A strain of Aspergillus fumigatus, which was observed to rapidly degrade poly-3-hydroxybutyrate (PHB) in a leaf compost, was found to secrete an extracellular hydrolase when grown on PHB as the sole carbon source. Isolation and characterization of the PHB hydrolase (depolymerase) from this fungus revealed that the enzyme had a molecular weight of 57 kDa, an isoelectric point of 7.2, and a PHB hydrolysis activity maxima which occurred at 70°C and pH 8.0. Affinity labeling experiments suggested that this fungal hydrolase is a type of serine esterase. The cyclic trimers of 3-hydroxybutyrate were found to reversibly inhibit the enzymes. 相似文献
The populations of many species are structured such that mating is not random and occurs between members of local patches.
When patches are founded by a single female and all matings occur between siblings, brothers may compete with each other for
matings with their sisters. This local mate competition (LMC) selects for a female-biased sex ratio, especially in species
where females have control over offspring sex, as in the parasitic Hymenoptera. Two factors are predicted to decrease the
degree of female bias: (1) an increase in the number of foundress females in the patch and (2) an increase in the fraction
of individuals mating after dispersal from the natal patch. Pollinating fig wasps are well known as classic examples of species
where all matings occur in the local patch. We studied non-pollinating fig wasps, which are more diverse than the pollinating
fig wasps and also provide natural experimental groups of species with different male morphologies that are linked to different
mating structures. In this group of wasps, species with wingless males mate in the local patch (i.e. the fig fruit) while
winged male species mate after dispersal. Species with both kinds of male have a mixture of local and non-local mating. Data
from 44 species show that sex ratios (defined as the proportion of males) are in accordance with theoretical predictions:
wingless male species<wing-dimorphic male species<winged male species. These results are also supported by a formal comparative
analysis that controls for phylogeny. The foundress number is difficult to estimate directly for non-pollinating fig wasps
but a robust indirect method leads to the prediction that foundress number, and hence sex ratio, should increase with the
proportion of patches occupied in a crop. This result is supported strongly across 19 species with wingless males, but not
across 8 species with winged males. The mean sex ratios for species with winged males are not significantly different from
0.5, and the absence of the correlation observed across species with wingless males may reflect weak selection to adjust the
sex ratio in species whose population mating structure tends not to be subdivided. The same relationship is also predicted
to occur within species if individual females adjust their sex ratios facultatively. This final prediction was not supported
by data from a wingless male species, a male wing-dimorphic species or a winged male species.

Objectives: Mixed-use urban environments, such as arterial roads with adjacent commercial land uses, represent crash locations with the highest risk. These locations are often characterized by high volumes of motor vehicle traffic, on-street parking, and interactions with multiple road user groups such as pedestrians, cyclists, and public transportation. The objective of this study was to investigate previously identified crash risk factors for mixed-use urban environments and assess how parking occupancy, center medians, and cyclist volume influence performance and workload in a driving simulator study.
Methods: Thirty participants were recruited for the study. Participants completed 6 drives that presented different combinations of cyclist volume, median condition, and parking occupancy. Incorporated into the simulator drives was a secondary peripheral detection task (PDT) designed to measure mental workload. Participants provided subjective assessments of workload using the Rating Scale Mental Effort (RSME).
Results: Mean lateral lane position was found to significantly vary across the 3 independent variables of parking occupancy, cyclist volume, and median conditions. No significant changes were identified for mean speed across the conditions. Subjective and objective measures of workload identified changes due to the presence of cyclists with slower reaction times for the PDT task when cyclists were present.
Conclusion: The findings provide insight into the interaction of road design elements in mixed-use urban road environments and demonstrate that increasingly complex environments increase driver demand. This has important road design implications for mixed-use arterial roads, which are often characterized by complex interactions between multiple road user groups. 相似文献
With the implementation of the new EU environmental framework directives, high tier risk assessments of chemicals will be increasingly needed. For high production chemicals, additional tests will complement the standard battery for aquatic toxicity assessments (daphnids, algae, and fish). In the context of a new chemical notification at the European Union level, we have developed a seed germination and root elongation toxicity test with the freshwater aquatic plant Rorippa nasturtium-aquaticum (water cress) to confirm the low environmental risk of the chelant [S,S]-EDDS. A 14 day semi-static growth inhibition test was conducted with daily renewal of the test solution. No concentration related inhibition was found on the basis of any of the criteria investigated, i.e., time and extent of germination, biomass, number of leaves, stalk and root lengths. The no-observed effect concentration was considered to be >or=387 mg SS-EDDS/l. Although germination was selected as an appropriate endpoint to assess the effect of a chelant on an aquatic plant (other endpoints would have been dependant on essential metals that are chelated in standard culture tests), the absence of dose related effects requires further tests with higher exposure concentrations and/or other toxicant(s) to assess the validity of the test as a general tool for aquatic risk assessment. 相似文献
