Organic solvents, such as dimethylsulfoxide (DMSO) and methanol are widely used as vehicles to solubilise lipophilic test compounds in toxicity testing. However, the effects of such solvents upon innate detoxification processes in aquatic organisms are poorly understood. This study assessed the effect of solvent exposure upon cytochrome P450 (CYP)-mediated xenobiotic metabolism in Daphnia magna and zebrafish larvae (4 d post fertilisation). Adult D. magna were demonstrated to have a low, but detectable, metabolism of ethoxyresorufin in vivo and this activity was not modulated by pre-exposure to DMSO or methanol (24 h, up to 0.1% and 0.05% v/v, respectively). In contrast, the metabolism of ethoxyresorufin in zebrafish larvae was significantly reduced by both solvents (0.1% and 0.05% v/v, respectively) after 24 h of exposure. In zebrafish, these observed decreases in activity towards ethoxyresorufin were accompanied by decreased expression of a variety of genes coding for drug metabolising enzymes (corresponding to CYP1, CYP2, CYP3 and UDP-glucuronyl transferase [UGT] family enzymes), measured by quantitative PCR. Reduction of gene expression and CYP1 enzyme activities by methanol (0.05% v/v) in zebrafish larvae was partially reversed by co-exposure with Aroclor 1254 (100 μg L−1). Overall this study suggests that relatively low concentrations of organic solvents can impact upon the biotransformation of certain xenobiotics in zebrafish larvae, and that this warrants consideration when assessing compounds for metabolism and toxicity in this species.  相似文献

At the former nuclear weapon production site in Hanford, WA, caustic radioactive tank waste leaks into subsurface sediments and causes dissolution of quartz and aluminosilicate minerals, and precipitation of sodalite and cancrinite. This work examines changes in pore structure due to these reactions in a previously-conducted column experiment. The column was sectioned and 2D images of the pore space were generated using backscattered electron microscopy and energy dispersive X-ray spectroscopy. A pre-precipitation scenario was created by digitally removing mineral matter identified as secondary precipitates. Porosity, determined by segmenting the images to distinguish pore space from mineral matter, was up to 0.11 less after reaction. Erosion-dilation analysis was used to compute pore and throat size distributions. Images with precipitation had more small and fewer large pores. Precipitation decreased throat sizes and the abundance of large throats. These findings agree with previous findings based on 3D X-ray CMT imaging, observing decreased porosity, clogging of small throats, and little change in large throats. However, 2D imaging found an increase in small pores, mainly in intragranular regions or below the resolution of the 3D images. Also, an increase in large pores observed via 3D imaging was not observed in the 2D analysis. Changes in flow conducting throats that are the key permeability-controlling features were observed in both methods.  相似文献

Due to worldwide restrictions on polybrominated diphenyl ethers (PBDEs), the demand for alternative flame retardants (AFRs), such as organophosphate flame retardants (OPFRs), novel brominated FRs (NBFRs) and hexabromocyclododecanes (HBCDs), has recently increased. Little is known about human exposure to NBFRs and OPFRs and that their levels in dust have been scarcely evaluated worldwide. To increase the knowledge regarding these chemicals, we measured concentrations of five major NBFRs, ten OPFRs and three HBCD isomers in indoor dust from New Zealand homes. Dust samples were taken from living room floors (n=34) and from mattresses of the same houses (n=16). Concentrations (ngg(-1)) of NBFRs were: 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE) (<2-175), decabromodiphenyl ethane (DBDPE) (<5-1430), 2-ethylhexyl-2,3,4,5-tetrabromobenzoate (TBB) (<2-2285) and bis(2-ethylhexyl)-3,4,5,6-tetrabromophthalate (TBPH) (<2-640). For OPFRs, concentrations (ngg(-1)) ranged between: tri-ethyl-phosphate (TEP) (<10-235), tri-n-butyl-phosphate (TnBP) (<20-7545), tris-(2-chloroethyl)-phosphate (TCEP) (<20-7605), tris-(1-chloro-2-propyl) phosphate (TCPP) (20-7615), tri-(2-butoxyethyl)-phosphate (TBEP) (50-27325), tris-(2,3-dichloropropyl)-phosphate (TDCPP) (20-16560), tri-phenyl-phosphate (TPhP) (20-35190), and tri-cresyl-phosphate (TCP) (<50-3760). HBCD concentrations fell in the range <2-4100ngg(-1). BTBPE, DBDPE, TBPH, TBEP, and TnBP showed significant positive correlation (p<0.05) between their concentrations in mattresses and the corresponding floor dust (n=16). These data were used to derive a range of plausible exposure scenarios. Although the estimated exposure is well below the corresponding reference doses (RfDs), caution is needed given the likely future increase in use of these FRs and the currently unknown contribution to human exposure by other pathways such as inhalation and diet.  相似文献

