Pendrin mediates uptake of perchlorate in a mammalian in vitro system   总被引:1,自引:0,他引:1  
Perchlorate is a known endocrine disruptor present in groundwater, vegetables and dairy food products in many regions of the United States. It interferes with the uptake of iodide into the thyrocyte by the sodium-iodide symporter at the basolateral surface, thus potentially disrupting the synthesis of thyroid hormone. Because transport of iodide from the thyroid follicular cells to the follicular lumen is mediated by the protein pendrin at the apical surface, we hypothesized that perchlorate may also interact with this protein. Therefore, HeLa cells were transfected with the human SLC26A4 gene, which encodes pendrin, to generate an in vitro mammalian system expressing the recombinant pendrin protein (HeLa-PDS). The HeLa-PDS cells, along with untransfected cells, were then cultured in presence of iodide and/or perchlorate. Intracellular levels of these two chemicals were measured by ion chromatography tandem mass spectrometry. Results from this study show that iodide and perchlorate uptake increases significantly in HeLa-PDS cells as compared to untransfected cells. Thus, recombinant HeLa cells expressing pendrin protein accumulate iodide and perchlorate intracellularly, indicating that pendrin is involved in the uptake of perchlorate. Additional results from this study suggest that iodide and perchlorate competitively inhibit each other for uptake by pendrin. The ability of perchlorate to compete with iodide for uptake by both basal and apical transporters may increase the potential of perturbation of thyroid homeostasis and therefore the estimated risk posed to susceptible human populations.  相似文献

Winter application of manure poses environmental risks. Seven continuous corn, instrumented watersheds (approximately 1 ha each) at the USDA-ARS North Appalachian Experimental Watershed research station near Coshocton, Ohio were used to evaluate the environmental impacts of winter manure application when using some of the Ohio Natural Resources Conservation Service recommendations. For 3 yr on frozen, sometimes snow-covered, ground in January or February, two watersheds received turkey litter, two received liquid swine manure, and three were control plots that received N fertilizer at planting (not manure). Manure was applied at an N rate for corn; the target level was 180 kg N ha(-1) with a 30-m setback from the application area to the bottom of each watershed. Four grassed plots (61 x 12 m) were used for beef slurry application (9.1 Mg ha(-1) wet weight); two plots had 61 x 12 m grassed filter areas below them, and two plots had 30 x 12 m filter areas. There were two control plots. Nutrient concentrations were sometimes high, especially in runoff soon after application. However, most events with high concentrations occurred with low flow volumes; therefore, transport was minimal. Applying manure at the N rate for crop needs resulted in excess application of P. Elevated P losses contributed to a greater potential of detrimental environmental impacts with P than with N. Filter strips reduced nutrient concentrations and transport, but the data were too limited to compare the effectiveness of the 30- and 61-m filter strips. Winter application of manure is not ideal, but by following prescribed guidelines, detrimental environmental impacts can be reduced.  相似文献

Large dams have been constructed on rivers to meet human demands for water, electricity, navigation, and recreation. As a consequence, flow and temperature regimes have been altered, strongly affecting river food webs and ecosystem processes. Experimental high-flow dam releases, i.e., controlled floods, have been implemented on the Colorado River, U.S.A., in an effort to reestablish pulsed flood events, redistribute sediments, improve conditions for native fishes, and increase understanding of how dam operations affect physical and biological processes. We quantified secondary production and organic matter flows in the food web below Glen Canyon dam for two years prior and one year after an experimental controlled flood in March 2008. Invertebrate biomass and secondary production declined significantly following the flood (total biomass, 55% decline; total production, 56% decline), with most of the decline driven by reductions in two nonnative invertebrate taxa, Potamopyrgus antipodarum and Gammarus lacustris. Diatoms dominated the trophic basis of invertebrate production before and after the controlled flood, and the largest organic matter flows were from diatoms to the three most productive invertebrate taxa (P. antipodarum, G. lacustris, and Tubificida). In contrast to invertebrates, production of rainbow trout (Oncorhynchus mykiss) increased substantially (194%) following the flood, despite the large decline in total secondary production of the invertebrate assemblage. This counterintuitive result is reconciled by a post-flood increase in production and drift concentrations of select invertebrate prey (i.e., Chironomidae and Simuliidae) that supported a large proportion of trout production but had relatively low secondary production. In addition, interaction strengths, measured as species impact values, were strongest between rainbow trout and these two taxa before and after the flood, demonstrating that the dominant consumer-resource interactions were not necessarily congruent with the dominant organic matter flows. Our study illustrates the value of detailed food web analysis for elucidating pathways by which dam management may alter production and strengths of species interactions in river food webs. We suggest that controlled floods may increase production of nonnative rainbow trout, and this information can be used to help guide future dam management decisions.  相似文献

A historical contaminant impact assessment was conducted at 48 heavy manufacturing facilities located in 20 different states for a U.S.-based company. The facilities evaluated were industrial manufacturing sites that operated for as long as 100 yr and used several types of hazardous substances, including solvents and degreasers, oils and other petroleum products, paints and pigments, and heavy metals. The purpose of conducting the impact assessment was to provide direction and guidance on future environmental objectives and pollution prevention initiatives. The impact assessment involved examining historical investigative and remediation costs since 1985, types of contaminants, subsurface geology, hydrology, and regulatory requirements. The results reveled that 85% of the historical environmental costs were associated with hexavalent chromium and chlorinated solvents. Other contaminants, such as oils and other petroleum products and other heavy metals, were far more commonly detected but only accounted for the remaining 15% of costs. The results also indicated that the costs are also strongly associated with the type of geologic environment to which the chemicals were released. As a result of these findings, an aggressive pollution prevention program has been initiated to eliminate the use of those contaminants that are especially expensive to remediate and to develop stronger and more effective engineering controls at facilities located in sensitive ecological areas.  相似文献

