Even though petroleum-degrading microorganisms are widely distributed in soil and water, they may not be present in sufficient numbers to achieve contaminant remediation. In such cases, it may be useful to inoculate the polluted area with highly effective petroleum-degrading microbial strains to augment the exiting ones. In order to identify a microbial strain for bioaugmentation of oil-contaminated soil, we isolated a microbial strain with high emulsification and petroleum hydrocarbon degradation efficiency of diesel fuel in culture. The efficacy of the isolated microbial strain, identified as Candida catenulata CM1, was further evaluated during composting of a mixture containing 23% food waste and 77% diesel-contaminated soil including 2% (w/w) diesel. After 13 days of composting, 84% of the initial petroleum hydrocarbon was degraded in composting mixes containing a powdered form of CM1 (CM1-solid), compared with 48% of removal ratio in control reactor without inoculum. This finding suggests that CM1 is a viable microbial strain for bioremediation of oil-contaminated soil with food waste through composting processes. 相似文献

Polychlorodibenzothiophenes (PCDTs) are sulfur analogues of polychlorodibenzofurans (PCDFs) and have been detected in environmental samples. We used density functional theory calculations (Gaussian 98W) to predict the photodechlorination pathways of octachlorodibenzothiophene (OCDT) and octachlorodibenzofuran (OCDF) in hexane, and we compared the predicted pathways with those observed during UV irradiation experiments. OCDT and OCDF were observed to degrade through first-order dechlorination processes, and the rate constant for OCDT was less than one-third that for OCDF. The main experimental photodechlorination pathways of OCDT and OCDF led to hexachlorinated and tetrachlorinated congeners, respectively; that is, the photodechlorination pathway of OCDT differed from that of OCDF. On the assumption that the dechlorination mechanisms involved radical reactions, we used DFT calculations to estimate bond-dissociation energies and single-point energies of OCDT and OCDF and their dechlorinated congeners, and we used the resulting information, along with hypotheses regarding the rate-controlling step of the degradations, to predict theoretical degradation pathways. We propose that reaction of dechlorinated radicals with a hydrogen donor was the rate-controlling step for OCDT and that C-Cl bond dissociation by UV light was the rate-controlling step for OCDF. 相似文献

This study aims at understanding flood risks and their impact on a community, in order to enhance communities’ resilience and adaptive capacity to these threats. It also investigates the possibility of looking at and handling risk from a resilience point of view. Therefore, while a conventional risk management process is employed in this study, social, physical, economic, and institutional dimensions of resilience are also included in order to grasp the extent of risks and the ways in which communities face, cope with, and recover from flooding. Findings showed that there was no significant difference in the perception of flood risk among household heads educated up to secondary school level, suggesting that they believe floods are purely natural events. Those with a higher level of education (high school and above) (82.7 % of respondents) were aware that flood disasters are the result of hazard and vulnerability combined. In addition, social dynamics were apparently strengthened by such disasters, which resulted in cohesion and mutual help following floods in some wards. Also, households with more sources of income and more savings appear to recover faster than others after a flooding event. With regard to governance and networks, greater efforts have to be made by local institutions to ensure basic functioning during and after disaster events and to invest more into risk reduction activities. However, further studies need to be conducted to clarify the understanding of the impact flood disasters have on the environment and community lives and livelihoods in general, as traditional coping strategies, although still practical, no longer suffice in the face of changes in climate and environment. 相似文献

Six products were formed by reaction of ethynylestradiol (EE2) with sodium hypochlorite in buffered solutions. 4-Chloroethynylestradiol (4-ClEE2) and 2,4-dichloroethynylestradiol (2,4-diClEE2) were identified as the two major reaction products, using preparative HPLC, MS, and NMR. When EE2 reacted with chlorine at different pHs (pH 5, 7, and 9) or chlorine concentrations (0.2, 1, 2, and 5 mmol/l, corresponding to molar ratios to EE2, 1, 5, 10, and 25, respectively), the formation of 4-ClEE2 and 2,4-diClEE2 was observed under the above conditions, and the highest yields were 20 and 52 mol%, respectively. EE2 was consumed almost completely within 5 min of chlorination by addition of chlorine of more than 1 mmol/l (molar ratio to EE2, 5). On the other hand, the two products existed in highly chlorinated solutions after 60 min (4ClEE2, 1-6 mol%; 2,4-diClEE2, 3-25 mol%). The estrogenic activities of 4-ClEE2 by estrogen receptor alpha or beta binding assay were similar to those of the parent EE2, and the activities of 2,4-diClEE2 were lower about 10 times. 相似文献