Contaminants in settled indoor dust are potentially health hazardous to human. Thus, identification and quantification of toxic chemicals in settled indoor dust is of great concern. In this study, the levels of major anions ( $ \mathrm{C}{{\mathrm{l}}^{-}},\mathrm{N}{{\mathrm{O}}_2}^{-},\mathrm{B}{{\mathrm{r}}^{-}},\mathrm{N}{{\mathrm{O}}_3}^{-},\mathrm{P}{{\mathrm{O}}_4}^{3-}\,\mathrm{and}\,\mathrm{S}{{\mathrm{O}}_4}^{2- } $ ), trace metals (Cr, Mn, Fe, Co, Ni, Cu, Zn, Cd, As and Pb) and polybromodiphenyl ethers (PBDEs) in settled office and home dust were determined and correlations between the contaminants investigated. Depending on the available materials in both microenvironments, the most possible sources were identified. The results showed that the settled office dusts (n?=?6 pooled samples from 85 offices) were more contaminated than home dusts (n?=?8 homes). For anions, $ \mathrm{S}{{\mathrm{O}}_4}^{2- } $ and Cl accounted for 87 and 97 % of the total office and home dust contaminants, respectively. For trace metals, Fe, Cu, Zn and Mn, accounted for 98 % of the contaminants in both office and home dust samples. Fe exhibited the highest percentage of 76.7 and 87.3 % in office and home dust samples, respectively. For PBDEs, the mean concentrations detected in office and home dust ranged between 5.8–86.3 and 1.5–20.6 ng?g?1, respectively. The log-transformed correlation between the total concentrations of trace metals and major anions detected in offices and homes was positive for offices and negative for homes with a statistically significant values (r?=?0.73, p?<?0.01; r?= ?0.22, p?<?0.01, respectively). The daily exposure rates determined for the most hazardous such as As, Cd, Pb and PBDEs congeners, relative to the individual concentrations reported in the literature in settled indoor dust, were found very lower. Therefore, maybe it is possible to expect less potential health risk. Investigation of formation of coordination compounds between trace metals and PBDEs congeners is possible; however, this requires further study.  相似文献

The compositional changes of saturates, aromatics, resins and asphaltenes (SARA) fractions in aqueous clay/oil microcosm experiments with a hydrocarbon-degrading microorganism community were analysed using Iatroscan. The clay mineral samples used in this study were organomontmorillonite, acid-activated montmorillonite and K, Ca, Zn and Cr montmorillonites produced by modifying the original montmorillonite sample. The evaluation and quantification of biodegradation and adsorption were carried out using a combination of the Iatroscan and gravimetric analysis. The SARA compositions in the presence of organomontmorillonite and acid-activated montmorillonite after incubation follow the same pattern in which the aromatic fraction is higher than the other fractions unlike in the presence of unmodified, K, Ca and Zn montmorillonites, where the saturates fraction is higher than the other fractions. Changes in SARA fractions due to biodegradation seemed to occur most in the presence of unmodified and calcium montmorillonites; hence, the removal of SARA fractions due to biodegradation was significant and enhanced in the presence of these two clay samples. However, biodegradation in the presence of organomontmorillonite and acid-activated and Cr montmorillonites was hindered. The study indicated that Cr montmorillonite adsorbed resins most, whereas Zn and K montmorillonites adsorbed aromatics most after incubation.  相似文献

Invasive species, range-expanding species, genetically modified organisms (GMOs), synthetic organisms, and emerging pathogens increasingly affect the human environment. We propose a framework that allows comparison of consecutive stages that such novel organisms go through. The framework provides a common terminology for novel organisms, facilitating knowledge exchange among researchers, managers, and policy makers that work on, or have to make effective decisions about, novel organisms. The framework also indicates that knowledge about the causes and consequences of stage transitions for the better studied novel organisms, such as invasive species, can be transferred to more poorly studied ones, such as GMOs and emerging pathogens. Finally, the framework advances understanding of how climate change can affect the establishment, spread, and impacts of novel organisms, and how biodiversity affects, and is affected by, novel organisms.  相似文献

