redshank faeces were not sampled. Significant correlations between the number of cysts in a pellet or faecal sample and the proportion of that sample constituted by Artemia adults suggested that most cysts were ingested while in the ovisacs of gravid females. The proportion of cysts destroyed during digestion increased when accompanied by harder food items or grit, and when fewer cysts were ingested. The median number of intact cysts was higher in redshank faeces than in their pellets, but cysts extracted from pellets were more likely to hatch. A higher proportion of redshank pellets contained Artemia cysts in spring than in autumn, but more redshank migrated through the area in autumn. Significantly fewer cysts were recorded in redshank pellets in winter than in spring or autumn. Our results confirm that there is potential for long-distance dispersal of Artemia cysts via waders during both northwards (spring) and southwards (autumn) migrations.  相似文献

We recently demonstrated whole genome sequencing of a human fetus using only parental DNA samples and plasma from the pregnant mother. This proof-of-concept study demonstrated how samples obtained noninvasively in the first or second trimester can be analyzed to yield a highly accurate and substantially complete genetic profile of the fetus, including both inherited and de novo variation. Here, we revisit our original study from a clinical standpoint, provide an overview of the scientific approach, and describe opportunities and challenges along the path toward clinical adoption of noninvasive fetal whole genome sequencing. © 2013 John Wiley & Sons, Ltd.  相似文献
