The present study was to evaluate heavy metal toxicity stress symptoms in fish blood during long-term exposure of sublethal concentration of copper sulphate (CuSO4). The effects of copper on various heamatological parameters were evaluated exposing Indian fresh water fish, Channa punctatus to a sub-lethal concentration of copper sulphate (0.36 mg l(-1)) for different periods. Exposure of fish to copper showed a significant decrease in the haemoglobin (Hb) content from 10.73 to 6.60%, red blood cells (RBC) from 2.86 to 1.84 x 10(6)/mm3 and packed cell volume (PCV) from 31.00 to 23.33% at the end of 45th day as compared to control. Whereas the white blood cells (WBC) increased from 60.00 to 92.48 x10(3)/mm3, clotting time (CT) from 27.66 to 43.00 second, erythrocyte sedimentation rate from 5.0 to 13.66 mm/hr and mean corpuscular volume from 108.11 to 126.85 cmu, significantly, with increase in exposure periods. Although mean corpuscular haemoglobin concentration (MCHC) and mean corpuscular haemoglobin (MCH) values showed significant increase during 15 and 30 days exposures, both the values were found significantly increased at the end of the experimental period. The differential leucocytes count (DLC) showed a significant increase in the populations of lymphocytes and eosinophils, whereas neutrophils, monocytes and basophiles were found decreased after exposure to copper.  相似文献

Bacillus megaterium F-8 exhibited an intracellular acetamide hydrolyzing activity (AHA) when cultivated in modified nutrient broth with 3 % tryptone, 1.5 % yeast extract, and 0.5 % sodium chloride, at pH 7.2, 45 °C for 24 h. Maximum AHA was recorded in the culture containing 0.1 M of sodium phosphate buffer, (pH 7.5) at 45 °C for 20 min with 0.2 % of acetonitrile and resting cells of B. megaterium F-8 equivalent to 0.2 ml culture broth. This activity was stable up to 55 °C and was completely inactivated at or above 60 °C. Maximum acyl transferase activity (ATA) was recorded in the reaction medium containing 0.1 M of potassium phosphate buffer, (pH 8.0) at 55 °C for 5 min with 0.85 mM of acetamide as acyl donor and hydroxylamine hydrochloride as acyl acceptor and resting cells of B. megaterium F-8 equivalent to 0.94 mg cells (dry weight basis). This activity was stable up to 60 °C and a rapid decline in enzyme activity was recorded above it. Under the optimized conditions, this organism hydrolyzed various nitriles and amides such as propionitrile, propionamide, caprolactam, acetamide, and acrylamide to corresponding acids. Acyl group transfer capability of this organism was used for the production of acetohydroxamic acid. ATA of B. megaterium F-8 showed broad substrate specificity such as for acetamide followed by propionamide, acrylamide, and lactamide. This amide hydrolyzing and amidotransferase activity of B. megaterium F-8 has potential applications in enzymatic synthesis of hydroxamic acids and bioremediation of nitriles and amides contaminated soil and water system.  相似文献

Plants have to counteract unavoidable stress-caused anomalies such as oxidative stress to sustain their lives and serve heterotrophic organisms including humans. Among major enzymatic antioxidants, catalase (CAT; EC 1.11.1.6) and ascorbate peroxidase (APX; EC 1.11.1.11) are representative heme enzymes meant for metabolizing stress-provoked reactive oxygen species (ROS; such as H2O2) and controlling their potential impacts on cellular metabolism and functions. CAT mainly occurs in peroxisomes and catalyzes the dismutation reaction without requiring any reductant; whereas, APX has a higher affinity for H2O2 and utilizes ascorbate (AsA) as specific electron donor for the reduction of H2O2 into H2O in organelles including chloroplasts, cytosol, mitochondria, and peroxisomes. Literature is extensive on the glutathione-associated H2O2-metabolizing systems in plants. However, discussion is meager or scattered in the literature available on the biochemical and genomic characterization as well as techniques for the assays of CAT and APX and their modulation in plants under abiotic stresses. This paper aims (a) to introduce oxidative stress-causative factors and highlights their relationship with abiotic stresses in plants; (b) to overview structure, occurrence, and significance of CAT and APX in plants; (c) to summarize the principles of current technologies used to assay CAT and APX in plants; (d) to appraise available literature on the modulation of CAT and APX in plants under major abiotic stresses; and finally, (e) to consider a brief cross-talk on the CAT and APX, and this also highlights the aspects unexplored so far.  相似文献