Bone metabolism is a tightly regulated process that controls bone remodeling and repair in addition to maintaining circulating calcium and phosphate levels. It has been shown that certain organohalogen contaminants may adversely impact bone tissue metabolism and structure in wildlife species. However, exceedingly few studies have addressed the bone-related effects of organohalogen exposure in birds. The objective of the present study was to investigate the associations between markers of bone metabolism and structural integrity, and concentrations of established and current-use halogenated flame retardants (FRs) in ring-billed gulls (Larus delawarensis) nesting in a known FR hotspot area in the St. Lawrence River (Montreal, Canada). Bone metabolism was assessed using plasma calcium and inorganic phosphate levels, and alkaline phophatase activity, while bone (tarsus; trabecular and cortical sections) structure quality was examined using the percentage of bone tissue comprised in the total bone volume (Bv/Tv) and bone mineral density (BMD). Bv/Tv and BMD of the tarsus tended (not significant) to be positively associated with circulating calcium levels in male ring-billed gulls. Moreover, concentrations of FRs in male bird liver (brominated diphenyl ether (BDE)-154, -183, -201, and -209) and plasma (BDE-209) were negatively correlated with trabecular and cortical BMD of the tarsus. These correlative associations may suggest light demineralization of bone tissue associated with FR exposure in male ring-billed gulls. Present findings provide some evidence that bone (tarsus) metabolism and mineral composition may be impacted in high FR-exposed (mainly to PBDEs) ring-billed gulls breeding in the highly urbanized Montreal region.  相似文献

While snowflakes fell and Christmas carols heralded the imminent holiday, over 3000 delegates to the Third National Conference on Air Pollution were told to “Control Now for Clean Air.”

They were told this in many ways by many people—by the Vice President of the United States; by the Secretary of the Department of Health, Education, and Welfare who summoned the Conference; and by senators, congressmen, governors, and mayors.

Then eight panel sessions convened to discuss and to determine how to control now for clean air. Speakers from every section of American life addressed themselves to this subject; when they were finished, the eight session chairmen summarized what had been said, what had been, learned. Many species of conservation concern are spatially structured and require dispersal to be persistent. For such species, altering the distribution of suitable habitats on the landscape can affect population dynamics in ways that are difficult to predict from simple models. We argue that for such species, individual-based and spatially explicit population models (SEPMs) should be used to determine appropriate levels of off-site restoration to compensate for on-site loss of ecologic resources. Such approaches are necessary when interactions between biologic processes occur at different spatial scales (i.e., local [recruitment] and landscape [migration]). The sites of restoration and habitat loss may be linked to each other, but, more importantly, they may be linked to other resources in the landscape by regional biologic processes, primarily migration. The common management approach for determining appropriate levels of off-site restoration is to derive mitigation ratios based on best professional judgment or pre-existing data. Mitigation ratios assume that the ecologic benefits at the site of restoration are independent of the ecologic costs at the site of habitat loss. Using an SEPM for endangered red-cockaded woodpeckers, we show that the spatial configuration of habitat restoration can simultaneously influence both the rate of recruitment within breeding groups and the rate of migration among groups, implying that simple mitigation ratios may be inadequate.  相似文献

The levels and distribution of polynuclear aromatic hydrocarbons (PAHs) were determined in soil samples from background locations in the UK and Norway, to investigate their spatial distribution and the controlling environmental factors. Concentrations ranged between 42 and 11200 microg kg(-1) (geometric mean 640 microg kg(-1)) and 8.6 and 1050 microg kg(-1) (150 microg kg(-1)) dry weight in the UK and Norwegian soil, respectively. Proximity to sources and locations susceptible to high atmospheric depositional inputs resulted in higher concentrations. Statistically significant relationships were observed between PAH and total organic carbon (TOC) in the Norwegian samples. High molecular weight PAHs correlated with black carbon (BC) in UK-woodland soil. These observations support the hypothesis that TOC plays an important role in the retention of PAHs in soil and that PAHs are often combined with BC during combustion emissions. PAHs with 4 and more rings comprised approximately 90% of total PAHs in the UK soil, but only 50% in the Norwegian soil. The mixture of PAHs implied that fractionation occurred during long-range atmospheric transport and deposition. The lighter PAHs with lower K(ow) values more readily reached the most remote sites. The heavier PAHs with higher K(ow) values remained in closer proximity to sources.  相似文献