A model to estimate the entry length to a fully developed diffusive boundary layer above a sediment bed, such as those found in lakes, reservoirs, rivers, and estuaries, is presented. The model is used to determine how the length of a sediment bed in mass-transfer experiments influences the measured vertical diffusive flux at the sediment-water interface. A nondimensional local mass flux is introduced in the form of a Sherwood number (Sh) and expressed as a function of both the distance from the leading edge of the sediment bed (x) and the Schmidt number (Sc). Similarly, a mean Sherwood number (Sh(ave)) for a sediment bed of length (L) is introduced. The diffusive boundary layer grows with distance, and its thickness depends on the Schmidt number (i.e., the diffusive boundary layer gets thicker and develops more quickly as the Schmidt number decreases). For Schmidt numbers greater than or equal to 100, the diffusive boundary layer begins to develop slowly but is fully developed when the nondimensional horizontal coordinate (x+) is approximately 1000. The Sherwood number is largest (i.e., infinity) near the leading edge of the sediment bed (i.e., at x = 0), decreases as the distance from the bed increases, and, finally, approaches a constant value for a fully developed diffusive boundary layer (Sh(infinity)). In this paper, the distance to a fully developed diffusive boundary layer (L99) and the required length of a sediment bed are related explicitly to Sc, sheer velocity (U*), and the relative errors of local or average Sherwood numbers (Sh or Sh(ave), respectively) against the Sherwood number for the fully developed diffusive boundary layer (Sh(infinity)). The lengths L99 and L decrease as the Schmidt number increases and become independent of the Schmidt number when Sc is greater than 1000. A longer sediment bed is needed when the shear velocity or the Schmidt number is small (e.g., L99 and L approximately 1.0 m and 8.0 m, respectively, for Sc = 500, U* = 0.1 cm/s, and a 3% acceptable error). Experimental studies may not be able to meet these requirements and an adjustment of measured mass-transfer rates at a sediment-water interface may be necessary. Background, Goals and Scope In response to concerns that have been raised about chemical substances that may alter the function of endocrine systems and
result in adverse effects on human health, an OECD initiative was undertaken to develop and validate in vitro and in vivo assays to identify chemicals that may interfere with endocrine systems of vertebrates. Here we report on studies that were
conducted to develop and standardize a cell-based screening assay using the H295R cell line to prioritize chemicals that may
act on steroidogenic processes in humans and wildlife. These studies are currently ongoing as part of the ‘Special Activity
on the Testing and Assessment of Endocrine Disruptors’ within the OECD Test Guidelines Program to review, develop, standardize,
and validate a number of in vitro and in vivo toxicological assays for testing and assessment of chemicals concerning their potential to interact with the endocrine system
of vertebrates.
Study Design Six laboratories from five countries participated in the pre-validation studies. Each laboratory tested the effects of three
model chemicals on the production of testosterone (T) and estradiol (E2) using the H295R Steroidogenesis Assay. Chemicals
tested were well described inducers or inhibitors of steroidogenic pathways (forskolin, prochloraz and fadrozole). All experiments
were conducted in 24 well plates following standard protocols. Six different doses per compound were analyzed in triplicate
per plate. A quality control (QC) plate was run in conjunction with the chemical exposure plate to account for inter-assay
variation. Each chemical exposure was conducted two or three times.
Results All laboratories successfully detected increases and/or decreases in hormone production by H295R cells after exposure to the
different model compounds and there was good agreement in the pattern of response for all groups. Forskolin increased both
T and E2 while fadrozole and prochloraz decreased production of both hormones. All chemicals affected hormone production in
a dose-dependent manner with the exception of fadrozole which caused maximum inhibition of E2 at the two least concentrations
tested. Some inter-laboratory differences were noted in the alteration of hormone production measured in chemically exposed
cells. However, with the exception of the production of T measured at one laboratory in cells exposed to forskolin, the EC50s calculated were comparable (coefficients of variation 34–49%) for all hormones.
Discussion and Perspectives The results indicated that the H295R Steroidogenesis Assay protocol was robust, transferable and reproducible among all laboratories.
However, in several instances that were primarily related to one laboratory there were unexplained minor uncertainties related
to the inter-laboratory hormone production variation. Based on the findings from this Phase 2 prevalidation study, the H295R
Steroidogenesis Assay protocol is currently being refined. The next phase of the OECD validation program will test the refined
protocol among the same group of laboratories using an extended set of chemicals (∼30) that will include positive and negative
chemical controls as well as a broad spectrum of different potential inducers and inhibitors of steroidogenic pathways.

We investigated the decolorization of Orange II with and without the addition of co-substrates and nutrients under an anaerobic sequencing batch reactor (ASBR). The increase in COD concentrations from 900 to 1750 to 3730 mg/L in the system treating 100 mg/L of Orange II-containing wastewater enhanced color removal from 27% to 81% to 89%, respectively. In the absence of co-substrates and nutrients, more than 95% of decolorization was achieved by the acclimatized anaerobic microbes in the bioreactor treating 600 mg/L of Orange II. The decrease in mixed liquor suspended solids concentration by endogenous lysis of biomass preserved a high reducing environment in the ASBR, which was important for the reduction of the Orange II azo bond that caused decolorization. The maximum decolorization rate in the ASBR was approximately 0.17 g/hr in the absence of co-substrates and nutrients. 相似文献