A majority of small-scale renewable energy sources including the solar PV modules, fuel cells gives out output voltage in the range of around 15–40 V DC. This needs to be stepped up to suit load requirements using a high voltage gain converter. Since renewable sources inherently generate sudden variations in input voltage, a good output voltage profile even during such random variations in input conditions is essential. This paper presents modeling of a high step-up converter configuration with closed loop control. The converter topology is designed to operate with moderate duty ratios and the simple coupled inductor. The converter is capable of high step-up and can find application in solar PV systems. The controller response is good with low steady state error and required dynamics. The modeling and simulation is carried out using MATLAB/Simulink software package.  相似文献

We examine the dynamics and spatial determinants of land change in India by integrating decadal land cover maps (1985–1995–2005) from a wall-to-wall analysis of Landsat images with spatiotemporal socioeconomic database for ~630,000 villages in India. We reinforce our results through collective evidence from synthesis of 102 case studies that incorporate field knowledge of the causes of land change in India. We focus on cropland–fallow land conversions, and forest area changes (excludes non-forest tree categories including commercial plantations). We show that cropland to fallow conversions are prominently associated with lack of irrigation and capital, male agricultural labor shortage, and fragmentation of land holdings. We find gross forest loss is substantial and increased from ~23,810 km2 (1985–1995) to ~25,770 km2 (1995–2005). The gross forest gain also increased from ~6000 km2 (1985–1995) to ~7440 km2 (1995–2005). Overall, India experienced a net decline in forest by ~18,000 km2 (gross loss–gross gain) consistently during both decades. We show that the major source of forest loss was cropland expansion in areas of low cropland productivity (due to soil degradation and lack of irrigation), followed by industrial development and mining/quarrying activities, and excessive economic dependence of villages on forest resources.

An experimental study has been carried out to analyze the effect of inclined ribs used as roughness element on heat transfer along its friction characteristics. The roughness parameters include relative roughness pitch (P/e) ranges from 4–16, relative roughness height (e/Dh) ranges from 0.021–0.043 and angle of attack (α) ranges from 30° to 75°. The Reynolds number (Re) lies in the range of 4500–28000. The heat transfer and friction factor data obtained from the experiment and it is compared with the data obtained from smooth duct under the same conditions. Considerable augmentation was observed in heat transfer and friction over the smooth duct. Correlation was also developed for Nusselt number (Nu) and friction factor (f) as a function of roughness and flow parameter.  相似文献

The present study investigated the comprehensive chemical composition [organic carbon (OC), elemental carbon (EC), water-soluble inorganic ionic components (WSICs), and major & trace elements] of particulate matter (PM2.5) and scrutinized their emission sources for urban region of Delhi. The 135 PM2.5 samples were collected from January 2013 to December 2014 and analyzed for chemical constituents for source apportionment study. The average concentration of PM2.5 was recorded as 121.9 ± 93.2 μg m?3 (range 25.1–429.8 μg m?3), whereas the total concentration of trace elements (Na, Ca, Mg, Al, S, Cl, K, Cr, Si, Ti, As, Br, Pb, Fe, Zn, and Mn) was accounted for ~17% of PM2.5. Strong seasonal variation was observed in PM2.5 mass concentration and its chemical composition with maxima during winter and minima during monsoon seasons. The chemical composition of the PM2.5 was reconstructed using IMPROVE equation, which was observed to be in good agreement with the gravimetric mass. Source apportionment of PM2.5 was carried out using the following three different receptor models: principal component analysis with absolute principal component scores (PCA/APCS), which identified five major sources; UNMIX which identified four major sources; and positive matrix factorization (PMF), which explored seven major sources. The applied models were able to identify the major sources contributing to the PM2.5 and re-confirmed that secondary aerosols (SAs), soil/road dust (SD), vehicular emissions (VEs), biomass burning (BB), fossil fuel combustion (FFC), and industrial emission (IE) were dominant contributors to PM2.5 in Delhi. The influences of local and regional sources were also explored using 5-day backward air mass trajectory analysis, cluster analysis, and potential source contribution function (PSCF). Cluster and PSCF results indicated that local as well as long-transported PM2.5 from the north-west India and Pakistan were mostly pertinent.  相似文献