This paper presents the results from study investigating the efficiency with which different surfactants remove crude oil from contaminated soil using a soil washing process. The surfactants studied were aqueous solutions of rhamnolipid, saponin and sodium dodecyl sulfate (SDS). The efficiency of surfactants' removal was quantified and then GC/MS analysis conducted to investigate the distribution of hydrocarbons remaining on the washed soil samples compared to those on a control. The results showed that SDS removed the most crude oil from soil, followed by rhamnolipid and then saponin. However, the different surfactants showed preferences in terms of which crude oil components they removed from the contaminated soil. SDS removed more of the aliphatics than aromatic hydrocarbons whereas saponin removed the aromatic hydrocarbon preferentially to the aliphatic hydrocarbons. Clearly these results provide important information for the selection of surfactants used to remove crude oil from contaminated soils. 相似文献
This study is the first to be conducted in Lebanon on the isolation and molecular characterization and the antimicrobial resistance profile of environmental pathogenic bacterial strains. Fifty-seven samples of seawater, sediment, crab, and fresh water were collected during the spring and summer seasons of 2003. The isolation of Escherichia coli and Salmonella using appropriate selective media revealed that 94.7% of the tested samples were contaminated with one or both of the tested bacteria. The polymerase chain reaction (PCR) was then used to identify the species of both bacteria using various sets of primers. Many pathogenic E. coli isolates were detected by PCR out of which two were identified as O157:H7 E. coli. Similarly, the species of many of the Salmonella isolates was molecularly identified. The confirmed isolates of Salmonella and E. coli were then tested using the disk diffusion method for their susceptibility to four different antimicrobials revealing high rates of antimicrobial resistance. 相似文献
U.S. Environmental Protection Agency (EPA) Emission Measurement Center in conjunction with EPA Regions VI and VIII, the state of Utah, and the U.S. Department of Defense have conducted a series of long-term pilot and field tests to determine the accuracy and reliability of a visible opacity monitoring system consisting of a conventional digital camera and a separate computer software application for plume opacity determination. This technology, known as the Digital Opacity Compliance System (DOCS), has been successfully demonstrated at EPA-sponsored Method-9 "smoke schools", as well as at a number of government and commercially operated industrial facilities. Results from the current DOCS regulatory pilot study demonstrated that, under regulatory enforcement conditions, the average difference in opacity measurement between the DOCS technology and EPA Reference Method 9 (Method 9) was 1.12%. This opacity difference, which was computed from the evaluation of 241 regulated air sources, was found to be statistically significant at the 99% confidence level. Hazard assessment is an essential element in the evaluation of the potential effects of chemical substances on the environment. To date, most work has focused on hazard assessment schemes for the aquatic environment, but in recent years, a number of proposals have been developed for other environmental compartments. Due to limited datasets, the suitability of the toxicity cut-off values in these schemes has not been fully determined and the practicalities associated with using these approaches have not been fully established. This study, which focused on the soil compartment, was performed to examine cut-off values proposed by two terrestrial hazard assessment schemes and establish the availability of data. Data on earthworms indicated that current proposals for toxicity cut-off values are appropriate. However, analysis of IUCLID (International Uniform ChemicaL Information Database) indicates that even for commonly used high production volume chemicals, insufficient data are available to enable classification. Whilst the necessary data may already be available for selected groups of substances (e.g. pesticides and veterinary medicines), a significant experimental testing programme would therefore be required before a terrestrial classification system could be applied widely. PROBLEM: The National Institute for Occupational Safety and Health's [NIOSH] National Occupational Research Agenda (DHHS Publication No. 96-115) reports that approximately 50% of miners will experience hearing loss by age 50, compared to only 9% of the general population. The present investigation examines three antecedents believed to be associated with miner's use of hearing protection. METHOD: A posttest-delayed-posttest-control group field research design was employed to assess antecedents toward wearing hearing protection. RESULTS: Following the initial posttest, miners' attitudes and subjective norms were antecedents to intentions to wear hearing protection devices. Also, intentions toward wearing hearing protection predicted hearing protection behaviors. Approximately six weeks later, miners' attitudes and perceived behavioral control were each significant predictors of intentions to wear hearing protection and again, intentions were positively associated with hearing protection behaviors. IMPACT ON INDUSTRY: Our results indicate that appeals to normative influences may be the most effective antecedent to employ when persuading coal miners to wear hearing protection. However, messages designed to impact attitudes and perceived behavioral control were also effective. 相似文献
INTRODUCTION: This longitudinal field study was designed to encourage Appalachian coal miners in West Virginia and Pennsylvania to engage in hearing-protection behaviors. METHOD: Participants were mailed postcards that featured either a positive, negative, or neutral message on the outside of the postcard and a message encouraging hearing protection behaviors on the inside. The first posttest measurement of the effectiveness of the persuasive messages was conducted about a week after the postcards were mailed. The delayed posttest measurement was conducted six weeks later. RESULTS: Responses from 307 coal miners revealed that the positive or neutral messages generated significantly more self-reported hearing protection behaviors than the negative message. Identical results were obtained in a delayed posttest assessment of miners' self-reported hearing protection behaviors. The positive message was also more effective than either the neutral or negative message in preventing defensive mechanisms from emerging over time. IMPACT ON INDUSTRY: Positive and neutral messages were convincingly more successful than negative messages in facilitating self-reported hearing protection behaviors among coal miners.
