INTENTION, GOAL, SCOPE, BACKGROUND: Cyanobacteria have the natural ability to degrade moderate amounts of organic pollutants. However, when pollutant concentration exceeds the level of tolerance, bleaching of the cells and death occur within 24 hours. Under stress conditions, cyanobacterial response includes the short-term adaptation of the photosynthetic apparatus to light quality, named state transitions. Moreover, prolonged stresses produce changes in the functional organization of phycobilisomes and in the core-complexes of both photosystems, which can result in large changes in the PS II fluorescence yield. The localization of ferredoxin-NADP+ reductase (FNR) at the ends of some peripheral rods of the cyanobacterial phycobilisomes, makes this protein a useful marker to check phycobilisome integrity. OBJECTIVE: The goal of this work is to improve the knowledge of the mechanism of action of a very potent pesticide, lindane (gamma-hexaclorociclohexane), in the cyanobacterium Anabaena sp., which can be considered a potential candidate for bioremediation of pesticides. We have studied the effect of lindane on the photosynthetic apparatus of Anabaena using fluorescence induction studies. As ferredoxin-NADP+ reductase plays a key role in the response to oxidative stress in several systems, changes in synthesis, degradation and activity of FNR were analyzed. Immunolocalization of this enzyme was used as a marker of phycobilisome integrity. The knowledge of the changes caused by lindane in the photosynthetic apparatus is essential for rational further design of genetically-modified cyanobacteria with improved biorremediation abilities. METHODS: Polyphasic chlorophyll a fluorescence rise measurements (OJIP) have been used to evaluate the vitality and stress adaptation of the nitrogen-fixing cyanobacterium Anabaena PCC 7119 in the presence of increasing concentrations of lindane. Effects of the pesticide on the ultrastructure have been investigated by electron microscopy, and FNR has been used as a marker of phycobilisome integrity. RESULTS AND DISCUSSION: Cultures of Anabaena sp. treated with moderate amounts of lindane showed a decrease in growth rate followed by a recovery after 72 hours of pesticide treatment. Concentrations of lindane below 5 ppm increased the photosynthetic performance and activity of the cells. Higher amounts of pesticide caused a decrease in these activities which seems to be due to a non-competitive inhibition of PS II. Active PS II units are converted into non-QA reducing, so called heat sink centers. Specific activity and amount of FNR in lindane-treated cells were similar to the values measured in control cultures. Release of FNR from the thylakoid after 48 hours of exposure to 5 ppm of lindane towards the cytoplasm was detected by immunogold labeling and electron microscopy. CONCLUSIONS: From these results, we conclude that the photosynthetic performance and activity of the cells are slightly increased in the presence of lindane up to 5 ppm. Moreover, in those conditions, lindane did not produce significant changes in the synthesis, degradation or activity of FNR. The high capability of Anabaena to tolerate lindane makes this cyanobacterium a good candidate for phytoremediation of polluted areas. RECOMMENDATION AND OUTLOOK: The results of this study show that cultures of Anabaena PCC 7119 tolerate lindane up to 5 ppm, without significant changes in the photosynthetic vitality index of the cells. However, a slight increase in phycobiliprotein synthesis is observed, which is related to total protein content. This change might be due to degradation of proteins less stable than phycobiliproteins. An identification of the proteins with altered expression pattern in the presence of the pesticide remains the subject of further work and will provide valuable information for the preparation of strains which are highly tolerant to lindane.  相似文献

Several collocated semicontinuous instruments measuring particulate matter with particle sizes < or =2.5 microm (PM2.5) sulfate (SO4(2-)) and nitrate (NO3-) were intercompared during two intensive field campaigns as part of the PM2.5 Technology Assessment and Characterization Study. The summer 2001 urban campaign in Queens, NY, and the summer 2002 rural campaign in upstate New York (Whiteface Mountain) hosted an operation of an Aerosol Mass Spectrometer, Ambient Particulate Sulfate and Nitrate Monitors, a Continuous Ambient Sulfate Monitor, and a Particle-Into-Liquid Sampler with Ion Chromatographs (PILS-IC). These instruments provided near real-time particulate SO4(2-) and NO3- mass concentration data, allowing the study of particulate SO4(2-)/NO3- diurnal patterns and detection of short-term events. Typical particulate SO4(2-) concentrations were comparable at both sites (ranging from 0 to 20 microg/m3), while ambient urban particulate NO3- concentrations ranged from 0 to 11 microg/m3 and rural NO3- concentration was typically less than 1 microg/m3. Results of the intercomparisons of the semicontinuous measurements are presented, as are results of the comparisons between the semicontinuous and time-integrated filter-based measurements. The comparisons at both sites, in most cases, indicated similar performance characteristics. In addition, charge balance calculations, based on major soluble ionic components of atmospheric aerosol from the PILS-IC and the filter measurements, indicated slightly acidic aerosol at both locations.  相似文献