The continuous rise in the cost of fossil fuels as well as in environmental pollution has attracted research in the area of clean alternative fuels for improving the performance and emissions of internal combustion (IC) engines. In the present work, n-butanol is treated as a bio-fuel and investigations have been made to evaluate the feasibility of replacing diesel with a suitable n-butanol-diesel blend. In the current research, an experimental investigation was carried out on a variable compression ratio CI engine with n-butanol-diesel blends (10–25% by volume) to determine the optimum blending ratio and optimum operating parameters of the engine for reduced emissions. The best results of performance and emissions were observed for 20% n-butanol-diesel blend (B20) at a higher compression ratio as compared to diesel while keeping the other parameters unchanged. The observed deterioration in engine performance was within tolerable limits. The reductions in smoke, nitrogen oxides (NO x ), and carbon monoxide (CO) were observed up to 56.52, 17.19, and 30.43%, respectively, for B20 in comparison to diesel at rated power. However, carbon dioxide (CO2) and hydrocarbons (HC) were found to be higher by 17.58 and 15.78%, respectively, for B20. It is concluded that n-butanol-diesel blend would be a potential fuel to control emissions from diesel engines.

Human populations are using oceans as their household dustbins, and microplastic is one of the components which are not only polluting shorelines but also freshwater bodies globally. Microplastics are generally referred to particles with a size lower than 5 mm. These microplastics are tiny plastic granules and used as scrubbers in cosmetics, hand cleansers, air-blasting. These contaminants are omnipresent within almost all marine environments at present. The durability of plastics makes it highly resistant to degradation and through indiscriminate disposal they enter in the aquatic environment. Today, it is an issue of increasing scientific concern because these microparticles due to their small size are easily accessible to a wide range of aquatic organisms and ultimately transferred along food web. The chronic biological effects in marine organisms results due to accumulation of microplastics in their cells and tissues. The potential hazardous effects on humans by alternate ingestion of microparticles can cause alteration in chromosomes which lead to infertility, obesity, and cancer. Because of the recent threat of microplastics to marine biota as well as on human health, it is important to control excessive use of plastic additives and to introduce certain legislations and policies to regulate the sources of plastic litter. By setup various plastic recycling process or promoting plastic awareness programmes through different social and information media, we will be able to clean our sea dustbin in future.  相似文献

The article reports on the first year of the Blackstone Project, a joint pilot project of the Massachusetts Department of Environmental Protection (DEP) and the Department of Environmental Management (DEM). The project is being used to test the best approach to promoting toxics use reduction (similar to ”source reduction“ as defined by EPA). In the project, the two state agencies are testing different methods of inspecting compliance across media (i.e. compliance to air, water, hazardous waste, and Right-to-Know requirements), biasing enforcement actions towards toxics use reduction, and coordinating regulatory activity with technical assistance activity. The Blackstone Project Team finds that the approach tested in the Blackstone Project was successful in its setting. A wide range of environmental protection objectives was cost effectively met through the Blackstone approach. In FY91, DEP will test the approach in the other DEP Regional Offices, with more facilities of a wider variety.  相似文献
