Miscible-displacement experiments were conducted to examine the impact of microbial lag and bacterial cell growth on the transport of salicylate, a model hydrocarbon compound. The impacts of these processes were examined separately, as well as jointly, to determine their relative effects on biodegradation dynamics. For each experiment, a column was packed with porous medium that was first inoculated with bacteria that contained the NAH plasmid encoding genes for the degradation of naphthalene and salicylate, and then subjected to a step input of salicylate solution. The transport behavior of salicylate was non-steady for all cases examined, and was clearly influenced by a delay (lag) in the onset of biodegradation. This microbial lag, which was consistent with the results of batch experiments, is attributed to the induction and synthesis of the enzymes required for biodegradation of salicylate. The effect of microbial lag on salicylate transport was eliminated by exposing the column to two successive pulses of salicylate, thereby allowing the cells to acclimate to the carbon source during the first pulse. Elimination of microbial lag effects allowed the impact of bacterial growth on salicylate transport to be quantified, which was accomplished by determining a cell mass balance. Conversely, the impact of microbial lag was further investigated by performing a similar double-pulse experiment under no-growth conditions. Significant cell elution was observed and quantified for all conditions/systems. The results of these experiments allowed us to differentiate the effects associated with microbial lag and growth, two coupled processes whose impacts on the biodegradation and transport of contaminants can be difficult to distinguish.  相似文献

A water-sampling device to monitor the quality of water periodically and temporarily flowing out of concrete tubes, sewers or channels is described. It inexpensively and easily enables a qualitative characterization of contamination via these point-source entry routes. The water sampler can be reverse engineered with different sizes and materials, once installed needs no maintenance, passively samples the first surge, and the emptying procedure is short. In an agricultural catchment area in Germany we monitored an emergency overflow of a sewage sewer, an outlet of a rainwater sewer and two small drainage channels as input sources to a small stream. Seven inflow events were analysed for 20 pesticide agents (insecticides, fungicides and herbicides). All three entry routes were remarkably contaminated. We found parathion-ethyl concentrations of 0.3 microg l(-1), diuron up to 17.3 microg l(-1), ethofumesate up to 51.1 microg l(-1), metamitron up to 92 microg l(-1) and prosulfocarb up to 130 microg l(-1).  相似文献

An increasing percentage of agricultural land in Germany is used for oil seed plants. Hence, rape has become an important agricultural plant (in Saxony 1998: 12% of the farmland) in the recent years. During flowering of rape along with intensive radiation and high temperatures, a higher production and emission of biogenic VOC was observed. The emissions of terpenes were determined and more importantly, high concentrations of organic carbonyl compounds were observed during this field experiment. All measurements of interest have been carried out during two selected days with optimal weather conditions. It is found that the origin or the mechanism of formation of different group of compounds had strong influence on the day to day variation of their concentrations. The emission flux of terpenes from flowering rape plants was determined to be 16–32 μg h−1 m−2 (30–60 ng h−1 per g dry plant––540–1080 ng h−1 per plant), in total. Limonene, α-thujene and sabinene were the most important compounds (about 60% of total terpenes). For limonene and sabinene reference emission rates (MS) and temperature coefficients were determined: βlimonene=0.108 K−1 and MS=14.57 μg h−1 m−2; βsabinene=0.095 K−1 and MS=5.39 μg h−1 m−2.The detected carbonyl compound concentrations were unexpectedly high (maximum formaldehyde concentration was 18.1 ppbv and 3.4 ppbv for butyraldehyde) for an open field. Possible reasons for these concentrations are the combination of primary emission from the plants induced by high temperature and high ozone stress, the secondary formation from biogenically and advected anthropogenically emitted VOC at high radiation intensities and furthered by the low wind speeds at this time.  相似文献