In recent years, ambient measurements of hourly ozone precursor concentrations, namely speciated and total nonmethane organic compounds (NMOCs), have become available through the Photochemical Assessment Monitoring Stations (PAMS) program. Prior to this, NMOCs were measured in the central business district using a canister to obtain the 3-hr integrated sample for the 6:00 a.m.-9:00 a.m. period. Such sampling had been carried out annually for nearly a decade at three locations in the New York City metropolitan area. The intent of these measurements, along with measurements of the other ozone precursor, NO(x), was to provide an understanding of ozone formation and the emissions loading and mix in the urban area. The analysis of NMOC and NO(x) measurements shows a downward trend in the case of NMOC. In addition, we compared the canister-based NMOC concentrations with data obtained from the PAMS program for the 6:00 a.m.-9:00 a.m. period. Analysis of the NMOC concentrations reveals poor spatial correlation between the various monitors, reflecting the effect of localized emissions. This suggests that NMOC measurements made at a single location cannot be viewed as representative of the entire region. We investigate whether long-term exposure to heavy metals, including immunosuppressive metals like mercury (Hg), is associated with infectious disease in a wild cetacean. Post-mortem investigations on 86 harbour porpoises, Phocoena phocoena, found dead along the coasts of England and Wales revealed that 49 of the porpoises were healthy when they died as a consequence of physical trauma (most frequently entrapment in fishing gear). In contrast, 37 porpoises died of infectious diseases caused by parasitic, bacterial, fungal and viral pathogens (most frequently pneumonia caused by lungworm and bacterial infections). We found that mean liver concentrations of Hg, selenium (Se), the Hg:Se molar ratio, and zinc (Zn) were significantly higher in the propoises that died of infectious disease compared to healthy porpoises that died from physical trauma. Liver concentrations of lead (Pb), cadmium (Cd), copper (Cu) and chromium (Cr) did not differ between the two groups. Hg, Se, and the Hg:Se molar ratio were also positively correlated with age. The association between Zn concentration and disease status may result from Zn redistribution in response to infection. Further work is required to evaluate whether chronic exposure to Hg may have presented a toxic challenge to the porpoises that succumbed to infectious disease.  相似文献

Sorbent materials consisting of organoclay immobilized onto the surface of a solid support were evaluated for use in pentachlorophenol (PCP) and polycyclic aromatic hydrocarbon (PAH) remediation of groundwater at a creosote-contaminated Superfund site. Cetylpyridinium-exchanged low pH montmorillonite clay (CP-LPHM) was bonded to either sand (CP-LPHM/sand) or granular activated carbon (GAC) (CP-LPHM/GAC) using the free acid form of carboxymethylcellulose as an adhesive. Effluent from an oil-water separator was eluted through equal bed volumes of composite (4 g 3:2 CP-LPHM/GAC or 13 g CP-LPHM/sand), affinity-extracted, and quantitatively analyzed by GC/MS. PCP, naphthalene, fluorene, phenanthrene, pyrene, and total PAHs were initially reduced by both CP-LPHM/GAC (> or =99%, 61%, 99%, > or =99%, 97%, and 94%, respectively) and CP-LPHM/sand (90%, 70%, 94%, 95%, 93%, and 86%, respectively). Complete breakthrough of naphthalene occurred after approximately 15 h of elution through 3:2 CP-LPHM/GAC and 22 h through CP-LPHM/sand. PCP showed complete breakthrough following 18 h of elution through 3:2 CP-LPHM/GAC and 26 h through CP-LPHM/sand. However, 50% breakthrough was not attained for higher molecular weight PAHs, as fluoranthene, pyrene, benzo[a]anthracene, and chrysene continued to be greatly reduced with both 3:2 CP-LPHM/GAC (98%, 95%, 94%, and 95%, respectively) and CP-LPHM/sand (75%, 73%, 76%, and 78%, respectively) after 48 h of continuous elution. Results confirm prior studies, indicating that these organoclay-containing composites have a high capacity for contaminants found in wood preserving waste. Further, results suggest that the inclusion of CP-LPHM may be useful as part of an effective strategy for groundwater remediation of high concentrations of PCP and PAHs, in particular high molecular weight and carcinogenic PAHs.  相似文献

We have determined the congener compositions of nine commercial Aroclor products of polychlorinated biphenyls (PCBs) to the sub-part-per-million level using high-resolution gas chromatography combined with high-resolution mass spectrometry according to US Environmental Protection Agency (EPA) Method 1668A. These Aroclor composition data should allow improved characterization and risk assessment of PCB contamination at hazardous waste sites, particularly for dioxin-like PCB congeners. By combining the data on the concentrations of each dioxin-like congener with its World Health Organization toxicity equivalency factor, we have established dioxin toxic equivalent concentrations for each pure Aroclor product.  相似文献