Chlorophenols and chloroguaiacols were quantified in juvenile chinook salmon captured near bleached kraft mills in the upper Fraser River in December 1987. Fish captured in April 1988 showed up to 55-fold induction of hepatic mixed function oxidase activity (EROD), and were contaminated with up to 370 ng·kg−1 of 2,3,7,8-TCDF and 68 ng·kg−1 of 2,3,7,8-TCDD. Harvest restrictions and stock enhancement are commonly proposed management responses for sustaining degraded fisheries, but comparisons of their relative effectiveness have seldom been considered prior to making policy choices. We built a population model that incorporated both size-dependent harvest restrictions and stock enhancement contributions to explore trade-offs between minimum length limits and stock enhancement for improving population sustainability and fishery metrics (e.g., catch). We used a Murray cod Maccullochella peelii peelii population as a test case, and the model incorporated density-dependent recruitment processes for both hatchery and wild fish. We estimated the spawning potential ratio (SPR) and fishery metrics (e.g., angler catch) across a range of minimum length limits and stocking rates. Model estimates showed that increased minimum length limits were much more effective than stock enhancement for increasing SPR and angler catches in exploited populations, but length limits resulted in reduced harvest. Stocking was predicted to significantly increase total recruitment, population sustainability, and fishery metrics only in systems where natural reproduction had been greatly reduced via habitat loss, fishing mortality was high, or both. If angler fishing effort increased with increased fish abundance from stocking efforts, fishing mortality was predicted to increase and reduce the benefits realized from stocking. The model also indicated that benefits from stock enhancement would be reduced if reproductive efficiency of hatchery-origin fish was compromised. The simulations indicated that stock enhancement was a less effective method to improve fishery sustainability than measures designed to reduce fishing mortality (e.g., length limits).  相似文献

Air pollution control devices (APCDs) are not compulsory for medical waste incinerators (MWIs) in developing countries. In South Africa, combustion gases are usually vented directly to the atmosphere at temperatures greater than the formation temperature of dioxin. The possibility of dioxin formation outside the incinerator stack has been hypothesized. A plume model has been developed and tested in the wind tunnel with a scale model of an incinerator stack. The plume temperature and trajectory predictions of the plume model were verified within a +/- 3% experimental accuracy. Using South African data, the plume model predicts that the residence time of gases in the temperature range of 150-450 degrees C in a plume is 1.3 sec on average for 5% of a year (18 days) at meteorological conditions resulting in wind speeds of less than 1 m/sec. Two published dioxin formation models were used to assess the probability of dioxin formation in the plume. The formation models predict that the average polychlorinated dibenzodioxins/furans (PCDD/Fs) formed in the plume will exceed the stack emission regulations in South Africa of 0.2 ng/Nm3 toxic equivalent quotient (TEQ) by between 2 and 40 times. The calculated concentrations do not include additional gaseous PCDD/F compounds that may be formed at high-temperature post-combustion zones through pyrosynthesis mechanisms.  相似文献

A model is created for assessing the redevelopment potential of brownfields. The model is derived from a space and time conceptual framework that identifies and measures the surface and subsurface risk factors present at brownfield sites. The model then combines these factors with a contamination extent multiplier at each site to create an index of redevelopment potential. Results from the application of the model within an urbanized watershed demonstrate clear differences between the redevelopment potential present within five different near-surface geologic units, with those units containing clay being less vulnerable to subsurface contamination. With and without the extent multiplier, the total risk present at the brownfield sites within all the geologic units is also strongly correlated to the actual costs of remediation. Thus, computing the total surface and subsurface risk within a watershed can help guide the remediation efforts at broad geographic scales, and prioritize the locations for redevelopment.  相似文献

To improve safety one has to know where risks are. For determining risks, hazards have to be identified and representative accident scenarios defined. This needs effort and technique. Man is quite limited in foresight without having experience and lessons from the past. For knowing the risk of an incidental, undesired event both its severity and probability has to be estimated. Then ways to reduce risk become clear. In a process plant risks are many and it is not possible to remove them all. One has to attribute priorities. Intuitive and qualitative methods can do much, but plant complexity may be large and communication on risk may become difficult without formal methodology. Quantitative risk analysis offers much, but has its weaknesses and drawbacks. The required effort is considerable, specialists are needed, and variability in answers is large. Yet, a model built to go along with the life of an installation and updated periodically may be very useful. This paper presents an overview of the demand, problems encountered, possible remedies, and an outlook on useful improvement and extension of risk analysis methodology, including decision making.  相似文献

This paper examines the relationship between perceived risk and hazard events in two communities in terms of the perception and acceptability of risk. One community experienced an acute risk event comprised of a fire at a chemical plant that resulted in a large-scale evacuation (an acute hazard). The other community was involved in a seven-year controversy over the siting of a hazardous waste incinerator (a chronic risk). While the results are broadly consistent with a learning model of perceived risk, the support is more limited than envisioned; hence, the static forces of risk perception are stronger than anticipated.  相似文献