Biosurfactant-enhanced solubilization of NAPL mixtures   总被引:5,自引:0,他引:5  
Remediation of nonaqueous phase liquids (NAPLs) by conventional pump-and-treat methods (i.e., water flushing) is generally considered to be ineffective due to low water solubilities of NAPLs and to mass-transfer constraints. Chemical flushing techniques, such as surfactant flushing, can greatly improve NAPL remediation primarily by increasing the apparent solubility of NAPL contaminants. NAPLs at hazardous waste sites are often complex mixtures. However, the equilibrium and nonequilibrium mass-transfer characteristics between NAPL mixtures and aqueous surfactant solutions are not well understood. This research investigates the equilibrium solubilization behavior of two- and three-component NAPL mixtures (containing akylbenzenes) in biosurfactant solutions. NAPL solubilization is found to be ideal in water (i.e., obeys Raoult's Law), while solubilization in biosurfactant solutions was observed to be nonideal. Specifically, the relatively hydrophobic compounds in the mixture experienced solubility enhancements that were greater than those predicted by ideal enhanced solubilization theory, while the solubility enhancements for the relatively hydrophilic compounds were less than predicted. The degree of nonideality is shown to be a nonlinear function of the NAPL-phase mole fraction. Empirical relationships based on the NAPL-phase mole fraction and/or micelle-aqueous partition coefficients measured in single-component NAPL systems are developed to estimate values for the multicomponent partition coefficients. Empirical relationships that incorporate both the NAPL-phase mole fraction and single-component partition coefficients yield much improved estimates for the multicomponent partition coefficient.  相似文献

In this paper, semi-analytical expressions of the effective hydraulic conductivity ( $K^{E})$ and macrodispersivity ( $\alpha ^{E})$ for 3D steady-state density-dependent groundwater flow are derived using a stationary spectral method. Based on the derived expressions, we present the dependence of $K^{E}$ and $\alpha ^{E}$ on the density of fluid under different dispersivity and spatial correlation scale of hydraulic conductivity. The results show that the horizontal $K^{E}$ and $\alpha ^{E}$ are not affected by density-induced flow. However, due to gravitational instability of the fluid induced by density contrasts, both vertical $K^{E}$ and $\alpha ^{E}$ are found to be reduced slightly when the density factor ( $\gamma $ ) is less than 0.01, whereas significant decreases occur when $\gamma $ exceeds 0.01. Of note, the variation of $K^{E}$ and $\alpha ^{E}$ is more significant when local dispersivity is small and the correlation scale of hydraulic conductivity is large.  相似文献

The etiology of salivary gland malignancies still remains unclear. Metal compounds are of special interest since they show ubiquitous presence in the environment, are present in many working places, and are accepted (co-)carcinogens in some other malignancies. Metals enter the body as xenobiotics by inhalation or ingestion. This study investigated the genotoxic potential of sodium dichromate (Na2Cr2O7), nickel sulfate (NiSO4), cadmium sulfate (CdSO4) and zinc chloride (ZnCl2) on human salivary gland cells and lymphocytes. Macroscopically healthy tissue of salivary glands was harvested from 46 patients during surgery and isolated to single cells by enzymatic digestion. The cells were incubated with Na2Cr2O7, NiSO4, CdSO4 or ZnCl2. Na2Cr2O7 was also incubated in combination with the other metal compounds listed. Carcinogenic and co-carcinogenic effects of cadmium were tested by incubation with Na2Cr2O7 and consecutive repair intervals. DNA damage and repair were evaluated by the Comet assay, determining DNA-strand breaks. The extent of damage was quantified using a digital analysis system. Na2Cr2O7 produced significantly enhanced DNA-strand breaks in human salivary gland tissue and lymphocytes. All other metal compounds exerted no damaging effect on both cell types. Co-incubation of Na2Cr2O7 with the other metals revealed a significant additive effect only for CdSO4. Specific analysis of the influence of cadmium showed a reduction of DNA-repair after Na2Cr2O7-induced strand breaks in salivary gland cells. This study provides evidence that exposure to distinct metals may significantly contribute to malignant salivary gland tumors. In consequence, further studies as epidemiological and toxicological data are warranted to determine the role of distinct metals as potential (co-) carcinogens.  相似文献

SUMMARY

This article presents the heuristic ‘Restrictions and Options’ that was created in an interdisciplinary research team to identify restricting and supporting factors regarding sustainable ways of actions for different institutional and individual actors in the need-field of nutrition. The heuristic format is first presented on a general level. Then it is combined with two different approaches: first, with a systemic constructivist approach to organization theory, and second, with the ipsative theory of action (psychology). Its abilities are illustrated by examples from two research projects.

The heuristic tool was developed to help identify relevant factors that restrict or support sustainable development in two empirical settings and also could be used to compare the findings. Combining the two approaches remains for future research.  相似文献

Purpose

The purpose of the study is the enhancement of criticality assessments for resources in order to address function specific factors like dissipation, recycling, bio-activity and toxicity. The developed methodology is applied to platinum-containing cytostatic drugs and automotive catalytic converters.