The U.S. Environmental Protection Agency in 1997 revised the 1-hr ozone (O3) National Ambient Air Quality Standard (NAAQS) to one based on an 8-hr average, resulting in potential nonattainment status for substantial portions of the eastern United States. The regulatory process provides for the development of a state implementation plan that includes a demonstration that the projected future O3 concentrations will be at or below the NAAQS based on photochemical modeling and analytical techniques. In this study, four photochemical modeling systems, based on two photochemical models, Community Model for Air Quality and the Comprehensive Air Quality Model with extensions, and two emissions processing models, Sparse Matrix Optimization Kernel for Emissions and Emissions Modeling System, were applied to the eastern United States, with emphasis on the northeastern Ozone Transport Region in terms of their response to oxides of nitrogen and volatile organic carbon-focused controls on the estimated design values. With the 8-hr O3 NAAQS set as a bright-line test, it was found that a given area could be termed as being in or out of attainment of the NAAQS depending upon the modeling system. This suggests the need to provide an estimate of model-to-model uncertainty in the relative reduction factor (RRF) for a better understanding of the uncertainty in projecting the status of an area's attainment. We investigated the relative roles of natural factors and shoreline harvest leading to recent declines of the black leather chiton (Katharina tunicata) on the outer Kenai Peninsula, Alaska (U.S.A.). This intertidal mollusk is a strongly interacting grazer and a culturally important subsistence fishery for Sugpiaq (Chugach Alutiiq) natives. We took multiple approaches to determine causes of decline. Field surveys examined the significant predictors of Katharina density and biomass across 11 sites varying in harvest pressure, and an integrated analysis of archaeological faunal remains, historical records, traditional ecological knowledge, and contemporary subsistence invertebrate landings examined changes in subsistence practices through time. Strong evidence suggests that current spatial variation in Katharina density and biomass is driven by both human exploitation and sea otter (Enhydra lutris) predation. Traditional knowledge, calibrated by subsistence harvest data, further revealed that several benthic marine invertebrates (sea urchin, crab, clams, and cockles) have declined serially beginning in the 1960s, with reduced densities and sizes of Katharina being the most recent. The timing of these declines was coincident with changes in human behavior (from semi-nomadic to increasingly permanent settlement patterns, improved extractive technologies, regional commercial crustacean exploitation, the erosion of culturally based season and size restrictions) and with the reestablishment of sea otters. We propose that a spatial concentration in shoreline collection pressure through time, increased harvest efficiency, and the serial depletion of alternative marine invertebrate prey have led to intensified per capita predator impacts on Katharina and thus its recent localized decline.  相似文献

Continuous cropping in the absence of external nutrient inputs to soils has led to the expression of poorly productive patches in farmers’ fields of western Kenya. Farmers attempting to correct these conditions are often confused by the spatial and symptomatic irregularity of affected plants and, until recently, no soil management product was commercially available that is specifically formulated to restore soil fertility to these patches. PREP-PAC consists of 2.0 kg of Minjingu rock phosphate (RP), 200 g of urea, seeds of various symbiotic nitrogen-fixing food legumes, rhizobial inoculant, gum arabic seed adhesive, lime for seed pelleting, and instructions for the use of these materials. It is intended for addition to 25 m2 and produced at a cost of $ 0.56 per unit. The general principle is to apply slowly available RP sufficient for several cropping seasons with readily available nitrogenous fertilizer and to intercrop farmer’s maize (Zea mays) with a legume that provides residual fixed-nitrogen and organic inputs to the soil. This approach was tested in on-farm experiments conducted in collaboration with several grassroots rural development organizations. An experiment examined interactions between PREP-PAC components in a maize–soybean (Glycine max) intercrop in nutrient-depleted soils with sandy and clayey surface horizons. The treatments included ±RP, ±urea, and ±inoculants arranged as a 23 factorial with four replicates at each location. Total value of the intercrops ranged between $ 0.83 in the unamended plots and $ 2.44 in plots treated with PREP-PAC. Significant positive effects were observed with the addition of RP (P<0.001), urea (P=0.04), and inoculant (P=0.01) and in interactions between RP and urea (P=0.02) or inoculant (P=0.07). The return ratio to PREP-PAC investment was 2.6 in the sandy soil and 3.7 in the clay. PREP-PACs were tested on-farm in 52 symptomatic patches containing maize–bean intercrops with and without an improved variety of climbing Phaseolus vulgaris cv. Flora. Unamended patches (25 m2=0.0025 ha) produced 1.6 kg maize and 0.08 kg bean. With addition of PREP-PAC containing Flora, yields increased to 4.1 kg maize and 1.1 kg bean (P<0.001 for both crops). Improvement in bean yield during the first cropping season nearly offset PREP-PAC’s investment costs. PREP-PAC is a strategic approach because all of its ingredients, except for urea, originate from East Africa, and are relatively inexpensive; the product is intended for distribution through existing retail and development networks.  相似文献