Methods

The study is methodically based on an analysis of resource specific factors like exploration rates, reserves-to-production ratio and regional distribution of exploration areas as well as on the investigation of product/functional depending factors like recycling rates, dissipation rates, bio-diversity and toxicity. Taking into account that economic and ecological risks may occur at any stage of the supply, consumption and dissipation processes, the whole life cycles of the two analyzed products (cytostatic drugs and automotive catalytic converters) are considered. As an approach to reduce potential economic and ecological risks the study is especially focused on recycling strategies.

Background

In order to get a better understanding of platinum as an essential resource for the development of our society the history and the cultural impact of the term "resource" are introduced.

Results & Discussion

The availability of platinum is crucial for several products of our modern society. Areas of application are e.g. jewellery, automotive catalytic converters, investments (coins, bars), computers, mobile devices, fertilizers and cytostatic drugs. Economic risks are caused by limited sources and dynamic demand of new application areas like fuel cells and drugs. Platinum-containing drugs are used for the treatment of several kinds of cancer such as testicular, breast, colon and prostate. Currently the pharmaceutical industry requires 6,9 tons per year (3 percent of the total demand of platinum). Due to the improvement of medical standards and the ageing society, especially in developing countries, the demand of platinum-containing drugs will rise significantly. The dissipation of toxic soluble salts and the lack of advanced waste-water treatment and recycling systems is a problem of the usage of platinum-containing drugs. Compared with cytostatic drugs (14.6 kg/a for Germany) the dissipation of platinum particles of mobile catalytic converters is much higher (5-20 tons/a only in North America, Europe and Japan; 184,2 kg/a in Germany). On the other hand the emission of platinum nano-particles of automotive catalytic converters seems less critical. Furthermore advanced recycling technologies are already available for them.

Conclusions

The evaluation of resources has to consider resource specific and functional depending factors. In order to reduce economic and ecological risks of the dissipation of platinum salts in cytostatic drugs three principle options can be identified: the development of ecologically compatible and cost-effective substitutes, efficient recycling systems and future-oriented sanitary-systems as well as waste-water treatment facilities, which are able to separate platinum salts. Concerning the dissipation of platinum particles of mobile converters relevant research questions are the identification of adequate substitutes as well as the extension of recycling capacities.

Recommendations and perspectives

The study demonstrates a research gap concerning substitutes for automotive catalytic converters as well as for platinum-containing cytostatic drugs. The development and implementation of recycling concepts, technologies and capacities to reuse platinum as a secondary resource is an essential issue. As a consequence the economic and ecological risks can be reduced by increasing the resource efficiency. Special research has to be done concerning the development of future-oriented sanitary-systems and waste-water treatment facilities in order to separate platinum in waste water. Furthermore, if the future demand of new products like fuel cells is considered, a resource conflict concerning the potential functional applications will be evident. In order to address these challenges the criticality assessment has to be enhanced by ethic and social factors.  相似文献

Objective

Omphalocele is known to be associated with genetic anomalies like trisomy 13, 18 and Beckwith–Wiedemann syndrome, but not with Turner syndrome (TS). Our aim was to assess the incidence of omphalocele in fetuses with TS, the phenotype of this association with other anomalies, their karyotype, and the fetal outcomes.

Method

Retrospective multicenter study of fetuses with confirmed diagnosis of TS. Data were extracted from a detailed questionnaire sent to specialists in prenatal ultrasound.

Results

680 fetuses with TS were included in this analysis. Incidence of small omphalocele in fetuses diagnosed ≥12 weeks was 3.1%. Including fetuses diagnosed before 12 weeks, it was 5.1%. 97.1% (34/35) of the affected fetuses had one or more associated anomalies including increased nuchal translucency (≥3 mm) and/or cystic hygroma (94.3%), hydrops/skin edema (71.1%), and cardiac anomalies (40%). The karyotype was 45,X in all fetuses. Fetal outcomes were poor with only 1 fetus born alive.

Conclusion

TS with 45,X karyotype but not with X chromosome variants is associated with small omphalocele. Most of these fetuses have associated anomalies and a poor prognosis. Our data suggest an association of TS with omphalocele, which is evident from the first trimester.  相似文献
